[Periodic respiration during sleep at high altitude. Effects of a hypnotic benzodiazepine, loprazolam].

Sleep and respiration studies were carried out in 12 subjects (9 males, 3 females) at an altitude of 4,800 metres, during effect of a French expedition in the Himalayas. The effect of loprazolam, a hypnotic benzodiazepine, was investigated in a double-blind, 2 parallel group, 1 mg loprazolam versus placebo trial. Sleep was evaluated by means of electroencephalographic recordings and questionnaires. The effects of altitude in each subject were intercurrent wakefulness increase, slow wave sleep and paradoxical sleep decrease and nocturnal periodic breathing. The mean duration of sleep apnea epis

The timing of sleep spindles is modulated by the respiratory cycle in humans.

OBJECTIVE: Coupling of sleep spindles with cortical slow waves and hippocampus sharp-waves ripples is crucial for sleep-related memory consolidation. Recent literature evidenced that nasal respiration modulates neural activity in large-scale brain networks. In rodents, this respiratory drive strongly varies according to vigilance states. Whether sleep oscillations are also respiration-modulated in humans remains open. In this work, we investigated the influence of breathing on sleep spindles during non-rapid-eye-movement sleep in humans. METHODS: Full night polysomnography of twenty healthy p

Sleep fragmentation, impaired glymphatic clearance, and long-term cognitive impairment after critical illness.

Sleep disruption is nearly universal during critical illness and is increasingly linked to long-term cognitive impairment among intensive care unit survivors. Advances in neuroscience have highlighted the glymphatic system - a brain-wide perivascular network that clears metabolic waste such as β-amyloid and phosphorylated tau - as a plausible mechanism connecting sleep disturbance to adverse neurocognitive outcomes. Glymphatic transport is highly state-dependent, functioning optimally during slow-wave sleep and diminishing with wakefulness or fragmented sleep. This invited review synthesizes p

Is there a relationship between delta sleep at night and afternoon cerebral blood flow, assessed by HMPAO-SPECT in depressed patients and normal control subjects? Preliminary data.

We wished to explore the relationships between waking HMPAO uptake and visually scored polysomnography. We hypothesized that HMPAO activity would correlate positively with slow wave sleep measures the same night. Eight unmedicated unipolar patients with current DSM-IV major depression (17-item Hamilton Depression Rating Scale score 21.5+/-2.9) and seven control subjects received polysomnography on 2 consecutive nights. On the afternoon following the adaptation night, subjects received cerebral SPECT, with 15 mCi Tc-99m-HMPAO injected while subjects performed the Continuous Performance Task. Pa

[Effects of neurotropin and other drugs on EEG alterations in SART-stressed (repeated cold-stressed) rats].

SART-stressed (repeated cold-stressed) rats, experimental model animals for vagotonic-type dysautonomia, have been reported to show EEG with lower-amplitude fast waves during resting-arousal and higher-amplitude slow waves during slow-wave sleep compared to normal rats. In this report, the effects of certain drugs on EEG alterations were investigated using the power spectral analysis. EEG was measured 60 min after a single dose of drugs and on the day following the final dose of 6 administrations. Neurotropin, a sedative analgesic, slightly increased faster waves on resting-arousal EEG and slo

Neurostimulation techniques to enhance sleep and improve cognition in aging.

Sleep plays a critical role in the process of memory consolidation. In particular, during non-rapid eye movement (NREM) slow wave sleep, slow-oscillations, spindles, hippocampal sharp wave ripples, and their phase coupling are involved in the process of transferring and consolidating information recently encoded and temporarily stored in the hippocampus into long-term memory stored in the neocortex. There is evidence that aging and neurodegenerative conditions, in particular Alzheimer's disease, are associated with changes to this transient grouping of NREM oscillations. Therefore, methods to

Sleep stage dynamics in young patients with sleep bruxism.

STUDY OBJECTIVES: We hypothesized that sleep stage dynamics are different in patients with sleep bruxism (SB) and that these changes are associated with the occurrence of rhythmic masticatory muscle activity (RMMA). METHODS: Fifteen healthy controls and 15 patients with SB underwent overnight polysomnography. Sleep variables and survival curves of continuous runs of each sleep stage were compared between the groups. Stage transition dynamics and the probability of stage fragmentation were analyzed for three epochs before and after the epoch with RMMA. Survival curves of continuous runs of eac

Clinical, EEG, electromyographic and polysomnographic studies in restless legs syndrome caused by magnesium deficiency.

The present paper reports biochemical and neurophysiological investigations in ten cases with restless leg syndrome. Other neuropsychiatric affections and factors which could generate the symptomatology of restless legs syndrome were not included. The EEG recordings demonstrated evident reticular neuronal hypersynchrony generated by hyperpnoea (sinusoidal slow waves). The classical EEG investigations pointed out neuromuscular hyperexcitability, but some modifications of the functional parameters of the neuromuscular excitability and conductibility (signs of neuropathy) were also noticed. Sleep promotes illusory word compositions, a distinct form of false memory.

Extensive evidence supports the beneficial effects of sleep on memory and learning, including the consolidation and reorganization of memories and the extraction of regularities from encoded experiences. Nevertheless, some studies suggest that sleep may also increase false memories, potentially as a byproduct of regularities extraction. Physiologically, time-compressed memory replay in the hippocampus during non-rapid-eye-movement (nREM) sleep is believed to contribute to the consolidation process, although the functional significance of time compression remains elusive. Recently, we proposed

Sleep architecture, cocaine and visual learning.

BACKGROUND: Disturbances in sleep associated with chronic cocaine use may underlie abstinence-related cognitive dysfunction. We hypothesized that sleep-related cognitive function would be impaired in chronic cocaine users, and that this impairment would be associated with abstinence-related changes in sleep architecture. METHODS: Twelve chronic cocaine users completed a 23-day in-patient study that included randomized, placebo-controlled, cocaine self-administration sessions. We report polysomnographic measurement of rapid eye-movement (REM) sleep and slow-wave activity, and performance on a

Neonatal cortical activity organizes into transient network states that are affected by vigilance states and brain injury.

Early neurodevelopment is critically dependent on the structure and dynamics of spontaneous neuronal activity; however, the natural organization of newborn cortical networks is poorly understood. Recent adult studies suggest that spontaneous cortical activity exhibits discrete network states with physiological correlates. Here, we studied newborn cortical activity during sleep using hidden Markov modeling to determine the presence of such discrete neonatal cortical states (NCS) in 107 newborn infants, with 47 of them presenting with a perinatal brain injury. Our results show that neonatal cort

Spatial memory performance is associated with region-specific coordination of hippocampo-cortical sleep oscillations.

Sleep plays a critical role in memory consolidation, yet how coordination among sleep oscillations across hippocampo-cortical circuits relates to behavioral performance remains incompletely understood. Here, we examined how cross-regional coordination of cardinal sleep oscillations during non-rapid eye movement (nREM) sleep relates to spatial memory performance. Adult rats were trained in an object-place recognition task and allowed to sleep while local field potentials were recorded simultaneously from dorsal and ventral hippocampus (CA1d and CA1v) and retrosplenial, prefrontal, and lateral e

Slow wave oscillations in Schizophrenia First-Degree Relatives: A confirmatory analysis and feasibility study on slow wave traveling.

Abnormal sleep oscillations have recently been proposed as endophenotypes of schizophrenia. However, optimization of methodological approaches is still necessary to standardize analyses of their microstructural characteristics. Additionally, some relevant features of these oscillations remain unexplored in pathological conditions. Among others, slow wave traveling is a promising proxy for diurnal processes of brain connectivity and excitability. The study of slow oscillations propagation appears particularly relevant when schizophrenia is conceptualized as a dys-connectivity syndrome. Given th

Sleep oscillations in the thalamocortical system induce long-term neuronal plasticity.

Long-term plasticity contributes to memory formation and sleep plays a critical role in memory consolidation. However, it is unclear whether sleep slow oscillation by itself induces long-term plasticity that contributes to memory retention. Using in vivo prethalamic electrical stimulation at 1 Hz, which itself does not induce immediate potentiation of evoked responses, we investigated how the cortical evoked response was modulated by different states of vigilance. We found that somatosensory evoked potentials during wake were enhanced after a slow-wave sleep episode (with or without stimulatio

Sleep disorders in Tourette's syndrome.

Sleep disorders have been reported in approximately 80% of Tourette's syndrome (TS) patients. Sleep studies in TS patients have demonstrated a 30% reduction in delta-sleep (slow-wave sleep) in nontreated subjects, decreased percentage of REM-sleep and the presence of tics during sleep. A subgroup of young TS patients was reported to have an increased percentage of delta sleep. Although these findings were initially thought to result from deranged dopaminergic and serotoninergic functions in TS, we suggest that abnormalities of hypothalamic-mediated control mechanisms involving the intrinsic op

Disturbances of the sleep-waking cycle in patients with vascular brain stem lesions.

Continuous polygraphic recordings of six patients with vascular brain stem lesions were evaluated to investigate the relationship between 1) the localization or extent of the lesions and the alteration of the sleep-waking cycle, and 2) the evolution of the clinical condition and the sleep-waking cycle. Absent REM sleep and severely disorganized NREM sleep were associated with extensive pontine lesions involving the pontine tegmentum. In "locked-in" syndrome, the neural mechanisms responsible for both REM and NREM sleep continued to function, but the sleep-waking cycle was disturbed and marked
