Tuberculosis is caused by Mycobacterium tuberculosis (Mtb), a pathogen with a remarkable ability to survive within its human host by balancing replication and persistence throughout the course of infection. The success of Mtb depends on its ability to adapt to diverse and hostile environments imposed by the host. This adaptation is primarily mediated through transcriptional regulation of gene expression. While Mtb encodes several transcription factors essential for growth, it remains unclear which transcription factors directly regulate the bacterial cell cycle. In this work, we characterized

The ability to proliferate is a common feature of most T-cell populations. However, proliferation follows different cell-cycle dynamics and is coupled to different functional outcomes according to T-cell subsets. Whether the mitotic machineries supporting these qualitatively distinct proliferative responses are identical remains unknown. Here, we show that disruption of the microtubule-associated protein LIS1 in mouse models leads to proliferative defects associated with a blockade of T-cell development after β-selection and of peripheral CD4+ T-cell expansion after antigen priming. In contras

Investigation of primitive human haemopoietic cell behaviour requires methodologies for monitoring asynchronously activated cells over several generations. We describe a high-resolution procedure for tracking 5- (and 6-) carboxyfluorescein diacetate succinimidyl ester (CFSE)- labelled human haemopoietic cells through six cell cycles based on the precise halving of their CFSE-fluorescence at each mitosis. Using this approach in combination with DNA or surface antigen staining, we show that the addition of Flt3-ligand (FL) to a cytokine cocktail consisting of Steel factor, IL-3, IL-6 and G-CSF i

Hematopoietic stem cells (HSCs) self-renew to expand their pool but often differentiate upon division. Thus, it remains unclear how HSCs maintain their stemness during or after division. Here, we show that the suppression of glutamate dehydrogenase (Glud1), a key enzyme responsible for glutamate (Glu) catabolism, supports the maintenance of HSCs during division. We find that self-renewing mouse HSCs during hematopoietic regeneration exhibited lower intracellular Glu levels compared to HSCs differentiating during ex vivo culture. We therefore focus on the negative regulation of Glu catabolic re

Weed control is crucial for global food production, as weeds cause economic losses. Chalcones are multifunctional compounds with diverse molecular targets, strong phytotoxic potential, and easy synthesis. This work investigated the herbicidal potential and mode of action of methoxychalcones, as well as their precursor, cinnamaldehyde. Herbicidal activity was assessed via dose-response curves at 0-100 µmol L- 1. Mode of action experiments included phenylalanine ammonia lyase (PAL) activity, electrolyte leakage, chlorophyll a fluorescence, photosynthetic oxygen evolution, and mitotic index. Resu

Chromosome segregation in female germ cells and early embryonic blastomeres is known to be highly prone to errors. The resulting aneuploidy is therefore the most frequent cause of termination of early development and embryo loss in mammals. And in specific cases, when the aneuploidy is actually compatible with embryonic and fetal development, it leads to severe developmental disorders. The main surveillance mechanism, which is essential for the fidelity of chromosome segregation, is the Spindle Assembly Checkpoint (SAC). And although all eukaryotic cells carry genes required for SAC, it is not

In mammalian cell lines, the endosomal sorting complex required for transport (ESCRT)-III mediates abscission, the process that physically separates daughter cells and completes cell division. Cep55 protein is regarded as the master regulator of abscission, because it recruits ESCRT-III to the midbody (MB), the site of abscission. However, the importance of this mechanism in a mammalian organism has never been tested. Here we show that Cep55 is dispensable for mouse embryonic development and adult tissue homeostasis. Cep55-knockout offspring show microcephaly and primary neural progenitors req

Little is known about the factors that regulate the asymmetric division of cancer stem-like cells (CSC). Here, we demonstrate that EGFL6, a stem cell regulatory factor expressed in ovarian tumor cells and vasculature, regulates ALDH+ ovarian CSC. EGFL6 signaled at least in part via the oncoprotein SHP2 with concomitant activation of ERK. EGFL6 signaling promoted the migration and asymmetric division of ALDH+ ovarian CSC. As such, EGFL6 increased not only tumor growth but also metastasis. Silencing of EGFL6 or SHP2 limited numbers of ALDH+ cells and reduced tumor growth, supporting a critical r

Chromosome aberrations are a sensitive indicator of genetic change, and the measurement of chromosome aberration frequency in peripheral blood lymphocytes is often used as a biological dosimeter of exposure (1,4). The length of time that cells are maintained in culture before cytogenetic analysis is probably the most important in vitro factor that influences both the frequency and types of aberrations that are seen following exposure to mutagens. Therefore, for accurate cytogenetic measurements of genetic damage, cells must be analyzed in their first mitosis following exposure. As cells progre

Genes coding for homologs of the highly conserved cell division protein FtsZ were isolated from Bartonella henselae and Bartonella quintana, the causative agents of cat scratch disease and trench fever, respectively. DNA fragments coding for the ftsZ open reading frames (ORFs) were cloned into Escherichia coli following PCR amplification with primers based on the ftsZ sequence of the closely related species Bartonella bacilliformis. The amino acid sequences predicted from the cloned B. henselae and B. quintana ftsZ ORFs are 81 to 83% identical to the corresponding protein in B. bacilliformis.

Single cell suspension from dermal leprosy granulomas (10 tuberculoid and 10 lepromatous) was prepared and an assessment of the division and protein synthesis by the cells was made. The cells of tuberculoid granulomas showed a high incorporation of 3H-thymidine and 14C-leucine. On the contrary, the cells of the lepromatous granulomas exhibited poor division but their protein synthesis remained unimpaired. These observations suggest that the epithelioid cell granuloma of tuberculoid leprosy appears to be more active and secretory than the macrophage granuloma of lepromatous leprosy.

Various metabolites of arachidonic acid are now known to influence cell division. In this paper the effects on cell proliferation of arachidonic acid, some inhibitors of arachidonic acid metabolism and some analogs of arachidonic acid metabolites is described. The epithelial cell proliferation rate in the jejunum, in the descending colon and in dimethylhydrazine-induced tumors of rat colon was measured using a stathmokinetic technique. Administration of arachidonic acid resulted in retardation of cell proliferation in each of the tissues examined. A cyclooxygenase inhibitor (Flurbiprofen) prev

The intestinal immune system is constantly challenged to distinguish between innocuous dietary antigens, commensal microbiota and intestinal self-antigens (self-Ags) versus harmful pathogens and malignant cells. It resides in the intestinal wall itself, Peyer's patches (PPs) and the draining lymph nodes (LNs). Dendritic cells (DCs) are found in all of these structures and are professional antigen-presenting cells (APCs) dictating and maintaining T cell fate. Here, we review how DCs contribute to immune homeostasis in the gastrointestinal system through multiple strategies: division in labor an

Circadian rhythms and the cell cycle are two fundamental biological oscillators. They are linked through molecular mechanisms creating coupled oscillators where the outcomes of this coupling depend on numerous factors, such as the directionality of coupling (i.e., unidirectional vs. bidirectional), periods of the two oscillators, and the number and strength of connections. In this review, we provide a general overview of the links between circadian rhythms and the cell cycle and discuss potential implications of this connection in stem cell proliferation and differentiation of mammalian cells.

Although lifelong renewal and additional compensatory growth in response to demand are undeniable facts, so far, no specific stem cells have been found for pancreatic cells. According to the consensus model, the development of pancreas results from the hierarchical differentiation of pluripotent stem cells towards the appearance of the first endocrine and exocrine cells at approximately 7.5 to 8th gestation week (GW) of human embryo. However, the primitive endocrine cells arising from the embryonic phase of development do not appear to be mature or fully functional. Asymmetric localization of

The trabecular meshwork's (TM) physiological role is to maintain normal intraocular pressure by regulating aqueous humor outflow. With age, and particularly in eyes with primary open angle glaucoma, the number of cells residing within the TM is markedly decreased and the function of the tissue is compromised. Here we evaluate if transplantation of induced pluripotent stem cell derived TM like cells (iPSC-TM) restores TM cellularity and function in human eyes obtained from older human donors. Human iPSC were differentiated into iPSC-TM and compared to primary TM cells by RNAseq. iPSC-TM were th

Adaptive Foxp3(+)CD4(+) regulatory T (iTreg) cells develop outside the thymus under subimmunogenic antigen presentation, during chronic inflammation, and during normal homeostasis of the gut. iTreg cells are essential in mucosal immune tolerance and in the control of severe chronic allergic inflammation, and most likely are one of the main barriers to the eradication of tumors. The Foxp3(+) iTreg cell repertoire is drawn from naive conventional CD4(+) T cells, whereas natural Treg (nTreg) cells are selected by high-avidity interactions in the thymus. The full extent of differences and similari

Recent studies, showing that cell cycle-related nuclear proteins p105 and Ki-67 are associated with Alzheimer's disease (AD)-related cytoskeletal pathology, suggested that these proteins, in addition to their functions in regulating the cell cycle, may have more specialised functions in the adult nervous system. In order to test this hypothesis we studied the expression of the cell cycle-related proteins Ki-67, pCNA and p53 in the hippocampi of 33 subjects, including some with AD or other neurodegenerative disorders and some with no neurological disease. By immunohistochemistry we found nuclea

Cultures of JU-56 cells were irradiated in either G1 or G2 and examined either in their first post-irradiation metaphase (in diploids) or in their second post-irradiation metaphase (in colcemid-induced tetraploids). The timing of fixation, together with tritiated thymidine pulse labelling, allowed selection for scoring only metaphases of cells that were in the G1 or G2 phase of the cell cycle during irradiation. With G1 irradiation it was found that many of the aberrations observed at the first division were not present (as derivatives) at the second division, and also that new aberrations wer

The molecular circadian clock and melatonin secretion are pivotal for coordinating homeostasis pathways in many animals. Melatonin has been shown to regulate reproduction and neoblast proliferation in triclad flatworms, yet the extent to which these mechanisms operate in other platyhelminths remains unknown. We examined light sensing, endogenous melatonin production, and circadian control of asexual reproduction in the early-branching catenulid Stenostomum virginianum. Microscopy revealed refractile bodies with photoreceptor-like structures, blue-light autofluorescence, and PAX6 immunoreactivi
