Antibiotic resistance in exopolysaccharide-forming Staphylococcus epidermidis clinical isolates from orthopaedic implant infections.

The opportunistic pathogen Staphylococcus epidermidis is able to produce biofilm and to frequently cause implant infections. In recent years, it has also exhibited an increasing antimicrobial drug resistance. Here, the resistance to a panel of 16 different antibiotics in 342 clinical strains of S. epidermidis from orthopaedic implant infections has been investigated. The isolates were pheno- and genotyped for extracellular polysaccharide production, relevant to staphylococcal biofilm formation, in order to ascertain possible associations with antibiotic resistance. Approximately 10% of the iso

Detecting methicillin-resistant Staphylococcus epidermidis--disc diffusion, broth breakpoint or polymerase chain reaction?

Growth conditions are important for the expression of resistance to methicillin among staphylococci. Consequently a phenotypic susceptibility test has to be chosen carefully to avoid false susceptible results. In this study we wanted to devise rapid and simple phenotypic tests whose results completely correlate with the presence of the methicillin resistance gene, mecA. A simplified polymerase chain reaction (PCR) method not needing separate DNA extraction from the tested bacteria was used to amplify a 449 bp region of the mecA gene. One hundred and ten strains of S. epidermidis were tested. T

Glycan-Dependent Corneocyte Adherence of Staphylococcus epidermidis Mediated by the Lectin Subdomain of Aap.

Staphylococcus epidermidis and other coagulase-negative staphylococci (CoNS) that colonize skin are known to promote skin immunity and inhibit colonization of pathogens that cause skin and soft tissue infections, including Staphylococcus aureus. However, S. epidermidis adherence to corneocytes, the cells that constitute the uppermost layer of the skin epidermis, remains poorly understood. Our study documents that S. epidermidis corneocyte adherence is dependent upon the accumulation-associated protein (Aap). Aap is composed of two distinct A and B domains. The A domain is comprised of a repeat

Staphylococcus epidermidis meningitis and an intraspinal abscess associated with a midthoracic dermal sinus tract.

Congenital neuroectodermal defects are associated with meningitis that is unresponsive to conventional antibiotic therapy, recurrent bacterial meningitis, or meningitis due to an organism that is not usually the cause of this disease. Midthoracic dermoid and epidermoid cysts occur rarely and are easily overlooked. We report the case of a 13-month-old boy with meningitis that was unresponsive to antimicrobial therapy. We subsequently identified an intraspinal abscess, an infection due to Staphylococcus epidermidis, and a midthoracic dermoid cyst. We emphasize the need for clinicians to have a h

The use of nonsteroidal antiinflammatory drugs to prevent adherence of Staphylococcus epidermidis to medical polymers.

The effect of salicylates and other nonsteroidal antiinflammatory drugs on the production of Staphylococcus epidermidis extracellular slime was studied. A dose-related decrease in slime production was observed with increasing concentrations of salicylic acid. S. epidermidis grown in 5 mM salicylic acid were less likely to adhere to Silastic, polyvinyl chloride, polyurethane, and Teflon catheters (P less than .006); strains grown in 2 mM salicylic acid, ibuprofen, indomethacin, or phenylbutazone were less adherent to Silastic catheters (P less than .001). Similar results were obtained with poly

The activity of rifampin and analogs against Staphylococcus epidermidis biofilms in a CAPD environment model.

Rifampin has been noted to exhibit exceptional antimicrobial activity against Staphylococcus epidermidis biofilms as compared to commonly used antibiotics. To further explore this unique effect of rifampin, we evaluated the antimicrobial activity of three commercially available preparations of rifampin, two rifampin analogs (CGP29861 and rifapentine) and the parent compound rifamycin SV. These were tested against standardized S. epidermidis biofilms in various milieus. All six members of the rifamycin group tested demonstrated marked antimicrobial activity but with minor foci of resisters when

Septic Arthritis of the Hip in a Premature Neonate Caused by Staphylococcus epidermidis: A Case Report.

Neonatal septic arthritis is a rare but potentially destructive condition requiring prompt diagnosis and intervention to prevent long-term sequelae. We reported a premature neonate who presented on day of life 20 with refusal to move the left hip and was diagnosed with septic arthritis caused by Staphylococcus epidermidis, an uncommon pathogen in neonates without indwelling devices. Diagnostic imaging confirmed left hip septic arthritis, and surgical irrigation and drainage were performed. Operative cultures grew S. epidermidis, and the patient completed targeted intravenous antimicrobial ther

Emergence of linezolid-resistant Staphylococcus epidermidis in the tertiary children's hospital in Cracow, Poland.

Coagulase-negative staphylococci, ubiquitous commensals of human skin, and mucous membranes represent important pathogens for immunocompromised patients and neonates. The increasing antibiotic resistance among Staphylococcus epidermidis is an emerging problem worldwide. In particular, the linezolid-resistant S. epidermidis (LRSE) strains are observed in Europe since 2014. The aim of our study was to genetically characterize 11 LRSE isolates, recovered mostly from blood in the University Children's Hospital in Krakow, Poland, between 2015 and 2017. For identification of the isolates at the spec

Relationship between quinolone use and resistance of Staphylococcus epidermidis in patients with acne vulgaris.

Staphylococcus epidermidis is a bacterium known to inhabit the skin. In treatment of acne vulgaris, the cutaneous milieu is exposed to oral or topical antimicrobials. We previously reported that the antimicrobial resistance of Cutibacterium acnes isolated from acne patients is affected by antimicrobial use. The aim of this study was to investigate the relationship between quinolone use and resistance in skin bacteria, particularly S. epidermidis, from acne patients. A total of 92 and 87 S. epidermidis strains isolated from clinic patients and hospital outpatients with acne vulgaris, respective

Slime layer formation and the prevalence of mecA and aap genes in Staphylococcus epidermidis isolates.

INTRODUCTION: Staphylococcus epidermidis strains are frequently associated with catheter-related infection, acute bacteremia, and hospital-acquired infection. Some isolates produce an extracellular matrix called slime that may make them more resistant to antibiotics. The aim of this study was to determine antimicrobial resistance patterns, the prevalence of slime production, and the distribution of genes (mecA and aap, respectively) associated with these phenotypes in S. epidermidis nasal isolates from health-care personnel. METHODOLOGY: A descriptive cross-sectional study was performed on 16

[Case of Staphylococcus epidermidis endocarditis with a bacterial vegetation suspected by intraoperative alternating pulse].

We report here a case of Staphylococcus epidermidis endocarditis with bacterial vegetation which was suspected by intraoperative alternating pulse on arterial pressure monitoring. A 65-year-old man was diagnosed as lumbar spinal canal stenosis and scheduled for operation. Preoperatively, he had chronic renal failure, mild aortic valve stenosis, and moderate mitral valve regurgitation. Neither inflammatory signs nor vegetations on TEE were observed preoperatively. General anesthesia was induced with propofol and vecuronium, and maintained with oxygen (33%), nitrous oxide and sevoflurane. Two ho

Effects on antibiotic resistance of Staphylococcus epidermidis following adhesion to polymethylmethacrylate and to silicone surfaces.

A number of studies appears to give emphasis to the role of prosthetic materials in determining microbial adherence and resistance to host defence and drug therapy. Aim of this study was to explore whether the direct contact with biomaterial substrata of different chemical nature could influence bacterial behaviour, determining possible changes in the bacteria population as far as antibiotic resistance is concerned. To this end, susceptibility to penicillin, erythromycin, clindamycin, cefamandole, imipenem, vancomycin, ciprofloxacin. ampicillin, cefazolin, trimethoprim-sulfamethoxazole, chlora

Staphylococcosis of turkeys. 6. Development of penicillin resistance in an interfering strain of Staphylococcus epidermidis.

Staphylococcus epidermidis strain 115, used as an interfering agent to help reduce the incidence of staphylococcosis in turkeys, was converted into a penicillin- and chloramphenicol-resistant strain designated 115R. This was accomplished by introducing a plasmid carrying the beta-lactamase (penicillinase) and chloramphenicol-resistance genes into S. epidermidis 115 by electroporation. The resultant strain, 115R, was an efficient producer of beta-lactamase and had marked increased resistance to penicillin and chloramphenicol. A beta-lactamase DNA probe was used to confirm the presence of the be

Evaluation of Omadacycline Alone and in Combination with Rifampin against Staphylococcus aureus and Staphylococcus epidermidis in an In Vitro Pharmacokinetic/Pharmacodynamic Biofilm Model.

Biofilm-associated infections lead to substantial morbidity. Omadacycline (OMC) is a novel aminomethylcycline with potent in vitro activity against Staphylococcus aureus and Staphylococcus epidermidis, but data surrounding its use in biofilm-associated infections are lacking. We investigated the activity of OMC alone and in combination with rifampin (RIF) against 20 clinical strains of staphylococci in multiple in vitro biofilm analyses, including an in vitro pharmacokinetic/pharmacodynamic (PK/PD) CDC biofilm reactor (CBR) model (simulating human exposures). The observed MICs for OMC demonstr

Molecular insights into efflux pump genes in ciprofloxacin- and methicillin-resistant Staphylococcus aureus and Staphylococcus epidermidis clinical isolates from Iraq.

Efflux pumps play a critical role in developing antimicrobial resistance (AMR) in S.aureus and S.epidermidis. Therefore, this study aimed to investigate the frequency and distribution of efflux pump resistance genes (norA, mepA, and mdeA), and potentially assess their association with antibiotic susceptibility profiles in ciprofloxacin- and methicillin-resistant S. aureus (CipR-MRSA) and S. epidermidis (CipR-MRSE) using both phenotypic methods and molecular detection. Among 250 clinical samples collected randomly from hospitalized Iraqi patients, 56% were Gram-positive, with 42.1% identified a
