Sporothrix species are thermally dimorphic fungi responsible for sporotrichosis, a globally prevalent subcutaneous mycosis and an emerging zoonotic threat, particularly in South America. The high virulence of Sporothrix brasiliensis and its efficient transmission from cats to humans have intensified recent outbreaks, underscoring the importance of understanding the pathogenic mechanisms. While several putative virulence factors have been identified, such as melanin production, cell wall remodeling, extracellular vesicles, and thermotolerance, functional studies remain hampered by limited molec

BACKGROUND: Sporotrichosis is a fungal infection caused by species of the Sporothrix schenckii complex. Antifungal treatment with itraconazole and amphotericin B is limited by increasing resistance, adverse effects, and prolonged treatment courses, highlighting the need for novel antifungal strategies.
METHODS: The antifungal activity of seven antimicrobial peptides derived from arthropod toxins against Sporothrix species was evaluated: six synthetic peptides derived from the spider Lycosa erythrognatha toxin and one peptide isolated from apitoxin (melittin). Minimum inhibitory concentrations

Sporothrix brasiliensis is the predominant etiological agent of feline sporotrichosis in Brazil, where zoonotic dissemination represents an escalating public health concern. Reports of therapeutic refractoriness and the emergence of itraconazole-resistant isolates underscore the pressing need for alternative antifungal strategies. This study evaluated in vitro antifungal activity of a biogenic silver nanoparticle against 24 Brazilian clinical isolates of S. brasiliensis (18 cats and 6 dogs). Broth microdilution assays revealed minimum inhibitory concentrations ranging from 0.47 to 3.75 µg/mL w

Zoonotic sporotrichosis, a subcutaneous mycosis caused by Sporothrix brasiliensis, has become hyperendemic and a serious public health issue in Brazil and an emerging disease throughout the world. Typical sporotrichosis is defined as fixed or lymphocutaneous lesion development, however, reports of atypical presentations have been described in hyperendemic areas, which may result in a worse prognosis. Thus, considering an increase in atypical cases and in more severe extracutaneous cases and hospitalizations reported in Brazil, we aimed to perform a systematic review to search for hypersensitiv

A 26-year-old woman presented an eyelid lesion, after being scratched by a cat that had a similar skin lesion. It evolved into a cervical lymph node enlargement. With a hypothesis of Parinaud´s oculoglandular syndrome (POS) due to cat scratch disease (CSD), doxycycline was prescribed. After two weeks of treatment without improvement, a biopsy and blood sample were obtained. Itraconazole was prescribed and the skin lesion improved, but not the lymph node enlargement. A Sporothrix schenckii complex was isolated from the skin sample. Also, a specie-specific polymerase chain reaction detected Bart

Sporotrichosis is a fungal disease caused by the members of the Sporothrix pathogenic clade, and one of the etiological agents is Sporothrix schenckii. The cell wall of this organism has been previously analyzed and thus far is known to contain an inner layer composed of chitin and β -glucans, and an outer layer of glycoproteins, which are decorated with mannose and rhamnose-containing oligosaccharides. The L-rhamnose biosynthesis pathway is common in bacteria but rare in members of the Fungi kingdom. Therefore, in this study, we aimed to disrupt this metabolic route to assess the contribution

Sporotrichosis is a serious public health problem in Brazil that affects human patients and domestic animals, mainly cats. Thus, the search for new antifungal agents is required also due to the emergence and to the lack of effective drugs available in the therapeutic arsenal. The aim of this study was to evaluate the in vitro antifungal profile of two synthetic series of coumarin derivatives against Sporothrix schenckii and Sporothrix brasiliensis. The three-components synthetic routes used for the preparation of coumarin derivatives have proved to be quite efficient and compounds 16 and 17 ha

Sporotrichosis is a global subcutaneous fungal infection caused by the Sporothrix schenckii complex. Sporotrichosis is an uncommon infection in Iran, and there have been no phenotypic, molecular typing or antifungal susceptibility studies of Sporothrix species. This study aimed to identify nine Iranian isolates of the S. schenckii complex to the species level using colony morphology, carbohydrate assimilation tests, and PCR-sequencing of the calmodulin gene. The antifungal susceptibilities of these Sporothrix isolates to five antifungal agents (amphotericin B (AMB), voriconazole (VRC), itracon

Epidemiological data on the aetiologic agents of feline sporotrichosis in Malaysia have not been reported, though human sporotrichosis in Malaysia is reported to be transmitted primarily via cat scratch. To the best of our knowledge, the present report is the first study of the molecular epidemiology of Sporothrix schenckii isolates from cats with sporotrichosis in Malaysia. In the present work, we characterised 18 clinical isolates from cats in Malaysia based on molecular properties, including sequence analyses of the calmodulin gene and the rDNA ITS region and selective PCR of mating type (M

Sporothrix schenckii is a human pathogen that causes sporotrichosis, a cutaneous subacute or chronic mycosis. Little is known about the innate immune response and the receptors involved in host recognition and phagocytosis of S. schenckii. Here, we demonstrate that optimal phagocytosis of conidia and yeast is dependent on preimmune human serum opsonisation. THP-1 macrophages efficiently ingested opsonised conidia. Competition with D-mannose, methyl α-D-mannopyranoside, D-fucose, and N-acetyl glucosamine blocked this process, suggesting the involvement of the mannose receptor in binding and pha

Although the usual form of sporotrichosis is a lymphocutaneous lesion, Sporothrix schenckii can cause articular disease that is difficult to diagnose. We describe two patients with sporotrichosis who presented with tenosynovitis and the carpal tunnel syndrome. A tissue specimen is required for a precise diagnosis; granulomatous tenosynovitis suggests an infectious cause. Unless appropriate cultures for bacteria, mycobacteria and fungi are obtained, the diagnosis may be missed or delayed. Mouse inoculations may be required to isolate S. schenckii from tissue, which rarely shows the delicate fun

The frequency of opportunistic fungal infections has been increasing, impacting agriculture, food, and health sectors. In this work, four thiosemicarbazone-chalcones (TC) were synthesized and evaluated by the radial diffusion method against filamentous fungi. All TCs were effective against Aspergillus parasiticus, especially the fluor-substituted one, with radial growth inhibition of 62,9% and 74,4% at the lower (0.209 µmol/ml) and highest (1.670 µmol/ml) concentrations tested, respectively. On the other hand, the non-substituted derivative was the most active against A. carbonarius, inhibitin

Sporotrichosis by Sporothrix brasiliensis is increasingly reported in South America. Here, we present the first autochthonous human case in Paraguay, transmitted by a local infected cat. After 63 days of clinical signs onset, the patient was correctly diagnosed and antifungal treatment started, highlighting the need to increase awareness for this emerging disease.

Sporothrix schenckii pathogenesis has been extensively studied to date, but several proteins of unknown function remain to be assigned to the infectious process. Previously, proteomic data showed that one protein was highly abundant (SPSK_02764) in cell wall peptidorhamnomannan, but no obvious homologs or sequence features hinted at the activity or function of this protein. In this work, in vitro analysis showed that this protein is highly glycosylated and is an adhesin with specificity for type-I and type-II collagen, renaming this protein as Collagen Binding Protein 1 or Cbp1. Yeast-like cel

We aimed to develop and validate a Loop-mediated Isothermal Amplification (LAMP) assay to Sporothrix brasiliensis. LAMP reaction was developed using six primers designed based on calmodulin gene. In the LAMP reaction, we tested twenty isolates of S. brasiliensis from animals and humans, along with ten tissue samples extracted from the left footpad of mice that had been experimentally infected with S. brasiliensis. In addition, it included DNA samples from various other fungal species for specificity evaluation. All S. brasiliensis isolates yielded positive results in the LAMP, and the limit of

BACKGROUND: Sporotrichosis, a cosmopolitan mycosis caused by dimorphic fungi of the Sporothrix complex, affects humans and animals. This study aimed to develop new molecular markers for Sporothrix genome detection in biological samples using PCR.
METHODS: A specific region of DNA sequences from the Sporothrix genus, publicly available in GenBank, was chosen for primer design. After testing the in silico specificity of these primers, in vitro specificity was evaluated using the PCR technique.
RESULTS: Three specific primers with 100% specificity for the Sporothrix genus were generated.

INTRODUCTION: Sporotrichosis is a subcutaneous and chronic infection caused by traumatic inoculation of pathogenic sporothrix species, usually infecting the skins and subcutaneous tissues of humans and animals. However, the lack of epidemiological data required further molecular identification to describe the distribution of this fungus in our region. In this study, forty-eight clinical sporothrix isolated from Sun Yat-Sen Memorial Hospital were classified, and the susceptibility of each strain to seven antifungal agents was determined.
METHODS: Forty strains of S. globosa and eight strains o
