[The effects of isoflurane and enflurane on cerebral hemodynamics and cerebral oxygen consumption in humans].

The effects of isoflurane (1 MAC) and enflurane (1 MAC) on cerebral blood flow and cerebral oxygen consumption were studied in 20 male patients without intracranial disease undergoing coronary artery bypass surgery (mean age 57 and 59 years respectively). The aim of the study was to investigate whether both agents diminish autoregulation of cerebral blood flow and CO2 reactivity of cerebral blood vessels. Patients were randomly assigned to one of two groups (10 patients each) receiving either isoflurane 1.15 vol.% or enflurane 1.68 vol.% endexpiratory. Measurements were performed and blood sam

Hepatic centrilobular necrosis in rats after exposure to halothane, enflurane, or isoflurane.

Exposure of phenobarbital-pretreated rats to low concentrations of halothane (0.5%) and reduced oxygen tension (FIO2 0.08) resulted in the development of liver necrosis in 51% of the animals. Fasting of rats for 24 hours before the same type of exposure increased the incidence of liver necrosis to 80%. Exposure of fed rats to enflurane (1.5%) and isoflurane (1.4%) in conjunction with low oxygen tensions resulted in no liver necrosis; however, in fasting animals, these same concentrations, when accompanied by low oxygen concentrations, produced an incidence of liver necrosis of 35% and 80%, res

Brain-penetrant microtubule-stabilizer epothilone B delays isoflurane-induced unconsciousness in mice.

Inhalational anesthetics are currently believed to cause unconsciousness by acting on multiple molecular targets including neural ion channels, receptors, mitochondria, synaptic proteins, and cytoskeletal proteins. Inhalational anesthetics including isoflurane bind to cytoskeletal microtubules (MTs), potentially contributing to causing unconsciousness. This possibility is supported by our demonstration of isoflurane resistance in rats treated once with the brain-penetrant MT-stabilizing drug epothilone B (epoB), and by a recent study in mice using a similar drug given daily over two weeks, whi

Isoflurane Postconditioning Alleviates Ischemic Neuronal Injury Via MiR-384-5p Regulated Autophagy.

The purpose of the study was to investigate the effect of isoflurane postconditioning on neuron injury in MCAO (middle cerebral artery occlusion) rats and its molecular mechanism of affecting autophagy through miR-384-5p/ATG5 (autophagy-related protein 5). HT22 cells (mouse hippocampal neuronal cell line) were exposed to 1.5% isoflurane for 30 min after OGD/R (oxygen-glucose deprivation/reoxygenation). Flow cytometry and CCK-8 kit were used to analyze changes in apoptosis and cell viability. The level of miR-384-5p was detected by qRT-PCR. Targetscan database prediction combined with dual luci

High Glucose Enhances Isoflurane-Induced Neurotoxicity by Regulating TRPC-Dependent Calcium Influx.

Isoflurane is a commonly used inhalational anesthetic that can induce neurotoxicity via elevating cytosolic calcium (Ca2+). High glucose regulates the expression of a family of non-selective cation channels termed transient receptor potential canonical (TRPC) channels that may contribute to Ca2+ influx. In the present study, we investigated whether high glucose enhances isoflurane-induced neurotoxicity by regulating TRPC-dependent Ca2+ influx. First, we evaluated toxic damage in mice primary cultured hippocampal neurons and human neuroblastoma cells (SH-SY5Y cells) after hyperglycemia and isof

Xenon and isoflurane improved biventricular function during right ventricular ischemia and reperfusion.

BACKGROUND: Although anesthetics have some cardioprotective properties, these benefits are often counterbalanced by their negative inotropic effects. Xenon, on the other hand, does not influence myocardial contractility. Thus, xenon may be a superior treatment for the maintenance of global hemodynamics, especially during right ventricular ischemia, which is generally characterized by a high acute complication rate. METHODS: The effects of 70 vol% xenon and 0.9 vol% isoflurane on biventricular function were assessed in a porcine model (n=36) using the conductance catheter technique, and the ex

Intracellular mechanism of mitochondrial adenosine triphosphate-sensitive potassium channel activation with isoflurane.

UNLABELLED: The precise mechanism of isoflurane and mitochondrial adenosine triphosphate-sensitive potassium channel (mitoK(ATP)) interaction is still unclear, although the mitoK(ATP) is involved in isoflurane-induced preconditioning. We examined the role of various intracellular signaling systems in mitoK(ATP) activation with isoflurane. Mitochondrial flavoprotein fluorescence (MFF) was measured to quantify mitoK(ATP) activity in guinea pig cardiomyocytes. To confirm isoflurane-induced MFF, cells were exposed to Tyrode's solution containing either isoflurane (1.0 +/- 0.1 mM) or diazoxide and

Propofol infusion vs thiopentone/isoflurane anaesthesia for prominent ear correction in children.

Postoperative nausea and vomiting (PONV) frequently follows prominent ear correction under general anaesthesia in children. In a prospective, single-blind study, we compared the incidence of PONV after propofol infusion anaesthesia with that following thiopentone induction and isoflurane maintenance in 30 children aged from four to 14 years randomly allocated to one of two groups. All the children were mechanically ventilated. Anaesthesia was supplemented in both groups with nitrous oxide and infiltration of the ears using a mixture of bupivacaine, adrenaline, and hyaluronidase. One child rece

Somatosensory evoked potential surgical monitoring. Observations during combined isoflurane-nitrous oxide anesthesia.

Somatosensory evoked potentials are used to monitor the integrity of the of the spinal cord during surgical procedures where the spinal cord is at risk. Changes in somatosensory evoked potential latencies and amplitudes can indicate impending structural damage. This use of somatosensory evoked potentials is complicated by the effects of nonsurgical factors, including anesthetics. Fifty patients were studied during surgery to determine the effects of coadministered isoflurane and nitrous oxide on cortical and subcortical somatosensory evoked potentials potentials following stimulation of the ti

Effects of isoflurane on feline intestinal blood flow during hemorrhage-induced hypovolemia.

The influence of isoflurane on intestinal reflex vasoconstriction during hemorrhage was investigated in cats (n = 10) during basal chloralose-nitrous oxide anesthesia. Intestinal blood flow (IBF) was studied in a model with controllable intestinal perfusion pressures to exclude local myogenic vascular responses related to changes in intraluminal pressure. A jejunal segment, which was dissected free in situ, was perfused via an extracorporeal arterial circuit which included a roller pump and a variable arterio-venous shunt. Intestinal perfusion pressure was controlled by adjusting the shunt flo

Quantification of Isoflurane Uptake for Immobilization of Ostrich Embryos for Preclinical In Ovo Imaging.

Preclinical imaging has recently been expanded through the use of ostrich embryos as an alternative in vivo model. In ovo experiments represent a promising substitute for conventional rodent-based animal testing. For artifact-free dynamic nuclear medicine imaging, reliable immobilization of embryos is essential. Although previous studies have demonstrated the feasibility of isoflurane anesthesia, the kinetics and uptake mechanisms of isoflurane in ostrich embryos remain insufficiently characterized. The aim of this study was to characterize gas exchange dynamics in ostrich eggs and to quantify

The Janus Face of Isoflurane: A Dose-Age-Response Model of Cognitive Impact.

Isoflurane remains a cornerstone of clinical anaesthesia, valued for its robust pharmacological profile. However, its long-term neurological impact, specifically its potential to trigger cognitive impairment, remains a subject of intense debate. As a highly lipophilic agent that readily penetrates the blood-brain barrier, isoflurane exerts complex modulatory effects on various neurotransmitter systems and signaling cascades. While traditional paradigms have predominantly focused on isoflurane-induced neurotoxicity, characterised by neuronal apoptosis, oxidative stress and neuroinflammation, em

Oxidative stress, inflammation and the metabolic EpOME are associated with occupational exposure to isoflurane.

This study assessed the occupational exposure to the waste anesthetic gas (WAG) isoflurane in veterinarians focusing on oxidative stress and inflammatory markers in addition to metabolomic profiling. This was a case-control study with matched individuals related to dosimeter data for WAG. Systemic oxidative stress markers (malondialdehyde, 8-iso-prostaglandin F2α and carbonylated proteins) were screened, as well as carbonylated protein degradation products and 8-hydroxydeoxyguanosine in urine. Cholesterol and fractions were also assessed. Antioxidant enzymes were evaluated in washed red blood

Isoflurane Exposure Influence on Sleep Architecture in a Rat Model of Alzheimer's Disease.

BACKGROUND: A clear understanding of how general anesthesia affects the brain in patients with Alzheimer disease will be crucial to optimize clinical care. We present results from laboratory investigations with a rat model used to study Alzheimer disease (TgF344-AD) to evaluate the effect of isoflurane anesthesia on the early recovery from anesthesia and postanesthesia sleep architecture in animals that exhibit mild symptoms of cognitive decline. METHODS: We used electroencephalogram (EEG) and electromyogram recordings to distinguish WAKE, NREM, and REM sleep in 8 rats from the transgenic mod

Comparison of Dexmedetomidine/Morphine and Xylazine/Morphine as Premedication in Isoflurane-Anesthetized Sheep.

In this study we investigated the sedative, anesthetic, and pulmonary histopathologic effects of dexmedetomidine/morphine (DM) and xylazine/morphine (XM) in sheep. We hypothesized that DM would provide profound sedation and better maintain physiologic parameters under anesthesia than XM in sheep undergoing laparoscopic surgery. Nineteen male sheep were premedicated with either DM (dexmedetomidine [0.006 mg/kg] and morphine [0.3 mg/kg]) or XM (xylazine [0.1 mg/kg] and morphine [0.3 mg/kg]). After DM or XM administration, 3 blinded veterinarians evaluated sedation scores (0 [no sedation], 1 [mil

TREK-1 and TREK-2 Knockout Mice Are Not Resistant to Halothane or Isoflurane.

BACKGROUND: A variety of molecular targets for volatile anesthetics have been suggested, including the anesthetic-sensitive potassium leak channel, TREK-1. Knockout of TREK-1 is reported to render mice resistant to volatile anesthetics, making TREK-1 channels compelling targets for anesthetic action. Spinal cord slices from mice, either wild type or an anesthetic- hypersensitive mutant, Ndufs4, display an isoflurane-induced outward potassium leak that correlates with their minimum alveolar concentrations and is blocked by norfluoxetine. The hypothesis was that TREK-1 channels conveyed this cur
