Claudin-2 downregulation by KSHV infection is involved in the regulation of endothelial barrier function.

BACKGROUND: Kaposi sarcoma (KS), caused by the infection of Kaposi sarcoma-associated herpesvirus (KSHV), is a disease manifested mainly by dark purple skin and mouth nodules. Cancer care studies showed that co-infection of KSHV and human immunodeficiency virus (HIV) was able to increase the patients' survival, but the underlying mechanisms are still elusive. METHODS: To understand the mechanism underlying the prolonged survival in KSHV-HIV co-infected patients, we performed microarray analysis on RNA extracted from biopsies from KS tumors and adjacent healthy tissues in four KS patients. Sub

Genomic organization of claudin-1 and its assessment in hereditary and sporadic breast cancer.

Human claudin-1 is an integral protein component of tight junctions, a structure controlling cell-to-cell adhesion and, consequently, regulating paracellular and transcellular transport of solutes across human epithelia and endothelia. Recently, a claudin-1 (CLDN1) cDNA has been isolated from human mammary epithelial cells (HMECs). CLDN1 expression in HMECs, in contrast to low or undetectable levels of expression in a number of breast tumors and breast cancer cell lines, points to CLDN1 as a possible tumor-suppressor gene. In order to evaluate the CLDN-1 gene in sporadic and hereditary breast

Pentoxifylline, possibly by modulating expression of claudins in the hippocampus attenuates autistic-related behaviours in male mice subjected to maternal separation stress.

OBJECTIVES: Maternal separation (MS) stress is a risk factor for abnormal behaviors including autism-like behaviors. The blood brain barrier (BBB) dysfunction is involved in the pathophysiology of autism spectrum disorder (ASD). Pentoxifylline (PTX), a methylxanthine commonly used for peripheral vascular disease, has been shown that exerted neuroprotective effects. This study aims to examine the effects of PTX on autistic-like behaviors in MS mice, considering its possible effects on gene expression of claudins (CLDNs), as main molecules regulates BBB integrity. METHODS: Forty male mice were

Kidney-Brain Axis: In Renal Failure, Cardiotonic Steroids Stimulate Collagen-I in the Hypothalamus and Claudins 1 and 7 in the Choroid Plexus.

INTRODUCTION: Uremic rats exhibit elevations of Na/K-ATPase inhibitor Marinobufagenin (MBG). The study investigated whether MBG-induced hypothalamic collagen-1 production occurs in uremic rats. Because choroid plexus cells are involved in transporting substances from the cerebrospinal fluid, the effects of MBG on claudin-1 and claudin-7 were also studied. Whether increased MBG would be associated with plasma procollagen type I C-peptide in patients with chronic kidney disease was also examined. METHODS: Male Sprague-Dawley rats underwent 5/6 nephrectomy. 12 patients with chronic kidney diseas

Modulating claudin-2 with CRISPR-Cas9 to improve photodynamic therapy outcomes in colorectal cancer.

Claudin-2 (CLDN2) is a tight junction protein that is overexpressed in colorectal cancer (CRC) and is associated with chemoresistance. Photodynamic therapy (PDT) is an emerging treatment that utilizes a photosensitizer (in this case, chlorin e6 [Ce6]) and light to generate cytotoxic reactive oxygen species (ROS). This paper investigated the influence of the combination of Ce6-PDT and CRISPR-Cas9-mediated CLDN2 knockout (KO) on the relative metabolic activity of the CRC cell line. CRISPR-Cas9 was used to produce HCT116 cells with CLDN2 KO. Ce6 was placed on the cells, and the red laser (659 nm,

Claudin 18.2 testing in gastroesophageal and pancreatobiliary cancers: current status, clinical implications and challenges.

Claudin (CLDN) 18.2 is a tight-junction protein predominantly expressed in normal gastric epithelium. Malignant transformation can disrupt epithelial cell polarity, resulting in the exposure and overexpression of CLDN18.2 on tumor cell surfaces. This characteristic makes CLDN18.2 a promising therapeutic target in subsets of patients with gastric and gastroesophageal junction (G/GEJ), and pancreatobiliary cancers. The recent approval by the US Food and Drug Administration (FDA) of the anti-CLDN18.2 monoclonal antibody zolbetuximab marks a significant advancement in the management of locally adv

Candida tropicalis Alters Barrier Permeability and Claudin-1 Organization in Intestinal Epithelial Cells.

Inflammatory bowel disease (IBD) is an autoimmune disorder characterized by chronic inflammation of the gut and compromised intestinal barrier function, resulting from aberrant immune responses targeting the intestinal microbiota. While the involvement of Candida albicans in IBD pathogenesis is well-documented, the role of non- albicans Candida species in IBD remains less understood. Recent studies have identified a correlation between elevated levels of Candida tropicalis , a notable non- albicans opportunistic fungus, and the development of IBD. However, the precise impact of C. tropicalis o

Claudin-1 expression in acne and its correlation with disease severity: clinical and in silico validation.

The pilosebaceous unit is the site of a common chronic inflammatory skin disorder called acne. Tight junctions (TJ) are present in sebocytes and are primarily composed of claudin-1. When claudin-1 is knocked down, the TJ barrier leaks, and holocrine secretion is compromised. This leads to the accumulation of partially degenerated sebocytes in the pilosebaceous ducts and the creation of microcomedos, which can cause acne lesions. The study's aim was to measure the claudin-1 gene and protein expression in acne patients' blood samples and establish a correlation with the severity of the condition

Exploring therapeutic potential of claudin in Flavivirus infection: A review on current advances and future perspectives.

Flavivirus such as Dengue, Zika, West Nile, Japanese encephalitis, and yellow fever virus, composed of single-stranded positive-sense RNA, predominantly contaminated through arthropods. Flavivirus infection characterises from asymptomatic signs to severe hemorrhagic fever and encephalitis. The host's immune system detects these viruses and provides a defence mechanism to sustain their life and growth. However, flaviviruses through different mechanisms compromise the host's immune defence. The current pharmacotherapeutic strategies against Flavivirus infection target different stages of the Fla

The claudin-like apicomplexan microneme protein is required for gliding motility and infectivity of Plasmodium sporozoites.

Invasion of host cells by apicomplexan parasites such as Toxoplasma and Plasmodium spp requires the sequential secretion of the parasite apical organelles, the micronemes and the rhoptries. The claudin-like apicomplexan microneme protein (CLAMP) is a conserved protein that plays an essential role during invasion by Toxoplasma gondii tachyzoites and in Plasmodium falciparum asexual blood stages. CLAMP is also expressed in Plasmodium sporozoites, the mosquito-transmitted forms of the malaria parasite, but its role in this stage is still unknown. CLAMP is essential for Plasmodium blood stage grow

Predictive Significance of Claudin-3 for Epithelial Barrier Dysfunction in Chronic Rhinosinusitis With Nasal Polyps.

PURPOSE: The abnormal expression of tight junction (TJ) plays a vital role in the pathogenesis of chronic rhinosinusitis with nasal polyps (CRSwNP). However, there is no appropriate tool to distinguish and diagnose epithelial barrier defects in clinical practice. This study aimed to evaluate the predictive value of claudin-3 for epithelial barrier dysfunction in CRSwNP. METHODS: In this study, TJ protein levels were evaluated by real-time quantitative polymerase chain reaction, immunofluorescent, and immunohistochemistry staining in control subjects and CRSwNP patients. The receiver operating

EPCAM and TROP2 share a role in claudin stabilization and development of intestinal and extraintestinal epithelia in mice.

Epithelial cell adhesion molecule (EPCAM) is a transmembrane glycoprotein expressed on the surface of most epithelial and epithelium-derived tumor cells and reported to regulate stability of epithelial tight junction proteins, claudins. Despite its widespread expression, loss of EPCAM function has so far only been reported to prominently affect intestinal development, resulting in severe early onset enteropathy associated with impaired growth and decreased survival in both humans and mice. In this study, we show that the critical role of EPCAM is not limited to intestinal tissues and that it s

Claudin-9 constitutes tight junctions of folliculo-stellate cells in the anterior pituitary gland.

The anterior pituitary gland regulates growth, metabolism, and reproduction by secreting hormones. Folliculo-stellate (FS) cells are non-endocrine cells located among hormone-producing cells in the anterior pituitary glands. They form follicular lumens, which are sealed by tight junctions (TJs). Although FS cells are hypothesized to contribute to fine-tuning of endocrine cells, little is known about the exact roles of FS cells. Here, we investigated the molecular composition of TJs in FS cells. We demonstrated that occludin is a good marker for TJs in the pituitary gland and examined the struc

Utility of Claudin-4 versus BerEP4 and B72.3 in pleural fluids with metastatic lung adenocarcinoma.

INTRODUCTION: Lung adenocarcinoma (LADC) is the most common occult primary in patients presenting with a malignant pleural effusion. Distinguishing metastatic LADC from reactive mesothelial cells (RMC) and malignant mesothelioma (MM) based on morphology alone has been a persistent diagnostic challenge in cytopathology. Claudin-4, a major functional constituent of tight junctions, has been shown to help distinguish LADC from RMC and MM in surgical specimens. Our goal was to further validate and assess the utility of Claudin-4 in comparison to BerEP4 and B72.3 in malignant effusions with a focus
