PHAEOHYPHOMYCOSIS ASSOCIATED WITH OSSIFICATION OF THE SKULL AND CERVICAL VERTEBRAE IN A SWELL SHARK (CEPHALOSCYLLIUM VENTRIOSUM).

A female, captive bred, juvenile swell shark ( Cephaloscyllium ventriosum ) was observed swimming in tight circles and rolling. Radiographs and computed tomography of this individual revealed extensive cartilage mineralization of the skull and cranial cervical vertebrae compared with diagnostic images of clinically healthy conspecifics. Gross necropsy and histopathologic examination revealed ossification and fibrosis of the cartilaginous matrix of the skull and cervical vertebrae with deep invasion by a pigmented hyphal fungus. There was no growth on fungal culture, but fungal polymerase chain

Anterior and pan-endophthalmitis caused by Candida albicans and Aspergillus fumigatus: report of two cases.

We report two posttraumatic fungal endophthalmitis cases with anterior and entire segment involvement caused, respectively, by Candida albicans and Aspergillus fumigatus. The anterior endophthalmitis induced by C. albicans presented multifocal lesions with hyphael structures in anterior chamber, but without involvement of posterior vitreous. The pan-endophthalmitis caused by A. fumigatus displayed significant hypopyon and vitritis. Treatment strategies are anterior chamber cleaning for anterior endophthalmitis and vitrectomy for pan-endophthalmitis, excepting systemic and topical antifungal dr

Emerging Fungal Pathogen Candida auris Evades Neutrophil Attack.

Candida auris has recently emerged as the first fungal pathogen to cause a global public health threat. The reason this species is causing hospital-associated outbreaks of invasive candidiasis with high mortality is unknown. In this study, we examine the interaction of C. auris with neutrophils, leukocytes critical for control of invasive fungal infections. We show that human neutrophils do not effectively kill C. auris Compared to Candida albicans, neutrophils poorly recruited to C. auris and failed to form neutrophil extracellular traps (NETs), which are structures of DNA, histones, and prot

Intravesical Fungus Ball Caused by Candida albicans in a Diabetic Patient With Septic Shock: A Case Report.

Fungal urinary tract infections (UTIs) are rare. In exceptional cases, fungal colonization can lead to the formation of a "Fungus ball" or fungal bezoar, which may cause urinary tract obstruction and sepsis. We report the case of a 61-year-old male with poorly controlled type 2 diabetes mellitus, presenting with septic shock and anuria. Imaging studies revealed an intravesical mass with air locules, and urine cultures grew Candida albicans. Despite systemic antifungal therapy, persistent obstruction required surgical management. A cystoscopy and open surgical approach allowed complete extracti

Allergy and immunity to fungal infections and colonization.

Innate and cell-mediated immunity are considered as the principal defence lines against fungal infections in humans. Most opportunistic mycoses occur in individuals with defective innate and/or adaptive cellular immunity. The morbidity and mortality rates associated with infections caused by fungal pathogens are high, and prevention, diagnosis and treatment of these infections remain quite difficult. A variety of pathological conditions, including impaired immune function, are believed to cause host susceptibility to fungal infections as well as to determine the severity and characteristics of

The changing face of dermatophytosis: from common skin disease to emerging resistant epidemic.

Dermatophytosis is a commonly encountered superficial fungal infection with an affinity for keratin-rich structures, such as skin, hair and nails. The recently emerging recalcitrant dermatophytosis is a recurrent, relapsing, chronic or persistent disease, which has a significant impact on the social, emotional and financial aspects of a patient's life. Trichophyton indotineae, a newly identified species, has become resistant to conventional therapies, unlike its predecessor. This review highlights the shift of this commonly encountered skin disease from an easily manageable clinical entity to

Decisive role of histopathology of lens capsule in the diagnosis of chronic fungal postoperative endophthalmitis.

We report three cases of refractory chronic endophthalmitis after cataract surgery presenting to a referral center, and with repeated negative cultures. Initial treatment consisted of intravitreal and systemic antibiotics, with partial improvement. After subsequent worsening, pars plana vitrectomy, intraocular lens explantation and en bloc capsulectomy were performed. Histopathological examination revealed multiple filamentous fungal structures, sequestered between anterior/posterior lens capsule in all cases. Chronic postoperative fungal endophthalmitis may manifest with negative cultures pos

Potential antifungal effects of human platelets against zygomycetes in vitro.

Zygomycosis is increasingly recognized in immunocompromised hosts. We investigated whether platelets become activated after contact with Zygomycetes and adhere to conidial and hyphal structures using immunofluorescence. The platelets' influence on fungal viability was evaluated by assessing hyphal elongation and hyphal damage. Platelets became activated and strongly adhered to conidia and hyphae of Zygomycetes. Platelets induced time dependent damage to hyphae and significantly reduced (P<.05) hyphal elongation. We found that platelets possess antifungal capacities against Zygomycetes based on

Surgical treatment of fungal mycetoma.

Chemotherapy for chronic fungal infections is often ineffective, and the associated delay in surgical treatment may result in radical procedures being performed to effect a cure. The author describes four patients with chronic fungal lesions, three involving a foot and one the abdominal wall, all of which failed to respond to antifungal chemotherapy. The lesions were excised, using procedures aimed at preserving function. Infection recurred in two patients during a 9-month follow-up. The author concludes that surgical therapy should be instituted early in the disease to forestall spread to dee

Pyridylethanol(phenylethyl)amines are non-azole, highly selective Candida albicans sterol 14α-demethylase inhibitors.

Sterol 14α-demethylase (CYP51) is the main drug target for the treatment of fungal infections. The worldwide increase in the incidence of opportunistic fungal infections and the emerging resistance to available azole-based antifungal drugs, raise the need to develop structurally distinct and selective fungal CYP51 inhibitors. In this work we have, for the first time, investigated the binding of pyridylethanol(phenylethyl)amines to any fungal CYP51. The comparison of the binding to Candida albicans and human CYP51 studied by spectroscopic and modeling methods revealed moieties decisive for sele

Candida albicans biofilms: comparative analysis of room-temperature and cryofixation for scanning electron microscopy.

Biofilms are frequently related to invasive fungal infections and are reported to be more resistant to antifungal drugs than planktonic cells. The structural complexity of the biofilm as well as the presence of a polymeric extracellular matrix (ECM) is thought to be associated with this resistant behavior. Scanning electron microscopy (SEM) after room temperature glutaraldehyde-based fixation, have been used to study fungal biofilm structure and drug susceptibility but they usually fail to preserve the ECM and, therefore, are not an optimised methodology to understand the complexity of the fun

Morphological Changes in Fungal Elements and Antifungal Drug Susceptibility in an Ex Vivo Model of Fungal Keratitis.

PURPOSE: Fungal keratitis is a serious eye infection that can cause irreversible vision loss. One of the causative fungal pathogens, Fusarium solani (Neocosmospora solani), shows limited susceptibility to antifungal drugs. Furthermore, differences between in vitro and clinical outcomes are encountered in some cases, complicating treatment. In this study, we compared in vitro and ex vivo antifungal susceptibility using a porcine corneal model of F. solani keratitis. METHODS: An ex vivo fungal keratitis model was developed using porcine corneas, which were inoculated with F. solani and assigned

Design, synthesis, and evaluation of novel 3,4-isoxazolediamide derivatives for the combination treatment of azole-resistant candidiasis.

Invasive fungal infections are emerging as serious infectious diseases worldwide. Due to the frequent emergence of resistance, the cure for invasive fungal infections is often unachievable. The molecular chaperone Hsp90 provides a promising target because it supports survival, virulence, and drug resistance in a variety of pathogens. Herein, we report on the structural optimization and structure-activity relationship studies of 3,4-isoxazolediamide analogs. As a new class of fungal Hsp90 inhibitor, compound B25 was found to have good synergistic effects with fluconazole and to avoid potential

Antifungal symbiotic peptide NCR044 exhibits unique structure and multifaceted mechanisms of action that confer plant protection.

In the indeterminate nodules of a model legume Medicago truncatula, ∼700 nodule-specific cysteine-rich (NCR) peptides with conserved cysteine signature are expressed. NCR peptides are highly diverse in sequence, and some of these cationic peptides exhibit antimicrobial activity in vitro and in vivo. However, there is a lack of knowledge regarding their structural architecture, antifungal activity, and modes of action against plant fungal pathogens. Here, the three-dimensional NMR structure of the 36-amino acid NCR044 peptide was solved. This unique structure was largely disordered and highly d

Synthesis, Antifungal Activity, and Potential Mechanism of Natural Product Pogostone Derivatives Containing an Acylhydrazone Scaffold.

The current study designed and synthesized a series of acylhydrazone derivatives based on the natural product pogostone. Among them, compound 2a15 exhibited potent fungicidal activity against Valsa mali (V. mali) (EC50 = 0.34 μg/mL) and Sclerotinia sclerotiorum (S. sclerotiorum) (EC50 = 0.45 μg/mL). At 200 μg/mL, 2a15 exhibited 81.26 and 82.08% efficacies against V. mali and S. sclerotiorum, respectively. Qualitative structure-activity analysis showed that when the benzene ring was substituted by a bromine atom, chlorine atom, or electron-withdrawing group, compounds with the substituent group

Novel Antifungal Agents and Their Activity against Aspergillus Species.

There is a need for new antifungal agents, mainly due to increased incidence of invasive fungal infections (IFI), high frequency of associated morbidity and mortality and limitations of the current antifungal agents (e.g., toxicity, drug-drug interactions, and resistance). The clinically available antifungals for IFI are restricted to four main classes: polyenes, flucytosine, triazoles, and echinocandins. Several antifungals are hampered by multiple resistance mechanisms being present in fungi. Consequently, novel antifungal agents with new targets and modified chemical structures are required

Spacer-free BODIPY fluorogens in antimicrobial peptides for direct imaging of fungal infection in human tissue.

Fluorescent antimicrobial peptides are promising structures for in situ, real-time imaging of fungal infection. Here we report a fluorogenic probe to image Aspergillus fumigatus directly in human pulmonary tissue. We have developed a fluorogenic Trp-BODIPY amino acid with a spacer-free C-C linkage between Trp and a BODIPY fluorogen, which shows remarkable fluorescence enhancement in hydrophobic microenvironments. The incorporation of our fluorogenic amino acid in short antimicrobial peptides does not impair their selectivity for fungal cells, and enables rapid and direct fungal imaging without

Necrosis of maxilla, nasal, and frontal bone secondary to extensive rhino-cerebral mucormycosis.

Mucormycosis is an opportunistic fulminant fungal infection caused by zygomycetes. This fungus can cause a variety of infections in human beings, particularly in the uncontrolled diabetes mellitus. Zygomycetes impinge into the vascular network, resulting in thrombosis and necrosis of the surrounding hard and soft tissues. The infection begins in the nose and paranasal sinuses due to inhalation of fungal spores and spread to orbital and intracranial structures either by direct invasion or through the blood vessels. Sinus mucormycosis is often accompanied by a poor prognosis and a high mortality

Structure of Saccharomyces cerevisiae chitinase 1 and screening-based discovery of potent inhibitors.

Chitinases hydrolyse the beta(1,4)-glycosidic bonds of chitin, an essential fungal cell wall component. Genetic data on a subclass of fungal family 18 chitinases have suggested a role in cell wall morphology. Specific inhibitors of these enzymes would be useful as tools to study their role in cell wall morphogenesis and could possess antifungal properties. Here, we describe the crystallographic structure of a fungal "plant-type" family 18 chitinase, that of Saccharomyces cerevisiae CTS1. The enzyme is active against 4-methylumbelliferyl chitooligosaccharides and displays an unusually low pH op
