Examination of the cerebrospinal fluid by isoelectric focusing was done in 2 cases of progressive multifocal leukoencephalopathy (PML). The diagnosis was confirmed at autopsy in one case. IgG oligoclonal bands were found in both, suggesting that there was intrathecal immunoglobulin synthesis. Production may be due to a specific intrathecal response to papova virus or to IgG oligoclonal formation by lymphoma cells confined to the central nervous system or to a specific immune response to tumor cell antigens.

Mono[3H]propionyl-alpha-bungarotoxin, prepared with N-succinimidyl [2,3-3H]propionate (sp. radioactivity 50 Ci/mmol) and purified to homogeneity by electrofocusing, retains its biological activity and stability. Rate constants for its binding to acetylcholine receptor were 4.4-fold lower than for unlabelled toxin; no dissociation was detectable. Analysis of enzymic digests of toxin showed 3H is located mainly of entirely in epsilon-propionyl-lysine.

Streptozotocin diabetes strongly reduced the N-acetyl-beta-glucosaminidase activity in the submaxillary glands of mice. A reciprocal change was observed between the enzyme activity and blood sugar in diabetes. Insulin treatment of th diabetic mice returned the reduced activity to the normal level. Isoelectric focusing analysis showed that the submaxillary gland enzyme was composed of two isozymes having isoelectric points (pI) of 4.8 and 8.8. Only the pI 8.8 isozyme was affected by the diabetes.

An antigen was prepared from metabolic products which were produced by maintaining first instar larvae of Hypoderma lineatum in RPMI tissue culture medium for 48 h. Three major proteins were identified in the secretory products and were characterised in terms of their molecular weights and iso-electric points. The antigen compared favourably with a soluble extract of larvae when used in an enzyme-linked immunosorbent assay (ELISA) against a panel of control sera and 2000 bovine sera collected from farms in Andalucia, Spain.

Patients affected with subacute sclerosing panencephalitis were found to show high measles antibody activity; γ-globulin M-components were detected in the serum, cerebrospinal fluid and brain tissue. The relative antibody activity was different in the different IgGs separated by gel filtration. The γ-globulins were separated into several components by isoelectric focussing on a sucrose gradient and each showed different degrees of measles antibody activity. Better separation was achieved by isoelectric focusing than by gel chromatography.

We have found that kallikrein-like proteins differ in their isoelectric point but share antigenic determinants. For identification of kallikrein-like proteins an initial separation was carried out in flat-bed isoelectrofocusing gels. The kallikrein-like nature was demonstrated by an immunological similarity to kallikrein-like proteins by immunoblotting using antiserum against a kallikrein family member for staining. We used this system to identify different kallikrein-like proteins during purification of both known as well as new enzymes.

An antimycin A-resistant derivative of the human cell line, D98, has been obtained by selective mutagenesis with N-methyl-N'-nitro-N-nitrosoguanidine. By means of DEAE-Sepharose CL-6B ion exchange chromatography and TSK-GEL G2000 SW high-performance gel filtration, a purified protein with fibrinolytic activity was obtained from the venom of Agkistrodon halys brevicaudus (Korean mamushi). Among vertebrates, turtles are able to tolerate exceptionally low oxygen tensions. We have investigated the compensatory mechanisms that regulate respiration and blood oxygen transport in snapping turtles during short exposure to hypoxia. Snapping turtles started to hyperventilate when oxygen levels dropped below 10% O(2). Total ventilation increased 1.75-fold, essentially related to an increase in respiration frequency. Anti-secretory factor (ASF) was determined by the rat intestinal loop model in the pituitary gland and in the blood at various intervals after experimental infection of mice with Schistosoma mansoni. In juvenile diabetics treated on an outpatient basis, the HbA1c value was determined by means of isoelectric focusing. This method proved to be relatively simple and reproducible. Sera and cerebrospinal fluid (CSF) from five subacute sclerosing panencephalitis (SSPE) patients were absorbed with a concentrated preparation of cell cultures infected with measles virus.
