Ginsenoside Rh4-regulated macrophage polarization and oxidative stress in septic myocardial injury.

Septic cardiomyopathy (SCM) is a severe complication of sepsis. The therapeutic potential of Ginsenoside Rh4 (Rh4) in SCM remains unclear. This study aimed to investigate the effects and mechanisms of Rh4 on SCM using both in vivo and in vitro approaches. In the in vivo experiments, Rh4 significantly reduced the mortality rate in SCM mice, improved ejection fraction and diastolic function, and protected myocardial cell mitochondria, thereby alleviating myocardial cell injury. Transcriptomic sequencing, ELISA, and PCR results suggested that Rh4 reduced inflammatory cytokines in myocardial tissu

The orexinergic system mediates the effects of ginsenoside Rg1 on cyclophosphamide-induced reproductive toxicity.

Cyclophosphamide (CP) is a widely prescribed chemotherapeutic and immunosuppressive agent known to cause significant reproductive toxicity. Ginseng effectively counteracts CP-induced reproductive toxicity, exhibiting pharmacological effects that resemble enhanced orexin signaling. In this study, molecular docking identified ginsenoside Rg1 as the candidate with the highest binding affinity for orexin receptor 1 (OX1R). In CP-treated male mice, Rg1 dose-dependently ameliorated reproductive toxicity, as evidenced by improved sexual behavior, sperm motility and DNA integrity, serum testosterone l

Multi-omics approaches to explore the therapeutic mechanism for ginsenoside Rg1 against MASLD.

Ginsenoside Rg1 (G-Rg1), a traditional Chinese medicine, alleviates metabolic dysfunction-associated steatotic liver disease (MASLD). However, the mechanism by which G-Rg1 improves metabolic disorders in MASLD by regulating the gut microbiota remains ambiguous‌. We constructed a diet-induced murine MASLD model and employed 16S rRNA sequencing and non-targeted metabolomic analysis to ‌investigate the mechanism of G-Rg1 in treating MASLD, focusing on its regulatory effect on the gut microbiota. Our results revealed that G-Rg1 significantly increased the 5-hydroxyindoleacetic acid levels and acti

Septin 2: A direct target of Ginsenoside Rg1 mediating its neuroprotective effects.

Ginsenoside Rg1 (Rg1), a representative saponin of ginseng, is evidenced with a remarkable neuroprotective effect; however, its direct target in neural cells has not been addressed. The present study investigated the reconstructing effect on the damaged neurite network of Rg1, and its direct target responsible for this bio-effect. The results showed that Rg1 can alleviate the Aβ25-35-induced neuronal damage and synaptic atrophy in primary neurons and PC12 cell models, respectively. Furthermore, the Septin 2, involved in cell division and cytoskeleton organization, was identified as a protein s

Research Progress on Effects of Ginsenoside Rg2 and Rh1 on Nervous System and Related Mechanisms.

Neurological-related disorders are diseases that affect the body's neurons or peripheral nerve tissue, such as Parkinson's disease (PD) and Alzheimer's disease (AD). The development of neurological disorders can cause serious harm to the quality of life and functioning of the patient. The use of traditional therapeutic agents such as dopamine-promoting drugs, anticholinergic drugs, cholinesterase inhibitors, and NMDA receptor antagonists is often accompanied by a series of side effects such as drug resistance, cardiac arrhythmia, liver function abnormalities, and blurred vision. Therefore, the

Ginsenoside Rc Modulates SIRT6-NRF2 Interaction to Alleviate Alcoholic Liver Disease.

Alcoholic liver disease (ALD) is a serious worldwide health problem. Ginsenoside Rc is a major active ingredient isolated from Panax ginseng, whose pharmacological effects counteract oxidative stress, inflammation, and lipid accumulation. However, it is still unclear whether ginsenoside Rc might exert beneficial effects on alcohol-induced liver injury. To this aim, mice primary hepatocytes (MPHs) were challenged with alcohol to test ginsenoside Rc's effects on their intracellular alcohol metabolism. C57BL/6J mice or SIRT6alb-/- mice were chronically fed a diet with added alcohol or given a sin

Efficacy of ginsenoside Rg3 nanoparticles against Ehrlich solid tumor growth in mice.

Solid tumors are fairly common and face many clinical difficulties since they are hardly surgically resectable and broadly do not respond to radiation and chemotherapy. The current study aimed to fabricate ginsenoside Rg3 nanoparticles (Rg3-NPs) and evaluate their antitumor effect against Ehrlich solid tumors (EST) in mice. Rg3-NPs were fabricated using whey protein isolates (WPI), maltodextrin (MD), and gum Arabic (GA). EST was developed by the injection of mice with Ehrlich ascites cells (2.5 × 106). The mice were divided into a control group, EST group, and the EST groups that were treated

Ginsenoside Compound K Enhances Fracture Healing via Promoting Osteogenesis and Angiogenesis.

Fractures have an extraordinarily negative impact on an individual's quality of life and functional status, particularly delayed or non-union fractures. Osteogenesis and angiogenesis are closely related to bone growth and regeneration, and bone modeling and remodeling. Recently Chinese medicine has been extensively studied to promote osteogenic differentiation in MSCs. Studies have found that Ginseng can be used as an alternative for tissue regeneration and engineering. Ginseng is a commonly used herbal medicine in clinical practice, and one of its components, Ginsenoside Compound K (CK), has

Ginsenoside Rb1 is an immune-stimulatory agent with antiviral activity against enterovirus 71.

ETHNOPHARMACOLOGICAL RELEVANCE: According to the theory of traditional Chinese medicine, the main pathogenesis of severe hand, foot and mouth disease (HFMD) is that the heat and wet poisons are deeply trapped in the viscera, which causes the deficiency of Qi and Yin in the patient's body. Ginsenoside Rb1 (Rb1) is the most abundant triterpenoid saponin in Panax quinquefolius L., which has the function of Qi-invigorating and Yin-nourishing. Enterovirus 71 (EV71) is one of the causative pathogens of HFMD, especially the form associated with some lethal complications. Therefore, the therapeutic ef

Ginsenoside Rh2 inhibits vascular endothelial growth factor-induced corneal neovascularization.

VEGF-induced neovascularization plays a pivotal role in corneal neovascularization (CoNV). The current study investigated the potential effect of ginsenoside Rh2 (GRh2) on neovascularization. In HUVECs, pretreatment with GRh2 largely attenuated VEGF-induced cell proliferation, migration, and vessel-like tube formation in vitro. At the molecular level, GRh2 disrupted VEGF-induced VEGF receptor 2 (VEGFR2)-Grb-2-associated binder 1 (Gab1) association in HUVECs, causing inactivation of downstream AKT and ERK signaling. Gab1 knockdown (by targeted short hairpin RNA) similarly inhibited HUVEC prolif

Ginsenoside Rh2 inhibits metastasis of glioblastoma multiforme through Akt-regulated MMP13.

Glioblastoma multiforme (GBM) is the most malignant type of primary brain tumor. Although the growth of the tumor cells in a relatively closed space may partially account for its malignancy, highly invasive nature of glioblastoma cells has been suggested to be the main reason for the failure of current therapeutic approaches. Ginsenoside Rh2 (GRh2) has recently been shown to significantly suppress the growth and survival of GBM through inhibiting epidermal growth factor receptor signaling, whereas its effects on the invasion and metastasis have not been examined. Here, we showed that GRh2 dose

Ginsenoside Rb1 inhibits fibrillation and toxicity of alpha-synuclein and disaggregates preformed fibrils.

Compelling evidence indicates that α-synuclein (α-syn) aggregation plays a central role in the pathogenesis of Parkinson's disease (PD) and other synucleinopathies. Identification of compounds that inhibit or reverse the aggregation process may thus represent a viable therapeutic strategy against PD and related disorders. Ginseng is a well-known medicinal plant that has been used in East Asia for more than two thousand years to treat several conditions. It is now understood that the pharmacological properties of ginseng can be attributed to its biologically active components, the ginsenosides,

Inhibition of Bcl-2/Bax/cleaved caspase-3 pathway activity by rare ginsenosides ameliorates cyclophosphamide-induced oligoasthenospermia (OA) in male mice.

ETHNOPHARMACOLOGICAL RELEVANCE: Oligoasthenozoospermia (OA) is the main factor leading to male infertility. According to traditional Chinese medicine, ginseng and its derivatives can improve male sperm quality by nourishing the kidneys and replenishing qi. Ginsenosides have been recognized as the principal pharmacologically active constituents of Panax ginseng, and are generally categorized into highly polar and rare ginsenosides. Rare ginsenosides can be generated through thermal transformation of the highly polar forms, yielding structurally related compounds collectively referred to as heat

Ginsenosides Rh2 and Rg3 in first-line combination therapy: Prospects for rapid clinical translation in drug-resistant triple-negative breast cancer.

Triple-negative breast cancer (TNBC), defined by the absence of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor-2 (HER2), remains one of the most aggressive breast cancer subtypes, characterized by high metastatic potential, poor prognosis, and limited therapeutic options. Despite extensive clinical efforts, durable improvements in survival have been limited due to the intrinsic heterogeneity of TNBC and the rapid emergence of multidrug resistance (MDR), primarily mediated by P-glycoprotein (P-gp) overexpression.
