Free amino acids in human tonsillar tissue.

Free amino acids in the tonsils of 20 individuals were measured column chromatographically. Those always found in readily detectable amounts included O-phosphoserine, taurine, O-phosphoethanolamine, aspartic acid, hydroxyproline, threonine, serine, glutamic acid, proline, glycine, alanine, alpha-amino-n-butyric acid, valine, cystine, methionine, isoleucine, leucine, tyrosine, phenylalanine, ornithine, gamma-amino-butyric acid, lysine, histidine, and arginine. Results were compared for three clinical pathological groups and for four age groups. Some abnormal values may result from the pathologi

Oral supplementation with sesaminol, a sesame-related lignan, ameliorates ethanol-induced dysbiosis of the gut microbiota and increases the gut luminal short-chain fatty acid concentrations of mice.

Chronic ethanol consumption significantly increases the risk of colorectal cancer. The pathogenesis of ethanol-related colorectal cancer involves oxidative stress and inflammation induced by ethanol in the colon and rectum, as well as dysfunction of the gut barrier and greater intestinal permeability. Previously, we demonstrated that chronic oral ethanol administration in mice leads to dysbiosis of the fecal microbiota, similar to that which characterizes human inflammatory bowel disease. In addition, this ethanol-induced gut pathophysiology was alleviated by the oral administration of sesamin

Organic aciduria in hypoxic premature newborns simulating an inborn error of metabolism.

A very unusual pattern of organic acid excretion was established in the urine of two premature newborns with severe respiratory distress and cerebral haemorrhages. By combined gas chromatography/mass spectrometry the following acids were identified: lactic acid, alpha-hydroxy-butyric acid, beta-hydroxy-butyric acid, alpha-hydroxy-isovaleric acid, and p-hydroxy-phenyllactic acid. Calculation of the concentration revealed an excessive excretion of lactic acid and also very high excretion of the other acids. A postmortem blood sample from one of the patients revealed a comparable pattern. The abn

Astragaloside IV alleviates ulcerative colitis via gut microbiota - butyrate metabolism axis to reshape Th17/Treg balance.

Gut microbiota dysbiosis and Th17/Treg cell imbalance play critical roles in the pathogenesis of ulcerative colitis (UC). Astragaloside IV (AS-IV) exhibits extensive anti-inflammatory and immunomodulatory activities; however, the crosstalk between gut microbiota and Th17/Treg cells modulated by AS-IV remains unreported. Here, chronic colitis was induced in mice by free access to 2.5 % dextran sulfate sodium (DSS) solution over three 7-day cycles, with concurrent AS-IV administration. AS-IV effectively alleviated DSS-induced chronic colitis in mice, as evidenced by increased body weight and col

Gas-liquid chromatography in routine processing of blood cultures for detecting anaerobic bacteraemia.

Gas-liquid chromatography was performed on 233 positive blood cultures and findings were compared with culture results. Obligate anaerobic bacteria were recovered from 78 out of 79 blood cultures containing butyric or iso-valeric acids, or both; from 28 out of 69 blood cultures containing succinic acid; and from only one out of 41 blood cultures containing succinic but not butyric or iso-valeric acid. Good correlations (88%) were found for the recovery of anaerobic bacteria and the detection of butyric and/or iso-valeric acids. Detecting volatile fatty acids by gas-liquid chromatography perfor

Gas-liquid chromatography of positive blood cultures for rapid presumptive diagnosis of anaerobic bacteremia.

Gas-liquid chromatographic analysis of volatile fatty acid patterns was performed when growth was initially detected in 87 positive blood cultures. Isovaleric acid, butyric acid, or both were found in 39 of 43 (90%) blood cultures that contained anaerobes but were absent in all 44 cultures that contained facultatively anaerobic or aerobic bacteria. The detection of isovaleric acid indicated the presence of Bacteroides fragilis, as this acid was found in 88% of unimicrobial and 75% of polymicrobial bacteremias due to B. fragilis. Butyric acid was detected most often in blood cultures which grew

Ferulic acid mediates microbial fermentation of arabinoxylan to enhance host immunity by suppressing TLR4/NF-κB signaling.

The study was conducted to explore the relationship between arabinoxylan (AX) structure and microbial fermentation characteristics, and reveal molecular mechanism of AX on regulating immune function of the host. Results indicated that the group of wheat bran AX showed greater activity of feruloyl esterase, production of short chain fatty acids and ferulic acid compared with the blank group (P < 0.05). The AX increased sIgA concentration and protein expression of protein expression of TLR4 and NF-κB (p65), but decreased mRNA expression of pro-inflammatory cytokines in the ileum of weaned pig mo

Gut Microbiota and Metabolomics Insights into the Gualou-Xiebai Herb Pair for Dyslipidemia and Atherosclerosis.

As a chronic lipid driven arterial disease, dyslipidemia is one of the most critical risk factors for atherosclerosis (AS). The gut microbiota plays an important role in regulating host lipid metabolism disorders. Studies have shown that the herb "Gualou-Xiebai" (GLXB) can effectively regulate the blood lipid levels of ApoE -/- mice and inhibit blood lipid accumulation. However, it is not yet clear whether GLXB herb pair can alleviate lipid abnormalities in AS diseases by inhibiting cholesterol absorption. Meanwhile, the regulatory effect of GLXB herb pair on gut microbiota metabolites needs f

Identification of characteristic volatiles and metabolomic pathways during the fermentation of red grapefruit by Monascus purpureus using HS-SPME-GC-MS and metabolomics.

Fermentation of red grapefruit by Monascus purpureus (M. purpureus) results in complex changes in flavor compounds and metabolic profiles, but the specifics of these alterations are not well understood. This study aimed to investigate the changes in flavor compounds and metabolomic traits during this fermentation process. Using Headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) with non-targeted metabolomics, we analyzed flavor compounds and measured physicochemical indices throughout the fermentation period. We identified 23 volatile flavor metabolites

The growth-stimulating factor of Treponema phagedenis from bovine digital dermatitis lesions.

Bovine digital dermatitis (BDD) is an infectious skin disease of the hoof characterized by painful ulcerations that cause lameness in dairy cattle. Cell-free supernatants (CFS) of Falsiporphyromonas endometrii predominantly isolated from BDD lesions had the highest growth-stimulating effect on Treponema phagedenis among BDD-associated bacteria. Butyric acid was detected at a concentration of 45.4 mM in CFS of F. endometrii, and the growth of T. phagedenis was significantly promoted by butyric acid supplementation.

Phenylalanine Butyramide Is a New Cosmetic Ingredient with Soothing and Anti-Reddening Potential.

Human skin is colonized by diverse commensal microbes, making up the skin microbiota (SM), contributing to skin integrity and homeostasis. Many of the beneficial effects aroused by the SM are exerted by microbial metabolites such as short-chain fatty acids (SCFAs), including butyric acid. The SCFAs can be used in cosmetic formulations against skin diseases to protect SM by preserving and/or restoring their natural balance. Unpleasant sensorial properties and unfavorable physico-chemical properties of butyrate strongly limit its cosmetic use. In contrast, some butyrate derivatives, including ph

Integrated co-cultivation and subsequent esterification: Harnessing Saccharomyces cerevisiae and Clostridium tyrobutyricum for streamlined ester production.

The rising demand for natural products is accelerating research into sustainable methods for producing bio-based flavourings like ethyl butyrate. In this study, ethyl butyrate was successfully produced through the enzymatic esterification of butyric acid and ethanol, which were derived from the co-cultivation of Clostridium tyrobutyricum and Saccharomyces cerevisiae. Initial monoculture experiments with both strains were performed to investigate compromised fermentation conditions for co-cultivation. Based on these findings, anaerobic co-cultivation conditions were established at 37 °C and 150

Rat fibroblast-derived interferon: production in serum-free medium and enhancement by theophylline.

Incubation of rat fibroblasts (RFA-1) with theophylline after Newcastle disease virus (NDV) induction, increased rat fibroblast interferon (RfIFN) yields 3 to 10-fold. However, incubation with butyric acid for 24 or 48 h before NDV addition resulted in no change or decrease in IFN production. Combined treatment with butyric acid and theophylline did not improve the yields obtained with the methylxanthine alone. Production of IFN by RFA-1 cells was the same in serum-free and in serum-containing media. These findings will facilitate RfIFN production and purification.
