Assessing the accumulation of microplastics in earthworms (Eisenia fetida) using traditional bioaccumulation modeling and synchrotron-based microcomputed tomography.

Microplastics (MPs) are recognized as a ubiquitous contaminant in terrestrial ecosystems. Earthworms, which play key roles in soil structure, nutrient cycling, and ecosystem function, are important bioindicators for assessing MP exposure and fate. However, uncertainties remain regarding MP toxicokinetics in earthworms, including ingestion dynamics, retention, and the potential for translocation across the gut epithelium. This study investigated the movement and fate of MPs in Eisenia fetida by quantifying uptake and elimination across five common MP morphologies (films, fibers, foams, fragment

Monitoring pH-induced lipid dynamics in mRNA-lipid nanoparticles by synchrotron small-angle X-ray scattering.

Lipid nanoparticles (LNPs), a promising drug delivery system, face key limitations in terms of a poor understanding toward their assembling mechanism and intracellular dynamics. Conventional characterization methods like dynamic light scattering and spectroscopy fail to capture the dynamic mesoscopic structural changes of LNPs under pH gradient. To address this issue, here we employed synchrotron small-angle X-ray scattering (SR-SAXS) to ex-situ monitor the dynamic self-assembly process of mRNA-encapsulated LNPs at different physiologically-relevant pH. By performing time-resolved SAXS samplin

Synchrotron tomography-based inversion pipeline for estimating elastic properties for rat vertebral endplate finite element models.

Lower back pain is linked to vertebral biomechanics, with vertebral endplates (VEPs) playing a key role. Finite element modelling (FEM) is a powerful tool for studying VEP biomechanics but relies on accurate material property inputs, which remain difficult to obtain. Synchrotron computed tomography (sCT) enables detailed visualisation of intact VEP microstructure under near-physiological loads in situ, with three-dimensional strain fields obtained by digital volume correlation (DVC) providing experimental reference data for FEM validation. We applied inverse finite element methodologies to est

Ultra-high-resolution synchrotron phase-contrast CT enables microstructural pulmonary imaging at clinical dose levels.

BACKGROUND: Ultra-high-resolution propagation-based synchrotron phase-contrast CT is an emerging technique for lung imaging. However, its feasibility and diagnostic potential at radiation doses comparable to those used in standard clinical procedures has yet to be established. This study aims to evaluate the performance of phase-contrast CT in comparison with state-of-the-art high-resolution multislice CT and bronchoscopy, and to validate its diagnostic accuracy histologically using porcine and, for the first time, human lung specimens. METHODS: Phase-contrast CT experiments were conducted at

Fast Degradation of Solid Electrolyte in Initial Cycling Processes, Tracked in 3D by Synchrotron X-ray Computed Tomography.

Solid-state lithium batteries are developing rapidly as a promising next-generation battery, while challenges still persist in understanding their degradation processes during cycling due to the difficulties in characterization. In this study, the 3D morphological evolution of the Li3PS4 solid electrolyte was tracked during electrochemical cycles (plating and stripping) until short circuit by utilizing in situ synchrotron X-ray computed tomography with sufficient spatial and temporal resolution. During the degradation process, cracks in the electrolyte alternately generated from the two electr

Comparison of Thiel-fixed and fresh-frozen temporal bon biomechanics with dynamic synchrotron-based X-ray imaging.

BACKGROUND: The Thiel embalming method offers lifelike tissue preservation, yet its suitability for biomechanical studies remains debated. METHODS: Here, we investigated two human temporal bones, one Thiel-fixed and one fresh-frozen, using dynamic synchrotron-based X-ray imaging under acoustic stimulation. FINDINGS: Both specimens exhibited a linear response at the umbo, but the Thiel-fixed specimen displayed discontinuity after the incudo-malleolar joint, suggesting ossicular chain stiffening. Our findings align well with previous studies indicating potential tissue degradation induced by T

Deformation and Failure Mechanisms of Natural/Waste Rubber Blends Under Mechanical and Thermal Cycles: A Synchrotron In Situ Study.

This study investigates the deformation and failure mechanisms of natural rubber (NR) and waste rubber blends under mechanical and thermal cycles using in situ synchrotron wide-angle X-ray diffraction (WAXD). The incorporation of ground tire rubber (GTR) into NR enhances strain-induced crystallization (SIC), reducing the onset strain for crystallization and increasing mechanical reinforcement. Cyclic loadings reveal significant hysteresis and residual deformation, influenced by GTR content. Moreover, the strain at complete melting of SIC crystals is found to decrease with GTR content, suggesti

The human middle ear in motion: 3D visualization and quantification using dynamic synchrotron-based X-ray imaging.

The characterization of the vibrations of the middle ear ossicles during sound transmission is a focal point in clinical research. However, the small size of the structures, their micrometer-scale movement, and the deep-seated position of the middle ear within the temporal bone make these types of measurements extremely challenging. In this work, dynamic synchrotron-based X-ray phase-contrast microtomography is used on acoustically stimulated intact human ears, allowing for the three-dimensional visualization of entire human eardrums and ossicular chains in motion. A post-gating algorithm is u

Microarchitecture of the penis bone (baculum) of a seal: A 3D morphometric examination using synchrotron and laboratory micro-computed tomography.

We examined the ultrastructure of the mammalian os penis at the high-resolution synchrotron level. Previously, bacular microanatomy had only been investigated histologically. We studied the baculum of the harp seal (Pagophilus groenlandicus), in which the baculum varies more in size and shape than does a mechanically constrained bone (humerus). We (1) investigated the microarchitecture of bacula and humeri from the same seal specimens, and (2) described changes in bone micro- and macro-morphology associated with age (n = 15, age range = 1-35 years) and bone type. We analyzed cross-sectional ge

Morphological and lipid metabolism alterations in macrophages exposed to model environmental nanoplastics traced by high-resolution synchrotron techniques.

The release of nanoplastics (NPs) in the environment is a significant health concern for long-term exposed humans. Although their usage has certainly revolutionized several application fields, at nanometer size, NPs can easily interact at the cellular level, resulting in potential harmful effects. Micro/Nanoplastics (M/NPs) have a demonstrated impact on mammalian endocrine components, such as the thyroid, adrenal gland, testes, and ovaries, while more investigations on prenatal and postnatal exposure are urgently required. The number of literature studies on the NPs' presence in biological sam

A hybrid machine-learning approach for analysis of methane hydrate formation dynamics in porous media with synchrotron CT imaging.

Fast multi-phase processes in methane hydrate bearing samples pose a challenge for quantitative micro-computed tomography study and experiment steering due to complex tomographic data analysis involving time-consuming segmentation procedures. This is because of the sample's multi-scale structure, which changes over time, low contrast between solid and fluid materials, and the large amount of data acquired during dynamic processes. Here, a hybrid approach is proposed for the automatic segmentation of tomographic data from time-resolved imaging of methane gas-hydrate formation in sandy granular

Structural properties in ruptured mitral chordae tendineae measured by synchrotron-based X-ray phase computed tomography.

The link between the structural properties and the rupturing of chordae tendineae in the mitral valve complex is still unclear. Synchrotron-radiation-based X-ray phase computed tomography (SR-XPCT) imaging is an innovative way to quantitatively analyze three-dimensional morphology. XPCT has been employed in this study to evaluate the chordae tendineae from patients with mitral regurgitation and to analyze structural changes in the ruptured chordae tendineae in patients with this condition. Six ruptured mitral chordae tendineae were obtained during surgical repairs for mitral regurgitation and

Time-Lapse In Situ 3D Imaging Analysis of Human Enamel Demineralisation Using X-ray Synchrotron Tomography.

Caries is a chronic disease that causes the alteration of the structure of dental tissues by acid dissolution (in enamel, dentine and cementum) and proteolytic degradation (dentine and cementum) and generates an important cost of care. There is a need to visualise and characterise the acid dissolution process on enamel due to its hierarchical structure leading to complex structural modifications. The process starts at the enamel surface and progresses into depth, which necessitates the study of the internal enamel structure. Artificial demineralisation is usually employed to simulate the proce

Successive phase transitions of the spin-orbit-coupled metal Cd2Re2O7probed by high-resolution synchrotron x-ray diffraction.

The 5dpyrochlore oxide superconductor Cd2Re2O7(CRO) has attracted significant interest as a spin-orbit-coupled metal (SOCM) that spontaneously undergoes a phase transition to an odd-parity multipole phase by breaking the spatial inversion symmetry due to the Fermi liquid instability caused by strong spin-orbit coupling. Despite the significance of structural information during the transition, previous experimental results regarding lattice deformation have been elusive. We have conducted ultra-high resolution synchrotron radiation x-ray diffraction experiments on a high-quality CRO single crys

Pressure-induced phase transitions in MBANP crystal: A study by synchrotron radiation X-ray diffraction and Raman spectroscopy.

Raman spectroscopy and synchrotron radiation X-ray diffraction have been used to study the effect of pressure on 2-(-α-methylbenzylamino)-5-dinitropyridine (MBANP). Several changes are observed with increasing pressure in the Raman spectra of this system, such as splitting of various bands and disappearance of bands. Discontinuous shifts in wavenumber vs pressure plot indicate that a conformational phase transition takes place around 0.5 GPa. The behavior of the Raman spectra indicates that MBANP molecules present conformational phase transition at high-pressure. X-ray diffraction, performed w

A Synchrotron and Micro-CT Study of the Human Endolymphatic Duct System: Is Meniere's Disease Caused by an Acute Endolymph Backflow?

Background: The etiology of Meniere's disease (MD) and endolymphatic hydrops believed to underlie its symptoms remain unknown. One reason may be the exceptional complexity of the human inner ear, its vulnerability, and surrounding hard bone. The vestibular organ contains an endolymphatic duct system (EDS) bridging the different fluid reservoirs. It may be essential for monitoring hydraulic equilibrium, and a dysregulation may result in distension of the fluid spaces or endolymphatic hydrops. Material and Methods: We studied the EDS using high-resolution synchrotron phase contrast non-invasive

Elucidating the Drug Release from Metal-Organic Framework Nanocomposites via In Situ Synchrotron Microspectroscopy and Theoretical Modeling.

Nanocomposites comprising metal-organic frameworks (MOFs) embedded in a polymeric matrix are promising carriers for drug delivery applications. While understanding the chemical and physical transformations of MOFs during the release of confined drug molecules is challenging, this is central to devising better ways for controlled release of therapeutic agents. Herein, we demonstrate the efficacy of synchrotron microspectroscopy to track the in situ release of 5-fluorouracil (5-FU) anticancer drug molecules from a drug@MOF/polymer composite (5-FU@HKUST-1/polyurethane). A Micro-CT and Synchrotron Imaging Study of the Human Endolymphatic Duct with Special Reference to Endolymph Outflow and Meniere's Disease.

Meniere's disease remains enigmatic, and has no treatment with sufficient evidence. The characteristic histopathological finding is endolymphatic hydrops, suggesting either an overproduction or decreased reabsorption of endolymph in the human inner ear. This study presents the first analysis of the vascular plexus around the human endolymphatic duct using micro computed tomography and coherent synchrotron radiation with phase contrast imaging. Using a software program, data were processed by volume-rendering with scalar opacity mapping to create transparent three-dimensional reconstructions. A

Visualization and Pathological Characteristics of Hepatic Alveolar Echinococcosis with Synchrotron-based X-ray Phase Sensitive Micro-tomography.

Propagation-based phase-contrast computed tomography (PPCT) utilizes highly sensitive phase-contrast technology applied to X-ray micro-tomography, especially with the extensive use of synchrotron radiation (SR). Performing phase retrieval (PR) on the acquired angular projections can enhance image contrast and enable quantitative imaging. We employed the combination of SR-PPCT and PR for the histopathological evaluation of hepatic alveolar echinococcosis (HAE) disease and demonstrated the validity and superiority of PR-based SR-PPCT. Branching fractions of the CN + C3H6 reaction using synchrotron photoionization mass spectrometry: evidence for the 3-cyanopropene product.

The gas-phase CN + propene reaction is investigated using synchrotron photoionization mass spectrometry (SPIMS) over the 9.8-11.5 eV photon energy range. Experiments are conducted at room temperature in 4 Torr of He buffer gas. The CN + propene addition reaction produces two distinct product mass channels, C(3)H(3)N and C(4)H(5)N, corresponding to CH(3) and H elimination, respectively. The CH(3) and H elimination channels are measured to have branching fractions of 0.59 ± 0.15 and 0.41 ± 0.10, respectively. The absolute photoionization cross sections between 9.8 and 11.5 eV are measured for th
