Technological advances in microfluidic electroporation for biomedical applications.

Intracellular delivery is a fundamental enabling technology in biomedicine, allowing exogenous nucleic acids, proteins, drugs and nanomaterials to access intracellular targets and regulate cellular functions. This capability supports disease mechanism studies, cell engineering, therapeutic development and diagnostic analysis. However, conventional bulk electroporation is limited by high voltages, non-uniform electric fields, Joule heating, electrochemical by-products and reduced cell viability. Microfluidic electroporation integrates microscale cell handling with spatially confined electric fi

Microfluidic Mechanical Reactivation of Senescent Stem Cells.

Stem cell aging significantly impairs therapeutic efficacy, requiring innovative strategies to restore potency. We present a microfluidic cell-compressing platform for reactivation (µ-CPR) designed to apply controlled hydrodynamic deformation to late-passage stem cells. This mechanical stimulation facilitates functional reactivation without external chemical cues. Within a defined window, µ-CPR reduces oxidative stress, SA-β-gal activity, and γ-H2A.X foci, while enhancing proliferation and increasing the expression of canonical stemness-associated markers, including OCT4, SOX2, and KLF4. Quantification of Viruses in Wastewater on a Centrifugal Microfluidic Disc.

Rapid and onsite detection of pathogens in water is a critical first step in preventing the spread of infectious diseases from the environment to humans. However, current microbial monitoring practices are tedious, expensive, and slow. These limitations significantly impede our ability to promptly identify potential risks to public health, leading to delays in implementing the necessary interventions. In this study, we report the development of a portable centrifugal microfluidic disc (CD) that integrates sample concentration, purification, and a droplet digital reverse transcription LAMP (ddR

Suspended Tissue Open Microfluidic Patterning (STOMP).

Free-standing tissue structures tethered between pillars are powerful mechanobiology tools for studying cell contraction. To model interfaces ubiquitous in natural tissues and upgrade existing single-region suspended constructs, we developed Suspended Tissue Open Microfluidic Patterning (STOMP), a method to create multi-regional suspended tissues. STOMP uses open microfluidics and capillary pinning to pattern subregions within free-standing tissues, facilitating the study of complex tissue interfaces, such as diseased-healthy boundaries (e.g., fibrotic-healthy) and tissue-type interfaces (e.g.

Current microfluidic platforms for reverse engineering of cornea.

According to the World Health Organization, corneal blindness constitutes 5.1% of global blindness population. Surgical outcomes have been improved significantly in the treatment of corneal blindness. However, corneal transplantation is limited by global shortage of donor tissue, prompting researchers to explore alternative therapies such as novel ocular pharmaceutics to delay corneal disease progression. Animal models are commonly adopted for investigating pharmacokinetics of ocular drugs. However, this approach is limited by physiological differences in the eye between animals and human, eth

Operando NMR metabolomics of a microfluidic cell culture.

In this work we demonstrate the use of microfluidic NMR for in situ culture and quantitative analysis of metabolism in hepatocellular carcinoma (HCC) cell lines. A hydrothermal heating system is used to enable continuous in situ NMR observation of HCC cell culture over a 24 h incubation period. This technique is nondestructive, non-invasive and can measure millimolar concentrations at microlitre volumes, within a few minutes and in precisely controlled culture conditions. This is sufficient to observe changes in primary energy metabolism, using around 500-3500 cells per device, and with a time

On-site bioaerosol sampling and detection in microfluidic platforms.

As the recent coronavirus disease (COVID-19) pandemic and several severe illnesses such as Middle East respiratory syndrome coronavirus (MERS-CoV), Influenza A virus (IAV) flu, and severe acute respiratory syndrome (SARS) have been found to be airborne, the importance of monitoring bioaerosols for the control and prevention of airborne epidemic diseases outbreaks is increasing. However, current aerosol collection and detection technologies may be limited to on-field use for real-time monitoring because of the relatively low concentrations of targeted bioaerosols in air samples. The Application of Microfluidic Techniques on Tissue Engineering in Orthopaedics.

BACKGROUND: Tissue engineering (TE) is a promising solution for orthopaedic diseases such as bone or cartilage defects and bone metastasis. Cell culture in vitro and scaffold fabrication are two main parts of TE, but these two methods both have their own limitations. The static cell culture medium is unable to achieve multiple cell incubation or offer an optimal microenvironment for cells, while regularly arranged structures are unavailable in traditional cell-laden scaffolds, which results in low biocompatibility. To solve these problems, microfluidic techniques are combined with TE. By provi

Microfluidic chip fabrication and method to detect influenza.

Fast and effective diagnostics play an important role in controlling infectious disease by enabling effective patient management and treatment. Here, we present an integrated microfluidic thermoplastic chip with the ability to amplify influenza A virus in patient nasopharyngeal (NP) swabs and aspirates. Upon loading the patient sample, the microfluidic device sequentially carries out on-chip cell lysis, RNA purification and concentration steps within the solid phase extraction (SPE), reverse transcription (RT) and polymerase chain reaction (PCR) in RT-PCR chambers, respectively. Microfluidic immunoassay for rapid detection of cotinine in saliva.

A microfluidic immunoassay is successfully developed for rapid analysis of cotinine saliva samples, which is a metabolite of nicotine and is widely used as a biomarker to evaluate the smoking status and exposure to tobacco smoke. The core microfluidic chip is fabricated by polydimethylsiloxane (PDMS) with standard soft lithography. Each chip is capable of eight parallel analyses of cotinine samples. The analyses can be completed within 40 min with 12 μl sample consumption. The linear detection range is 1 ~ 250 ng/ml and the minimum detectable concentration is 1 ng/ml respectively. The correlat

Stiffness dependent separation of cells in a microfluidic device.

Abnormal cell mechanical stiffness can point to the development of various diseases including cancers and infections. We report a new microfluidic technique for continuous cell separation utilizing variation in cell stiffness. We use a microfluidic channel decorated by periodic diagonal ridges that compress the flowing cells in rapid succession. The compression in combination with secondary flows in the ridged microfluidic channel translates each cell perpendicular to the channel axis in proportion to its stiffness. We demonstrate the physical principle of the cell sorting mechanism and show t

Microfluidic platform for combinatorial synthesis in picolitre droplets.

This paper presents a droplet-based microfluidic platform for miniaturized combinatorial synthesis. As a proof of concept, a library of small molecules for early stage drug screening was produced. We present an efficient strategy for producing a 7 × 3 library of potential thrombin inhibitors that can be utilized for other combinatorial synthesis applications. Picolitre droplets containing the first type of reagent (reagents A(1), A(2), …, A(m)) were formed individually in identical microfluidic chips and then stored off chip with the aid of stabilizing surfactants. These droplets were then mix

A switchable digital microfluidic droplet dye-laser.

Digital microfluidic devices allow the manipulation of droplets between two parallel electrodes. These electrodes can act as mirrors generating a micro-cavity, which can be exploited for a droplet dye-laser. Three representative laser-dyes with emission wavelengths spanning the whole visible spectrum are chosen to show the applicability of this concept. Sub-microlitre droplets of laser-dye solution are moved in and out of a lasing site on-chip to down-convert the UV-excitation light into blue, green and red laser-pulses.

[Study on temperature measurement and control for microfluidic systems].

A technique for temperature control and measurement in microfluidic systems was developed. The technique is easy to be implemented in an analytical lab and the experimental setup is relatively simple and inexpensive. The parts for temperature measurement include a CCD camera, a fluorescence microscope and an image acquisition card. The spatial distribution and the temporal variation of temperature in microfluids were measured. The method is non-invasive and the spatial and temporal resolutions were 0. 8 microm and 40 ms, respectively a thermal control device was built using indium-tin-oxide (I

Sickle cell vasoocclusion and rescue in a microfluidic device.

The pathophysiology of sickle cell disease is complicated by the multiscale processes that link the molecular genotype to the organismal phenotype: hemoglobin polymerization occurring in milliseconds, microscopic cellular sickling in a few seconds or less [Eaton WA, Hofrichter J (1990) Adv Protein Chem 40:63-279], and macroscopic vessel occlusion over a time scale of minutes, the last of which is necessary for a crisis [Bunn HF (1997) N Engl J Med 337:762-769]. Using a minimal but robust artificial microfluidic environment, we show that it is possible to evoke, control, and inhibit the collect

A microfluidic device for measuring cellular membrane potential.

Recent developments in microfluidics have enabled the design of a lab-on-a-chip system capable of measuring cellular membrane potential. The chip accesses liquid samples sequentially by sipping from a microplate through a capillary, mixes the samples with cells flowing through a microchannel, contacts the cells with potential-sensitive dyes, and reads out cellular responses using fluorescence detection. The rate of cellular uptake of membrane-permeable, ionic fluorophores by THP-1 cells was found to depend strongly on membrane potential. The ratio of the fluorescence of the anionic dye DiBAC(4

Responsive Open Microfluidic Logic for Mechanical Computing.

Logic circuits, a cornerstone of digital computation, have been increasingly adapted to microfluidic systems to enable autonomous, programmable fluidic operations that can also interact with the environment. Unlike traditional microfluidics, which rely on enclosed channels and external pumps, open microfluidics leverage capillary spontaneous flow to manipulate liquids. Integrating logic into these systems facilitates sequential dispensing, mixing, and reaction triggering, without the need for external electronic inputs or pumps. This study leverages elasto-capillary coupling to implement logic

Modular microfluidic probe for addressable fluidic landscapes.

Applications in chemistry, biology, and diagnostics increasingly require precise and reconfigurable delivery of chemical cues on open and accessible surfaces. Achieving such spatially resolved and dynamically programmable control remains challenging with conventional microfluidic systems, whose fixed and enclosed architectures limit adaptability, scalability, and sample access. Here, we introduce a modular, 3D-printed input-processing-output-flow (IPOF) framework that enables plug-and-play reconfiguration of open-space microfluidic functions without redesigning the entire probe. This strategy

Microfluidic valves for wearable sweat analysis: a review.

Wearable sweat analysis can extend health monitoring from clinics to daily life. However, sweat is produced slowly and at changing rates, which makes sweat handling difficult. During collection and transport, sweat may remain in the device, flow backward, mix with older sweat, or become contaminated. The liquid that reaches the sensor may therefore differ from freshly secreted sweat. Microfluidic valves can control when sweat moves, where it flows, and how it reaches the sensing region. This review compares five valve classes relevant to wearable sweat systems. They are capillary, Tesla, mecha
