Measuring emotional memory in the elevated T-maze using a training-to-criterion procedure.

The elevated T-maze, an ethologically based test, has been used to investigate the effects of anxiolytic drugs on memory and the relationships between neural systems involved in such modulation. This test allows the measurement in the same rat of two kinds of aversively motivated behaviors--inhibitory avoidance and one-way escape. The apparatus consists of three arms of equal dimensions, elevated 50 cm from the floor. One arm is enclosed by walls and stands perpendicular to the two open arms. Placing the rat at the end of the enclosed arm and recording the time to withdraw from this arm during

Neuroinflammation increases GABAergic tone and impairs cognitive and motor function in hyperammonemia by increasing GAT-3 membrane expression. Reversal by sulforaphane by promoting M2 polarization of microglia.

BACKGROUND: Hyperammonemia induces neuroinflammation and increases GABAergic tone in the cerebellum which contributes to cognitive and motor impairment in hepatic encephalopathy (HE). The link between neuroinflammation and GABAergic tone remains unknown. New treatments reducing neuroinflammation and GABAergic tone could improve neurological impairment. The aims were, in hyperammonemic rats, to assess whether: (a) Enhancing endogenous anti-inflammatory mechanisms by sulforaphane treatment reduces neuroinflammation and restores learning and motor coordination. (b) Reduction of neuroinflammation

Retrograde amnesia and selective damage to the hippocampal formation: memory for places and object discriminations.

Using a within-subjects design, rats were trained on two place-memory problems and five object-discrimination problems at different intervals prior to receiving either ibotenate lesions of the hippocampal formation or sham surgery. Places # 1 and 2 were fixed-platform water-maze tasks that were run in different rooms and they were learned during the 14th and 2nd week before surgery, respectively. Object-discrimination problems # 1-5 were learned during the 13th, 10th, 7th, 4th, and 1st week before surgery, respectively. Rats with hippocampal lesions displayed impaired retention of both Place p

Medial septal lesions in rats produce permanent deficits for strategy selection in a spatial memory task.

Rats with medial septal (MS) lesions have been shown to consistently use a stereotypic response strategy rather than a nonstereotypic spatial learning strategy when solving a radial maze task. The present study examined the long-term effects of MS lesions on spatial memory performance to determine whether MS lesions permanently impair rats from using a nonstereotypic strategy. Male rats, initially trained on a radial maze, were given either MS or sham surgeries and were subsequently retested on the maze. Consistent with previous studies, all rats with MS lesions used a stereotypic strategy dur

The behavior of vasopressin-deficient rats (Brattleboro strain).

Seven Brattleboro rats homozygous for diabetes insipidus (DI) and seven normal Long-Evans (LE) rats were tested on a neuropsychological test battery comprised of the following tasks: time-spent-eating in two adaptation boxes, time-to-emerge into an open field, adaptation to a T-maze, contingently reinforced T-maze alternation, olfactory and visual discrimination, runway learning, approach-avoidance conflict, step-through passive avoidance, prod burying, and stress-induced interference. It was found that DI rats adapted more slowly than LE rats to novel environments (e.g., adaptation box, T-maz

(R)-Ketamine attenuates LPS-induced endotoxin-derived delirium through inhibition of neuroinflammation.

RATIONALE: (R)-Ketamine produced beneficial effects in a variety of models of inflammatory diseases, including low dose of bacterial lipopolysaccharide (LPS) (0.5-1.0 mg/kg)-induced endotoxemia. LPS-treated mice have been used as animal model of delirium. OBJECTIVES: We investigated the effects of (R)-ketamine in neuroinflammation and cognitive impairment in rodents after administration of high dose of LPS. METHODS: LPS (5 mg/kg) or saline was administered intraperitoneally (i.p.) to mice. (R)-Ketamine (10 mg/kg) was administrated i.p. 24 h before and/or 10 min after LPS injection. RESULTS:

Shallow water (paddling) variants of water maze tests in mice.

When Richard Morris devised his water maze in 1981(7), most behavioral work was done in rats. However, the greater understanding of mouse genetics led to the mouse becoming increasingly important. But researchers found that some strains of mutant mice were prone to problems like passively floating or diving when they were tested in the Morris water maze(11). This was unsurprising considering their natural habitat; rats swim naturally (classically, the "sewer rat"), whereas mice evolved in the dry areas of central Asia. To overcome these problems, it was considered whether shallow water would b

Cognitive dysfunction in schizophrenia: comparison of treatment with atypical antipsychotic agents and conventional neuroleptic drugs.

Impaired cognitive function is both a feature of schizophrenia and a side effect of conventional neuroleptics. Maze tests were used to determine the effects on cognition of conventional dopamine antagonist neuroleptics (haloperidol and fluphenazine) and the newer serotonin-dopamine antagonist antipsychotics (risperidone and clozapine). Patients on clozapine or risperidone showed better performance on the maze tasks than untreated patients or patients taking conventional neuroleptics. In particular, patients treated with risperidone or clozapine were better able to maintain motor coordination w

Chronic interleukin-6 mediated neuroinflammation decreases anxiety, and impaires spatial memory in aged female mice.

INTRODUCTION: Neuroinflammation is a common feature of many psychiatric disorders as well as a common underlying mechanism of neurodegenerative diseases. Sex has been shown to strongly influence the development as well as the clinical expression of these pathologies. However, there is still a neglect regarding the consideration of sex effects in rodent experiments, and a substantial underrepresentation of females in studies. This work set out to expand our knowledge of neuroinflammatory mechanisms in female mice, at both a behavioral and molecular level. METHODS: This study used GFAP-IL6 mice

Neonatal exposure to BDE 209 impaired learning and memory, decreased expression of hippocampal core SNAREs and synaptophysin in adult rats.

Polybrominated diphenyl ethers (PBDEs) are a class of flame retardants. While the mechanism remains unknown, the potential neurotoxic effects of PBDEs remain a relevant issue. In the present study, neonatal Sprague-Dawley rats of both sexes were administered BDE 209 (1, 10, or 20mg/kg body weight) or peanut oil once daily from postnatal day (PND) 5 to PND 10. We examined the spatial learning and memory by Morris water maze and the working and reference memory by eight-arm radial maze in the stage of adulthood. Compared with controls, significantly longer escape latencies and fewer platform-cro

Practical viewpoints on ethical questions and dilemmas in schema therapy.

OBJECTIVE: Methylmercuric chloride (MMC) has neurotoxicity, while hydrogen sulfide (H2S) has shown inhibitory properties against nerve damage induced by various factors. The study aimed to investigate the impact of H2S on MMC-induced learning and memory impairment in mice and to explore the underlying mechanisms. METHODS: A mouse model of learning and memory impairment was established by MMC gavage, and sodium hydrosulfide (NaHS) was used as an H2S donor for intervention. Cell viability and live/dead cell ratio in HT22 neuronal cells were assessed by CCK-8 assay and Calcein/PI staining, respe

Neonatal hippocampal damage in rats: long-term spatial memory deficits and associations with magnitude of hippocampal damage.

This study investigated the effects of neonatal hippocampal ablation on the development of spatial learning and memory abilities in rats. Newborn rats sustained bilateral electrolytic lesions of the hippocampus or were sham-operated on postnatal day 1 (PN1). At PN20-25, PN50-55, or PN90-95, separate groups of rats were tested in a Morris water maze on a visible "cue" condition (visible platform in a fixed location of the maze), a spatial "place" condition (submerged platform in a fixed location), or a no-contingency "random" condition (submerged platform in a random location). Rats were tested

Netrin-3 rectifies diabetes-induced cognitive impairment by counteracting hippocampal oxidative stress and NLRP3 inflammasome activation.

Diabetes frequently causes substantial cognitive decline, yet available treatments offer minimal benefit. Netrin-3, a laminin-like protein, regulates nervous system development and inflammatory responses. However, its involvement in diabetes-associated cognitive impairment remains poorly understood. The escalating global diabetes burden, combined with the absence of disease-modifying therapies for cognitive complications, creates an urgent need to identify novel molecular targets. Our research evaluated whether Netrin-3 could counter cognitive deterioration using db/db mice as a type 2 diabete

Focal lesions of human hippocampal CA1 neurons in transient global amnesia impair place memory.

A critical role in place learning has been attributed to place cells within the cornu ammonis 1 (CA1) sector of the hippocampus in rodents. The role of CA1 cells in the human hippocampus with regard to place learning remains elusive. Using a virtual Morris water maze, we investigated patients with acute transient global amnesia (TGA), a rare self-limiting dysfunction of the hippocampal system. Fourteen individuals with selective and focal lesions in the CA1 sector of the hippocampus showed a profound impairment in place learning. The size of the lesions and the duration of the TGA correlated w

[A physiological analysis of dissociative learning against a background of physostigmine and pentobarbital].

Physostigmine (0.7-0.8 mg/kg, i.p.) decreased and pentobarbital (13.4-14.6 mg/kg) increased the locomotor and emotional activity of rats in the "open field". Both drugs induced the reversible amnesia to a conditioned reaction in a double T-maze with positive (nutritional) reinforcement. These changes in behavioral activity were correlated with dissociated learning of rats after the injection of the drugs: physostigmine largely decreased the number of errors during learning as compared with pentobarbital. However, in both cases rats reached the learning criterion sooner than the control animals

Influence of different levels of neonatal mild/moderate hypoxia-ischemia in learning abilities of rats at the age of one month.

We studied the effects of mild/moderate hypoxia-ischemia in 90 newborn Wistar rats divided into 3 groups. Two of the three groups were submitted to two different levels of hypoxia (FiO2 = 0.05, group F5 and FiO2 = 0.1, group F10) and the third to normoxia (FiO2 = 0.21, group F21) in a thermoneutral and controlled environment. We examined their influence in a maze test 1 month later after two training trials, a 48-hour fasting period and handling stress just before starting the trial. The learning ability in animals exposed to FiO2 = 0.1 was lower (significantly higher number of squares crossed

Mice deficient for the synaptic vesicle protein Rab3a show impaired spatial reversal learning and increased explorative activity but none of the behavioral changes shown by mice deficient for the Rab3a regulator Gdi1.

Rab proteins are small GTPases involved in intracellular trafficking. Among the 60 different Rab proteins described in mammals, Rab3a is the most abundant in brain, where it is involved in synaptic vesicle fusion and neurotransmitter release. Rab3a constitutive knockout mice (Rab3a(-/-)) are characterized by deficient short- and long-term synaptic plasticity in the mossy fiber pathway and altered circadian motor activity, while no effects on spatial learning have been reported so far for these mice. The goals of this study were to analyse possible behavioral consequences of the lack of synapti

Working Memory Y-Maze Test for Xenopus Tadpoles.

Working memory refers to the multifaceted cognitive systems that are considered necessary for retaining information while performing complex tasks, such as reasoning and learning in humans. In vertebrates, working memory is evident as a conserved alternating searching strategy. We can use automated tracking tools to measure this search strategy in Xenopus tadpoles. The free-movement pattern Y-maze assay is validated for assessing spatial working memory and cognitive flexibility in Xenopus. Deviations from randomness in search strategies are quantified by comparing relative alternating and repe

Involvement of dopamine D1 receptors of the hippocampal dentate gyrus in spatial learning and memory deficits in a rat model of vascular dementia.

We investigated the involvement of dopamine (DA) and its D1 receptors of the hippocampal dentate gyrus (DG) in spatial learning and memory deficits in a rat model of vascular dementia (VD) established by permanent bilateral carotid occlusion. Spatial learning and memory abilities of rats were measured by Morris water maze, and extracellular concentrations of DA in the DG were determined by in vivo microdialysis. The DA concentrations in the DG decreased in the VD rats compared with sham-operated group. Microinjection of SFK38393 (D1 receptor agonist) into the DG attenuates spatial learning and

Decreased investigatory head scanning during exploration in learning-impaired, aged rats.

"Head scanning" is an investigatory behavior that has been linked to spatial exploration and the one-trial formation or strengthening of place cells in the hippocampus. Previous studies have demonstrated that a subset of aged rats with normal spatial learning performance show head scanning rates during a novel, local-global cue-mismatch manipulation that are similar to those of young rats. However, these aged rats demonstrated different patterns of expression of neural activity markers in brain regions associated with spatial learning, perhaps suggesting neural mechanisms that compensate for a
