Hippo cooperates with p53 to regulate lung airway mucous cell metaplasia.

Airway mucous cell metaplasia is a significant feature of many chronic airway diseases, such as chronic obstructive pulmonary disease, cystic fibrosis and asthma. However, the mechanisms underlying this process remain poorly understood. Here, we employed in vivo mouse genetic models to demonstrate that Hippo and p53 (encoded by Trp53) cooperate to modulate the differentiation of club cells into goblet cells. We revealed that ablation of Mst1 (Stk4) and Mst2 (Stk3), encoding the core components of Hippo signaling, significantly reduces mucous metaplasia in the lung airways in a lipopolysacchari

STK4 protein expression pattern follows different trends in endometrioid and serous endometrial adenocarcinoma upon tumor progression.

Источник пометил публикацию как отозванную

In a previous study, we showed that serine/threonine-protein kinase 4 (STK4) is involved in the control on proliferation and migration of endometrial cancer (EC) cells in vitro. In the present paper, we studied STK4 expression in EC tissues from a large cohort of patients to determine whether STK4 can serve as a marker for the aggressiveness and prognosis of EC. Tissue samples from patients with EC were examined for tumor type, grade, and stage. The STK4 protein expression in EC cells was assessed by immunohistochemistry and related to clinicopathological data of patients, such as progression

Effects of melatonin in polycystic ovary syndrome: is there Hippo pathway crosstalk?

OBJECTIVE: Polycystic ovary syndrome (PCOS) is a prevalent endocrine disorder among reproductive women, characterized by hyperandrogenism, oligo-ovulation and polycystic ovarian morphology. Incorporating complementary medicine alongside traditional lifestyle therapies for PCOS may offer additional benefits for affected women. Melatonin (MT), a hormone secreted by the pineal gland, has emerged as a potential treatment for regulating ovarian function in PCOS. However, the specific effects and underlying mechanisms of MT on PCOS need to be elucidated. METHODS: This review consolidates evidence f

HIPPO Stampede in Nerve Sheath Tumors.

Current therapies for malignant peripheral nerve sheath tumors (MPNSTs) are ineffective. The study by Wu et al. in this issue of Cancer Cell provides evidence that the HIPPO pathway is overactive in human MPNSTs and that combined modulation of LATS1/2-YAP/TAZ and PDGFR signaling in Schwann cells reduces MPNST growth.

The Hippo signalling pathway regulates the timing of epithelial cell replacement during Drosophila abdominal morphogenesis.

Developmental apoptosis plays an important role in shaping the organs of developing animals, such as occurs during the development of the adult fly abdomen, in which the apoptosis of larval epidermal cells (LECs) is delicately balanced with the proliferation of the presumptive adult epithelial cells. The signalling pathways involved in the coordinated regulation of these processes are at present poorly understood. Here, we demonstrate a role for the Hippo signalling pathway in regulating the timing of LEC clearance during abdominal development. Downregulating Hippo signalling promotes LEC surv

The emerging role of the Hippo signaling pathway in interorgan crosstalk.

Interorgan communication has emerged as a fundamental mechanism for maintaining systemic homeostasis, and its disruption contributes to the development of metabolic diseases, chronic inflammation, and cancer. The Hippo signaling pathway, traditionally known for controlling organ size, has recently been redefined as a context-dependent coordinator of systemic cues. By translating hormonal, metabolic, and microbial signals into coordinated cellular responses, Hippo signaling serves as a bidirectional communication hub that not only interprets systemic inputs but also generates "outputs" that con

Hippo signaling pathway in polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) is a complex endocrine-metabolic disorder syndrome, that predominantly affects women of reproductive age. It is characterized by marked clinical heterogeneity involving multiple systems including reproductive, metabolic and immune systems, while existing diagnostic protocols remain inadequate for clinical needs. Moreover, the incomplete understanding of PCOS etiology has limited therapeutic strategies for symptomatic management rather than interventions targeting core pathological mechanisms, resulting in PCOS frequently persisting as a chronic condition with a

Hippo signaling in mammalian reproduction.

ABSTRACT: Chronic endometritis, defined by chronic inflammation of the endometrium, remains a clinical and biologic challenge even using hysteroscopy allowing a direct vision of the uterine cavity without anesthesia, and conventional histology using Hematoxylin and Eosin staining. Our primary objectives were to evaluate the relevance of hysteroscopy and conventional histology compared to immunohistochemical expression of syndecan-1 (CD138, a marker of plasma cells), which is a heparan sulfate proteoglycan involved in inflammation and enables diagnosis of chronic endometritis. The second object

Hippo/YAP1 inhibition by verteporfin attenuates osteophyte formation in a mouse surgical osteoarthritis model.

Osteophyte is an outgrowth of cartilage formed at the margins of the affected joint through endochondral ossification-like processes, and is one of the most common radiographic features of osteoarthritis (OA) that has been used to define the stage of disease. Osteophyte has been regarded to adapt the joint to the altered biomechanics of OA patients, limits joint movement and represent a source of joint pain, however, the mechanism of osteophyte formation, the morphology characteristics and biomechanical properties of osteophyte cells are remained unclear. In the present study, we isolated oste

The role of the Hippo pathway in heart disease.

Heart disease, including coronary artery disease, myocardial infarction, heart failure, cardiac hypertrophy, and cardiomyopathies, is the leading causes of death worldwide. The Hippo pathway is a central controller for organ size and tissue growth, which plays a pivotal role in determining cardiomyocytes and nonmyocytes proliferation, regeneration, differentiation, and apoptosis. In this review, we summarize the effects of the Hippo pathway on heart disease and propose potential intervention targets. Especially, we discuss the molecular mechanisms of the Hippo pathway involved in maintaining c

Translating current basic research into future therapies for neurofibromatosis type 1.

Neurofibromatosis type 1 (NF1) is a hereditary tumour syndrome that predisposes to benign and malignant tumours originating from neural crest cells. Biallelic inactivation of the tumour-suppressor gene NF1 in glial cells in the skin, along a nerve plexus or in the brain results in the development of benign tumours: cutaneous neurofibroma, plexiform neurofibroma and glioma, respectively. Despite more than 40 years of research, only one medication was recently approved for treatment of plexiform neurofibroma and no drugs have been specifically approved for the management of other tumours. Work c

Chinese medicine monomers for hepatocellular carcinoma: New ideas related to autophagy.

Hepatocellular carcinoma (HCC) represents the most prevalent form of primary liver cancer, characterized by high mortality rates, frequent recurrence and metastasis, poor clinical prognosis, and a complex pathogenesis with limited therapeutic options. Autophagy plays a pivotal role in the immune response and functions as a lysosome-mediated degradation mechanism essential for recycling cellular components and eliminating aggregated proteins, damaged organelles, and invasive pathogens, thereby maintaining cellular function and dynamic homeostasis. Additionally, autophagy regulates several criti

Inhibition of cyclin-dependent kinase 7 down-regulates yes-associated protein expression in mesothelioma cells.

Cyclin-dependent kinase 7 (CDK7) is a protein kinase that plays a major role in transcription initiation. Yes-associated protein (YAP) is a main effector of the Hippo/YAP signalling pathway. Here, we investigated the role of CDK7 on YAP regulation in human malignant pleural mesothelioma (MPM). We found that in microarray samples of human MPM tissue, immunohistochemistry staining showed correlation between the expression level of CDK7 and YAP (n = 70, r = .513). In MPM cells, CDK7 expression level was significantly correlated with GTIIC reporter activity (r = .886, P = .019). Inhibition of CDK7

Deficiency of MST1 in endometriosis related peritoneal macrophages promoted the autophagy of ectopic endometrial stromal cells by IL-10.

Changes in the function of peritoneal macrophages contribute to the homeostasis of the peritoneal immune microenvironment in endometriosis. The mechanism by which ectopic tissues escape phagocytic clearance by macrophages to achieve ectopic colonization and proliferation is unknown. The expression of CD163 in peritoneal macrophages in patients with endometriosis is increased, with the overexpression of MAPK, which can promote the M2-type polarization of macrophages and reduce their ability to phagocytose ectopic endometrial cells. As an upstream regulator of MAPK, MST1 expression is deficient
