What genes are differentially expressed in individuals with schizophrenia? A systematic review.

Schizophrenia is a severe, complex mental disorder characterized by a combination of positive symptoms, negative symptoms, and impaired cognitive function. Schizophrenia is highly heritable (~80%) with multifactorial etiology and complex polygenic genetic architecture. Despite the large number of genetic variants associated with schizophrenia, few causal variants have been established. Gaining insight into the mechanistic influences of these genetic variants may facilitate our ability to apply these findings to prevention and treatment. Though there have been more than 300 studies of gene expr

Boosting Schizophrenia Genetics by Utilizing Genetic Overlap With Brain Morphology.

BACKGROUND: Schizophrenia is a complex polygenic disorder with subtle, distributed abnormalities in brain morphology. There are indications of shared genetic architecture between schizophrenia and brain measures despite low genetic correlations. Through the use of analytical methods that allow for mixed directions of effects, this overlap may be leveraged to improve our understanding of underlying mechanisms of schizophrenia and enrich polygenic risk prediction outcome. METHODS: We ran a multivariate genome-wide analysis of 175 brain morphology measures using data from 33,735 participants of

Comprehensive epithelial tubo-ovarian cancer risk prediction model incorporating genetic and epidemiological risk factors.

BACKGROUND: Epithelial tubo-ovarian cancer (EOC) has high mortality partly due to late diagnosis. Prevention is available but may be associated with adverse effects. A multifactorial risk model based on known genetic and epidemiological risk factors (RFs) for EOC can help identify women at higher risk who could benefit from targeted screening and prevention. METHODS: We developed a multifactorial EOC risk model for women of European ancestry incorporating the effects of pathogenic variants (PVs) in BRCA1, BRCA2, RAD51C, RAD51D and BRIP1, a Polygenic Risk Score (PRS) of arbitrary size, the eff

Breast and prostate cancer risk: The interplay of polygenic risk, rare pathogenic germline variants, and family history.

PURPOSE: We aimed to investigate to what extent polygenic risk scores (PRS), rare pathogenic germline variants (PVs), and family history jointly influence breast cancer and prostate cancer risk. METHODS: A total of 200,643 individuals from the UK Biobank were categorized as follows: (1) heterozygotes or nonheterozygotes for PVs in moderate to high-risk cancer genes, (2) PRS strata, and (3) with or without a family history of cancer. Multivariable logistic regression and Cox proportional hazards models were used to compute the odds ratio across groups and the cumulative incidence through life.

Genetic liability to major depression and risk of childhood asthma.

OBJECTIVE: Major depression and asthma frequently co-occur, suggesting shared genetic vulnerability between these two disorders. We aimed to determine whether a higher genetic liability to major depression was associated with increased childhood asthma risk, and if so, whether such an association differed by sex of the child. METHODS: We conducted a population-based cohort study comprising 16,687 singletons born between 1991 and 2005 in Denmark. We calculated the polygenic risk score (PRS) for major depression as a measure of genetic liability based on the summary statistics from the Major De

Increased Burden of Common Risk Alleles in Children With a Significant Fracture History.

Extreme presentations of common disease in children are often presumed to be of Mendelian etiology, but their polygenic basis has not been fully explored. We tested whether children with significant fracture history and no osteogenesis imperfecta (OI) are at increased polygenic risk for fracture. A childhood significant fracture history was defined as the presence of low-trauma vertebral fractures or multiple long bone fractures. We generated a polygenic score of heel ultrasound-derived speed of sound, termed "gSOS," which predicts risk of osteoporotic fracture. We tested if individuals from t

Autoinflammatory diseases in childhood, part 2: polygenic syndromes.

Autoinflammatory diseases are a family of disorders characterized by aberrant stimulation of inflammatory pathways without involvement of antigen-directed autoimmunity. They can be further divided in monogenic and polygenic types. Those without an identified genetic mutation are known as polygenic and include systemic-onset juvenile idiopathic arthritis, idiopathic recurrent acute pericarditis, Behçet syndrome, chronic recurrent multifocal osteomyelitis and inflammatory bowel disease among others. Autoinflammatory diseases are characterized by recurrent flares or persistent systemic inflammati

Associations between an educational attainment polygenic score with educational attainment in an African American sample.

Polygenic propensity for educational attainment has been associated with higher education attendance, academic achievement and criminal offending in predominantly European samples; however, less is known about whether this polygenic propensity is associated with these outcomes among African Americans. Using an educational attainment polygenic score (EA PGS), the present study examined whether this score was associated with post-secondary education, academic achievement and criminal offending in an urban, African American sample. Three cohorts of participants (N = 1050; 43.9% male) were initial

OligoPVP: Phenotype-driven analysis of individual genomic information to prioritize oligogenic disease variants.

An increasing number of disorders have been identified for which two or more distinct alleles in two or more genes are required to either cause the disease or to significantly modify its onset, severity or phenotype. It is difficult to discover such interactions using existing approaches. The purpose of our work is to develop and evaluate a system that can identify combinations of alleles underlying digenic and oligogenic diseases in individual whole exome or whole genome sequences. Information that links patient phenotypes to databases of gene-phenotype associations observed in clinical or no

Oligogenic inheritance in photosensitive juvenile myoclonic epilepsy?

The interplay of multiple genetic factors, as opposed to monogenic inheritance, is suspected to play a role in many idiopathic generalized epilepsies. This leads to a digenic or oligogenic inheritance model, which although rather simplified, may explain at least some of the clinical observations. Here we describe a family in which the clinical phenotype in the offspring can be explained by a combination of photosensitivity and epilepsy traits that segregated independently of each other. This case history demonstrates the need to evaluate family histories in more detail in order to uncover pote

Advances in the genetics of cerebrovascular disease and stroke.

MEDLINE searches identified epidemiologic, experimental, and clinical studies on the genetics of cerebrovascular disease and stroke, including the following topics: genetic epidemiology of stroke; genetics of systemic disorders that cause ischemic stroke, including coagulation disorders, connective tissue disorders, vasculopathies, metabolic disorders, and disorders of unknown etiology; and genetics of systemic disorders that cause hemorrhagic stroke. Recent discoveries in stroke genetics involve the genetic basis of monogenic disorders such as cerebral autosomal dominant arteriopathy with sub

[Autoinflammatory diseases in children: The contribution of genetics].

THE CONTRIBUTION OF GENETICS. Autoinflammatory diseases (AIDs) have benefited from modern genetic tools, in particular the widespread use of high-throughput sequencing, which has helped to improve our knowledge of the inheritance patterns of AIDs, whether hereditary or multifactorial. The case for genetic testing has become compelling. However, the new modes of inheritance revealed in recent years complicate genetic counselling and justify close collaboration between clinicians and biologists during multidisciplinary clinical meetings.

Familial Zenker's diverticulum.

CONCLUSION: The aetiology of Zenker's diverticulum is likely to be multifactorial. The geographical and racial variation in the prevalence and the rare familial cases suggest that genetic predisposition might have a role in the pathogenesis. The mode of inheritance in the rare possible familial cases seems to be autosomal dominant; recessive inheritance is non-existent or extremely rare. OBJECTIVES: To study the inheritance of Zenker's diverticulum. SUBJECTS AND METHODS: A detailed questionnaire concerning physical health and family history was sent to 122 patients with Zenker's diverticulum

Uncomplicated familial hypospadias: evidence for autosomal recessive inheritance.

Uncomplicated hypospadias was found in eight members of a large, consanguineous Bedouin family. Virilization and fertility were normal in the only postpubertal individual. The inheritance is most likely autosomal recessive and we suggest that in some of the familial cases in which polygenic or multifactorial inheritance was previously proposed, homozygosity for recessive genes may be responsible for the increased risk to siblings. Dominantly transmitted hypertelorism with diastema was an independent and coincidental finding in this family.

The Genes We Inherit and Those We Don't: Maternal Genetic Nurture and Child BMI Trajectories.

Recently, methods have been introduced using polygenic scores (PGS) to estimate the effects of genetic nurture, the environmentally-mediated effects of parental genotypes on the phenotype of their child above and beyond the effects of the alleles which are transmitted to the child. We introduce a simplified model for estimating genetic nurture effects and show, through simulation and analytical derivation, that our method provides unbiased estimates and offers an increase in power to detect genetic nurture of up to 1/3 greater than that of previous methods. Subsequently, we apply this method t

DiscoDivas: Leveraging genetic-ancestry continuum information to interpolate PRS for admixed populations.

Genome-wide association study (GWAS) summary statistics for training and individual-level cohorts for fine-tuning are essential for constructing predictive polygenic risk score (PRS) models. However, the relatively low representation of admixed populations in both GWAS summary statistics and individual-level datasets hinders the development of PRSs and equitable clinical translation for admixed populations. Prior work indicates that the most informative PRS model for a genetically homogeneous sample varies linearly in an ancestry continuum space. Guided by these observations, we introduce a ge

ASTWAS: modeling alternative polyadenylation and SNP effects in kernel-driven TWAS reveal novel genetic associations for complex traits.

Alternative polyadenylation (APA) of $3^{\prime}$untranslated regions ($3^{\prime}$UTRs) is a pervasive mechanism that regulates mRNA stability, localization, and translational efficiency by generating isoforms with distinct $3^{\prime}$UTR lengths and regulatory element composition. Despite its critical role in fine-tuning gene expression, APA has been largely overlooked in transcriptome-wide association studies (TWAS), which traditionally rely on linear models of SNP effects. To bridge this gap, we developed ASTWAS, a two-stage framework that first trains APA usage prediction models (BLUP, E
