The dynamic change of flavor characteristics in Pacific oyster (Crassostrea gigas) during depuration uncovered by mass spectrometry-based metabolomics combined with gas chromatography-ion mobility spectrometry (GC-IMS).

The flavor of Pacific oyster (Crassostrea gigas) significantly changed during the depuration process. This work aimed to explore the mechanism of flavor changes during the 72 h depuration by metabolomics combined with gas chromatography-ion mobility spectrometry (GC-IMS). The metabolomics analysis indicated that carbohydrate metabolism was more affected in the early stage of depuration, including the citrate cycle, glyoxylae and dicarboxylate metabolism, etc. After 72 h depuration, it affected mainly the metabolism of global and overview maps and nucleoside metabolism, etc. The equivalent umam

Associations of mortality with leisure-time physical activity and sitting time in postmenopausal women: Insights from NHANES.

OBJECTIVE: Postmenopausal women are vulnerable to insufficient physical activity and prolonged sitting. While both behaviors have been individually associated with mortality, their joint effects remain unclear. METHODS: We analyzed 5355 postmenopausal women (median age 63 years [interquartile range 56-71]) from the National Health and Nutrition Examination Survey (NHANES) 2007-2018. Leisure-time physical activity was categorized as insufficient (<600 metabolic equivalent of task minutes per week) or sufficient (≥600 metabolic equivalent of task minutes per week). Sitting time was classified a

Assessment of safety through mutagenicity and subchronic toxicity studies with black pepper extract preparation.

INTRODUCTION: Black pepper (Piper nigrum) is a rich source of natural and bioactive components such as N-trans-feruloyltyramine (NFT). In this paper, we discuss the results of the subchronic toxicity and mutagenicity studies conducted to understand the potential for adverse effects if any, of Black Pepper Extract Preparation (BPE). METHODS: To evaluate mutagenicity, an Ames test was conducted with BPE in the presence and absence of S9 metabolic activation. Long-term safety was inferred through a 90-day subchronic toxicology study using adult rats. Dose levels were selected with expected human

Does adding physical activity improve spinal bone mineral density in postmenopausal women?: A cross-sectional analysis of NHANES 2011 to 2018.

Physical activity (PA) is recommended for managing postmenopausal osteoporosis, yet evidence-based prescriptions for optimizing bone mineral density (BMD) remain undefined. To identify beneficial PA thresholds, this study examined the relationship between PA and total lumbar BMD in a cross-sectional analysis of 9339 postmenopausal women from the National Health and Nutrition Examination Survey 2011 to 2018. PA was quantified as Physical Activity Metabolic Equivalent of Task (PA-MET)-hours/week. The association between PA-MET-hours/week and total lumbar BMD was assessed using weighted multivari

Oxaliplatin-induced cardiotoxicity in mice is connected to the changes in energy metabolism in the heart tissue.

Oxaliplatin is a platinum-based alkylating chemotherapeutic agent used for cancer treatment. At high cumulative dosage, the negative effect of oxaliplatin on the heart becomes evident and is linked to a growing number of clinical reports. The aim of this study was to determine how chronic oxaliplatin treatment causes the changes in energy-related metabolic activity in the heart that leads to cardiotoxicity and heart damage in mice. C57BL/6 male mice were treated with a human equivalent dosage of intraperitoneal oxaliplatin (0 and 10 mg/kg) once a week for eight weeks. During the treatment, mic

Comparable Metabolic and Histopathological Observations of Enzymatic and Non-Enzymatic Thai Shallot Extracts in High-Fat Diet-Induced Obese Mice.

Thai shallot (Allium cepa L. var. aggregatum) is rich in quercetin glycosides, which can be enzymatically hydrolyzed into aglycone forms with potentially higher bioavailability. However, whether this structural modification enhances metabolic efficacy in vivo remains unclear. This study aimed to compare the metabolic and histopathological effects of enzymatic (ESE) and non-enzymatic (NES) Thai shallot extracts in a high-fat diet (HFD)-induced obese mouse model. Male C57BL/6 mice were fed HFD for 12 weeks to induce obesity, followed by a 12-week treatment with NES or ESE (1000 and 2000 mg/kg/da

Do metabolic responses to solar radiation scale directly with intensity of irradiance?

Endotherms exposed to air temperatures below thermal neutrality reduce their metabolic heat production when exposed to sunlight. The physiological effects of this additional source of heat gain from the environment usually are assumed to be proportional to the intensity of irradiance if other factors are held constant. We test this assumption by measuring changes in metabolic heat production produced by exposing a small mammal, the Siberian hamster (Phodopus sungorus) to four intensities of simulated solar radiation (0 W m-2, 317 W m-2, 634 W m-2 and 950 W m-2). In the absence of solar radiati

Atypical energy-related symptoms define biologically distinct subtypes of major depressive disorder.

Major depressive disorder (MDD) is a complex psychiatric disorder, characterized by a range of mood, cognitive, and neurovegetative symptoms. Current diagnostic criteria treat opposite symptom directions as equivalent; weight gain or loss, and increased or decreased sleep, each count toward a single diagnosis. We defined three subgroups of individuals meeting criteria for MDD: AERS+ (hypersomnia with increased appetite/weight gain), AERS- (insomnia with appetite/weight loss), and an Uncategorized group, and conducted genome-wide association meta-analyses for each (Neff = 47,858, 156,624, and 2

Dual impact of microplastic exposure in a mouse model: Impaired uterine receptivity and altered maternal-offspring metabolism.

With the increasing environmental presence of microplastics (MPs), concerns about their potential health risks have intensified. Our previous study detected MP deposition in the human uterus. However, its impact on uterine receptivity and the underlying mechanisms remains poorly understood, and the systemic metabolic consequences of MP exposure are yet to be elucidated. In this study, we investigated the reproductive and metabolic consequences of MP exposure using a mouse model. Female C57BL/6 mice were orally administered polystyrene microplastics (PS-MPs) at 50 mg/kg/day for 14 days (n = 30

Thermoneutrality Accelerates a Distinctive Metabolic Dysfunction-Associated Steatohepatitis Phenotype in a Diet-Induced Male Mouse Model With Similarities to Human Disease.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatohepatitis is a prevalent chronic liver disease characterized by steatosis, inflammation, and hepatocyte damage, with or without fibrosis. Robust preclinical models that represent the clinical features of metabolic dysfunction-associated steatohepatitis are critical for drug development. We aimed to examine the effects of 2 widely used Western-type diets and thermoneutral housing on the induction of metabolic dysfunction-associated steatohepatitis and liver fibrosis in mice. METHODS: Male mice were fed either a Gubra-Amylin metabolic dy

Development and qualification of 3 L scale-down model for large scale vaccine process on Vero cell culture using microcarriers.

Scale-down models (SDM) are pivotal tools for process understanding and improvement to accelerate the development of vaccines from laboratory research to global commercialization. In this study, a 3 L SDM representing a 50 L scale Vero cell culture process of a live-attenuated virus vaccine using microcarriers was developed and qualified based on the constant impeller power per volume principle. Both multivariate data analysis (MVDA) and the traditional univariate data analysis showed comparable and equivalent cell growth, metabolic activity, and product quality results across scales. Genetic variants in phosphatidylethanolamine N-methyltransferase and methylenetetrahydrofolate dehydrogenase influence biomarkers of choline metabolism when folate intake is restricted.

Choline is a required nutrient with roles in liver and brain function, lipid metabolism, and fetal development. Recent data suggest that choline requirements may be altered by polymorphisms in the phosphatidylethanolamine N-methyltransferase (PEMT) gene (ie, 5465G-->A; rs7946 and -744G-->C; rs12325817) and in the methylenetetrahydrofolate dehydrogenase (MTHFD1) gene (ie, 1958G-->A; rs2236225). This controlled feeding study, conducted in 2000-2001, examined the effects of the PEMT and MTHFD1 genetic variants on biomarkers of choline metabolism in premenopausal Mexican-American women (N=43) afte

Bone marrow and renal injury associated with haloalkene cysteine conjugates in calves.

Almost 40 years ago, it was reported that cattle-feed which had been extracted with hot trichloroethylene and then fed to calves produced renal injury and a fatal aplastic anaemia. The toxic factor was subsequently identified as S-(1,2-dichlorovinyl)-L-cysteine (DCVC). These original findings have been confirmed, a single intravenous dose of DCVC at 4 mg/kg, or 0.4 mg/kg intravenously per day administered for 10 days to calves produced aplastic anaemia, and renal injury after a single dose of 4 mg/kg. The toxicity to calves of a number of other haloalkene cysteine conjugates has been examined

Some physiological and metabolic effects of sodium nitroprusside and cyanide in the dog.

The cardiovascular and acid-base changes following equivalent i.v. bolus doses of sodium nitroprusside (SNP) and potassium cyanide (KCN) have been studied in two groups of anaesthetized dogs. In a third group, the metabolic changes produced by i.v. infusion of SNP 1.5 mg kg-1 at a constant rate over 1 h have been studied. In contrast to a decrease in arterial pressure following SNP, hypertension and tachycardia occurred after the administration of KCN, with hyperventilation and an increase in packed cell volume. During infusion of SNP, increases in plasma cyanide concentrations were associated
