o-Toluidine and its metabolites induce oxidative and nitrative DNA damage in human urinary bladder epithelial cells.

o-Toluidine has been used for multiple industrial purposes, but causes bladder carcinogenesis in exposed workers. However, its underlying mechanism is still unclear. In this study, we examined oxidative and nitrative DNA damage caused by o-toluidine and its metabolites, 2-amino-m-cresol, 4-amino-m-cresol (4AC) and N-(4-hydroxy-2-methylphenyl) acetamide, using T24 human bladder epithelial cells. MTT assay showed that 4AC induced the most potent cytotoxicity among o-toluidine and its metabolites. In flow cytometry, 4AC showed the strongest ability of generating reactive oxygen species (ROS) and

Causal associations between chronic obstructive pulmonary disease, pulmonary tuberculosis, asthma, and plasma metabolites: A bidirectional Mendelian randomization study.

BACKGROUND: Epidemiological evidence has clarified circulating metabolite-respiratory pathology associations, but their fundamental causal mechanisms remain unclear. METHODS: A systematic two-sample Mendelian randomization analysis was performed to explore causal relationships between 1400 plasma metabolites and three major respiratory disorders ((chronic obstructive pulmonary disease (COPD), pulmonary tuberculosis (PTB), and asthma)) using genome-wide association studies (GWASs) data. Inverse variance-weighted (IVW) regression was the primary method, with MR-Egger and weighted median as supp

Integrated multi-omics analysis identifies microbial and metabolic signatures and drivers of CNS autoimmunity.

Multiple sclerosis (MS) is an autoimmune disease of the central nervous system (CNS) driven by genetic and environmental determinants. The gut microbiome of people with MS (pwMS) is distinct and influences disease through immunomodulatory metabolite production. Circulating metabolites are altered in pwMS, but identifying microbial-metabolic drivers remains challenging. We previously showed that colonization by the gut commensal Limosilactobacillus reuteri (L. reuteri) exacerbates disease in the experimental autoimmune encephalomyelitis (EAE) model of MS, in a tryptophan-dependent manner. Here,

Functional analysis of RYR1 variants in Australian and New Zealand patients at risk of susceptibility to malignant hyperthermia.

BACKGROUND: Muscle biopsy and in vitro contracture testing remain the gold standard for presymptomatic diagnosis of malignant hyperthermia susceptibility. DNA analysis can be used when a familial variant is classified as pathogenic or likely pathogenic. For this, variants must meet a range of criteria, one of which is functional analysis in an approved system that demonstrates hypersensitivity of calcium release when exposed to an appropriate agonist. METHODS: Seven patients with a clinical history of malignant hyperthermia were screened for the presence of variants in RYR1 by amplicon or who

Cellulose acetate, a source from discarded cigarette butts for the development of mixed matrix loose nanofiltration membranes for selective separation.

This study presents an environmentally friendly method for extracting cellulose acetate (CA) from discarded cigarette filters, which is then utilized in the fabrication of cellulose-based membranes designed for high flux and rejection rates. CA membranes are likeable to separate dyes and ions, but their separation efficiency is exposed when the contaminant concentration is very low. So, we have integrated graphene oxide (GO) and carboxylated titanium dioxide (COOH-TiO2) in CA to develop mixed matrix membranes (MMMs) and studied them against dyes and most used salts. The CA has been extracted f

Altered blood microbiome in patients with HCV-related Child-Pugh class B cirrhosis.

BACKGROUND: Altered bacterial translocation is associated with changes in hepatic function and the progression from compensated to decompensated cirrhosis. Child-Turcotte-Pugh (CTP) score is an essential indicator of liver severity. Thus, we aimed to study differences in the blood microbiome together with metabolome profile between HCV-infected patients with CTP class B (CTP-B, significant functional compromise) and patients with CTP class A (CTP-A, well-compensated cirrhosis). METHODS: We conducted a cross-sectional study in patients with advanced HCV-related cirrhosis (n = 88) stratified by

Plasma metabolites of aromatic amino acids associate with clinical severity and gut microbiota of Parkinson's disease.

Gut microbial proteolytic metabolism has been reportedly altered in Parkinson's disease (PD). However, the circulating aromatic amino acids (AAA) described in PD are inconsistent. Here we aimed to investigate plasma AAA profiles in a large cohort of PD patients, and examine their correlations with clinical severity and gut microbiota changes. We enrolled 500 participants including 250 PD patients and 250 neurologically normal controls. Plasma metabolites were measured using liquid chromatography mass spectrometry. Faecal samples were newly collected from 154 PD patients for microbiota shotgun

Rapid and sustainable HPLC method for the determination of uremic toxins in human plasma samples.

Protein-bound uremic toxins, mainly indoxyl sulfate (3-INDS), p-cresol sulfate (pCS), and indole-3-acetic acid (3-IAA) but also phenol (Pol) and p-cresol (pC), are progressively accumulated during chronic kidney disease (CKD). Their accurate measurement in biomatrices is demanded for timely diagnosis and adoption of appropriate therapeutic measures. Multianalyte methods allowing the establishment of a uremic metabolite profile are still missing. Hence, the aim of this work was to develop a rapid and sensitive method based on high-performance liquid chromatography with fluorescence detection fo

Analysis of human cultured myotubes responses mediated by ryanodine receptor 1.

Malignant hyperthermia is a life-threatening condition caused by autosomal dominant mutations in the ryanodine receptor type 1 gene. Identifying patients predisposed to malignant hyperthermia is done through the Ca-induced Ca release test in Japan. We examined the intracellular calcium concentration in human cultured muscle cells and compared the sensitivity of myotubes to ryanodine receptor type 1 activators based on the Ca-induced Ca release rate. We assessed the utility of this method as an identifying test for predisposition to malignant hyperthermia. Muscle specimens were obtained from 34

Combination of cell culture and quantitative PCR (cc-qPCR) to assess disinfectants efficacy on Cryptosporidium oocysts under standardized conditions.

Oocysts of Cryptosporidium parvum are resistant to environmental conditions and many disinfectants. A combination of cell culture and quantitative real time PCR (cc-qPCR) is established for evaluation of anticoccidial disinfectants against C. parvum. C. parvum oocysts were treated with disinfectants, washed and oocysts were incubated with HCT-8 cell monolayers in the presence of excystation medium for 3h. Subsequently, unbound parasites were removed by washing with growing medium and the infected monolayers were further maintained in fresh growing medium for 48h. Genomic DNA was extracted from

Site-Tailored bimetallic Co-Ce catalyst on N-Doped carbon for highly selective hydrodeoxygenation of vanillin.

The efficient conversion of lignin-derived oxygenates into fuels and high-value chemicals is pivotal for the development of renewable energy systems. A central challenge lies in the selective cleavage of the inherently recalcitrant C-O bonds in bio-oil model compounds under mild conditions. In this study, a multi-interface Co-Ce catalyst supported on nitrogen-doped carbon was successfully constructed via a bimetallic synergy and metal-organic framework (MOF)-derived strategy. The introduction of Ce exerted a multifunctional synergistic effect beyond that of a conventional promoter, realizing m

Reversible H atom abstraction catalyzed by the radical S-adenosylmethionine enzyme HydG.

The organometallic H-cluster at the active site of [FeFe]-hydrogenases is synthesized by three accessory proteins, two of which are radical S-adenosylmethionine enzymes (HydE, HydG) and one of which is a GTPase (HydF). In this work we probed the specific role of H atom abstraction in HydG-catalyzed carbon monoxide and cyanide production from tyrosine. The isotope distributions of 5'-deoxyadenosine and p-cresol were evaluated using deuterium-labeled tyrosine substrates in H2O and D2O. The observation of multiply deuterated 5'-deoxyadenosine and deuterated S-adenosylmethionine when the reaction

Functional analysis of RYR1 variants in patients with confirmed susceptibility to malignant hyperthermia.

BACKGROUND: A major bottleneck to the introduction of noninvasive presymptomatic diagnostic tests for the pharmacogenetic disorder malignant hyperthermia is the lack of functional data for associated variants. METHODS: We screened 50 genes having a potential role in skeletal muscle calcium homeostasis using the HaloPlex™ (Agilent Technologies, Santa Clara, CA, USA) target enrichment system and next-generation sequencing. Twenty-one patients with a history of a clinical malignant hyperthermia reaction together with a positive in vitro contracture test were included. Eight variants in RYR1 were

Evaluation of TiO2 Based Photocatalytic Treatment of Odor and Gaseous Emissions from Swine Manure with UV-A and UV-C.

It is essential to mitigate gaseous emissions that result from poultry and livestock production to increase industry sustainability. Odorous volatile organic compounds (VOCs), ammonia (NH3), hydrogen sulfide (H2S), and greenhouse gases (GHGs) have detrimental effects on the quality of life in rural communities, the environment, and climate. This study's objective was to evaluate the photocatalytic UV treatment of gaseous emissions of odor, odorous VOCs, NH3, and other gases (GHGs, O3-sometimes considered as by-products of UV treatment) from stored swine manure on a pilot-scale. The manure emis
