The protective mechanisms underlying Ginsenoside Rg1 effects on rat sciatic nerve injury.

Ginsenoside Rg1 (GsRg1), derived from the herb Ginseng, was found to exert protective effects in nerve injury; however, the mechanisms underlying these effects remain to be determined. Oxidant stress and apoptosis are known to be involved in sciatic nerve injury. Thus, the aim of this study was to examine whether GsRg1 was able to modify sciatic nerve injury in a rat model. The following parameters were measured: (1) number of spinal cord motoneurons by Nissl staining, (2) oxidation parameters including spinal cord malondialdehyde (MDA) levels and activities of superoxide dismutase (SOD) and g

Peripheral immunomodulation with ginsenoside Rg1 ameliorates neuroinflammation-induced behavioral deficits in rats.

Neuroinflammatory disturbances have been closely associated with depression and many other neuropsychiatric diseases. Although targeting neuroinflammatory mediators with centrally acting drugs has shown certain promise, its translation is faced with several challenges especially drug delivery and safety concerns. Here, we report that neuroinflammation-induced behavioral abnormality could be effectively attenuated with immunomodulatory agents that need not to gain brain penetration. In a rat model with intracerebral lipopolysaccharide (LPS) challenge, we validated that ginsenoside Rg1 (Rg1), a

Anti-diabetic effect of ginsenoside Rb(3) in alloxan-induced diabetic mice.

As one of the main active component of protopanaxdiol type ginsenosides, ginsenoside Rb(3) is rarely reported in the treatment of diabetes. The anti-diabetic activity of ginsenoside Rb(3) was investigated in a model of alloxan-induced diabetic mice in the present study. The physiological parameter such as fasting blood glucose level, oral glucose tolerance, body weight, food intake and water intake were measured. Glucose consumption in C2C12 myotubes was also determined in order to investigate the molecular mechanism of ginsenoside Rb(3) in anti-diabetes. The alloxan-induced diabetic mice were

Integrative analyses of network pharmacology and bioinformatics reveal the synergistic antitumor effects of cantharidin and ginsenosides Rg3 on hepatocellular carcinoma.

Cantharidin (CTD) has been traditionally used in Traditional Chinese Medicine (TCM) and exhibits pronounced antitumor activity, particularly in hepatocellular carcinoma (HCC). However, its preclinical translation has been substantially constrained by dose-limiting toxicity. Inspired by traditional Mylabris and Ginseng combinations, we conducted an investigation into the synergistic efficacy of CTD and Ginsenoside-Rg3 (Rg3), one of the most prevalent and bioactive ginsenosides, in combating HCC. We find that CTD/Rg3 co-treatment synergistically suppressed HCC cell proliferation, migration, and

Affinity-guided and activity-oriented consecutive-CCC/UNIFAC of acetylcholinesterase-binding ginsenosides from Black ginseng: Extraction optimization and activity characterization.

The preparative isolation of acetylcholinesterase (AChE)-binding ginsenosides from Black ginseng is challenged by matrix complexity, broad polarity distribution, and redundant solvent-phase preparation during repeated countercurrent chromatography (CCC). A crude extract prepared by RSM-GA-BP-guided UAE was evaluated by concentration-resolved UF-LC-MS at seven bovine-AChE concentrations, with enzyme-free and thermally inactivated-AChE controls used to distinguish system background and nonspecific protein-associated retention. Binding degree was used as the operational UF-LC-MS response, and wei

Ginseng extract and total ginsenosides protect the function of hematopoietic stem cells by activating the Notch and Wnt signal pathways.

ETHNOPHARMACOLOGICAL RELEVANCE: Panax ginseng C.A. Meyer (ginseng), a traditional Chinese medicine, is famous for "Qi-tonifying" effect and widely used for healthcare and therapeutic effects in China. Modern pharmacology showed that Ginseng had a potential impact on hematopoietic stem cells (HSCs) that promote the regeneration of all blood cells in the bone marrow. The "Qi-tonifying" effect of ginseng might have close correlation with hematopoietic function. However, the protective effect of ginseng on HSCs has been rarely studied. AIM OF THE STUDY: To elucidate the difference of chemical com

The antioxidant activities of Korean Red Ginseng (Panax ginseng) and ginsenosides: A systemic review through in vivo and clinical trials.

A wide range of studies have steadily pointed out the relation of oxidative stress to the primary and secondary causes of human disease and aging. As such, there have been multiple misconceptions about oxidative stress. Most of reactive oxygen species (ROS) generated from chronic diseases cause oxidative damage to cell membrane lipids and proteins. ROS production is increased by abnormal stimulation inside and outside in the body, and even though ROS are generated in cells in response to abnormal metabolic processes such as disease, it does not mean that they directly contribute to the pathoge

Ginsenoside Rd-functionalized hydrogel for immune modulation and enhanced diabetic wound healing.

Persistent inflammation and oxidative stress are central barriers to effective diabetic wound repair. Successful healing requires restoration of a coordinated local microenvironment, yet achieving this remains a major clinical challenge. Herein, a composite ginsenoside Rd-functionalized hydrogel (Gel@Rd) with dynamic and static dual networks was fabricated using extracellular matrix-mimicking biomaterials. Gel@Rd exhibited favorable biocompatibility, self-healing capacity, and mechanical stability, facilitating prolonged retention and release of Rd at the wound site. In diabetic wounds, Gel@Rd

Ginsenoside Rh4-regulated macrophage polarization and oxidative stress in septic myocardial injury.

Septic cardiomyopathy (SCM) is a severe complication of sepsis. The therapeutic potential of Ginsenoside Rh4 (Rh4) in SCM remains unclear. This study aimed to investigate the effects and mechanisms of Rh4 on SCM using both in vivo and in vitro approaches. In the in vivo experiments, Rh4 significantly reduced the mortality rate in SCM mice, improved ejection fraction and diastolic function, and protected myocardial cell mitochondria, thereby alleviating myocardial cell injury. Transcriptomic sequencing, ELISA, and PCR results suggested that Rh4 reduced inflammatory cytokines in myocardial tissu

The orexinergic system mediates the effects of ginsenoside Rg1 on cyclophosphamide-induced reproductive toxicity.

Cyclophosphamide (CP) is a widely prescribed chemotherapeutic and immunosuppressive agent known to cause significant reproductive toxicity. Ginseng effectively counteracts CP-induced reproductive toxicity, exhibiting pharmacological effects that resemble enhanced orexin signaling. In this study, molecular docking identified ginsenoside Rg1 as the candidate with the highest binding affinity for orexin receptor 1 (OX1R). In CP-treated male mice, Rg1 dose-dependently ameliorated reproductive toxicity, as evidenced by improved sexual behavior, sperm motility and DNA integrity, serum testosterone l

Multi-omics approaches to explore the therapeutic mechanism for ginsenoside Rg1 against MASLD.

Ginsenoside Rg1 (G-Rg1), a traditional Chinese medicine, alleviates metabolic dysfunction-associated steatotic liver disease (MASLD). However, the mechanism by which G-Rg1 improves metabolic disorders in MASLD by regulating the gut microbiota remains ambiguous‌. We constructed a diet-induced murine MASLD model and employed 16S rRNA sequencing and non-targeted metabolomic analysis to ‌investigate the mechanism of G-Rg1 in treating MASLD, focusing on its regulatory effect on the gut microbiota. Our results revealed that G-Rg1 significantly increased the 5-hydroxyindoleacetic acid levels and acti

Septin 2: A direct target of Ginsenoside Rg1 mediating its neuroprotective effects.

Ginsenoside Rg1 (Rg1), a representative saponin of ginseng, is evidenced with a remarkable neuroprotective effect; however, its direct target in neural cells has not been addressed. The present study investigated the reconstructing effect on the damaged neurite network of Rg1, and its direct target responsible for this bio-effect. The results showed that Rg1 can alleviate the Aβ25-35-induced neuronal damage and synaptic atrophy in primary neurons and PC12 cell models, respectively. Furthermore, the Septin 2, involved in cell division and cytoskeleton organization, was identified as a protein s
