Xyloglucan, an abundant polysaccharide bearing galactose side-chains within the cell wall of cotyledons from leguminous seeds, exhibits superior immunomodulatory properties. However, the contribution of its galactose side-chains remains unclear. This study aimed to elucidate the role of the galactose side-chains in the immunomodulatory activity by comparing the effects of oligosaccharides with different degrees of galactosyl substitution on RAW 264.7 mouse macrophages. Three structurally distinct xyloglucan fractions were prepared from tamarind seed: native high-molecular-weight xyloglucan (XG

Alzheimer's disease (AD) is the most common neurodegenerative disorder and the primary cause of dementia. Although natural polysaccharides exhibit significant therapeutic potential against AD due to their multi-target activities, the structure-activity relationship (SAR) of uronic acid oligosaccharides-particularly the influence of specific glycosidic linkages and monosaccharide compositions-remains insufficiently characterized. To address this, we utilized a highly selective TEMPO oxidation system to prepare four structurally distinct uronic acid oligosaccharides (α1,4-G, β1,4-G, β1,3-G, and

Carbohydrate simulations are highly sensitive to the treatment of hydroxyl electrostatics because hydration, conformational sampling, and reactivity are governed by dense hydrogen-bonding networks. Here, we examine how oxygen quadrupole scaling affects AMOEBA d-glucose models generated with Poltype2 by comparing the standard 0.7 scaling with full 1.0 scaling for α- and β-d-glucose monosaccharides, 1→4-linked oligomers, and concentrated solutions. Increasing the oxygen quadrupole scaling from 0.7 to 1.0 improves monosaccharide hydration free energies, increases d-glucose-water hydrogen-bond cou

Carrageenan is a sulfated polysaccharide extracted from red algae, widely utilized in the food and pharmaceutical industries due to its unique gelling properties and biological activities. However, its high molecular weight and low solubility limit certain high-value applications. Carrageenases specifically degrade carrageenan to produce low-molecular-weight oligosaccharides with excellent bioactivities, thereby overcoming this bottleneck. This review systematically summarizes the microbial sources of carrageenases and the classification characteristics of their three types (κ, ι, and λ) withi

The objective of this study was to investigate the effect of mannose oligosaccharides (MOS) against cadmium (Cd)-induced hepatic oxidative damage and analyze its underlying antioxidant mechanism. Thirty male Sprague-Dawley (SD) rats were randomly divided into five groups: control group and four others treated with cadmium chloride (CdCl2) (2 mg/kg body weight (b.w.)) and different MOS levels at 0, 100, 300, 500 mg/kg b.w.. The results demonstrated that administration of MOS at a dose of 500 mg/kg significantly reduced Cd-induced oxidative damage in rat livers. This was evidenced by an increase

Galacto-oligosaccharides (GOS) and fructo-oligosaccharides (FOS) are food ingredients that improve human health, but their degradation throughout the human small intestine is not well understood. We studied the breakdown kinetics of FOS and GOS in the intestines of seven healthy Dutch adults. Subjects were equipped with a catheter in the distal ileum or proximal colon and consumed 5 g of chicory-derived FOS (degree of polymerization (DP) DP2-10), and 5 g of GOS (DP2-6). Postprandially, intestinal content was frequently collected until 350 min and analyzed for mono-, di-, and oligosaccharides.

Brucellosis is a dangerous zoonotic disease caused by bacteria of the genus Brucella. Diagnosis of brucellosis is based on the detection in animal and human sera of antibodies to the O-polysaccharide of Brucella lipopolysaccharide. The currently employed serodiagnosis of brucellosis relies on the use of the Brucella O-polysaccharide as a diagnostic antigen. However, the existence of bacterial species, which also express O-polysaccharides structurally similar to that of Brucella, may decrease the specificity of the brucellosis detection due to false-positive test results. It has been shown that

Chitosan oligosaccharide and its derivatives are known for their diverse biological activities. In this study, we communicate a convenient one-pot synthesis of N,N-dimethyl chitosan oligosaccharide (DMCOS) from chitin via acid-catalyzed tandem depolymerization-deacetylation-N-methylation pathway using formaldehyde as the methylation reagent. The synthesis protocol offers 77 % DMCOS that features a high degree of deacetylation, a high degree of methylation, and a low average molecular weight. Compared to chitosan, DMCOS exhibits superior antifungal activity against Candida species. Mechanism st

Necrotizing enterocolitis (NEC) is the leading cause of morbidity and mortality in premature infants. One of the most devastating complications of NEC is the development of NEC-induced brain injury, which manifests as impaired cognition that persists beyond infancy and which represents a proinflammatory activation of the gut-brain axis. Given that oral administration of the human milk oligosaccharides (HMOs) 2'-fucosyllactose (2'-FL) and 6'-sialyslactose (6'-SL) significantly reduced intestinal inflammation in mice, we hypothesized that oral administration of these HMOs would reduce NEC-induce

Emerging antimicrobial resistance in infections asks for novel intervention strategies. Galacto-oligosaccharides (GOS) might be attractive alternatives to antibiotics due to their anti-inflammatory and anti-adhesive properties. Mannheimia haemolytica is one of the major Pasteurellaceae associated with bovine lung infections. Using M. haemolytica, we demonstrated that GOS have the capacity to reduce bacterial viability and can be used as adjuvant to improve antibiotic efficacy. Using M. haemolytica-treated primary bronchial epithelial cells (PBECs) of calves, we identified the anti-adhesive and

Free oligosaccharides (fOSs) are soluble oligosaccharide species generated during N-glycosylation of proteins. Although little is known about fOS metabolism, the recent identification of NGLY1 deficiency, a congenital disorder of deglycosylation (CDDG) caused by loss of function of an enzyme involved in fOS metabolism, has elicited increased interest in fOS processing. The catabolism of fOSs has been linked to the activity of a specific cytosolic mannosidase, MAN2C1, which cleaves α1,2-, α1,3-, and α1,6-mannose residues. In this study, we report the clinical, biochemical, and molecular feature

Neisseria meningitidis (Nm) serogroup W (NmW) is one of the six meningococcal serogroups that cause majority of invasive meningococcal diseases (IMD). Its capsular polysaccharide (CPS) is a virulence factor and is a key component in NmW CPS-protein conjugate vaccines. The current clinically used NmW CPS-protein conjugate vaccines are effective but the costs are high and the products are heterogeneous at both the CPS and the conjugate levels. Towards the development of potentially better NmW CPS vaccines, herein we report the synthesis of homogeneous oligosaccharides of NmW CPS in a size-contro

Sepsis induces heparanase-mediated degradation of the endothelial glycocalyx, a heparan sulfate-enriched endovascular layer critical to vascular homeostasis, releasing highly sulfated domains of heparan sulfate into the circulation. These domains are oligosaccharides rich in heparin-like trisulfated disaccharide repeating units. Using a chemoenzymatic approach, an undecasaccharide containing a uniformly 13C-labeled internal 2-sulfoiduronic acid residue was synthesized on a p-nitrophenylglucuronide acceptor. Selective periodate cleavage afforded a heparin nonasaccharide having a natural structu

This study evaluated the immunostimulatory activity of curdlan oligosaccharides (GOS) in cyclophosphamide (CTX)-induced immunosuppressed mice and in RAW264.7 cells. GOS was able to stimulate the release of nitric oxide (NO), cytokines (IL-1β, IL-6 and TNF-α) and improve the phagocytic rate of peritoneal macrophages and RAW264.7 cells. It further enhanced immunoglobulins (Ig) release (IgG by 50.6%-74.7%, IgA by 31.3%-34.9%, IgM by 28.3%-66.7%), splenic lymphocyte proliferation (by 74.8%-91.3%), nature killer cells cytotoxicity (by 32.0%-49.6%), immunophenotypes of splenic lymphocytes (from 1.7

Milk oligosaccharides (MOs) are complex carbohydrates with multifunctional health benefits for the neonate. Poor reproductive performance in primiparous gilts limits their productivity. Changes in the structure and abundance of porcine MO (PMOs) through lactation with parity remains unknown and may explain superior new-born growth in litters from multiparous sows relative to gilts. We report 55 PMOs structures, of which 25 are new (17 sialylated and 8 neutral). Their incidence in gilt and sow colostrum was almost identical (53 vs. 54), but not in transitional milk (48 vs. 53) nor mature milk (
