Traditions of Czech and Slovak developmental physiology.

Since the beginning of the 19th century, the comparative and ontogenetic branches of developmental physiology were cultivated in our country. Evidence was given that development of the gastrointestinal tract in tadpoles is dependent on the quality of proteins in their food. A complete metamorphosis of Amblystoma mexicanum, was entirely accomplished by feeding with powderized thyroid gland. The definition and chronological delimitation of both the suckling and weaning period in experimental animals opened the investigation of the effect of disturbance of the natural environment caused by premat

Sildenafil in bronchopulmonary dysplasia-associated pulmonary hypertension: developmental and translational insights.

Bronchopulmonary dysplasia-associated pulmonary hypertension (BPD-PH) is increasingly viewed as a developmental pulmonary vascular disorder rather than a purely reactive vasoconstrictive complication of chronic lung disease. It involves impaired angiogenesis, reduced pulmonary vascular surface area, alveolar simplification, and progressive right ventricle-pulmonary artery uncoupling. Sildenafil, a phosphodiesterase type 5 inhibitor, is widely used because it enhances nitric oxide-cyclic guanosine monophosphate signaling and may reduce pulmonary vascular tone. However, neonatal-specific evidenc

Light sheet microscopy with acoustic sample confinement.

Contactless sample confinement would enable a whole host of new studies in developmental biology and neuroscience, in particular, when combined with long-term, wide-field optical imaging. To achieve this goal, we demonstrate a contactless acoustic gradient force trap for sample confinement in light sheet microscopy. Our approach allows the integration of real-time environmentally controlled experiments with wide-field low photo-toxic imaging, which we demonstrate on a variety of marine animal embryos and larvae. To illustrate the key advantages of our approach, we provide quantitative data for

Spatial gradients of protein-level time delays set the pace of the traveling segmentation clock waves.

The vertebrate segmentation clock is a gene expression oscillator controlling rhythmic segmentation of the vertebral column during embryonic development. The period of oscillations becomes longer as cells are displaced along the posterior to anterior axis, which results in traveling waves of clock gene expression sweeping in the unsegmented tissue. Although various hypotheses necessitating the inclusion of additional regulatory genes into the core clock network at different spatial locations have been proposed, the mechanism underlying traveling waves has remained elusive. Here, we combined mo

Chlamydia trachomatis: a model for intracellular bacterial parasitism.

Chlamydia comprises a diverse group of obligate intracellular bacteria that cause infections in animals, including humans. These organisms share fascinating biology, including distinct developmental stages, non-canonical cell surface structures, and adaptations to intracellular parasitism. Chlamydia trachomatis is of particular interest due to its significant clinical importance, causing both ocular and sexually transmitted infections. The strain L2/434/Bu, responsible for lymphogranuloma venereum, is the most common strain used to study chlamydial molecular and cell biology because it grows r

Revealing the biological features of the axolotl pancreas as a new research model.

INTRODUCTION: The pancreas plays a crucial role in digestion and blood glucose regulation. Current animal models, primarily mice and zebrafish, have limited the exploration of pancreatic biology from an evolutionary-developmental perspective. Tetrapod vertebrate axolotl (Ambystoma mexicanum) serves as a valuable model in developmental, regenerative, and evolutionary biology. However, the fundamental biology of the axolotl pancreas remains underexplored. This study aims to characterize the unique developmental, functional, and evolutionary features of the axolotl pancreas to expand the understa

From germline immortality to somatic rejuvenation: Unlocking the ovarian blueprint for longevity.

Aging is typically framed as a one-way, irreversible accumulation of molecular damage in cells and tissues, leading to progressive functional decline. Yet mammalian reproduction, and particularly female reproduction, reveals a striking exception to this rule. Despite residing within an aging organism and within a fast-aging ovarian tissue environment, oocytes give rise to embryos that begin life with restored developmental potential and youthful molecular organization. By reframing ovarian biology as a model for rejuvenation rather than solely as a site of reproductive decline, this Essay prop

Post-translational modifications as key regulators of apicomplexan biology: insights from proteome-wide studies.

Parasites of the Apicomplexa phylum, such as Plasmodium spp. and Toxoplasma gondii, undergo complex life cycles involving multiple stages with distinct biology and morphologies. Post-translational modifications (PTMs), such as phosphorylation, acetylation and glycosylation, regulate numerous cellular processes, playing a role in every aspect of cell biology. PTMs can occur on proteins at any time in their lifespan and through alterations of target protein activity, localization, protein-protein interactions, among other functions, dramatically increase proteome diversity and complexity. In add

Emerging methods to generate artificial germ cells from stem cells.

Germ cells are responsible for the transmission of genetic and epigenetic information across generations. At present, the number of infertile couples is increasing worldwide; these infertility problems can be traced to environmental pollutions, infectious diseases, cancer, psychological or work-related stress, and other factors, such as lifestyle and genetics. Notably, lack of germ cells and germ cell loss present real obstacles in infertility treatment. Recent research aimed at producing gametes through artificial germ cell generation from stem cells may offer great hope for affected couples

The placental maturation inflection point: an integrative biological framework for understanding early-onset versus late-onset preeclampsia.

BACKGROUND: Preeclampsia (PE) is clinically classified into early-onset (EOPE, <34 weeks) and late-onset (LOPE, ≥34 weeks) phenotypes that differ in epidemiology, angiogenic profiles, and outcomes. Although the 34-week cutoff is widely adopted, its biological rationale remains formally unarticulated in terms of placental developmental biology. OBJECTIVE: To propose and evaluate the Placental Maturation Inflection Point (PMIP) - the transition from parenchymal expansion (Phase I) to functional optimization (Phase II) at approximately 34-36 weeks - as an integrative biological framework explain

Embryonic bauplans and the developmental origins of facial diversity and constraint.

A central issue in biology concerns the presence, timing and nature of phylotypic periods of development, but whether, when and why species exhibit conserved morphologies remains unresolved. Here, we construct a developmental morphospace to show that amniote faces share a period of reduced shape variance and convergent growth trajectories from prominence formation through fusion, after which phenotypic diversity sharply increases. We predict in silico the phenotypic outcomes of unoccupied morphospaces and experimentally validate in vivo that observed convergence is not due to developmental lim

Systems biology analysis of the lung cancer‑related secretome.

Tumorigenesis is closely and highly associated with developmental biology. The present study aimed to discover and identify marker proteins strongly associated with the occurrence and development of non‑small cell lung cancer (NSCLC) in humans and to provide new ideas for investigating lung cancer markers by combining biological analyses of embryonic development. We established primary cultures for samples of tumor and control tissues from 9 patients with NSCLC and collected conditioned medium (CM). Subsequently, we used liquid chromatography and linear ion trap (LTQ) mass spectrometry to isol

Toward adaptive cyborg tissues.

Organoids and engineered tissues have emerged as powerful models of human biology, yet their development and function are often characterized through endpoint measurements that provide only static snapshots of dynamic biological processes. Recent advances in stretchable mesh electronics, developmental biointegration, and organoid engineering have enabled the creation of cyborg tissues with embedded sensing, stimulation, and computational capabilities. This review highlights recent progress in developmental bioelectronic integration, multifunctional tissue mapping, biological state reconstructi

Developmental neurogenetics and neuro-ophthalmology.

The field of developmental neurogenetics has burgeoned over the past decade. Through the combined efforts of developmental biologists, geneticists, and clinicians, genetic defects resulting in neuro-ophthalmic disorders such as holoprosencephaly, microphthalmia, dominant optic atrophy, and optic nerve colobomas have been identified and characterized at the molecular level. Experimental studies in model organisms are continuing to identify novel genes critical for ocular and central nervous system development. Mutations in some of these genes have revealed a spectrum of pathology similar to tha

Case of Plasmodium knowlesi Malaria in Poland Linked to Travel in Southeast Asia.

We report a case of Plasmodium knowlesi malaria imported to central Europe from Southeast Asia. Laboratory suspicion of P. knowlesi infection was based on the presence of atypical developmental forms of the parasite in Giemsa-stained microscopic smears. We confirmed and documented the clinical diagnosis by molecular biology techniques.

Embryonic stem cell lines derived from human blastocysts.

Human blastocyst-derived, pluripotent cell lines are described that have normal karyotypes, express high levels of telomerase activity, and express cell surface markers that characterize primate embryonic stem cells but do not characterize other early lineages. After undifferentiated proliferation in vitro for 4 to 5 months, these cells still maintained the developmental potential to form trophoblast and derivatives of all three embryonic germ layers, including gut epithelium (endoderm); cartilage, bone, smooth muscle, and striated muscle (mesoderm); and neural epithelium, embryonic ganglia, a

"Muscling" Throughout Life: Integrating Studies of Muscle Development, Homeostasis, and Disease in Zebrafish.

The proper development and function of skeletal muscle is vital for health throughout the lifespan. Skeletal muscle function enables posture, breathing, and locomotion; and also impacts systemic processes-such as metabolism, thermoregulation, and immunity. Diseases of skeletal muscle (myopathies, muscular dystrophies) and even some neurological, age-related, and metabolic diseases compromise muscle function and negatively affect health span and quality of life. There have been numerous, recent examples of studies on skeletal muscle development with exciting, therapeutic implications for muscle

Amphibian teeth: current knowledge, unanswered questions, and some directions for future research.

Elucidation of the mechanisms controlling early development and organogenesis is currently progressing in several model species and a new field of research, evolutionary developmental biology, which integrates developmental and comparative approaches, has emerged. Although the expression pattern of many genes during tooth development in mammals is known, data on other lineages are virtually non-existent. Comparison of tooth development, and particularly of gene expression (and function) during tooth morphogenesis and differentiation, in representative species of various vertebrate lineages is

Human pluripotent stem cells for the modelling of diseases of the retina and optic nerve: toward a retina in a dish.

Human pluripotent stem cells can be differentiated into specific, relevant cell types of interest including the cells of the retina and optic nerve. These cells can then be used to study fundamental biology as well as disease modelling and subsequent screening of potential treatments. Many models of differentiation and modelling have relied on two-dimensional monocultures of specific cell types, which are not representative of the complexity of the human retina and optic nerve. Hence, more complex models of the human retina and optic nerve are required. Three-dimensional organoids and emerging
