Several tumor suppressor genes have been shown to regulate cellular susceptibility to proliferation or apoptotic cell death. An essential first step in studies with the long-range goal of determining the effect of a tumor suppressor gene on cellular susceptibility to apoptosis is careful characterization of the cell's response to an apoptotic stimulus. The goals of this study were to characterize the apoptotic response of a tuberous sclerosis complex-2 (Tsc2) tumor suppressor gene-null cell line, to establish valid biochemical events that can be used as apoptosis markers, and to determine how

BACKGROUND: Chronic inflammatory demyelinating polyneuropathy (CIDP) is an immune-mediated peripheral neuropathy with heterogeneous and often incomplete responses to current immunotherapies, but the underlying immune basis remains poorly defined. Although CIDP shares features of immune-mediated demyelination with multiple sclerosis (MS), the two diseases affect distinct anatomical compartments and exhibit divergent therapeutic responses, suggesting fundamentally different underlying immune programs. Here, we address this gap by defining the peripheral immune architecture of CIDP using an integ

BACKGROUND: Hypertrophic cardiomyopathy (HCM) is defined clinically by pathological left ventricular hypertrophy. We have previously developed a plasma proteomics biomarker panel that correlates with clinical markers of disease severity and sudden cardiac death risk in adult patients with HCM. The aim of this study was to investigate the utility of adult biomarkers and perform new discoveries in proteomics for childhood-onset HCM.
METHODS: Fifty-nine protein biomarkers were identified from an exploratory plasma proteomics screen in children with HCM and augmented into our existing multiplexed

BACKGROUND: Reverse remodeling can occur after left ventricular assist device (LVAD) support, which is sufficient in some cases to allow explantation of the device without cardiac transplantation. The molecular mechanisms involved remain unknown. A specific pattern of expression of sarcomeric and non-sarcomeric proteins in the myocardium is thought to be essential for normal myocardial function. However, a detailed protein analysis of their role in recovery has not been performed previously.
METHODS: Myocardial samples were collected at implantation and explantation in 7 patients with dilated

The talin2-β-integrin interaction modulates focal adhesion dynamics, promoting tumor cell migration and invasion, and thus represents an attractive anticancer target. Here, we identified the first talin2-β-integrin peptide inhibitor using structure-based virtual screening. Peptide-2 exhibited nanomolar binding affinity for talin2 (Kd = 8.05 ± 0.17 nM) and favorable in vitro selective inhibition. Molecular dynamics (MD) simulations indicated that Peptide-2 binds stably to talin2. Further studies showed that Peptide-2 significantly suppressed the proliferation, migration, and invasion of MDA-MB-

BACKGROUND: The lack of specificity and high degree of false positive and false negative rates when using mammographic screening for detecting early-stage breast cancer is a critical issue. Blood-based molecular assays that could be used in adjunct with mammography for increased specificity and sensitivity could have profound clinical impact. Our objective was to discover and independently verify a panel of candidate blood-based biomarkers that could identify the earliest stages of breast cancer and complement current mammographic screening approaches.
METHODS: We used affinity hydrogel nanop

Actin cytoskeleton dynamics depend on a tight regulation of actin filament formation from an intracellular pool of monomers, followed by their linkage to each other or to cell membranes, followed by their depolymerization into a fresh pool of actin monomers. The ubiquitous requirement for continuous actin remodeling that is necessary for many cellular functions is orchestrated in large part by actin binding proteins whose affinity for actin is altered by inositol phospholipids, most prominently PI(4,5)P2 (phosphatidylinositol 4,5-bisphosphate). The kinetics of PI(4,5)P2 synthesis and hydrolysi

In this study, an injectable matrix was fabricated using electrospun collagen/Hyaluronic acid/GOQD nanofiber (Collagen-HA-GOQD) and thermoresponsive N-succinyl chitosan-grafted-carboxyl-terminated poly-N-isopropyl acrylamide (SCS-g-PNIPAAm-COOH) hydrogel. Different characterization tools authenticated the synthesis of SCS-g-PNIPAAm-COOH, GOQD (size ∼5 nm), and Collagen-HA-GOQD nanofibers with average diameter of -153 nm. The nanocomposite exhibited a 3D honeycomb-shaped porous structure, and its sol-gel transition occurred at 33.4 °C with G' of 186 ± 0.5 Pa at body temperature. The scaffold's

The nicotinic acetylcholine receptor α1 (nAChRα1) was investigated as a potential proinflammatory molecule in the kidney, given a recent report that it is an alternative urokinase plasminogen activator (uPA) receptor, in addition to the classical receptor uPAR. Two animal models and in vitro monocyte studies were involved: (1) In an ApoE(-/-) mouse model of chronic kidney disease, glomerular-resident cells and monocytes/macrophages were identified as the primary cell types that express nAChRα1 during hypercholesterolemia/uninephrectomy-induced nephropathy. Silencing of the nAChRα1 gene for 4 m

Whenever there is heavy proteinuria, the glomerular epithelial cells, the podocytes, show dramatic morphological changes which clearly demonstrate changes in cell adhesion. However, there is little information on the types of cell adhesion molecules expressed in the normal human glomerulus. Assessments of changes in cell adhesion molecules in human proteinuria have been confined to semi-quantitative immunostaining for integrins, and the results have not been entirely consistent. This study sought first to define which cell adhesion molecules are present in the normal glomerulus, using indirect

Caries is a disease which on the basis of numerous epidemiological data it should be possible to control. The preventive interventions which have proved to have the greatest effect on the diffusion of this disease are essentially: fluoroprophylaxis, oral hygiene, food hygiene and periodic dental examination. The common denominator, which has the greatest effect on success, is a good level of health education of the populations affected by the programme, with specific reference to the teeth. The importance of the diet as a possible element predisposing to caries is an ascertained fact by now, a

T cell-APC contact initiates T cell activation and is maintained by the integrin LFA-1. Talin1, an LFA-1 regulator, localizes to the immune synapse (IS) with unknown roles in T cell activation. In this study, we show that talin1-deficient T cells have defects in contact-dependent T cell stopping and proliferation. Although talin1-deficient T cells did not form stable interactions with APCs, transient contacts were sufficient to induce signaling. In contrast to prior models, LFA-1 polarized to T cell-APC contacts in talin1-deficient T cells, but vinculin and F-actin polarization at the IS was i

The leukocyte integrin LFA-1 plays a critical role in T cell trafficking and T cell adhesion to APCs. It is known that integrin-mediated adhesion is regulated by changes in integrin ligand-binding affinity and valency through inside-out signaling. However, the molecular mechanisms involved in TCR-mediated LFA-1 regulation are not well understood. In this study, we show that the cytoskeletal protein talin1 is required for TCR-mediated activation of LFA-1 through regulation of LFA-1 affinity and clustering. Depletion of talin1 from human T cells by small interfering RNAs impairs TCR-induced adhe

Corneal stromal fibrosis represents a major cause of visual impairment, primarily driven in part by the TGF-β1-mediated transition of stromal fibroblasts into contractile myofibroblasts. FARP2, a mediator of Rho-GTPase signaling and cytoskeletal organization, has been proposed as a potential mediator of this phenotypic transition; however, the specific functional contribution stromal remodeling remains largely undefined) its functional role in corneal stromal remodeling remains unclear. CSFs were isolated from New Zealand rabbits and cultured under standard conditions. Myofibroblast differenti

Integrins are a group of transmembrane glycoprotein receptors that are responsible for platelet activation through bidirectional signalling. These receptors have left their footprints in various cellular events and have intrigued many groups of scientists that have led to some significant discoveries. A lot of the recent understanding of haemostasis has been possible due to the integrins filling the gaps in between several cellular mechanism. Apart from this, other important functions carried out by integrins are growth and maturation of cardiomyocytes, mechano-transduction, and interaction wi

INTRODUCTION: Obesity is a major public health concern worldwide and contributes to multiple metabolic disorders. Low carbohydrate diets (LCDs) have been proposed as an effective strategy for weight management and metabolic improvement; however, the mechanisms underlying LCD-induced molecular changes in individuals with obesity remain incompletely understood. This study aimed to assess the impact of a three-month LCD on the proteomic profile of adults with obesity.
METHODS: Thirty adults (18-50 years) with obesity (BMI ≥ 30 kg/m2) followed an LCD (60-110 g/day) for 3 months. Anthropometric an

Stromal components not only help form the structure of neoplasms such as melanomas, but they also functionally contribute to their malignant phenotype. Thus, uncovering signaling pathways that integrate the behavior of both tumor and stromal cells may provide unique opportunities for the development of more effective strategies to control tumor progression. In this regard, extracellular matrix-mediated signaling plays a role in coordinating the behavior of both tumor and stromal cells. Here, evidence is provided that targeting a cryptic region of the extracellular matrix protein collagen (HU17

BACKGROUND: Atherosclerosis is a chronic vascular inflammatory disease caused by multiple factors. Anti-inflammatory treatment is an effective approach to treat atherosclerosis. Talin1 is a cell membrane-associated cytoskeletal protein that is widely expressed in mammals and plays essential roles in angiogenesis and endothelial cell barrier function. However, the role of Talin1 in atherosclerosis and the related mechanisms remains unclear.
METHODS: ApoE-KO mice were subjected to partial carotid artery ligation to establish an atherosclerosis model, and the expression of Talin1 in atherosclero
