The Effect of Co-occurring Substance Use on Gamma-hydroxybutyric Acid Withdrawal Syndrome.

OBJECTIVES: Gamma-hydroxybutyric acid (GHB) withdrawal is a complex syndrome which can be potentially life-threatening. Additionally, GHB-dependent patients frequently report co-occurring substance use of other psychoactive drugs. We assessed the add-on effect of co-use on GHB withdrawal symptoms. METHODS: We conducted an open-label, pretest-posttest design study with 95 patients selected from 229 inpatients admitted for detoxification, who were divided into GHB only (GO, n = 40), GHB plus sedatives (GSE, n = 38), and GHB plus stimulants (GST, n = 17) groups. GHB withdrawal was evaluated by m

Lipase catalyzed ultrasonic synthesis of poly-4-hydroxybutyrate-co-6-hydroxyhexanoate.

Four different lipases were compared for ultrasound-mediated synthesis of the biodegradable copolymer poly-4-hydroxybutyrate-co-6-hydroxyhexanoate. The copolymerization was carried out in chloroform. Of the enzymes tested, Novozym 435 exhibited the highest copolymerization rate, in fact the reaction rate was observed to increase with about 26-fold from 30 to 50°C (7.9×10(-3)Ms(-1)), sonic power intensity of 2.6×10(3)Wm(-2) and dissipated energy of 130.4Jml(-1). Copolymerization rates with the Candida antarctica lipase A, Candida rugosa lipase, and Lecitase Ultra™ were lower at 2.4×10(-4), 1.3×

[Gamma-hydroxybutyric acid (GHB) dependence and the GHB withdrawal syndrome: diagnosis and treatment].

Gamma-hydroxybutyric acid (GHB) is a neurotransmitter that occurs naturally in the brain and is increasingly being used as a 'party drug' because of its relaxing and euphoria-inducing effects. GHB has a limited medical use in the treatment of narcolepsy. GHB-intoxications occur often in non-medical use, and generally result in a coma of short duration. GHB use several times a day can lead to tolerance and dependence. After sudden cessation or reduction of intensive GHB use, a severe withdrawal syndrome may occur with symptoms varying from tremor, anxiety and agitation to autonomic instability,

A review of tolerability and abuse liability of gamma-hydroxybutyric acid for insomnia in patients with schizophrenia.

BACKGROUND: Approved therapeutic uses for gamma-hydroxybutyric acid (GHB) (or sodium oxybate), a gamma-aminobutyric acid type B and GHB receptor agonist, include narcolepsy in the United States and Europe and alcohol abuse treatment in Italy. Possible efficacy of GHB in schizophrenia has also been proposed. A tolerability concern regarding use of GHB is its abuse potential. Given the high comorbidity of substance disorders and schizophrenia, a systematic assessment of the published literature is crucial. OBJECTIVE: The aim of this review was to assess the tolerability and abuse liability of G

Effect of genetically caused excess of brain gamma-hydroxybutyric acid and GABA on sleep.

BACKGROUND: Exogenous gamma-hydroxybutyrate (GHB) increases slow-wave sleep and reduces daytime sleepiness and cataplexy in patients with primary narcolepsy. OBJECTIVE: To examine nighttime sleep and daytime sleepiness in a 13-year-old girl homozygous for succinic semialdehyde dehydrogenase (SSADH) deficiency, a rare recessive metabolic disorder that disrupts the normal degradation of 4-aminobutyric acid (GABA), and leads to an accumulation of GHB and GABA within the brain. METHODS: Sleep interview, nighttime polysomnography, Multiple Sleep Latency Tests, and continuous 24-hour in-lab record

Displacement of [3H] gamma-hydroxybutyrate binding by benzamide neuroleptics and prochlorperazine but not by other antipsychotics.

Since gamma-hydroxybutyrate receptor agonists exhibit dopaminergic regulatory properties and neuroleptic-like effects in neuropharmacological tests, the common neuroleptics were tested for [3H] gamma-hydroxybutyrate binding activity on rat brain membranes. (-)-Sulpiride, sultopride, amisulpride and prochlorperazine possess affinity for the gamma-hydroxybutyrate site(s), consistent with their therapeutic dosage. This study has revealed that gamma-hydroxybutyrate receptors represent an additional target for antipsychotics.

The effects of 3-hydroxybutyrate and glucose on human T cell responses to Candida albicans.

Diabetic patients are particularly susceptible to mucocutaneous candidosis. T lymphocytes are central to the induction of antigen-specific immune responses and may be sensitive to the biochemical abnormalities associated with poorly controlled diabetes; namely, hyperglycaemia and/or ketonemia. To examine this we have studied the effect of varying concentrations of glucose and 3-hydroxybutyrate (3-HB) in cultures of human T cells stimulated with Candida albicans antigen. Proliferation of T cells from six type 1 diabetic and six non-diabetic control subjects was significantly inhibited (both P <

Sickle cell crisis as evaluated from measurements of hydroxybutyrate dehydrogenase and myoglobin in plasma.

Data on plasma hydroxybutyrate dehydrogenase activity (I) and myoglobin concentration were used to evaluate painful sickle cell crises. I was increased during non-crisis steady state in patients with sickle cell disease as compared to normal values (232, SD 79.7 vs 85, SD 33 Sigma units/mL). During crisis, the mean value for I increased further to 379 (SD 139) Sigma units/mL. For 12 patients evaluated both during steady state and crisis, there was a mean increase in plasma I of 131% (SD 76%). Repeated determinations of I in sickle cell disease patients during several months while they were in

Gamma hydroxybutyrate. Correlation of serum and cerebrospinal fluid levels with electroencephalographic and behavioral effects.

Gamma hydroxybutyrate was administered to adult cats by intravenous infusion at varying dosages while an electroencephalogram (EEG) was recorded from electrodes placed stereotactically in the right and left hippocampus and thalamic intralaminar nucleus and from cortical electrodes. Blood cerebrospinal fluid samples were assayed for gamma hydroxybutyrate. The first EEG change, slowing was occasional spikes, was seen at serum levels of 75 mug per milliliter. Changes in the recordings progressed through a number of stages, culminating in bursts of poly spiking interspersed among periods of electr

Chemicals Affecting Cyanobacterial Poly(3-hydroxybutyrate) Accumulation: 2-Phenylethanol Treatment Combined with Nitrogen Deprivation Synergistically Enhanced Poly(3-hydroxybutyrate) Storage in Synechocystis sp. PCC6803 and Anabaena sp. TISTR8076.

Various photoautotrophic cyanobacteria increase the accumulation of bioplastic poly(3-hydroxybutyrate) (PHB) under nitrogen deprivation (-N) for energy storage. Several metabolic engineering enhanced cyanobacterial PHB accumulation, but these strategies are not applicable in non-gene-transformable strains. Alternatively, stimulating PHB levels by chemical exposure is desirable because it might be applied to various cyanobacterial strains. However, the study of such chemicals is still limited. Here, 19 compounds previously reported to affect bacterial cellular processes were evaluated for their

β-Hydroxybutyrate Maintains Energetically Demanding Neural Functions During Exogenous Glucose Deprivation.

Ketone bodies are a major source of cerebral energy during fasting and the ketogenic diet, but whether they can sustain brain tissue metabolism when glucose is absent remains uncertain. This question is difficult to resolve in vivo because circulating glucose is maintained, even during starvation, through endogenous production. We therefore used an ex vivo brain preparation to examine the metabolic capacity of tissue following incubation with β-hydroxybutyrate (BHB) as the only exogenous fuel, isolating brain tissue from its primary endogenous glucose sources. Mitochondrial respiration was mon

A review of capillary β-hydroxybutyrate cut-off levels for the exclusion and identification of diabetic ketoacidosis.

AIMS/INTRODUCTION: Diabetic ketoacidosis (DKA) is an acute, life-threatening emergency that can arise as a complication of diabetes mellitus. We sought to examine the literature regarding capillary β-OHB testing levels that can inform cut-offs and help clinicians to safely and effectively exclude or identify DKA. MATERIALS AND METHODS: For this narrative review with a systematic approach, MEDLINE, Embase, and Emcare databases were searched for primary studies. Included studies described capillary β-OHB levels in relation to suspected DKA, for populations aged ≥16 years. Exclusion criteria wer

β-hydroxybutyrate as an early predictive biomarker of diabetes remission following intensive dietary intervention.

PURPOSE: Although intensive weight management achieves considerable success in diabetes remission, individual response varies substantially. We aimed to evaluate the predictive ability of early β-hydroxybutyrate (BHB) for type 2 diabetes (T2D) remission following an intensive dietary intervention in Southeast Asian individuals, and explore the metabolomic signatures associated with remission. METHODS: We analysed 45 adults with obesity and T2D (age:42 ± 10 years, 66.7% male, body mass index:33.7 ± 4.2 kg/m²) who completed 12 weeks of total diet replacement (TDR; ~800 kcal/day) in a secondary

β-Hydroxybutyrate-induced mitochondrial dysfunction and oxidative stress in BMSCs are ameliorated by mitophagy activation.

Ketosis is a common metabolic disorder in periparturient dairy cows and is characterized by elevated circulating BHBA concentrations. Although the effects of ketosis on hepatic metabolism have been extensively studied, its impact on skeletal muscle remains poorly understood. This study investigated the effects of BHBA on bovine muscle satellite cells (BMSCs) and the role of mitochondrial quality control in BHBA-induced cellular injury. BHBA treatment significantly inhibited BMSC proliferation, promoted apoptosis, increased intracellular and mitochondrial ROS accumulation, reduced antioxidant e

β-Hydroxybutyrate Attenuates Diabetic Kidney Disease Partially Via β-hydroxybutyrylation of Nrf2.

Diabetic kidney disease (DKD) is one of the predominant microvascular complications in diabetes. In recent years, the nuclear factor E2-related factor 2 (Nrf2)-mediated anti-oxidative stress pathway is tightly regulated by multiple post-translational modifications, governing its role in countering oxidative stress and DKD pathogenesis. Recent studies have found that the principal constituent of ketone bodies, β-hydroxybutyrate (β-HB), not only serves as an alternative energy source, but also induced lysine β-hydroxybutyrylation (Kbhb), which has various unknown pathophysiological functions. A direct comparison of butyrate, 4-phenylbutyrate, and β-hydroxybutyrate in an in vitro tumor therapy model.

BACKGROUND: Cancer remains the second leading cause of death worldwide, emphasizing the urgent need for more effective therapies. One promising approach is the use of naturally occurring molecules. The short-chain fatty acid butyrate has attracted attention for its protective and anticancer properties in colorectal cancer. However, the beneficial effects of butyrate are restricted to colonocytes. To determine whether systemically available butyrate derivatives could elicit similar effects in other cell types, we investigated 4-phenylbutyrate and β-hydroxybutyrate treatment regimens in a non-co
