Effect of Lidocaine on Quinine-Induced Tinnitus in Guinea Pigs: A Focus on the Auditory Cortex.

HYPOTHESIS: This study investigated whether intravenous lidocaine reverses auditory cortex changes induced by systemic quinine administration. BACKGROUND: Quinine, like salicylate, is known to induce tinnitus. Previous studies have shown that salicylate-induced changes in the auditory cortex can be reversed by lidocaine, a compound known to suppress tinnitus. If lidocaine also reverses quinine-induced changes, it would further support the association between these neural changes and the presence of tinnitus. METHODS: Healthy male albino Hartley guinea pigs received systemic quinine, followed

Quinine sulfate as a therapeutic option in a patient with slow channel congenital myasthenic syndrome.

Slow channel congenital myasthenic syndrome is caused by a genetically determined kinetic anomaly of the acetylcholine receptor at the neuromuscular junction leading to its prolonged open state. Patients typically present with fatigability and static weakness of neck, hand and finger extensors. The open-channel blockers fluoxetine and quinidine have been used as standard treatment, although the former is limited by its side effects. We describe a patient with a novel "de novo" mutation in the α subunit of acetylcholine receptor with clinical and electrophysiological hallmarks of the disease. T

Amplification of quinine cardiac effects by the resistance-reversing agent prochlorperazine in falciparum malaria.

The use of reversing agents to overcome drug resistance is a potential new treatment strategy for both malaria and cancer. Laboratory studies have raised questions about the safety of this therapeutic approach, but data in humans are lacking. We therefore assessed the toxic potential of reversing agent therapy in Thai patients receiving quinine (17 mg/kg given over 4.5 hr) for falciparum malaria by serial measurements of the QTc interval, an electrocardiographic (ECG) marker of the effect of quinine. Six patients were randomly assigned to receive intravenous quinine alone while another six rec

Comparison of artemisinin suppositories with intravenous artesunate and intravenous quinine in the treatment of cerebral malaria.

Seventy-nine comatose cerebral malaria patients given standard supportive treatment were randomized to receive specific antimalarial chemotherapy of intravenous quinine, intravenous artesunate, or artemisinin suppositories. Artesunate and artemisinin reduced peripheral asexual parasitaemia significantly more rapidly than quinine (90% clearance time 16 h, 18.9 h and 34.5 h respectively), but did not significantly reduce the duration of coma or mortality. The rapid lowering of peripheral parasitaemia may not ameliorate complications already present. These results demonstrate that artemisinin sup

Altered spontaneous and osmotically induced drinking for fowls with permanent access to dilute quinine.

Fowls were given a dilute quinine solution as their sole source of fluid, and effects on normal ingestion and on drinking responses to dipsogenic stimuli were examined. Compared to controls with water, daily fluid intakes were depressed by 25% with quinine. There was no significant effect of quinine on food intake, but growth was suppressed slightly. Drinking in response to hypertonic NaCl injections was attenuated with quinine, and whereas an initial peak in water intake was seen directly after hypertonic NaCl injection, this was absent with quinine. Moreover, increases in quinine intake afte

The effect of mefloquine-sulfadoxine-pyrimethamine vs quinine on patients with complicated falciparum malaria.

Sixty-six patients with complicated falciparum malaria (defined as anaemia, hyperpyrexia, jaundice, or more than 2% of RBC parasitised) were studied. Patients with cerebral signs and symptoms were not included in the study. Patients were randomised in pairs to receive either mefloquine 750 mg, sulfadoxine 1500 mg and pyrimethamine 75 mg (MSP) single oral dose or quinine (10 mg/kg tds X 7 days oral therapy). All the patients were admitted in hospital for 7 days and were followed on days 14, 21 and 28. All patients survived. The parasite clearance times in MSP treated patients were significantly

Saccharin- or quinine-induced changes in the rat pups following prolonged ingestion by the dam.

Effects presumably induced by a chronic free ingestion of saccharin (0.40 mg/ml) or of quinine (0.25 mg/ml) by female rats during the pregestative, gestative and lactating periods were investigated in the ensuing progeny. Preweaned pups were studied from birth up to weaning (21 days of age) by means of selected gross behavioral tests. The saccharin litters were mainly characterized by a slowering in the body growth evolution. The quinine pups demonstrate several physical anomalies: (1) congenital malformations in 5% of the animals; (2) a significantly reduced birth weight followed by a persist

Adsorption dynamics of dipeptides in a column with chiral zwitterionic quinine-based stationary phase.

Adsorption-desorption kinetics in chromatographic columns is difficult to study by the moment method because of interference with other mass transfer processes. The Eberly-Spencer method provides another way to determine the desorption rate constant. A combination of these techniques was applied to study the adsorption dynamics of almost unretained (Gly-Gly) and retained (L(D)-Leu-L(D)-Leu and L(D)-Leu-L(D)-Phe) dipeptides in a column packed with chiral stationary phase bearing quinine-l-Ala-l-Leu (AK 57) as a chiral selector. It was shown that eddy diffusion and slow adsorption-desorption kin

Regulation of Ras p21 and RalA GTPases activity by quinine in mammary epithelial cells.

Quinine, a bitter compound, can act as an agonist to activate the family of bitter taste G protein-coupled receptor family of proteins. Previous work from our laboratory has demonstrated that quinine causes activation of RalA, a Ras p21-related small G protein. Ral proteins can be activated directly or indirectly through an alternative pathway that requires Ras p21 activation resulting in the recruitment of RalGDS, a guanine nucleotide exchange factor for Ral. Using normal mammary epithelial (MCF-10A) and non-invasive mammary epithelial (MCF-7) cell lines, we investigated the effect of quinine

Effective Treatment of Cutaneous Mold Infections by Antimicrobial Blue Light That Is Potentiated by Quinine.

BACKGROUND: Cutaneous mold infections commonly result from an array of traumatic injuries that involve direct inoculation of contaminated soil into wounds. Here, we explored the use of antimicrobial blue light (aBL; 405 nm wavelength) and the combination of aBL with quinine hydrochloride (aBL + Q-HCL) for the treatment of cutaneous mold infections. METHODS: Efficacy of aBL and aBL + Q-HCL in killing clinically important pathogenic molds (Aspergillus fumigatus, Aspergillus flavus, and Fusarium oxyprorum) was investigated. Ultraperformance liquid chromatography identified and quantified endogen

Effect of quinine and artesunate combination therapy on platelet count of children with severe malaria.

BACKGROUND: There are several case reports of quinine-induced thrombocytopenia but no clinical trials to ascertain its incidence and significance in severe malaria. OBJECTIVES: The primary objective was to assess the effect of quinine on the platelet count in children with severe malaria and to compare it with artesunate combination therapy (ACT), and the secondary objective was to assess outcome of treatment with quinine and ACT. METHODS: An open-labelled, randomised, controlled trial was undertaken in 100 children aged 6 months to 12 years who were diagnosed with malaria by microscopy and/

Renal-limited thrombotic microangiopathy and acute interstitial nephritis with a single dose of quinine.

Quinine has been reported to cause acute kidney injury by various mechanisms. The response to quinine can result in a spectrum of problems ranging from isolated thrombocytopenia to thrombotic microangiopathy (TMA) to disseminated intravascular coagulation. Quinine has also been reported to cause acute interstitial nephritis (AIN). We report an unusual presentation where both of these entities of renal-limited TMA and AIN were precipitated by a single dose of quinine.

Quinine for chloroquine-resistant falciparum malaria in pregnant Sudanese women in the first trimester.

A prospective clinical study in eastern Sudan described the efficacy and toxicity of quinine in early pregnancy in mothers with chloroquine-resistant falciparum malaria. Twenty-six pregnant Sudanese women in their first trimester (mean gestational age 8.5 weeks) were given quinine 10 mg/kg 3 times per day for 7 days and followed up every 2 weeks until delivery. One patient aborted (3.8%) and 2 patients (7.7%) experienced threatened abortion but delivered term babies. Recrudescence or re-infection was observed on day 21 in 1 patient. One baby died aged 6 months. There were no detectable congeni
