Sodium channels Nav1.7, Nav1.8 and pain; two distinct mechanisms for Nav1.7 null analgesia.

Genetic deletion and pharmacological inhibition are distinct approaches to unravelling pain mechanisms, identifying targets and developing new analgesics. Both approaches have been applied to the voltage-gated sodium channels Nav1.7 and Nav1.8. Genetic deletion of Nav1.8 in mice leads to a loss of pain and antagonists are effective analgesics. The situation with Nav1.7 is more complex. Complete embryonic loss of Nav1.7 in humans or in mouse sensory neurons leads to anosmia as well as profound analgesia as a result of diminished neurotransmitter release. This is mediated by enhanced endogenous

Spider and scorpion knottins targeting voltage-gated sodium ion channels in pain signaling.

In sensory neurons that transmit pain signals, whether acute or chronic, voltage-gated sodium channels (VGSCs) are crucial for regulating excitability. NaV1.1, NaV1.3, NaV1.6, NaV1.7, NaV1.8, and NaV1.9 have been demonstrated and defined their functional roles in pain signaling based on their biophysical properties and distinct patterns of expression in each subtype of sensory neurons. Scorpions and spiders are traditional Chinese medicinal materials, belonging to the arachnid class. Most of the studied species of them have evolved venom peptides that exhibit a wide variety of knottins specifi

Phenotypes of mutations related to voltage-dependent sodium channels on children and adolescents.

Cardiac channelopathies are a heterogeneous group of inherited cardiac diseases that are associated with mutations in the genes that encode the expression of cardiac ion channels. In view of this, it can be mentioned that the main hereditary arrhythmias in children and adolescents, caused by dysfunction of the ion channels, are Brugada Syndrome (BrS) and Long QT Syndrome (LQTS). However, few studies address the physiological effects of these conditions on children and adolescents. Thus, the aim of this study is to describe the mutation phenotype related to voltage-gated sodium channels in chil

Dexmedetomidine Inhibits Voltage-Gated Sodium Channels via α2-Adrenoceptors in Trigeminal Ganglion Neurons.

Dexmedetomidine, an α2-adrenoceptor agonist, is widely used as a sedative and analgesic agent in a number of clinical applications. However, little is known about the mechanism by which it exerts its analgesic effects on the trigeminal system. Two types of voltage-gated sodium channels, Nav1.7 and Nav1.8, as well as α2-adrenoceptors are expressed in primary sensory neurons of the trigeminal ganglion (TG). Using whole-cell patch-clamp recordings, we investigated the effects of dexmedetomidine on voltage-gated sodium channel currents (INa) via α2-adrenoceptors in dissociated, small-sized TG neur

Sodium channels, the electrogenisome and the electrogenistat: lessons and questions from the clinic.

In the six decades that have followed the work of Hodgkin and Huxley, multiple generations of neuroscientists and biophysicists have built upon their pivotal contributions. It is now clear that, in mammals, nine genes encode nine distinct voltage-gated sodium channels with different amino acid sequences and different physiological and pharmacological properties. The different sodium channel isoforms produce a multiplicity of distinct sodium currents with different time-dependent characteristics and voltage dependencies, which interact with each other and with the currents produced by other cha

Leaky sodium channels from voltage sensor mutations in periodic paralysis, but not paramyotonia.

BACKGROUND: Hypokalemic periodic paralysis (HypoPP) is associated with mutations in either the Ca(V)1.1 calcium channel or the Na(V)1.4 sodium channel. Some Na(V)1.4 HypoPP mutations have been shown to cause an anomalous inward current that may contribute to the attacks of paralysis. Herein, we test whether disease-associated Na(V)1.4 mutations in previously untested homologous regions of the channel also give rise to the anomalous current. METHODS: The functional properties of mutant Na(V)1.4 channels were studied with voltage-clamp techniques in an oocyte expression system. RESULTS: The Hy

Involvement of voltage-gated sodium channels blockade in the analgesic effects of orphenadrine.

Orphenadrine is a drug acting on multiple targets, including muscarinic, histaminic, and NMDA receptors. It is used in the treatment of Parkinson's disease and in musculoskeletal disorders. It is also used as an analgesic, although its mechanism of action is still unknown. Both physiological and pharmacological results have demonstrated a critical role for voltage-gated sodium channels in many types of chronic pain syndromes. We tested the hypothesis that orphenadrine may block voltage-gated sodium channels. By using patch-clamp experiments, we evaluated the effects of the drug on whole-cell s

Antillatoxin is a marine cyanobacterial toxin that potently activates voltage-gated sodium channels.

Antillatoxin (ATX) is a lipopeptide derived from the pantropical marine cyanobacterium Lyngbya majuscula. ATX is neurotoxic in primary cultures of rat cerebellar granule cells, and this neuronal death is prevented by either N-methyl-d-aspartate (NMDA) receptor antagonists or tetrodotoxin. To further explore the potential interaction of ATX with voltage-gated sodium channels, we assessed the influence of tetrodotoxin on ATX-induced Ca2+ influx in cerebellar granule cells. The rapid increase in intracellular Ca2+ produced by ATX (100 nM) was antagonized in a concentration-dependent manner by tet

Homologation of mexiletine alkyl chain and stereoselective blockade of skeletal muscle sodium channels.

The optical isomers (-)-(S)- and (+)-(R)-3-(2, 6-dimethylphenoxy)-2-methyl-1-propanamine (Me2), homologues of the antiarrhythmic and antimyotonic drug mexiletine (Mex), were synthesized and assayed as new potential antimyotonic agents. As observed with Mex, Me2 exhibits an enantioselective behaviour. Tests carried out on sodium currents of single muscle fibres of Rana esculenta demonstrated that (-)-(S)- and (+)-(R)-Me2 were less potent than Mex in producing tonic block, but showed a higher use-dependent block. (-)-(S)-Me2 and (-)-(R)-Mex were also used to study the excitability of muscle fibr

Blockade by cAMP of native sodium channels of adult rat skeletal muscle fibers.

Although the skeletal muscle sodium channel is a good substrate for cAMP-dependent protein kinase (PKA), no functional consequence was observed for this channel expressed in heterologous systems. Therefore, we investigated the effect of 8-(4-chlorophenylthio)adenosine 3',5'-cyclic monophosphate (CPT-cAMP), a membrane-permeable cAMP analog, on the native sodium channels of freshly dissociated rat skeletal muscle fibers by means of the cell-attached patch-clamp technique. Externally applied CPT-cAMP (0.5 mM) reduced peak ensemble average currents by approximately 75% with no change in kinetics.

Activity-induced internalization and rapid degradation of sodium channels in cultured fetal neurons.

A regulatory mechanism for neuronal excitability consists in controlling sodium channel density at the plasma membrane. In cultured fetal neurons, activation of sodium channels by neurotoxins, e.g., veratridine and alpha-scorpion toxin (alpha-ScTx) that enhance the channel open state probability induced a rapid down-regulation of surface channels. Evidence that the initial step of activity-induced sodium channel down-regulation is mediated by internalization was provided by using 125I-alpha-ScTx as both a channel probe and activator. After its binding to surface channels, the distribution of 1

[3H]Phenytoin identifies a novel anticonvulsant-binding domain on voltage-dependent sodium channels.

The voltage-dependent sodium channel has been proposed as a specific target for the actions of the anticonvulsant drug phenytoin. Working at 0-4 degrees, we previously reported the existence of specific [3H]phenytoin binding sites in rat brain membranes. In the present study, the binding of [3H]phenytoin was assessed at 22 degrees, a temperature favorable to the binding of sodium channel ligands. At 22 degrees, the site had a Kd of 1.5 microM, which is in the relevant therapeutic concentration range for anticonvulsant activity (1-10 microM), and a calculated Bmax of 4.5 pmol/mg of protein, whi

Kinetics of chlorpromazine block of sodium channels in single guinea pig cardiac myocytes.

Block of sodium current by chlorpromazine in single ventricular myocytes isolated from guinea pigs was studied using the whole cell patch clamp technique. Chlorpromazine in micromolar concentrations reduced the amplitude of peak sodium current associated with step depolarizations from a holding potential of -140 mV. Concentration-response curves obtained with a holding potential of -140 mV were best fit by a 2:1 stoichiometry, and were shifted in the direction of lower concentrations when a holding potential of -100 mV was used. In agreement with this observation, the steady-state inactivation

Differential effects of halothane on adult and juvenile sodium channels in human muscle.

Myoballs were cultured from biopsies of adult human skeletal muscle. Transient currents through the sodium channels were elicited by depolarizing a myoball membrane with the whole-cell patch-clamp technique. The properties of the sodium channels were determined from the Hodgkin-Huxley parameters (INa max, tau m, tau h, h infinity-curve) derived from these transients. Halothane, when applied at 3.4 mmol/l (approximately 15 kPa), blocked about 50% of the current through the adult, TTX-sensitive sodium channels but had little effect on the current through the juvenile, TTX-insensitive sodium chan

Pharmacological characterization of the voltage-dependent sodium channels of rainbow trout brain synaptosomes.

Batrachotoxin, aconitine, and veratridine, alkaloid activators of voltage-dependent sodium channels, stimulated 22Na+ uptake by rainbow trout brain synaptosomes. The potency and efficacy of activation by these compounds decreased in the following order: batrachotoxin greater than aconitine much greater than veratridine. Aconitine-stimulated sodium uptake was completely inhibited by tetrodotoxin, a specific blocker of voltage-dependent sodium channels. Polypeptide toxins in the venom of the scorpion, Leiurus quinquestriatus, and the insecticide DDT enhanced veratridine-dependent sodium uptake b

Decoding voltage-gated sodium channels structure: Molecular mechanisms linking architecture, function, and disease.

Voltage-gated sodium (NaV) channels are critical transmembrane proteins responsible for the initiation and propagation of electrical signals in excitable tissues. They consist of a large α subunit and auxiliary β subunits. Together, the voltage sensing, ion selectivity, and rapid gating transitions regulate neuronal, muscular, and cardiac excitability. Recent cryo-electron microscopy breakthroughs have transformed the understanding of NaV structure-function relationships, revealing the asymmetric organization of voltage-sensing domains (VSD), the architecture of the DEKA selectivity filter, th

Hyperbaric Oxygen Attenuates Cerebral Ischemia-Reperfusion Injury in Rats by Modulating Sodium Channels.

Ischemic stroke accounts for the majority of stroke cases. Hyperbaric oxygen therapy (HBOT) is being increasingly used as a treatment for this condition, but its precise mechanisms of action remain incompletely elucidated. This study examined hyperbaric oxygen (HBO) effects on sodium channels in hippocampal CA1 pyramidal neurons in rats with cerebral ischemia-reperfusion injury (CIRI). Using the middle cerebral artery occlusion (MCAO) model and whole-cell patch-clamp technology, voltage-gated sodium channel (VGSC) currents were measured at varying HBOT time points and treatment durations. Epithelial sodium channels: contributions to the regulation of vascular function and blood pressure.

The epithelial sodium channel (ENaC) is a sodium channel expressed in renal tubules that, by selectively allowing the transmembrane passage of sodium ions, plays a crucial role in maintaining extracellular fluid volume and regulating blood pressure. Recent data have indicated that ENaC also exists in the cells of the vascular wall and contributes to the direct regulation of vascular function and blood pressure. The expression and activation of ENaC are both known to be regulated through mineralocorticoid receptor activation and serum glucocorticoid-regulated kinase 1. In addition, insulin meta
