Navigating the neuronal recycling bin: Another look at huntingtin in coordinating autophagy.

Neurons, as post-mitotic and long-lived cells, rely heavily on autophagy to maintain cellular homoeostasis and ensure proper function. Huntingtin (HTT), a protein central to Huntington's disease (HD), has emerged as a putative multifunctional regulator within the neuronal autophagy-lysosome pathway. This review explores normal HTT's multifaceted role in neuronal autophagy, from its potential involvement in autophagy induction, its capacity to influence cargo recognition and autophagosome formation, and its contribution to autophagosome-lysosome fusion and transport. We also discuss the unique

Lightweight YOLO object detectors for PET and HDPE classification in recycling facilities.

Growing volumes of plastic waste highlight the need for reliable sorting technologies. This study examines whether lightweight YOLO object detectors can distinguish polyethylene terephthalate (PET) bottles from high-density polyethylene (HDPE) bottles, two common packaging materials that must be separated for recycling. Two public PET and HDPE datasets were augmented offline to simulate the visual variation typical of industrial sorting lines, and eleven compact YOLO models-nano, tiny and small versions across versions 7-12-were fine-tuned on these data. Their accuracy was quantified using mea

Harnessing CO2 fixation and reducing power recycling for enhanced polyhydroxyalkanoates industrial bioproduction.

Palm oil is an attractive feedstock for bioproduction due to its high carbon content and low cost. However, its metabolism generates excess reducing power, leading to redox imbalances and reduced metabolic efficiency in industrial fermentations. Through a model-driven approach integrating flux balance analysis, we activated the Calvin-Benson-Bassham (CBB) cycle in Cupriavidus necator to recycle surplus reducing power and restore metabolic balance in polyhydroxyalkanoate (PHA) bioproduction. Computational simulations predicted that constitutive activation of the CBB cycle enhanced CO2 fixation

Closed-Loop Recycling of Poly(vinyl butyral) Interlayer Film via Restabilization Technology.

Polyvinyl butyral (PVB) is a specialty polymer primarily used as an interlayer in laminated glass applications with no current circularity plan after the end of its life. This study presents a comprehensive recycling strategy for postconsumed PVB wastes based on a remelting-restabilization approach. Thermo-oxidative degradation of PVB was analyzed under heat and shear stress conditions in an internal mixer apparatus. The degradation mechanism of plasticized PVB (p-PVB) and unplasticized PVB (u-PVB) was identified as chain scission through melt flow rate (MFR), intrinsic viscosity (IV), and yel

A sustainable approach to recycling of polylactic acid with environmentally friendly reagents.

An important aspect before large-scale production and application of bioplastics such as polylactic acid (PLA), is the need to close the life cycle of the material to reduce the need for first-generation biomass and to prevent waste accumulation in the environment. In this work, starting from a high-mass linear commercial PLA, a depolymerization route based on a bulk alcoholysis reaction in the molten state was developed. For this purpose two polyalcohols, pentaerythritol and dipentaerythritol, and an environmentally friendly catalyst, i.e., zinc stearate, were utilized. The formation and spec

Up-recycling of waste wood into value-added room temperature phosphorescent materials.

Up-recycling waste wood as a source for producing materials is crucial for sustainability. Here, we discovered that in situ thermal curing of melamine-formaldehyde resin (MF) with natural wood enhanced its room temperature phosphorescence (RTP) performance. This enhancement is due to the interaction of MF with the components in wood during the in situ thermal curing process, both covalently and non-covalently, promoting RTP emission of holocellulose and lignin in natural wood. With this discovery, we converted waste wood into RTP materials (R-wood@MF) with a lifetime of 332.5 ms at 530 nm via

Dynamic Covalent Chemistry Enabled Closed-Loop Recycling of Thermally Modified Polymer Membrane.

The increasing demand for sustainable solutions to oil-water separation and end-of-life membrane disposal has prompted the development of recyclable membrane technologies. In this study, we present an innovative approach to fabricating closed-loop, recyclable nanofibrous membranes (RFMs) utilizing reversible covalent networks based on the Diels-Alder reaction. A methacrylate-based copolymer was synthesized via free radical polymerization, combining hydrophobic monomers for enhanced separation performance, with furan-functionalized monomers for recyclability. This copolymer was electrospun into

Dual-Factor-Controlled Dynamic Precursors Enable On-Demand Thermoset Degradation and Recycling.

Thermosets are well known for their advantages such as high stability and chemical resistance. However, developing sustainable thermosets with degradability and recyclability faces several principal challenges, including reconciling the desired characteristics during service with the recycling and reprocessing properties required at the end of life, establishing efficient methods for large-scale synthesis, and aligning with current manufacturing process. Here a general strategy is presented for the on-demand degradation and recycling of thermosets under mild conditions utilizing dynamic precur

Anaerobic heme recycling by gut microbes: Important methods for monitoring porphyrin production.

Heme is the most abundant species of iron inside the human body and an essential cofactor for numerous electron/chemical group transfer reactions and catalyses, especially those involving O2. Whole anaerobic biomes exist that also depend on heme but lack widespread, O2-dependent pathways for heme synthesis and breakdown. The gastrointestinal tract is an anaerobic ecosystem where many microbes are auxotrophic for heme, and where the abundant members of the Bacteroidetes phylum convert heme into iron and porphyrins. Working with mixtures of these hydrophobic compounds presents challenges for ana

A facile crush-and-sieve treatment for recycling end-of-life photovoltaics.

The shift towards renewable energy mix has resulted in an exponential growth of the photovoltaic (PV) industry over the past few decades. Parallelly, new recycling technology developments are required to address the incoming volume of waste as they gradually approach their end-of-life (EoL) to realize the concept of a circular economy. Typical recycling processes involve high-temperature burning for separation and release of the PV cells for metal recovery processes. However, this thermal process generates gaseous by-products that cause serious health and environmental issues. Eschewing the ne

Strategies of organic phosphorus recycling by soil bacteria: acquisition, metabolism, and regulation.

Critical to meeting cellular phosphorus (P) demand, soil bacteria deploy a number of strategies to overcome limitation in inorganic P (Pi ) in soils. As a significant contributor to P recycling, soil bacteria secrete extracellular enzymes to degrade organic P (Po ) in soils into the readily bioavailable Pi . In addition, several Po compounds can be transported directly via specific transporters and subsequently enter intracellular metabolic pathways. In this review, we highlight the strategies that soil bacteria employ to recycle Po from the soil environment. We discuss the diversity of extrac

Integrin linked kinase regulates endosomal recycling of N-cadherin in melanoma cells.

Malignant transformation is characterized by a phenotype "switch" from E- to N-cadherin - a major hallmark of epithelial to mesenchymal transition (EMT). The increased expression of N-cadherin is commonly followed by a growing capacity for migration as well as resistance to apoptosis. Integrin Linked Kinase (ILK) is a key molecule involved in EMT and progression of cancer cells. ILK is known as a major signaling mediator involved in cadherin switch, but the specific mechanism through which ILK modulates N-cadherin expression is still not clear. Studies were carried out on human melanoma WM793

Challenges and opportunities of solvent-based additive extraction methods for plastic recycling.

Additives are ubiquitously used in plastics to improve their functionality. However, they are not always desirable in their 'second life' and are a major bottleneck for chemical recycling. Although research on extraction techniques for efficient removal of additives is increasing, it resembles much like uncharted territory due to the broad variety of additives, plastics and removal techniques. Today solvent-based additive extraction techniques, solid-liquid extraction and dissolution-precipitation, are considered to be the most promising techniques to remove additives. This review focuses on t

An electrochemical Hg2+ sensor based on signal amplification strategy of target recycling.

In this work, an unmodified homogeneous electrochemical sensor based on electrochemical bonding and catalytic hairpin assembly (CHA) was first constructed for the high sensitivity detection of Hg2+. Herein, tetraferrocene, a synthesized compound, was used as a signal marker that modified both ends of the hairpin probe to amplify the electrochemical signal. The interaction of T-Hg2+-T could induce the catalytic self-assembly of hairpins by means of auxiliary DNA. The rigid DNA triangle that was formed easily reaches the electrode and induced Au-S self-assembly assisted by potential, allowing te

Hcr1/eIF3j Is a 60S Ribosomal Subunit Recycling Accessory Factor In Vivo.

Hcr1/eIF3j is a sub-stoichiometric subunit of eukaryotic initiation factor 3 (eIF3) that can dissociate the post-termination 40S ribosomal subunit from mRNA in vitro. We examine this ribosome recycling role in vivo by ribosome profiling and reporter assays and find that loss of Hcr1 leads to reinitiation of translation in 3' UTRs, consistent with a defect in recycling. However, the defect appears to be in the recycling of the 60S subunit, rather than the 40S subunit, because reinitiation does not require an AUG codon and is suppressed by overexpression of the 60S dissociation factor Rli1/ABCE1

Impaired Transferrin Receptor Palmitoylation and Recycling in Neurodegeneration with Brain Iron Accumulation.

Neurodegeneration with brain iron accumulation (NBIA) is a genetically heterogeneous condition characterized by progressive dystonia with iron accumulation in the basal ganglia. How NBIA-associated mutations trigger iron overload remains poorly understood. After studying fibroblast cell lines from subjects carrying both known and unreported biallelic mutations in CRAT and REPS1, we ascribe iron overload to the abnormal recycling of transferrin receptor (TfR1) and the reduction of TfR1 palmitoylation in NBIA. Moreover, we describe palmitoylation as a hitherto unreported level of post-translatio

Liquid-liquid hierarchical separation and metal recycling of waste printed circuit boards.

Waste electrical and electronic equipment is rapidly increasing worldwide, resulting in a large quantity of waste printed circuit boards (WPCBs). There is a great challenge on how to efficiently separate mixed metals in WPCBs, which consists of more than 10 elements including hazardous Cr, Pb and Cd. In this work, based on atomic interactions, a method of liquid-liquid hierarchical separation is developed to separate the mixed metals dissociated from the pyrolyzed WPCBs of mobile phones. The hierarchical separation of L→LFe-rich + LCu,Pb-rich, LCu,Pb-rich→LCu-rich + LPb-rich and LPb-rich→SCu-d

Influenza A virus ribonucleoproteins modulate host recycling by competing with Rab11 effectors.

Influenza A virus assembly is an unclear process, whereby individual virion components form an infectious particle. The segmented nature of the influenza A genome imposes a problem to assembly because it requires packaging of eight distinct RNA particles (vRNPs). It also allows genome mixing from distinct parental strains, events associated with influenza pandemic outbreaks. It is important to public health to understand how segmented genomes assemble, a process that is dependent on the transport of components to assembly sites. Previously, it has been shown that vRNPs are carried by recycling
