Hippocampal and amygdalar structures preferentially target somatostatin-expressing interneurons in the auditory cortex.

The auditory cortex integrates sensory and contextual inputs in emotion and fear driven situations that can be consolidated over learning. Somatostatin-expressing (SST+) interneurons play a key role in inhibitory modulation of sensory processing, yet their long-range inputs remain poorly characterised. We performed rabies-based retrograde monosynaptic tracing to compare the specific inputs to SST+ interneurons with the general input connectome of neurons in the mouse auditory cortex. Overall, inputs were similar, with the majority of presynaptic neurons originating from the ipsilateral auditor

Disturbing the sound of silence: Bilateral temporal cortex stimulation and auditory mental imagery.

Auditory imagery depends on temporal-cortical mechanisms that generate and sustain internal sound representations. If these mechanisms are causally involved, externally perturbing temporal cortex should alter the quality of imagery. We tested whether bilateral high-frequency transcranial random noise stimulation (hf-tRNS) over temporal cortex alters the vividness and control of auditory imagery. Forty-nine healthy adults completed two sessions on separate days, receiving Active hf-tRNS in one session and Sham in the other (order counterbalanced). The Bucknell Auditory Imagery Scale (BAIS; Vivi

Music can be reconstructed from human auditory cortex activity using nonlinear decoding models.

Music is core to human experience, yet the precise neural dynamics underlying music perception remain unknown. We analyzed a unique intracranial electroencephalography (iEEG) dataset of 29 patients who listened to a Pink Floyd song and applied a stimulus reconstruction approach previously used in the speech domain. We successfully reconstructed a recognizable song from direct neural recordings and quantified the impact of different factors on decoding accuracy. Combining encoding and decoding analyses, we found a right-hemisphere dominance for music perception with a primary role of the superi

Focal tDCS of auditory cortex in chronic tinnitus: A randomized controlled mechanistic trial.

OBJECTIVE: The goal of this pilot study was to understand how focal transcranial direct current stimulation (tDCS) targeting auditory cortex changes brain function in chronic tinnitus using magnetic resonance imaging (MRI). METHODS: People with chronic tinnitus were randomized to active or sham tDCS on five consecutive days in this mechanistic trial (n = 10/group). Focal 4x1 tDCS (central anode, surround cathodes) targeted left auditory cortex, with single-blind 2 mA current during twenty-minute sessions. Arterial spin-labeled and blood oxygenation level dependent MRI occurred immediately bef

Salicylate increased ascorbic acid levels and neuronal activity in the rat auditory cortex.

IMPORTANCE: Clinical observations have implied a central origin for tinnitus and potential therapeutic effects of ascorbic acid (AA); however, the detailed mechanisms remain undetermined. OBJECTIVE: To investigate changes in the AA levels and neural activity in the auditory cortex (AC) during salicylate-induced tinnitus. METHODS: Rats were randomly divided into 3 groups: (1) saline group, which received an intraperitoneal saline injection; (2) SS group, which received an intraperitoneal sodium salicylate (SS) injection (350 mg/kg); and (3) SS+Lido group, which received an intraperitoneal SS

Electrocorticography reveals continuous auditory and visual speech tracking in temporal and occipital cortex.

During natural speech perception, humans must parse temporally continuous auditory and visual speech signals into sequences of words. However, most studies of speech perception present only single words or syllables. We used electrocorticography (subdural electrodes implanted on the brains of epileptic patients) to investigate the neural mechanisms for processing continuous audiovisual speech signals consisting of individual sentences. Using partial correlation analysis, we found that posterior superior temporal gyrus (pSTG) and medial occipital cortex tracked both the auditory and the visual

Selective Neuronal Activation by Cochlear Implant Stimulation in Auditory Cortex of Awake Primate.

UNLABELLED: Despite the success of cochlear implants (CIs) in human populations, most users perform poorly in noisy environments and music and tonal language perception. How CI devices engage the brain at the single neuron level has remained largely unknown, in particular in the primate brain. By comparing neuronal responses with acoustic and CI stimulation in marmoset monkeys unilaterally implanted with a CI electrode array, we discovered that CI stimulation was surprisingly ineffective at activating many neurons in auditory cortex, particularly in the hemisphere ipsilateral to the CI. Furthe

Spatial pattern of BOLD fMRI activation reveals cross-modal information in auditory cortex.

Recent findings suggest that neural representations in early auditory cortex reflect not only the physical properties of a stimulus, but also high-level, top-down, and even cross-modal information. However, the nature of cross-modal information in auditory cortex remains poorly understood. Here, we used pattern analyses of fMRI data to ask whether early auditory cortex contains information about the visual environment. Our data show that 1) early auditory cortex contained information about a visual stimulus when there was no bottom-up auditory signal, and that 2) no influence of visual stimula

Auditory cortex asymmetry, altered minicolumn spacing and absence of ageing effects in schizophrenia.

The superior temporal gyrus, which contains the auditory cortex, including the planum temporale, is the most consistently altered neocortical structure in schizophrenia (Shenton ME, Dickey CC, Frumin M, McCarley RW. A review of MRI findings in schizophrenia. Schizophr Res 2001; 49: 1-52). Auditory hallucinations are associated with abnormalities in this region and activation in Heschl's gyrus. Our review of 34 MRI and 5 post-mortem studies of planum temporale reveals that half of those measuring region size reported a change in schizophrenia, usually consistent with a reduction in the left hem

Inhibition of voltage-gated channel currents in rat auditory cortex neurons by salicylate.

Salicylate is a medicine for anti-inflammation with a side effect of tinnitus. To understand the mechanisms of tinnitus induced by salicylate, we studied the effects of salicylate on voltage-gated ion channels and action potential firing rates in freshly dissociated rat pyramidal neurons in auditory cortex (AC) using the whole-cell patch technique. We found that salicylate reduced the voltage-gated sodium current (I(Na)), the delayed rectifier potassium current (I(K(DR))) and the L-type voltage-gated calcium current (I(Ca,L)) in concentration-dependent manner. An amount of 1mM salicylate shift

Image-to-sound conversion: experience-induced plasticity in auditory cortex of blindfolded adults.

The ability to adapt to environmental changes is based on the impressive capacity of the central nervous system for plasticity changes. A better understanding of the requirements of neuroplasticity will help to apprehend and predict the success of sensory prostheses. To investigate neuroplastic changes associated with (1) blindfolding and (2) the use of a mobile visual-auditory substitution system, five normally sighted adults underwent weekly measurements of neuromagnetic activity using a 122-channel whole head neuromagnetometer. The substitution device converted visual images into sound patt

Inhibitory effect of tDCS on auditory evoked response: Simultaneous MEG-tDCS reveals causal role of right auditory cortex in pitch learning.

A body of literature has demonstrated that the right auditory cortex (AC) plays a dominant role in fine pitch processing. However, our understanding is relatively limited about whether this asymmetry extends to perceptual learning of pitch. There is also a lack of causal evidence regarding the role of the right AC in pitch learning.  We addressed these points with anodal transcranial direct current stimulation (tDCS), adapting a previous behavioral study in which anodal tDCS over the right AC was shown to block improvement of a microtonal pitch pattern learning task over 3 days. To address the

Alpha rhythms in the left auditory cortex set the speed limit for speech comprehension.

Human cognition operates within distinct temporal windows spanning from very short to extremely long. Each cognitive function has its own processing speed range, beyond which performance deteriorates or even becomes impossible. Speech comprehension, as one of the most important cognitive functions in civilized societies, exemplifies this constraint: Performance declines markedly when speech rates exceed about 10 syllables per second. However, the neural mechanisms underlying this speed limit remain unclear. Here, by combining psychophysics, scalp-electroencephalography, transcranial alternatin

Brain-wide mapping of neuroanatomical connections to the auditory cortex of hearing and deaf mice.

Remarkable therapeutic innovations have made it possible to establish hearing in congenitally deaf subjects. Despite these advances, a potential obstacle to restoring auditory function is that the absence of auditory experience alters the connectivity of the auditory cortex, a region that contributes to auditory perception and cognition. Here we used an intersectional genetic approach to map the brainwide inputs to the primary auditory cortex of congenitally deaf mice and their hearing littermates. We found that deaf mice displayed a significant reduction in afferents arising from the basomedi

Structural and functional atypicality in the temporal cortex are associated with auditory perception in maltreated children.

Child maltreatment adversely affects brain development, resulting in vulnerabilities in brain structure and function connectivity, as well as various psychiatric disorders. However, the relationship between structural changes in auditory-related regions and auditory function remains unclear. Therefore, this study investigated the relationship between auditory frequencies associated with brain atypicality and child maltreatment. T1-weighted magnetic resonance imaging (MRI) and functional MRI were used to assess differences in gray matter volume (GMV) and functional connectivity (FC) in maltreat

Effect of Lidocaine on Quinine-Induced Tinnitus in Guinea Pigs: A Focus on the Auditory Cortex.

HYPOTHESIS: This study investigated whether intravenous lidocaine reverses auditory cortex changes induced by systemic quinine administration. BACKGROUND: Quinine, like salicylate, is known to induce tinnitus. Previous studies have shown that salicylate-induced changes in the auditory cortex can be reversed by lidocaine, a compound known to suppress tinnitus. If lidocaine also reverses quinine-induced changes, it would further support the association between these neural changes and the presence of tinnitus. METHODS: Healthy male albino Hartley guinea pigs received systemic quinine, followed

Auditory processing control by the medial prefrontal cortex: A review of the rodent functional organisation.

Afferent inputs from the cochlea transmit auditory information to the central nervous system, where information is processed and passed up the hierarchy, ending in the auditory cortex. Through these brain pathways, spectral and temporal features of sounds are processed and sent to the cortex for perception. There are also many mechanisms in place for modulation of these inputs, with a major source of modulation being based in the medial prefrontal cortex (mPFC). Neurons of the rodent mPFC receive input from the auditory cortex and other regions such as thalamus, hippocampus and basal forebrain
