Signed, sealed, delivered: a generalizable model for living biotherapeutic dosing and metabolism.

Living Biotherapeutic Products (LBPs) offer a promising therapeutic strategy for metabolic disorders rooted in gut microbiome dysfunction, yet quantitative frameworks for predicting their efficacy remain underdeveloped. We introduce the Bacterial Compartment Absorption and Transit (BCAT) model, a pharmacokinetic-pharmacodynamic framework that couples probiotic transit, endogenous microbiome metabolism, and enzymatic transformation within a unified dose-optimization setting. Building on the classical CAT model, BCAT incorporates mechanistically-derived colon compartments and treats dosing time

Gut Microbial Choline TMA-Lyase CutC: From Metabolic Mechanism to a Novel Therapeutic Target for Diseases.

In recent years, the pivotal role of the gut microbiota and its metabolites in host health and disease has garnered increasing attention. Dietary phosphatidylcholine and choline are metabolized by gut bacteria to generate trimethylamine (TMA). Upon entering the bloodstream, TMA is oxidized by host liver enzymes to trimethylamine N-oxide (TMAO), a known independent risk factor for various systemic diseases, including atherosclerosis, thrombosis, and chronic kidney disease. Within this complex "diet-gut-host" metabolic axis, the microbial choline TMA-lyase (CutC) acts as the key rate-limiting en

Electric-Field Pixel Array Biosensor for Catalytic Ion Detection in Diabetic Nephropathy.

Trimethylamine N-oxide (TMAO) has recently emerged as a clinically relevant biomarker for the early diagnosis and risk stratification of chronic diabetic nephropathy and cardiovascular disease. Here, we report an electric-field-driven pixel array urine biosensor for the assessment of diabetic nephropathy and for elucidating the relationship among urine TMAO, glucose, and urine microalbumin-to-creatinine ratio (UACR). The purified trimethylamine N-oxide reductase (torA) enzyme catalyzes the reduction of TMAO to trimethylamine (TMA) and H2O, during which generated H+ ions modulate the local ioni

Dietary Nutrients, Gut Microbiota, and Cardiac Function: From Metabolic Mechanisms to Clinical Applications.

BACKGROUND: The heart depends on a continuous and flexible energy supply from fatty acids, glucose, and other substrates. Emerging evidence shows that gut microbiota-derived metabolites-such as trimethylamine-N-oxide (TMAO), short-chain fatty acids (SCFAs), secondary bile acids, indoles, phenylacetylglutamine (PAGln), and branched-chain amino acids-modulate cardiac metabolism and function. Although clinical evidence linking these metabolites to cardiovascular outcomes is expanding, most data remain associative, with limited causal or interventional proof. METHODS: A comprehensive narrative re

Proteomic and metabolomic analysis reveals new insights into quaternary amine metabolism in Citrobacter amalonaticus CJ25.

Gut microbiota and human physiology are closely linked, and our microbiota can influence disease through immunological and metabolic activity. Quaternary amines, such as choline and carnitine, are abundant in the human gut and are found in red meat, beef, eggs, seafood, wheat, and beets. Canonically, choline and carnitine are broken down into the pro-atherogenic metabolite, trimethylamine (TMA), by various microbes in the human gut. A glycyl radical enzyme, CutC, is involved in the breakdown of choline to TMA. Carnitine is broken down to TMA via a gamma-butyrobetaine intermediate. TMA from the

Carnitine: Its Crucial Role in Metabolic Health and Cardiovascular Function.

Carnitine is an amino acid derivative synthesized from lysine and methionine that is vital for fatty acid metabolism and energy production. Predominantly in its L-form, carnitine facilitates the transport of long-chain fatty acids into mitochondria for β-oxidation, which is necessary for adenosine triphosphate production. While carnitine deficiency is rare, even among vegetarians, its supplementation could be a promising avenue to address various cardiovascular issues, including ischemia, arrhythmias, and cardiomyopathy. Evidence strongly suggests that carnitine can remarkably improve cardiac

Targeted curation of the gut microbial gene content modulating human cardiovascular disease.

One of the most-cited examples of the gut microbiome modulating human disease is the microbial metabolism of quaternary amines from protein-rich foods. By-products of this microbial processing promote atherosclerotic heart disease, a leading cause of human mortality globally. Our research addresses current knowledge gaps in our understanding of this microbial metabolism by holistically inventorying the microorganisms and expressed genes catalyzing critical atherosclerosis-promoting and -ameliorating reactions in the human gut. This led to the creation of an open-access resource, the Methylated

Variable solid state aggregations in a series of (isocyanide)gold(I) halides with the novel trimethylamine-isocyanoborane adduct.

This paper provides evidence that the degree of aurophilic character and resulting solid state arrangement observed in (isocyanide)gold(I) halides is in part dependent on the nature of halide substituent. In particular, we report here the synthesis of a series of novel (isocyanide)gold(I) halides involving trimethylamine-isocyanoborane, an unusual 'zwitterionic' isocyanide that is comparable to the more familiar methyl isocyanide, but potentially more strongly Lewis basic. The reaction of chloro(dimethyl sulfide)gold(I) and the isocyanoborane species, (RNC)(R =(H3C)3NB(H)2), gives the novel cr

Metabolomics of tracheal wash samples and exhaled breath condensates in healthy horses and horses affected by equine asthma.

The present work characterized the metabolomic profile of tracheal wash (TW) and exhaled breath condensate (EBC) in healthy horses and horses with respiratory disease. Six asthma-affected horses (group A) and six healthy controls (group H) underwent clinical, endoscopic and cytologic examinations of upper airways to confirm the active phase of asthma. TW and EBC samples were collected from each animal and investigated by proton nuclear magnetic resonance (1H-NMR) metabolomic analysis. A total of ten out of 38 metabolites found in the TW were significantly different between the groups (p < 0.05

Methods for syntheses of N-methyl-DL-aspartic acid derivatives.

A novel practical method for the synthesis of N-methyl-DL-aspartic acid 1 (NMA) and new syntheses for N-methyl-aspartic acid derivatives are described. NMA 1, the natural amino acid was synthesized by Michael addition of methylamine to dimethyl fumarate 5. Fumaric or maleic acid mono-ester and -amide were regioselectively transformed into beta-substituted aspartic acid derivatives. In the cases of maleamic 11a or fumaramic esters 11b, the alpha-amide derivative 13 was formed, but hydrolysis of the product provided N-methyl-DL-asparagine 9 via base catalyzed ring closure to DL-alpha-methylamino

Photoinduced electron-transfer substitution reactions via unusual charge-transfer intermediates.

The photoinduced substitution reactions of halogenated alkanes (1-haloadamantanes, 1-haloronorbornanes, menthyl chloride) with a homologous series of amines or alcohols (methylamine, 2-methyl-2-aminopropane, methanol, or 2-methyl-2-propanol) to form the corresponding alkane-substituted amines or ethers and HCl were investigated. The geometry of the bridgehead carbons made S(N)2 reactions impossible. Nonpolar reaction conditions were employed which made classical and nonclassical carbocation S(N)1 reaction pathways unlikely. The reaction rates were measured. Trapping experiments indicated that

Effects of phenothiazines on binding and processing of epidermal growth factor in 3T3 cells.

Chlorpromazine (CPZ) or the functionally related N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide caused a rapid decrease in binding of 125I-epidermal growth factor (EGF) that was due to a specific decrease in receptor affinity. The decrease in ligand binding was observed when cells were exposed to CPZ at either 4 degrees C or 37 degrees C but a rapid reversal of CPZs effects was observed only during a 37 degrees C incubation. In contrast to the decrease in 125I-EGF binding seen after short (30 min) accumulations at 37 degrees C, the presence of CPZ caused a large increase in the amount of c

Characterization of the fourth component of complement in the serum of the clawed frog Xenopus laevis.

The 202,000 MW protein XC, which is distinct from C3 but is essential for antibody-dependent haemolytic activity against SRBC, was obtained from Xenopus plasma following polyethylene glycol precipitation, ion-exchange chromatography and gel-filtration. The protein required other components of the Xenopus serum in order to lyse sensitized SRBC, but did not lyse unsensitized RRBC through the alternative pathway. The protein, contained at 0.17 mg/ml in the original plasma, comprised three distinct subunits of 96,000, 76,000 and 26,000, which were linked by disulphide bonds. Digestion by trypsin r

In situ product monitoring in heterogeneous reaction of gaseous trimethylamine on Fe2O3/Fe(NO3)3: Effect of environmental factor and particle property.

Gas-particle reactions represent an important atmospheric heterogeneous transformation process for organic amines (OAs). Environmental factors and particle properties may impact the gas-particle reaction products. Although the products from gas-particle reactions can be monitored by various in situ techniques, related data remain scarce. Here, the interfacial and gaseous products from the reaction of trimethylamine on Fe2O3/Fe(NO3)3 particles under light irradiation with mixed NO2, O2, SO2 and H2O were monitored using in-situ diffuse reflectance Fourier transform infrared spectroscopy and prot

Trimethylamine N-oxide and Homocysteine in Kidney Function and Disease: Distinct Metabolic Origins, Shared Renal Determinants and Pathophysiological Pathways.

Trimethylamine N-oxide (TMAO) and homocysteine are key biomarkers linked to cardiovascular and chronic kidney disease (CKD). Although they originate from distinct metabolic pathways, their circulating levels are jointly shaped by renal clearance, systemic metabolic status, and nutrient availability, complicating their biological interpretation. TMAO derives from gut microbiota-dependent metabolism of dietary precursors followed by hepatic oxidation of trimethylamine, whereas homocysteine is a central intermediate of one-carbon metabolism governed by remethylation and transsulfuration. Choline

Effect of an acute session of intermittent exercise on trimethylamine N-oxide (TMAO) production following choline ingestion.

INTRODUCTION: Trimethylamine N-oxide (TMAO) is a gut bacteria-dependent metabolite associated with poor cardiovascular health. Exercise is a known cardioprotective activity but the impact of an acute bout of exercise on TMAO production is unknown. OBJECTIVES/METHODS: This study assessed choline-derived production of TMAO following a single bout of intermittent exercise in a young, healthy cohort. RESULTS: Choline supplemented after either exercise or a time-matched resting period demonstrated a similar increase in circulating TMAO across an 8-hour period. CONCLUSION: This suggests that a si

Gut dysbiosis-related thrombosis in inflammatory bowel disease: Potential disease mechanisms and emerging therapeutic strategies.

Patients with inflammatory bowel disease (IBD) have an increased risk of developing venous thromboembolic events, which have a considerable impact on morbidity and mortality. Chronic inflammation plays a crucial role in the pathogenesis of thrombotic events in patients with IBD. However, many unresolved questions remain, particularly regarding the mechanisms that determine the persistent inflammatory state independent of disease activity. This review explored the role of gut microbiota dysbiosis and intestinal barrier dysfunction, which are considered distinctive features of IBD, in determinin

Paper-based electrochemiluminescence device for the rapid estimation of trimethylamine in fish via the quenching effect of thioglycolic acid-capped cadmium selenide quantum dots.

A paper-based electrochemiluminescence device (µPAD-ECL) for the estimation of trimethylamine (TMA) concentration in fish was developed using tris(2,2'-bipyridyl)ruthenium(II) complex coupled with water soluble thioglycolic acid-capped CdSe quantum dots on the inkjet-printed paper-based device. The quenching effect of tertiary amines on the ECL intensity was found to be sensitive and concentration dependent. This effect allows the measurement of TMA at low concentrations. Under the optimal conditions, the linear concentration range was exhibited from 1 × 10-12 to 1 × 10-7 M and a detection lim

Dietary lipids, gut microbiota and lipid metabolism.

The gut microbiota is a central regulator of host metabolism. The composition and function of the gut microbiota is dynamic and affected by diet properties such as the amount and composition of lipids. Hence, dietary lipids may influence host physiology through interaction with the gut microbiota. Lipids affect the gut microbiota both as substrates for bacterial metabolic processes, and by inhibiting bacterial growth by toxic influence. The gut microbiota has been shown to affect lipid metabolism and lipid levels in blood and tissues, both in mice and humans. Furthermore, diseases linked to dy

Flavin-containing monooxygenase 3 (FMO3): genetic variants and their consequences for drug metabolism and disease.

The review focuses on genetic variants of human flavin-containing monooxygenase 3 (FMO3) and their impact on enzyme activity, drug metabolism and disease. The majority of FMO-mediated metabolism in adult human liver is catalyzed by FMO3. Some drugs are metabolized in human liver predominantly by FMO3, but most drug substrates of FMO3 are metabolized also by other enzymes, particularly cytochromes P-450, and the FMO3-catalyzed reaction is not the major route of metabolism.Rare variants that severely affect production or activity of FMO3 cause the disorder trimethylaminuria and impair metabolism
