[Free radicals and lipid peroxidation in cell biology: physiopathologic prospects].

Oxygen derived free radicals (OFR) arise in the course of normal cellular life, especially during cellular respiration. They are formed when molecular oxygen is reduced to water. These highly reactive species are controlled by a protective system both enzymatic and non enzymatic which helps to prevent the accumulation of peroxidative damage to the cell. Lipid peroxides result from the reaction of oxygen derived free radicals with polyinsatured fatty acids of membranes phospholipids and can be formed both non enzymatically and enzymatically (eicosanoids). Oxygen derived free radicals attack ove

Cell-free DNA fragmentomics: from chromatin biology to clinical stratification.

Plasma cell-free DNA (cfDNA) is fragmented nonrandomly, reflecting the chromatin architecture of its source cells and the nucleases that process it. The fragmentome can therefore provide an indirect readout of cellular regulatory state. This review examines how DNA methylation, chromatin accessibility, nucleosome positioning, protein occupancy, and nuclease activity shape cfDNA fragmentation and considers their implications for oncology. We organize fragmentomic measurements into four classes: coverage and protection, fragment size, fragment-end properties, and diversity and entropy metrics. W

Manufacturing as biology: How T-cell processing shapes therapeutic outcomes.

T-cell therapies have transformed the treatment landscape for hematologic malignancies and show growing promise for solid tumors and non-oncologic diseases. Unlike conventional drugs, these living therapeutics are shaped not only by genetic engineering but also by the ex vivo manufacturing process itself, which functions as a powerful biological determinant of cell fate and function. Here, we present a framework that conceptualizes manufacturing as a form of biological programming that governs therapeutic outcomes through its effects on T-cell phenotype, fitness, and functional durability. We

Redox biology from the perspective of the red blood cell.

Decades of research have generated extensive knowledge on oxidant production and antioxidant systems in red blood cells. This work has given us not only a detailed understanding of redox control in these cells but also fundamental information that has helped shape the broader field of redox biology. This review covers aspects of this history and also highlights unresolved issues regarding erythrocyte antioxidant defenses. Hemoglobin is both the principal source of reactive oxidants, produced by autoxidation or interaction with redox-active xenobiotics, and a major target of oxidants that escap

Chronic granulomatous disease: lessons in cell biology from monogenic immunodeficiency.

Reactive oxygen species (ROS) are produced in immune cells by the phagocyte NADPH oxidase (NOX2) system that carries out coordinated transfer of electrons to molecular oxygen. The importance of the system in host defence and immunoregulation is underlined by chronic granulomatous disease (CGD), a severe monogenic immunodeficiency caused by mutations in genes encoding individual components of NOX2. CGD also leads to inflammatory manifestations due to the regulatory role of ROS in multiple signalling processes. We describe the system in detail, from its discovery to our current understanding of

Recent Advances in Single-Cell Analysis of Atherosclerotic Plaque Biology.

Atherosclerosis, the leading cause of coronary artery disease, stroke, and peripheral arterial disease, is now recognized as a lipid-driven disease complicated by an immune response that plays a major role in its pathogenesis. The response-to-injury hypothesis proposed by Ross et al. laid the foundation for understanding atherosclerosis as a chronic inflammatory process, in which endothelial injury and lipid insudation trigger immune activation, smooth muscle cell proliferation, and plaque formation. Traditional approaches, such as immunohistochemistry, flow cytometry, and bulk RNA sequencing,

Uncovering the core genetic programs governing plant guard cell biology.

Guard cells (GCs) regulate gas exchange and water loss in plants and have been extensively studied in Arabidopsis thaliana. However, cross-species comparisons at single-cell resolution remain limited. To address this, we aimed to define conserved and divergent transcriptomic signatures of GCs by generating a comparative single-cell atlas encompassing five species: A. thaliana, soybean (Glycine max), tomato (Solanum lycopersicum), rice (Oryza sativa), and sorghum (Sorghum bicolor). We performed single-nucleus RNA sequencing (sNucRNA-seq) on leaf tissues from each species, followed by integrativ

Novel insights into Hodgkin lymphoma biology by single-cell analysis.

The emergence and rapid development of single-cell technologies mark a paradigm shift in cancer research. Various technology implementations represent powerful tools to understand cellular heterogeneity, identify minor cell populations that were previously hard to detect and define, and make inferences about cell-to-cell interactions at single-cell resolution. Applied to lymphoma, recent advances in single-cell RNA sequencing have broadened opportunities to delineate previously underappreciated heterogeneity of malignant cell differentiation states and presumed cell of origin, and to describe

Stem cell biology and regenerative medicine for neonatal lung diseases.

Lung diseases remain one of the main causes of morbidity and mortality in neonates. Cell therapy and regenerative medicine have the potential to revolutionize the management of life-threatening and debilitating lung diseases that currently lack effective treatments. Over the past decade, the repair capabilities of stem/progenitor cells have been harnessed to prevent/rescue lung damage in experimental neonatal lung diseases. Mesenchymal stromal cells and amnion epithelial cells exert pleiotropic effects and represent ideal therapeutic cells for bronchopulmonary dysplasia, a multifactorial disea

LGR5 and LGR6 in stem cell biology and ovarian cancer.

OBJECTIVES: Autoantibodies have been described in the post-infectious state, specifically after Lyme disease and COVID-19. We aimed to describe the prevalence and potential clinical utility of several commercially available autoantibodies after these infections. METHODS: Euroimmun panels (myositis, scleroderma and ANA5) were assayed using sera from patients with Lyme disease with return to health (RTH) (n=70), post-treatment Lyme disease (n=58), COVID-19 RTH (n=47) and post-acute symptoms of COVID-19 (n=22). The post-Lyme questionnaire of symptoms (PLQS) was used to determine symptom burden a

Cell biology and function of neuronal ceroid lipofuscinosis-related proteins.

Neuronal ceroid lipofuscinoses (NCL) comprise a group of inherited lysosomal disorders with variable age of onset, characterized by lysosomal accumulation of autofluorescent ceroid lipopigments, neuroinflammation, photoreceptor- and neurodegeneration. Most of the NCL-related genes encode soluble and transmembrane proteins which localize to the endoplasmic reticulum or to the endosomal/lysosomal compartment and directly or indirectly regulate lysosomal function. Recently, exome sequencing led to the identification of four novel gene defects in NCL patients and a new NCL nomenclature currently c

Cell biology and immunology of Listeria monocytogenes infections: novel insights.

Listeria monocytogenes is an intracellular bacterium responsible for a disease characterized by several clinical features, such as septicemia, brain infection, abortion, and perinatal infection. These clinical features are attributed to its amazing capacity to cross several barriers in susceptible hosts. Intracellular infection is a consequence of the bacterium's capacity to enter a wide variety of mammalian cells, to not only survive but also replicate therein, and to its faculty to spread from one cell to the next, thereby escaping the humoral immune response. Here, we review both the well-e

Designing vaccines based on biology of human dendritic cell subsets.

The effective vaccines developed against a variety of infectious agents, including polio, measles, and hepatitis B, represent major achievements in medicine. These vaccines, usually composed of microbial antigens, are often associated with an adjuvant that activates dendritic cells (DCs). Many infectious diseases are still in need of an effective vaccine including HIV, malaria, hepatitis C, and tuberculosis. In some cases, the induction of cellular rather than humoral responses may be more important because the goal is to control and eliminate the existing infection rather than to prevent it.

New aspects in the biology of cutaneous B-cell lymphomas.

Immunologic and molecular genetic studies greatly contributed to a better understanding and interpretation of the distinct clinico-pathologic features of primary cutaneous B-cell lymphomas (CBCL), which are the basis for the consensus WHO-EORTC classification. There is increasingly accumulating evidence that these well defined clinico-pathologic entities of CBCL have specific immunologic and molecular features, which further support their nosologic categorization as well as either interesting similarities with other extranodal B-cell lymphomas or definite peculiarities as compared to nodal B-c
