Effects of nickel, chromate, and arsenite on histone 3 lysine methylation.

Occupational exposure to nickel (Ni), chromium (Cr), and arsenic (As) containing compounds has been associated with lung cancer and other adverse health effects. Their carcinogenic properties may be attributable in part, to activation and/or repression of gene expression induced by changes in the DNA methylation status and histone tail post-translational modifications. Here we show that individual treatment with nickel, chromate, and arsenite all affect the gene activating mark H3K4 methylation. We found that nickel (1 mM), chromate (10 microM), and arsenite (1 microM) significantly increase t

Role of lysine residue at 7th position of wasp chemotactic peptides.

Most of chemotactic peptides isolated from various kinds of wasp venom have lysine residue at 7th (or 8th) position, but only a chemotactic peptide from Icaria sp. has no basic amino acid residues in the sequence. The relation of chemotaxis with other biochemical activities such as superoxide generation and lysosomal enzyme release from guinea pig neutrophils was studied by the use of substitution analogs of Icaria chemotactic peptide at 7th position. Findings revealed two distinct ways of chemoattractant signal transduction in neutrophils.

Inhibition of gas vesicle production in Microcyclus aquaticus by L-lysine.

The timing and degree of gas vesicle production in Microcyclus aquaticus was affected by nutritional conditions. If 50 microng L-lysine/ml was added to a glucose-mineral salts medium (DM), the organism did not form gas vesicles. This effect was specific for L-lysine, as neither D-lysine nor meso-diaminopimelic acid prevented gas vesicle production. Cells grown in the presence of L-lysine did not contain any immunologically detectable gas vesicle protein, which indicates that L-lysine affects expression of the structural gene for the gas vesicle protein rather than assembly of the protein into

Characterization of a lysine-specific active transport system in Rickettsia prowazeki.

Rickettsia prowazeki possesses an active transport system for lysine with a Kt of influx of 1 muM. Extraction and chromatographic analysis of the accumulated labeled material show the material to be lysine rather than a derivative. This intracellular lysine pool can be exchanged with external unlabeled substrates for at least 10 min; The lysine analogues L-aminoethyl cysteine, N-methyl lysine, hydroxylysine, and D-lysine competitively inhibit uptake of L-lysine, but cadaverine, diaminopimelate, arginine, ornithine, and epsilon-aminocaproate do not. Accumulation of lysine can be inhibited by th

Lysine-targeted covalent inhibitors: mechanistic advances, pharmacological perspectives and translational challenges.

Targeted covalent inhibitors can provide potent and durable target modulation, but their development has largely centered on cysteine. Lysine offers a complementary covalent handle because it is widely distributed across catalytic sites, ligand-binding pockets, allosteric regions, and protein-protein interaction interfaces. Its high protonation state and proteomic abundance, however, make selective engagement particularly challenging. Recent studies have shown that productive lysine targeting depends less on the intrinsic reactivity of an aminophilic warhead than on molecular recognition, reac

Lactate-Mediated Lysine Lactylation in Renal Fibrosis: Current Progress and Challenges.

Renal fibrosis constitutes the core pathological hallmark of chronic kidney disease (CKD) progression. Its intricate pathogenesis and the paucity of efficacious clinical interventions have elevated it to a critical global public health concern. Recently, the intersection of metabolic reprogramming and epigenetic regulation has illuminated that aberrant lactate accumulation associated with dysregulated glycolysis and the ensuing imbalance of histone and non-histone lysine lactylation (Kla) may represent an important molecular link between renal cellular metabolic perturbations and the fibrotic

Epigenetic roles of lysine-specific demethylase 1 (LSD1) in cancer metabolism.

Lysine-specific demethylase 1 (LSD1) is a histone demethylase with a crucial role in cancer initiation and progression. Several LSD1 inhibitors and degraders are undergoing clinical trials. Metabolic reprogramming, a key hallmark of cancer cells, allows them to survive in harsh environments. Studies have highlighted the roles of epigenetic regulators, particularly LSD1, in modulating metabolic pathways to promote cancer cell growth, metastasis, and invasion. In this review, we highlight the roles of glucose, glutamine (Gln), and fatty acid (FA) metabolism in tumor cells, as well as the signali

SLC7A2-Mediated Lysine Catabolism Inhibits Immunosuppression in Triple Negative Breast Cancer.

Breast cancer is one of the most common malignant tumors worldwide. SLC7A2 is abnormally expressed in multiple cancers. However, its potential in triple negative breast cancer (TNBC) is still unclear. The purpose of this study was to investigate the roles of SLC7A2 and its underlying molecular mechanisms in TNBC. mRNA expression was detected by RT-qPCR. Protein expression was detected by western blot. Co-localization of ACOX1 and TCF1 was determined using FISH assay. Histone crotonylation was performed using in vitro histone crotonylation assay. Functional analysis was performed using CCK-8 an

Lysine methyltransferase SMYD2 inhibits antiviral innate immunity by promoting IRF3 dephosphorylation.

Phosphorylation of IRF3 is critical to induce type I interferon (IFN-I) production in antiviral innate response. Here we report that lysine methyltransferase SMYD2 inhibits the expressions of IFN-I and proinflammatory cytokines in macrophages upon viral infections. The Smyd2-deficient mice are more resistant to viral infection by producing more IFN-I and proinflammatory cytokines. Mechanistically, SMYD2 inhibits IRF3 phosphorylation in macrophages in response to viral infection independent of its methyltransferase activity. We found that SMYD2 interacts with the DNA-binding domain (DBD) and IR

Strategic Design of Catalytic Lysine-Targeting Reversible Covalent BCR-ABL Inhibitors*.

Targeted covalent inhibitors have re-emerged as validated drugs to overcome acquired resistance in cancer treatment. Herein, by using a carbonyl boronic acid (CBA) warhead, we report the structure-based design of BCR-ABL inhibitors via reversible covalent targeting of the catalytic lysine with improved potency against both wild-type and mutant ABL kinases, especially ABLT315I bearing the gatekeeper residue mutation. We show the evolutionarily conserved lysine can be targeted selectively, and the selectivity depends largely on molecular recognition of the non-covalent pharmacophore in this clas

Lysine methyltransferase 2D regulates muscle fiber size and muscle cell differentiation.

Kabuki syndrome (KS) is a rare genetic disorder caused primarily by mutations in the histone modifier genes KMT2D and KDM6A. The genes have broad temporal and spatial expression in many organs, resulting in complex phenotypes observed in KS patients. Hypotonia is one of the clinical presentations associated with KS, yet detailed examination of skeletal muscle samples from KS patients has not been reported. We studied the consequences of loss of KMT2D function in both mouse and human muscles. In mice, heterozygous loss of Kmt2d resulted in reduced neuromuscular junction (NMJ) perimeter, decreas

Lysine demethylase 3a in craniofacial and neural development during Xenopus embryogenesis.

Epigenetic modifier lysine demethylase 3a (Kdm3a) specifically demethylates mono‑ and di‑methylated ninth lysine of histone 3 and belongs to the Jumonji domain‑containing group of demethylases. Kdm3a serves roles during various biological and pathophysiological processes, including spermatogenesis and metabolism, determination of sex, androgen receptor‑mediated transcription and embryonic carcinoma cell differentiation. In the present study, physiological functions of Kdm3a were evaluated during embryogenesis of Xenopus laevis. Spatiotemporal expression pattern indicated that kdm3a exhibited i

Histone lysine dimethyl-demethylase KDM3A controls pathological cardiac hypertrophy and fibrosis.

Left ventricular hypertrophy (LVH) is a major risk factor for cardiovascular morbidity and mortality. Pathological LVH engages transcriptional programs including reactivation of canonical fetal genes and those inducing fibrosis. Histone lysine demethylases (KDMs) are emerging regulators of transcriptional reprogramming in cancer, though their potential role in abnormal heart growth and fibrosis remains little understood. Here, we investigate gain and loss of function of an H3K9me2 specific demethylase, Kdm3a, and show it promotes LVH and fibrosis in response to pressure-overload. Cardiomyocyte

Proteome-wide analysis of lysine acetylation in adult Schistosoma japonicum worm.

UNLABELLED: Lysine acetylation, a ubiquitous and conserved posttranslational modification, has recently been shown to participate in many diverse non-chromatin-associated biological processes in prokaryotes and eukaryotes. However, the full extent and functional significance of acetylation in Schistosoma japonicum is still unknown. To investigate the nature, extent, and biological functions of lysine acetylation in schistosomes, immunoaffinity-based acetyl-lysine peptide enrichment, integrated with mass spectrometry, was used to comprehensively characterize the lysine-acetylated proteins in th

Global profiling of lysine acetylation in human histoplasmosis pathogen Histoplasma capsulatum.

Histoplasma capsulatum is the causative agent of human histoplasmosis, which can cause respiratory and systemic mycosis in immune-compromised individuals. Lysine acetylation, a protein posttranslational protein modification, is widespread in both eukaryotes and prokaryotes. Although increasing evidence suggests that lysine acetylation may play critical roles in fungus physiology, very little is known about its extent and function in H. capsulatum. To comprehensively profile protein lysine acetylation in H. capsulatum, we performed a global acetylome analysis through peptide prefractionation, a

Synthesis and antibacterial activity of amphiphilic lysine-ligated neomycin B conjugates.

Amphiphilic lysine-ligated neomycin B building blocks were prepared by reductive amination of a protected C5″-modified neomycin B-based aldehyde and side chain-unprotected lysine or lysine-containing peptides. It was demonstrated that a suitably protected lysine-ligated neomycin B conjugate (NeoK) serves as a building block for peptide synthesis, enabling incorporation of aminoglycoside binding sites into peptides. Antibacterial testing of three amphiphilic lysine-ligated neomycin B conjugates against a representative panel of Gram-positive and Gram-negative strains demonstrates that C5″-modif

The CHD3 remodeler PICKLE promotes trimethylation of histone H3 lysine 27.

CHD3 proteins are ATP-dependent chromatin remodelers that contribute to repression of developmentally regulated genes in both animal and plant systems. In animals, this repression has been linked to a multiple subunit complex, Mi-2/NuRD, whose constituents include a CHD3 protein, a histone deacetylase, and a methyl-CpG-binding domain protein. In Arabidopsis, PICKLE (PKL) codes for a CHD3 protein that acts during germination to repress expression of seed-associated genes. Repression of seed-associated traits is promoted in pkl seedlings by the plant growth regulator gibberellin (GA). We underto

Poly(L-lysine)-modified iron oxide nanoparticles for stem cell labeling.

New surface-modified iron oxide nanoparticles were developed by precipitation of Fe(II) and Fe(III) salts with ammonium hydroxide and oxidation of the resulting magnetite with sodium hypochlorite, followed by the addition of poly( L-lysine) (PLL) solution. PLL of several molecular weights ranging from 146 ( L-lysine) to 579 000 was tested as a coating to boost the intracellular uptake of the nanoparticles. The nanoparticles were characterized by TEM, dynamic light scattering, FTIR, and ultrasonic spectrometry. TEM revealed that the particles were ca. 6 nm in diameter, while FTIR showed that th
