STING agonists (e.g., cGAMP), which function as immunotherapeutic agents activating the stimulator of interferon genes (STING) pathway, hold promise for treating pediatric solid tumors including neuroblastoma. However, their clinical translation is hindered by limitations including burst release, short half-life, poor targeting, and low intracellular delivery efficiency. In this study, we engineered a peptide self-assembled nanofiber (NFSA) based on the specific interaction mechanism between STING protein and cGAMP. By hydrophobically modifying the cGAMP-binding amino acid sequences on STING,

Protein arginine methyltransferase 6 (Prmt6), an epigenetic regulator, plays an unclear role in bone homeostasis and osteoblast senescence. In this study, we employed Western blot, qPCR, immunofluorescence, SA-β-Gal staining, and RNA-seq to systematically investigate the regulatory mechanism of the Prmt6/H3R2me2a/Sting/Ifitm3 axis in osteoblast senescence, and further validated the association of relevant molecules with osteoporosis at the animal level using immunohistochemistry and micro-CT. Our results showed that Prmt6 deficiency led to bone loss and induced osteoblast senescence in mice. M

UNLABELLED: Although fish possess an effective interferon (IFN) system to defend against viral infection, grass carp reovirus (GCRV) still causes epidemic hemorrhagic disease and tremendous economic loss in grass carp. Therefore, it is necessary to investigate the immune escape strategies employed by GCRV. In this study, we show that the structural protein VP4 of GCRV (encoded by the S6 segment) significantly restricts IFN expression by degrading stimulator of IFN genes (STING) through the autophagy-lysosome-dependent pathway. First, overexpression of VP4 inhibited the expression of IFN induce

Both regulatory B (Breg) and myeloid cells in tumors and lymph nodes drive immune suppression in pancreatic cancer. Current strategies to counter immune suppression emphasize myeloid cells but overlook Breg cells. We discovered that STING agonist expanded Breg cells depended on PI3Kγ but not PI3Kδ in pancreatic cancer, whereas activating myeloid cells were independent of PI3Kγ. Inhibition of PI3Kγ, but not PI3Kδ, decreased STING-induced IRF3 phosphorylation and Breg cell expansion in pancreatic cancer, while sustaining STING-induced IRF3 phosphorylation to activate myeloid cells. We developed

Activation of the stimulator of interferon genes (STING) pathway has emerged as a promising strategy for cancer immunotherapy. However, the initial cyclic dinucleotide (CDN) analogs developed as STING agonists have shown limited efficacy in clinical trials, prompting interest in non-CDN small-molecule alternatives. In this study, we identified a novel series of bromophenol derivatives as effective STING agonists. Among these derivatives, OSBP63 robustly activated the STING signaling pathway, resulting in enhanced phosphorylation of interferon regulatory factor 3 (p-IRF3) and increased secretio

The DNA sensor cyclic GMP-AMP synthase (cGAS) is important for antiviral and anti-tumor immunity. cGAS generates cyclic GMP-AMP (cGAMP), a diffusible cyclic dinucleotide that activates the antiviral response through the adaptor protein stimulator of interferon genes (STING). cGAMP cannot passively cross cell membranes, but recent advances have established a role for extracellular cGAMP as an "immunotransmitter" that can be imported into cells. However, the mechanism by which cGAMP exits cells remains unknown. Here, we identifed ABCC1 as a direct, ATP-dependent cGAMP exporter in mouse and human

The cGAS-STING [cyclic GMP-AMP (cGAMP) synthase (cGAS)-stimulator of interferon genes (STING)] pathway is an evolutionarily conserved pathway for sensing and responding to cytosolic double-stranded (ds)DNA, whether pathogen-associated or endogenous. Detection and binding of dsDNA by cGAS enzymatically generates the second messenger cGAMP, which in turn binds the endoplasmic reticulum-resident signalling protein STING, activating IRF3 (interferon regulatory factor 3), leading to expression of type I interferons. This pathway is necessary for both viral infection and antitumoral responses. Vario

Bladder cancer (BLCA) is an aggressive malignancy characterized by metastasis and drug resistance, creating an urgent demand for novel therapeutic targets. The lysosomal protein LAMP2 acts as a key oncogenic driver in BLCA and modulates the degradation of STING. As reported in prior studies, mechanistically, p62-mediated macroautophagy and HSC70-dependent chaperone-mediated autophagy (CMA) facilitate STING degradation. LAMP2 increases STING ubiquitination, leading to its lysosomal degradation in BLCA. Moreover, LAMP2 knockdown suppresses BLCA cell proliferation and migration. In our experiment

Stimulator of interferon response cGAMP interactor (STING), the central transducer of the cGAS-STING signaling axis, governs type I interferon (IFN-I) production that is essential for antiviral innate immunity. Modulating STING activity and stability offers potential therapeutic strategies for viral and autoimmune diseases. Here, we demonstrate that testis-expressed protein 264 (TEX264), an endoplasmic reticulum-selective autophagy (ER-phagy) receptor, shows upregulated expression following Herpes simplex virus 1 (HSV-1) infection. Overexpression of TEX264 inhibits the activation of IFN-I sign

The expression patterns of stimulator of interferon genes (STING) were investigated in a cohort of 158 T- and natural killer (NK)-cell and 265 B-cell non-Hodgkin lymphomas (NHLs), as well as in control reactive lymph nodes and tonsils. STING expression was assessed by immunohistochemical methods using diagnostic biopsy specimens obtained prior to treatment. Using an arbitrary 10% cutoff, STING was differentially expressed among T/NK-cell NHLs; positive in 36 out of 38 (95%) cases of ALK+ anaplastic large cell lymphoma (ALCL), 23 out of 37 (62%) ALK-ALCLs, 1 out of 13 (7.7%) angioimmunoblastic

Cytosolic recognition of DNA has emerged as a critical cellular mechanism of host immune activation upon pathogen invasion. The central cytosolic DNA sensor cGAS activates STING, which is phosphorylated, dimerizes and translocates from the endoplasmic reticulum (ER) to a perinuclear region to mediate IRF-3 activation. Poxviruses are double-stranded DNA viruses replicating in the cytosol and hence likely to trigger cytosolic DNA sensing. Here, we investigated the activation of innate immune signaling by 4 different strains of the prototypic poxvirus vaccinia virus (VACV) in a cell line proficie

The stimulator of interferon genes (STING) orchestrates type I interferon (IFN) production in response to cytosolic DNA and plays essential roles in antiviral defense and autoimmune pathogenesis. The stability of STING determines the activation intensity of the pathway. Therefore, identifying proteins that govern its homeostatic regulation is needed. Here, we uncover the lysosomal protease cathepsin L (CTSL) as a critical stabilizer of STING in cells. CTSL deficiency selectively impairs STING-induced IFN responses without compromising overall lysosomal digestive function. Mechanistically, CTSL

BACKGROUND: Severe refractory, neutrophilic asthma remains an unsolved clinical problem. STING agonists induce a neutrophilic response in the airways, suggesting that STING activation may contribute to the triggering of neutrophilic exacerbations. We aim to determine whether STING-induced neutrophilic lung inflammation mimics severe asthma.
METHODS: We developed new models of neutrophilic lung inflammation induced by house dust mite (HDM) plus STING agonists diamidobenzimidazole (diABZI) or cGAMP in wild-type, and conditional-STING-deficient mice. We measured DNA damage, cell death, NETs, cGA

BACKGROUND: Macrophage proinflammatory activation contributes to the pathology of severe acute pancreatitis (SAP) and, simultaneously, macrophage functional changes, and increased pyroptosis/necrosis can further exacerbate the cellular immune suppression during the process of SAP, where cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) plays an important role. However, the function and mechanism of cGAS-STING in SAP-induced lung injury (LI) remains unknown.
METHODS: Lipopolysaccharide (LPS) was combined with caerulein-induced SAP in wild type, cGAS -/- and sting -/- mice.

By detecting and responding to cytosolic DNA, the cGAS STING pathway regulates the innate immune responses bymediatinginflammatory reactions and antiviral defense. Thederegulation and modification of this system have been linked to variousneurodegenerative diseases like AD, PD and ALS. Accumulation of tau protein and Aβ aggregates to activate the pathway and releases neuroinflammatory cytokines which accelerates neuronal dysfunction and cognitive impairment as the symptom of AD. Similarly, in PD Alpha-synuclein aggregates activate the cGAS STING pathway and regulate the neuroinflammation and o

An immunosuppressive tumour microenvironment is a major obstacle in the control of pancreatic and other solid cancers1-3. Agonists of the stimulator of interferon genes (STING) protein trigger inflammatory innate immune responses to potentially overcome tumour immunosuppression4. Although these agonists hold promise as potential cancer therapies5, tumour resistance to STING monotherapy has emerged in clinical trials and the mechanism(s) is unclear5-7. Here we show that the administration of five distinct STING agonists, including cGAMP, results in an expansion of human and mouse interleukin (I

Aberrant activation of innate immune pathways is associated with a variety of diseases. Progress in understanding the molecular mechanisms of innate immune pathways has led to the promise of targeted therapeutic approaches, but the development of drugs that act specifically on molecules of interest remains challenging. Here we report the discovery and characterization of highly potent and selective small-molecule antagonists of the stimulator of interferon genes (STING) protein, which is a central signalling component of the intracellular DNA sensing pathway1,2. Mechanistically, the identified

Gibel carp (Carassius gibelio) is an economically important aquaculture species, and the infection with cyprinid herpesvirus 2 (CyHV-2) can cause severe disease outbreaks and significant economic losses; however, the antiviral innate immune mechanisms in this species remain poorly understood. The stimulator of interferon (IFN) genes (STING) is a key adaptor in antiviral innate immunity and plays a central role in the induction of IFN responses. In this study, we identified and characterized two sting homeologs, sting-A and sting-B, in amphitriploid gibel carp. Although both STING proteins loca

Inflammation and pain are intertwined responses to injury, infection, or chronic diseases. While acute inflammation is essential in determining pain resolution and opioid analgesia, maladaptive processes occurring during resolution can lead to the transition to chronic pain. Here we found that inflammation activates the cytosolic DNA-sensing protein stimulator of IFN genes (STING) in dorsal root ganglion nociceptors. Neuronal activation of STING promotes signaling through TANK-binding kinase 1 (TBK1) and triggers an IFN-β response that mediates pain resolution. Notably, we found that mice expr
