Some observations on the migration of Proteus mirabilis and other urinary tract pathogens over foley catheters.

The ability of uropathogens to migrate along external surfaces of silicone Foley catheters was examined. Proteus mirabilis and Pseudomonas aeruginosa were the most motile organisms. P. aeruginosa migrated over both triclosan-impregnated and nitrofurazone-impregnated catheters, but these antibacterials inhibited the migration of P. mirabilis.

Identification of plasmid-encoded extended spectrum beta-lactamases produced by a clinical strain of Proteus mirabilis.

Emergence and dissemination of multiresistant strain of Proteus mirabilis have made infections treatment more difficult that this bacterium is responsible. The aim of this study is to determine the implication of the enzymatic mechanism and to describe the properties of ESBLs (extended spectrum beta-lactamases). A clinical strain of Proteus mirabilis SM514 isolated in the intensive care unit at the Military hospital in Tunisia during the period 2004 was found to be highly resistant to cephalosporins and penicilins. Cells sonicate of the isolate hydrolysed cefotaxime more efficiently than ceftr

A sensor to detect the early stages in the development of crystalline Proteus mirabilis biofilm on indwelling bladder catheters.

A simple sensor has been developed to detect the early stages of urinary catheter encrustation and avoid the clinical crises induced by catheter blockage. In laboratory models of colonization by Proteus mirabilis, the sensor signaled encrustation at an average time of 43 h before catheters were blocked with crystalline biofilm.

Natural antibiotic susceptibility of Proteus spp., with special reference to P. mirabilis and P. penneri strains.

The natural susceptibility of 102 Proteus mirabilis and 35 Proteus penneri strains to 71 antibiotics was examined. Minimum inhibitory concentrations (MICs) were determined by applying a microdilution procedure in IsoSensitest broth (for all strains) and cation-adjusted Mueller Hinton broth (for some strains). P. mirabilis and P. penneri were naturally resistant to penicillin G, oxacillin, all tested macrolides, lincosamides, streptogramins, glycopeptides, rifampicin and fusidic acid. Both species were uniformly, naturally sensitive to all tested aminoglycosides, acylureidopenicillins, some cep

A cell-surface polysaccharide that facilitates rapid population migration by differentiated swarm cells of Proteus mirabilis.

Swarming by Proteus mirabilis is characterized by cycles of rapid population migration across surfaces, following differentiation of typical vegetative rods into long, hyperflagellated, virulent swarm cells. A swarm-defective TnphoA insertion mutant was isolated that was not defective in cell motility, differentiation or control of the migration cycle, but was specifically impaired in the ability to undergo surface translocation as a multicellular mass. The mutation, previously shown to compromise urinary tract virulence, was located within a 1112 bp gene that restored normal swarming of the m

Isolation and characterization of a novel class of plant antimicrobial peptides form Mirabilis jalapa L. seeds.

We have isolated from seeds of Mirabilis jalapa L. two antimicrobial peptides, designated Mj-AMP1 and Mj-AMP2, respectively. These peptides are highly basic and consist of 37 and 36 residues for Mj-AMP1 and Mj-AMP2, respectively. Both peptides contain three disulfide bridges and differ from one another only by 4 amino acids. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of the reduced and unreduced peptides suggests that the peptides associate into dimers in their native form. The Mj-AMPs exhibit a broad spectrum of antifungal activity since they are active against all 13

Contribution of three tRNA modification enzymes to Proteus mirabilis fitness and catheter-associated urinary tract infection.

RNA modifications play essential roles in cellular physiology by modulating RNA structure, stability, and translation efficiency. Among these, dihydrouridine is a conserved tRNA modification synthesized by the flavin-dependent enzymes DusA, DusB, and DusC, which introduce flexibility at defined uridine positions in the D-loop. While structural and biochemical studies in Escherichia coli have clarified dihydrouridine synthase (Dus) enzyme substrate specificities, their functional relevance in other bacteria remains uncharacterized. Here, we present the first functional characterization of the c

Cyclodextrin inclusion complexes enhance the solubility and anti-virulence activity of metronidazole against uropathogenic Proteus mirabilis.

Proteus mirabilis is a major uropathogen implicated in catheter-associated urinary tract infections and infection-induced urolithiasis, often exhibiting multidrug resistance. Improving the solubility and bioactivity of existing drugs represents a promising pharmaceutical strategy to overcome these challenges. In this study, inclusion complexes of metronidazole with α-cyclodextrin (α-CD), β-cyclodextrin (β-CD), and hydroxypropyl-β-cyclodextrin (HP-β-CD) were prepared using physical mixing and kneading methods. Complexes were characterized using differential scanning calorimetry (DSC) and Fourie

Pangenome Analysis of Proteus mirabilis Reveals Lineage-Specific Antimicrobial Resistance Profiles and Discordant Genotype-Phenotype Correlations.

UNLABELLED: Urinary tract infections (UTIs) impose a substantial health care burden with increasing antimicrobial resistance and treatment failure rates. Proteus mirabilis is a challenging UTI pathogen due to intrinsic resistances coupled with formation of crystalline biofilms. We combined pangenome analysis, antimicrobial resistance gene (AMR) predication, and antimicrobial susceptibility testing (AST) to determine whether highly multidrug-resistant (MDR) isolates of P. mirabilis arise from distinct lineages and explored the clinical utility of multilocus sequence typing (MLST). The pangenome

Spontaneous Proteus mirabilis Meningitis in Adults Requiring an Extended Antibiotic Course: Case Report and Literature Review.

Most cases of gram-negative bacillary meningitis occur in neonates and infants. Meningitis in adults caused by Proteus mirabilis has been reported rarely. Evidence-based guidelines for the treatment of adult patients with gram-negative bacillus meningitis are scarce. The optimal duration of antibiotic therapy for these patients is an unanswered question in the medical literature. This article outlines a case of community-acquired meningitis caused by P. mirabilis in an adult patient who required an extended antimicrobial treatment, after failing to a three-week antibiotic regime. Our patient,

Induction of Different Apoptosis Pathways by Two Proteus mirabilis Clinical Isolates Strains in Prostatic Epithelial Cells.

Bacterial prostatitis is believed to be the leading cause of recurrent urinary tract infections (UTIs) in men under 50 years of age and occurs both as an acute febrile disease responsive to antibiotics and as a chronic infection that is often unresponsive to antibiotic treatment. Proteus mirabilis is more commonly associated with UTIs in these abnormalities, especially in patients undergoing catheterisation. This pathogen is able to colonise the host's tissues and to cause disease thanks to the production of many virulence factors such as fimbriae, flagella, immune avoidance, host-damaging fac

Molecular diversity of Proteus mirabilis isolates producing extended-spectrum beta-lactamases in a French university hospital.

Between February 1997 and December 2002, 3340 hospitalised patients yielded samples positive for Proteus mirabilis, of whom 45 (1.3%) were colonised/infected by P. mirabilis producing extended-spectrum beta-lactamases (ESBLs). The gross incidence of patients colonised/infected by ESBL-producing P. mirabilis was 1.61/10(5) days of hospitalisation, with 20% of isolates being collected from patients in urology wards, most frequently (53.3%) from urine samples. Seventeen (37.7%) of the 43 isolates were obtained from samples collected within 48 h of hospitalisation, indicating that they were commun

The Integrative and Conjugative Element ICEPmiW2 in Proteus mirabilis W2 Facilitates the Dissemination of Antibiotic-Resistance Genes.

BACKGROUND: The extensive use of antibiotics for treating infectious diseases leads to their release into the environment, which in turn results in antibiotic pollution and thereby facilitates the dissemination of antibiotic-resistance genes (ARGs). Recently, despite the implementation of strict antibiotic usage restrictions, the accumulation of ARGs and multidrug-resistant bacteria in the aquaculture environment continues to show a trend of persistent spread. METHODS: The W2 strain was isolated in the presence of 32 μg/mL doxycycline. A broth microdilution assay was employed to determine the
