Anticancer Activity of Triterpene Glycosides Cucumarioside A0-1 and Djakonovioside A Against MDA-MB-231 as A2B Adenosine Receptor Antagonists.

Breast cancer is the most prevalent cancer in women worldwide and presents a major therapeutic challenge, particularly triple-negative breast cancer (TNBC), a subtype characterized by an aggressive clinical course but heightened sensitivity to chemotherapy. Natural products, such as triterpene glycosides derived from sea cucumbers, have emerged as promising candidates with high anticancer potential against TNBC. This study investigated the pathways of anticancer action of cucumarioside A0-1 (Cuc A0-1) and djakonovioside A (Dj A), isolated from the sea cucumber Cucumaria djakonovi, triggered an

Chemical synthesis of marine saponins.

Covering: 1989-2017 Saponins are characteristic metabolites of starfish and sea cucumbers, and occasionally are also found in sponges, soft coral, and small fish. These steroid or triterpenoid glycosides often show remarkable biological and pharmacological activities, such as antifungal, antifouling, shark repellent, antitumor and anti-inflammatory activities. Over one thousand marine saponins have been characterized; the majority of them can be categorized into three major structural types, i.e., asterosaponins, polyhydroxysteroid glycosides, and holostane glycosides. Thus far, only 12 marine

Assessment of potentially toxic element contamination in commercially harvested invertebrates from the Beibu Gulf, China.

Marine pollutants, especially potentially toxic elements (PTEs), increasingly threaten the ecological environment and fishery resources of the Beibu Gulf due to their bioaccumulative nature, toxicity, and persistence. However, the occurrences of multiple PTEs in marine invertebrates within this region remains unclear. Hence, a total of 18 species of commercially harvested invertebrates (shrimp, crab, cephalopod, shellfish, and sea cucumber) were collected from the Beibu Gulf, and the concentrations of nine important PTEs (As, Cd, Cr, Cu, Hg, Mn, Ni, Pb, and Zn) were examined. Subsequent stable

Antidepressant effect of the peptide EIILEAGK from Apostichopus japonicus via inflammatory and mitochondrial regulations in a lipopolysaccharide mouse model.

An increasing number of studies support the hypothesis of inflammation-induced depression, and several anti-inflammatory drugs are known to target tumour necrosis factor ɑ (TNF-ɑ) and interleukin 6 (IL-6) to show antidepressant potential. Here, a novel peptide derived from the sea cucumber Apostichopus japonicus was found to have an anti-inflammatory effect, as demonstrated by inhibiting the activation of NF-κB and reducing the levels of pro-inflammatory cytokines (TNF-α, IL-6, and IL-1β) in the brain tissue and blood of lipopolysaccharide (LPS)-treated mice. Interestingly, LPS-induced depress

Chemical Synthesis of Fucosylated Chondroitin Sulfate Tetrasaccharide with Fucosyl Branch at the 6-OH of GalNAc Residue.

Fucosylated chondroitin sulfate is a unique glycosaminoglycan isolated from sea cucumbers, with excellent anticoagulant activity. The fucosyl branch in FCS is generally located at the 3-OH of D-glucuronic acid but, recently, a novel structure with α-L-fucose linked to the 6-OH of N-acetyl-galactosamine has been found. Here, using functionalized monosaccharide building blocks, we prepared novel FCS tetrasaccharides with fucosyl branches both at the 6-OH of GalNAc and 3-OH of GlcA. In the synthesis, the protective group strategy of selective O-sulfation, as well as stereoselective glycosylation,

Surf Redfish-Based ZnO-NPs and Their Biological Activity with Reference to Their Non-Target Toxicity.

The marine environment is a rich source of bioactive compounds. Therefore, the sea cucumber was isolated from the Red Sea at the Al-Ain Al-Sokhna coast and it was identified as surf redfish (Actinopyga mauritiana). The aqueous extract of the surf redfish was utilized as an ecofriendly, novel and sustainable approach to fabricate zinc oxide nanoparticles (ZnO-NPs). The biosynthesized ZnO-NPs were physico-chemically characterized and evaluated for their possible antibacterial and insecticidal activities. Additionally, their safety in the non-target organism model (Nile tilapia fish) was also inv

Antiviral activity of marine sulfated glycans against pathogenic human coronaviruses.

Great interest exists towards the discovery and development of broad-spectrum antivirals. This occurs due to the frequent emergence of new viruses which can also eventually lead to pandemics. A reasonable and efficient strategy to develop new broad-spectrum antivirals relies on targeting a common molecular player of various viruses. Heparan sulfate is a sulfated glycosaminoglycan present on the surface of cells which plays a key role as co-receptor in many virus infections. In previous work, marine sulfated glycans (MSGs) were identified as having antiviral activities. Their mechanism of actio

Diversity, mechanism and structure-activity relationships of marine anticoagulant-active polysaccharides: A review.

Thrombosis is a major complication of cardiovascular disease that can lead to fatal myocardial infarction, acute ischemic stroke and venous thromboembolism, posing a significant threat to human health and even life. Recent research showed that polysaccharides from marine organisms, including marine plants and marine animals, exhibit excellent anticoagulant activity. However, different marine anticoagulant-active polysaccharides (MAPs) exhibit significant differences in both the structure and anticoagulant activity. This review systematically summarizes the diversity and structure of MAPs from

Antifungal and Antibacterial Activities of Isolated Marine Compounds.

To combat the ineffectiveness of currently available pharmaceutical medications, caused by the emergence of increasingly resistant bacterial and fungal strains, novel antibacterial and antifungal medications are urgently needed. Novel natural compounds with antimicrobial activities can be obtained by exploring underexplored habitats such as the world's oceans. The oceans represent the largest ecosystem on earth, with a high diversity of organisms. Oceans have received some attention in the past few years, and promising compounds with antimicrobial activities were isolated from marine organisms

Oligosaccharide Blocks PAR1 (Proteinase-Activated Receptor 1)-PAR4-Mediated Platelet Activation by Binding to Thrombin Exosite II and Impairs Thrombosis.

BACKGROUND: Inappropriate activation and aggregation of platelets can lead to arterial thrombosis. Thrombin is the most potent platelet agonist that activates human platelets via two PARs (proteinase-activated receptors), PAR1 and PAR4. The aim is to study the activity and mechanism of an oligosaccharide HS-11 (the undecasaccharide, derived from sea cucumber Holothuria fuscopunctata) in inhibiting thrombin-mediated platelet activation and aggregation and to evaluate its antithrombotic activity. METHODS: Platelet activation was analyzed by detecting CD62P/P-selectin expression using flow cytom

Hepatocytes and mitochondria dual-targeted astaxanthin WPI-SCP nanoparticles for the alleviation of alcoholic liver injury.

Alcoholic liver injury is one of the most frequent liver diseases around the world, and nutritional intervention has been considered as an effective way to alleviate alcohol liver injury. To alleviate the liver damage caused by alcohol, a type of astaxanthin (AXT) loaded nanoparticles were designed for dual targeting of hepatocytes and mitochondria. Firstly, galactooligosaccharides (GOS) were conjugated to whey protein isolate (WPI) and sea cucumber peptide (SCP) via the Maillard reaction, achieving a grafting degree of 29 %, then triphenylphosphonium (TPP) was linked by amide reaction. Second

Fucosylated chondroitin sulfate oligosaccharides from Isostichopus badionotus regulates lipid disorder in C57BL/6 mice fed a high-fat diet.

Fucosylated chondroitin sulfate (fCS) and its depolymerized derivative (DfCS), prepared from sea cucumbers, are well-known for their anticoagulant activity. However, their other functional activities are poorly understood. Recently, we obtained fCS oligosaccharides from Isostichopus Badionotus by a modified controllable Fenton-system, named as DfCS-Ib. The functional activities of these oligosaccharides are still unclear. The present study investigated anti-hyperlipidemic activity of DfCS-Ib using a high-fat diet (HFD)-fed mice model. The results indicated that DfCS-Ib reduced obesity, hyperli

Sulfated polysaccharide from Apostichopus japonicus viscera exhibits anti-inflammatory properties in vitro and in vivo.

Polysaccharides from sea cucumbers are known for their biological activities, but little is known about those from sea cucumber viscera. The present study isolated a sulfated polysaccharide (SCVP-2) from the viscera of Apostichopus japonicas, which had a molecular weight of 209.1 kDa. SCVP-2 comprised 66.3 % total sugars, 2.1 % uronic acid, 4.5 % proteins, and 25.5 % sulfate groups, containing glucosamine, galactosamine, glucose, galactose, and fucose. FT-IR and NMR analyses identified SCVP-2 as a fucoidan sulfate with sulfation patterns of the fucose branches as Fuc2S, Fuc4S, and Fuc0S. SEM a

Isolation of low-molecular-weight heparin/heparan sulfate from marine sources.

The glycosaminoglycan (heparin and heparan sulfate) are polyanionic sulfated polysaccharides mostly recognized for its anticoagulant activity. In many countries, low-molecular-weight heparins have replaced the unfractionated heparin, owing to its high bioavailability, half-life, and less adverse effect. The low-molecular-weight heparins differ in mode of preparation (chemical or enzymatic synthesis and chromatography fractionations) and as a consequence in molecular weight distribution, chemical structure, and pharmacological activities. Bovine and porcine body parts are at present used for ma

A theoretical discourse on the pharmacology of toxic marine ingestions.

The known and theoretical pharmacology of poisons involved in toxic marine ingestions has assisted in the development of specific therapeutics for these afflictions. The clinical manifestations of several toxic marine ingestions have suggested toxins/cogeners that may be involved in the poisoning process, providing direction for the development of diagnostic laboratory tests, including those for cyclic ethers. Future investigations should involve utilization of acetaminophen and indomethacin for chronic ciguatera fish poisoning, and the evaluation of the role of polycyclic ethers in the temper
