Noise and age-related hearing loss: a study of 40 123 gold miners in South Africa.

OBJECTIVE AND DESIGN: This retrospective cohort study aimed to describe the differential effect of noise exposure and age-related hearing loss in a large sample of gold miners in South Africa. STUDY SAMPLE: Audiological data of 40 123 South African mine workers were investigated. Data of a non-noise-exposed control group (n = 6162) and group exposed to underground noise (≥ 85 dB A (TWA) (n = 33 961) were included. Within these two larger noise-exposed groups two homogenous exposure groups (HEG) were also selected for analyses, namely the driller group (n = 4399) and the administration group (

[Hearing disorders in aphasia].

Aphasia is an acquired communication disorder that often involves receptive language abilities. After clinical assessment it is often not clear if this is partially due to a hearing loss, which can be compensated by hearing aids facilitating the rehabilitative process. In the present study the hearing ability of 88 male and female patients with aphasia after stroke, all of whom suffered from a left-hemispheric ischemia was assessed in the rehabilitative setting. We found that a majority of patients (72, 82%) was able to perform pure tone audiometry. 15 aphasic patients (21%) showed a hearing los

Progress in cochlear physiology after Békésy.

In the fifty years since Békésy was awarded the Nobel Prize, cochlear physiology has blossomed. Many topics that are now current are things Békésy could not have imagined. In this review we start by describing progress in understanding the origin of cochlear gross potentials, particularly the cochlear microphonic, an area in which Békésy had extensive experience. We then review progress in areas of cochlear physiology that were mostly unknown to Békésy, including: (1) stereocilia mechano-electrical transduction, force production, and response amplification, (2) outer hair cell (OHC) somatic mo

Age-related primary cochlear neuronal degeneration in human temporal bones.

In cases of acquired sensorineural hearing loss, death of cochlear neurons is thought to arise largely as a result of sensory-cell loss. However, recent studies of acoustic overexposure report massive degeneration of the cochlear nerve despite complete hair cell survival (Kujawa and Liberman, J Neurosci 29:14077-14085, 2009). To assess the primary loss of spiral ganglion cells (SGCs) in human ears, neuronal counts were performed in 100 temporal bones from 100 individuals, aged newborn to 100 years, selected to include only cases with a normal population of inner and outer hair cells. Ganglion

Speech perception in noise using directional microphones in open-canal hearing aids.

BACKGROUND: Individuals with impaired hearing find it difficult to understand speech in the presence of background noise--a problem addressed effectively by directional microphones. As open-canal fittings have become increasingly popular in the recent past, so has the debate about the effective directional benefit available from these devices. PURPOSE: This study investigates the benefit of directional microphones in two commercially available open-canal behind-the-ear hearing aids using the Hearing in Noise Test (HINT). STUDY SAMPLE: Sixteen individuals, between 50 and 85 year of age, with

Effects of noise on inferior colliculus evoked potentials and cochlear anatomy in young and aged chinchillas.

Like many aging humans, the aging chinchilla tends to lose high-frequency sensitivity at a faster rate than low-frequency sensitivity. This feature, combined with its excellent low-frequency hearing, makes the chinchilla attractive as an animal model for studying the relationship between noise-induced hearing loss (NIHL) and age-related hearing loss (AHL). In the present study, we examined susceptibility to noise in 15 aged (10-15 years old) and 15 young chinchillas. Two levels of noise were used, with the aim of determining whether age-related differences exist in the magnitude and rate of re

Genetics of age-related hearing loss in mice. II. Strain differences and effects of caloric restriction on cochlear pathology and evoked response thresholds.

The effects of genotype and diet on age-related hearing loss were evaluated using auditory brainstem response (ABR) thresholds and post-mortem cochlear histopathology in 5 inbred mouse strains, CBA/H-T6J (CH), DBA/2J (D2), C57BL/6J (B6), BALB/cByJ (BY) and WB/ReJ (WB), and their 10 F1 hybrid strains. The mice had been maintained since weaning on either a high-energy (HE) control diet or low-energy (LE) calorically restricted diet. ABR thresholds were obtained when the mice were 23 months old; the mice were allowed to age until they died from natural causes prior to obtaining the histological m

Ageing and the auditory nerve: Hearing sensitivity and endbulb synapses.

Age-related hearing loss impairs speech understanding for socialization and music appreciation for enjoyment, both of which compromise quality of life and can lead to cognitive decline. It has previously been shown that while standard audiometric hearing thresholds can remain normal over time, speech understanding in noise is more difficult and there is the emergence of tinnitus. These specific hearing difficulties are not revealed by standard audiograms but we now know that they have been attributed to loss of high threshold auditory nerve fibers caused by the disappearance of terminal ending

Extension and evaluation of a near-end listening enhancement algorithm for listeners with normal and impaired hearing.

In many applications in which speech is played back via a sound reinforcement system such as public address systems and mobile phones, speech intelligibility is degraded by additive environmental noise. A possible solution to maintain high intelligibility in noise is to pre-process the speech signal based on the estimated noise power at the position of the listener. The previously proposed AdaptDRC algorithm [Schepker, Rennies, and Doclo (2015). J. Acoust. Soc. Am. 138, 2692-2706] applies both frequency shaping and dynamic range compression under an equal-power constraint, where the processing

Strain-specific differences in the development of neuronal excitability in the mouse ventral nucleus of the trapezoid body.

This investigation compared the development of neuronal excitability in the ventral nucleus of the trapezoid body (VNTB) between two strains of mice with differing progression rates for age-related hearing loss. In contrast to CBA/Ca (CBA) mice, the C57BL/6J (C57) strain are subject to hearing loss from a younger age and are more prone to damage from sound over-exposure. Higher firing rates in the medial olivocochlear system (MOC) are associated with protection from loud sounds and these cells are located in the VNTB. We postulated that reduced neuronal firing of the MOC in C57 mice could cont

Aging, hearing acuity, and the attentional costs of effortful listening.

A dual-task interference paradigm was used to investigate the effect of perceptual effort on recall of spoken word lists by young and older adults with good hearing and with mild-to-moderate hearing loss. In addition to poorer recall accuracy, listeners with hearing loss, especially older adults, showed larger secondary task costs while recalling the word lists even though the stimuli were presented at a sound intensity that allowed correct word identification. Findings support the hypothesis that extra effort at the sensory-perceptual level attendant to hearing loss has negative consequences

Auditory peripheral influences on calcium binding protein immunoreactivity in the cochlear nucleus during aging in the C57BL/6J mouse.

The C57BL/6J (C57) mouse was selected as a suitable model for early presbyacusis to determine if there were correlations between peripheral pathology (spiral ganglion loss, inner and outer hair cell loss) and calcium binding immunoreactivity in the cochlear nucleus during aging. The quantitative stereological method, the optical fractionator, was used for determining the total number of neurons and calcium binding immunopositive neurons (calbindin, parvalbumin and calretinin) during aging in the posteroventral- and dorsal cochlear nucleus (PVCN and DCN) in C57 mice. Morphology, GluR1 and GRIP-C localization differ in octopus cells of C57BL6 and B6Cast mice.

The C57BL6 mouse (B6) is homozygous for the gene for age-related hearing loss (ahl/ahl) and shows normal adult-like hearing before subtle changes in hearing begin at about 30 days of age. The B6Cast mouse is congenic to B6, having the wild type allele for normal hearing from Castaneous Ei on a B6 background. It has normal hearing throughout most of its lifespan. This study characterized the morphology of octopus cell (OC) somata in the posterior-ventral cochlear nucleus and of synaptic terminals on the OC somata in 8-week-old B6 and B6Cast mice, and the immunolocalization of antibodies to GluR

Hearing loss associated with the modifier of deaf waddler (mdfw) locus corresponds with age-related hearing loss in 12 inbred strains of mice.

The modifier of deaf waddler (mdfw) and age-related hearing loss (Ahl) loci were both discovered as inbred strain polymorphisms that affect hearing loss in mice. Both loci map to the same position on chromosome (Chr) 10. The mdfw locus interacts epistatically with the deaf waddler (dfw) mutation on Chr 6, and the Ahl locus is a major contributor to AHL in several inbred strains. To investigate the possibility of allelism, we examined the correspondence of mdfw and Ahl phenotypes among 12 inbred mouse strains. The effects of strain-specific mdfw alleles on hearing loss were assessed in dfw2J/+

Bioinformatics and experiments reveal the hub genes of age-related hearing loss and the mechanism of PLK1 silencing in the protection of aging cochlear hair cells.

OBJECTIVE: We aim to identify the hub genes and explore molecular mechanisms underlying age-related hearing loss (ARHL) through multi-omics approaches and experimental validation. METHODS: Differentially expressed genes (DEGs) were screened from the GSE6045 dataset for functional enrichment analysis, protein-protein interaction (PPI) network analysis, and gene set enrichment analysis (GSEA), along with an investigation into the immune microenvironment in ARHL. Hub genes were assessed using the receiver operating characteristic (ROC) curves and further validated using GSE49543 datasets and ARH

Age-related hearing loss involves mitochondrial DNA instability and copy number depletion in the cochlea: Insights from in vivo and in vitro models.

Age-related hearing loss (ARHL) is a highly prevalent sensory neurodegenerative disorder that involves various molecular mechanisms. The present study investigated if age-related oxidative stress (OS) induces alterations in mitochondrial DNA (mtDNA) copy number and heteroplasmy, using complementary in vivo and in vitro models. Aged (30-month) CBA/CaJ mice have elevated auditory brainstem response (ABR) thresholds amplitudes vs. young (3-month) CBA/CaJ mice, a key physiological characteristic of ARHL The in vitro experiments employed the strial SV-K1 cochlear cell line treated with hydrogen per
