Dopamine D4 receptors elevated in schizophrenia.

Although the biological basis of schizophrenia is not known, possible causes include genetic defects, viruses, amines, brain structure and metabolism, neuroreceptors, and G proteins. The hypothesis of dopamine overactivity in schizophrenia is based on the fact that neuroleptics block dopamine D2 receptors in direct relation to their clinical antipsychotic potencies. Moreover, dopamine D2 or D2-like receptors are elevated in postmortem schizophrenia brain tissue. This elevation, however, is only found in vivo using [11C]methylspiperone but not [11C]raclopride. The dopamine D4 receptor gene has

Dopamine agonists and pituitary tumor shrinkage.

The primary aim of this review has been to clarify the tumor shrinking effects of dopamine agonists on pituitary macroadenomas of different cell types. Shrinkage is most dramatic for macroprolactinomas and is due to cell size reduction. Seventy-nine percent of 271 definite macroprolactinomas were reduced in size by at least 25%, and 89% shrank to some degree. Most shrinkage occurs during the first 3 months of treatment, although in a minority shrinkage is delayed. Dopamine agonist resistance during long-term therapy is exceptional. Drug withdrawal nearly always leads to a return of hyperprolac

Dopamine D2 mechanisms in canine narcolepsy.

Narcolepsy is a sleep disorder characterized by abnormal manifestations of rapid-eye-movement (REM) sleep and excessive daytime sleepiness. Using a canine model of the disease, we found that central D2 antagonists suppressed cataplexy, a form of REM-sleep atonia occurring in narcolepsy, whereas this symptom was aggravated by D2 agonists. The effect on cataplexy was stereospecific for the S(-) enantiomer of sulpiride (a D2 antagonist) and the R(+) enantiomer of 3-PPP (a D2 agonist). There was also a significant correlation between the in vivo pharmacological potency and in vitro drug affinity f

Dopamine transporter: biochemistry, pharmacology and imaging.

In recent years, there has been substantial progress in studying the dopamine transporter, a unique component of the functioning dopaminergic nerve terminal. The transporter has been studied by direct binding techniques using a variety of ligands which function as inhibitors of transport. Analogues of these ligands have been used as photoaffinity labels to solubilize and further characterize the transporter. While a variety of drugs bind to the transporter, it is clear that the transporter may serve as an important drug receptor, particularly for the reinforcing properties of some psychostimul

Dopamine-producing endometrial carcinoma with argyrophilia.

A case of moderately differentiated adenocarcinoma with argyrophilia arising in the endometrium of a 58-year-old Japanese woman is reported. Although she showed no endocrine symptoms, neuro-secretory granules were documented in the cytoplasm of some tumor cells by electron microscopy. Moreover, dopamine production by the tumor was demonstrated by biochemical determination. The possible mechanism accounting for such a lesion is discussed.

Abnormal neuroleptic/dopamine receptors in schizophrenia.

In order to test the dopamine hypothesis of schizophrenia, the neuroleptic/dopamine receptors in three dopamine-rich regions of 53 postmortem normal human brains and 42 schizophrenic brains were measured using 3H-haloperidol and 3H-spiperone. The binding of 2 nM 3H-haloperidol was significantly elevated in the caudate nucleus (90 +/- 10%) and the putamen (61 +/- 6%) from schizophrenic brains. The binding of 1 nM 3H-spiperone was also elevated by 50 +/- 7% in schizophrenic caudate and 47 +/- 8% in the putamen. The nucleus accumbens from schizophrenic brains revealed an enhanced binding of 110 t

Impulse control disorders and dopamine agonists.

Impulse control disorders (ICDs) are a potentially detrimental group of behaviours with recognised and increasingly reported associations with dopamine agonist (DA) use. As DAs are an established and commonly prescribed treatment for patients with prolactinoma, further understanding of the relationship between DAs and ICDs in this group of patients is merited. In this narrative review, we discuss the evidence to date exploring the prevalence of ICDs in patients with pituitary tumours, examine reasons for considerable variability in reported findings, and appraise and summarise evidence attempt

Dopamine Dysregulation Syndrome in Parkinson's Disease.

This case study examines how patients with Parkinson's disease (PD) may develop dopamine dysregulation syndrome (DDS), a condition characterized by the misuse of dopamine replacement therapy through excessive self-directed dosing beyond what is necessary to control motor symptoms. While carbidopa/levodopa remains the mainstay therapy for many adults with PD, some patients may choose Mucuna pruriens, a complementary medicine containing levodopa, to manage their motor symptoms. This case describes a patient with PD who presents with DDS following co-administration of self-directed Mucuna prurien

Regulation of Dopamine Transporter Surface Expression.

The dopamine transporter (DAT) is a plasma membrane protein expressed in dopamine (DA) neurons of the central nervous system and is critical for regulating DA neurotransmission. The DAT is responsible for the reuptake of released DA back into the presynaptic neuron, resulting in the termination of DA transmission. This process also recycles the DA back into the dopaminergic neuron for subsequent release. DAT is the target of psychostimulants including cocaine and amphetamines and has been associated with several neuropsychiatric disorders. Only DAT proteins located on the plasma membrane can r

Nicotine addiction: More than just dopamine.

Despite decades of research and anti-tobacco messaging, nicotine addiction remains an important public health problem leading to hundreds of thousands of deaths each year. While fundamental studies have identified molecular, circuit-level and behavioral mechanisms important for nicotine reinforcement and withdrawal, recent studies have identified additional pathways that are important for both nicotine seeking and aversion. In particular, although dopaminergic mechanisms are necessary for nicotine-dependent reward and drug-seeking, novel glutamate and GABA signaling mechanisms in the mesolimbi

MicroRNAs in Dopamine Agonist-Resistant Prolactinoma.

Dopamine agonists (DAs) are preferred for the treatment of prolactinomas and are usually very effective. Nonetheless, 20-30% of bromocriptine- and approximately 10% of cabergoline-treated individuals exhibit resistance to DAs. In addition, the mechanism underlying this phenomenon remains elusive. In this study, we summarize the major findings regarding the role of microRNAs (miRNAs) in the pathogenesis of DA-resistant prolactinoma (DARP). Currently available evidence suggests that miRNAs are usually dysregulated in DARP and that, although controversial, the dysregulated miRNAs target the trans

Dopamine dysregulation syndrome induced by proxy.

Dopamine dysregulation syndrome is a rare complication of Parkinson's disease (PD) treatment. We present a 70-year-old woman with a long-standing PD and a clinical picture compatible with dopaminergic dysregulation, which was ultimately revealed to be induced by her companion. Patient's exuberant choreiform dyskinesia led to a potential financial advantage when performed outside the hospital but excessive dopamine intake also occurred during hospital admission, without any obvious reward for the abuser. Even in cases where there is no place for a definitive diagnosis, deceptive behaviours must

Dopamine Agonists Can Reduce Cystic Prolactinomas.

CONTEXT: Cystic prolactinomas are considered resistant to volume reduction by dopamine agonists (DAs). Although several individual case reports and small case series have suggested that DAs may reduce these lesions, larger series using standardized imaging metrics are lacking. OBJECTIVE: The objectives of the study were to assess the efficacy of DAs on cyst size in patients with predominantly cystic prolactinomas and to characterize the clinical course and treatment outcomes in these patients. DESIGN: This study was a retrospective review. SETTING: The study was conducted at a tertiary refe

Pathway-Specific Dopamine Abnormalities in Schizophrenia.

In light of the clinical evidence implicating dopamine in schizophrenia and the prominent hypotheses put forth regarding alterations in dopaminergic transmission in this disease, molecular imaging has been used to examine multiple aspects of the dopaminergic system. We review the imaging methods used and compare the findings across the different molecular targets. Findings have converged to suggest early dysregulation in the striatum, especially in the rostral caudate, manifesting as excess synthesis and release. Recent data showed deficit extending to most cortical regions and even to other e

Dopamine receptor partial agonists and addiction.

Many drugs abused by humans acutely facilitate, either directly or indirectly, dopamine neurotransmission in the mesolimbic pathway. As a consequence dopamine receptor agonists and antagonists have been widely investigated as putative pharmacological therapies for addiction. This general strategy, however, has had only limited success due in part to poor treatment adherence and efficacy and the significant adverse effects of dopaminergic medications. In this perspective, we discuss the potential therapeutic use of dopamine receptor partial agonists in addiction, developed initially as antipsyc

Imaging dopamine transporters in Parkinson's disease.

The dopamine transporter (DAT) is responsible for clearance of dopamine from the synaptic cleft after its release. Imaging DAT availability provides a measure of dopamine terminal function and a method for detecting striatal dopamine deficiency states present in idiopathic Parkinson's disease and atypical neurodegenerative Parkinsonian disorders such as multiple system atrophy and progressive supranuclear palsy. DAT imaging with PET or single photon emission computed tomography can be used to support a diagnosis of dopamine-deficient parkinsonism in cases where this is suspected and rationaliz
