The effects of ear-point stimulation on the contents of somatostatin and Amino acid neurotransmitters in brain of rat with experimental seizure.

The goal of this study was to elucidate the anti-convulsion mechanisms of ear-point stimulation in rat with experimental seizure. We prepared the epilepsy rats by intrahippocampal injection of penicillin. One hour later the lower 1/2 auricular lobules of seizure rats, containing ear-points Pizhixia and Shenmen etc., was electrically stimulated, which was imitated as ear-point electrical acupuncture in humans. Radioimmunoassay and biochemical techniques were used to determine the contents of somatostatin and amino acid neurotransmitters in hippocampus of rats. The outcomes revealed epileptiform

Interaction analysis of aspirin with selective amino acids.

This study was conducted to assess the compatibility of aspirin with selective amino acids by studying the effect of amino acids on the solubility of aspirin, so that the attention could be paid towards the use of proteinous foods along with aspirin. Two different types of dissolution media, i.e., 0.5% solution of each amino acid and 100 mL of distilled water (100 mL each), were prepared. Then, 1 g of aspirin was added in both media and shaked gently. Ten milliliters of sample was withdrawn at different time intervals, i.e., 10, 20, 30, 40, 50 and 60 min and analyzed spectrophotometrically at

Brain amino acids in conscious rats in chronic normocapnic and hypocapnic hypoxemia.

The influence of hypoxemia on the brain content of several organic acids and NH+4, AND ITS RELATIONship to the accompanying hypocapnia was studied in unanesthetized rats subjected to hypoxemia for periods ranging between 2 hours and 7 days. Under acute conditions, 'mild' hypoxemia (FO2 = 6--7%), these increases were greater and accompanied by increased gamma-aminobutyric acid (GABA) and decreased glutamic and aspartic acid levels; glutamine and NH+4 remained normal. When hypocapnia was prevented, 'severe' hypoxemia induced only a rise in GABA and slight elevations in lactic and alpha-ketogluta

Effect of sound stimulation at several levels on concentrations of primary amines, including neurotransmitter candidates, in perilymph of the guinea pig inner ear.

Perilymph, which bathes the sensory cells of the cochlea, was collected from guinea pigs exposed to noise and analyzed via two cation-exchange HPLC procedures with fluorescence detection, resolving 51 and 81 primary-amine compounds, respectively, at a sensitivity limit of 0.1 pmol relative to leucine. During a first period, each animal was either exposed to noise at 80, 90, or 115 decibels sound-pressure level or maintained in silence (controls), and during a second period, the same animal was maintained in silence. Perilymph was collected during both periods, and perilymphatic components were

Regional distribution and postnatal changes of D-amino acids in rat brain.

Regional distribution of D-amino acids in rat brain was studied by the modified highly sensitive analytical method which was previously developed. The method includes fluorogenic derivatization of each amino acid, isolation of each amino acid by reverse-phase HPLC, followed by enantiomeric separation with Pirkle-type chiral stationary phases. D-Amino acid contents were determined in the cerebrum, cerebellum, hippocampus, medulla oblongata, pituitary gland and pineal gland. D-Aspartic acid was observed in the pineal gland (3524 +/- 263 nmol/g, data are for male rats of 6 weeks of age) and the p

Direct and indirect cellular effects of aspartame on the brain.

The use of the artificial sweetener, aspartame, has long been contemplated and studied by various researchers, and people are concerned about its negative effects. Aspartame is composed of phenylalanine (50%), aspartic acid (40%) and methanol (10%). Phenylalanine plays an important role in neurotransmitter regulation, whereas aspartic acid is also thought to play a role as an excitatory neurotransmitter in the central nervous system. Glutamate, asparagines and glutamine are formed from their precursor, aspartic acid. Methanol, which forms 10% of the broken down product, is converted in the bod

Effects of constituent amino acids on tubular handling of microinfused angiotensin II.

[14C]angiotensin II ([14C]AII) was microinjected alone or with excess L-isoleucine (IIe) or L-aspartic acid (Asp) into renal tubules of anesthetized rats. Urinary excretion of 14C-labeled material was measured, and the intact peptide and its metabolites were identified and quantified. When isoleucine was administered with [14C]AII, urinary recovery of the 14C-labeled material increased directly with distance of infusion site from glomerulus, and most of the radioactivity in urine was [14C]Ile. The data suggest that isoleucine interfered with reabsorption of [14C]Ile derived from hydrolysis of

Guest-Induced Arylamide Polymer Helicity: Twist-Sense Bias and Solvent-Dependent Helicity Inversion.

A benzene/naphthalene alternately incorporated amide polymer was synthesized and characterized. (1) H NMR spectroscopy, fluorescence, and circular dichroism (CD) experiments indicated that, in chloroform, the polymer could be induced by the chiral l-aspartic acid dianion or one of its derivatives to form a helical tubular conformation with twist-sense bias. CD titration studies showed that the l-aspartic acid dianion (8 equiv.) could lead to a maximum Cotton effect. It was also revealed that the twist-sense bias obeyed the majority rule, and 70 % enantiomeric excess could realize the maximum h

Mutations in the E1 hydrophobic domain of rubella virus impair virus infectivity but not virus assembly.

Rubella virus (RV) virions contain three structural proteins, a capsid protein that interacts with viral genomic RNA to form a nucleocapsid and two membrane glycoproteins, E2 and E1. We found that substitution of either an aspartic acid residue at Gly93 (G93D) or a glycine residue at Pro104 (P104G) in the internal hydrophobic domain of E1 affected virus infectivity but not virus assembly. Viruses carrying G93D and P104G mutations had impaired infectivity, reduced 1,000-fold and 10-fold, respectively. A revertant was isolated from the G93D mutant. Sequencing analysis showed that the substituted

Cerebrospinal fluid amino acids in pathological gamblers and healthy controls.

Amino acids, such as valine, isoleucine and leucine compete with tyrosine and tryptophan for transport into the brain and might thus affect the central serotonin and catecholamine patterns. Furthermore, the excitatory amino acids glutamic acid, aspartic acid and glycine are known to act on the N-methyl-D-aspartate receptor, which is part of the reward system. Based on these facts, we have explored the role of cerebrospinal fluid (CSF) amino acids in pathological gambling. Concentrations of amino acids were determined in CSF obtained from one female and 11 pathological male gamblers and 11 heal

A missense mutation causes aspartase deficiency in Yersinia pestis.

It is established that cells of Yersinia pestis, the causative agent of bubonic plague, excrete l-aspartic acid at the expense of exogenous l-glutamic acid during expression of the low-calcium response. Results of enzymic analysis provided here suggest that a previously defined deficiency of aspartase (AspA) accounts for this phenomenon rather than an elevated oxaloacetate pool. The only known distinction between most sequenced isolates of aspA from Y. pestis and the active gene in Yersinia pseudotuberculosis (the immediate progenitor of Y. pestis) is a single base transversion (G.C-->T.A) cau

Dual acid-base catalysis with biologically modified graphene oxide: a sustainable route to polyhydroquinolines with antimicrobial properties.

This article conducts an in-depth examination of graphene oxide-aspartic acid (GO-As) as a novel bifunctional nano-organocatalyst distinguished by both catalytic and antibacterial properties. The research elucidates the synthesis of GO through Hummer's method, followed by the covalent attachment of aspartic acid to the surface of GO nanosheets. This innovative approach is particularly notable as it circumvents the use of hazardous chemicals, thereby promoting environmental sustainability. The newly developed catalyst underwent rigorous analysis employing a variety of spectroscopic techniques,

[Megalencephaly with dystonia revealing Canavan disease].

Canavan disease, or N-acetyl aspartic aciduria, is an autosomal recessive leukodystrophy characterized by spongy degeneration of the brain. The disease results from the accumulation of N-acetyl aspartic acid in the brain, due to aspartoacylase deficiency. We report the case of a 6-month-old girl who presented with megalencephaly, peripheral hypertonia, and a developmental delay noticeable after 4 months of age. Magnetic resonance imaging of the brain with spectroscopy was suggestive of Canavan disease, which was confirmed by chromatography of urinary organic acids.

Cloning, sequence analysis and homology modeling of a novel phospholipase A2 from Heterometrus fulvipes (Indian black scorpion).

We report the cloning and sequencing of group III phospholipaseA(2) from Heterometrus fulvipes (HfPLA(2)), Indian black scorpion. The cDNA sequence codes for the mature portion of the group PLA(2) of 103 amino acids. The sequence has 85% identity with Mesobuthus tamulus (Indian red scorpion) PLA(2) and a 40% identity with bee venom PLA(2) and human group III PLA(2). Most of the essential features of group III PLA(2) like Ca(2+) binding loop and catalytic residues are conserved. Homology modeling was done with the known structure of group III bee venom PLA(2). Effects of Amino Acid Side-Chain Length and Chemical Structure on Anionic Polyglutamic and Polyaspartic Acid Cellulose-Based Polyelectrolyte Brushes.

We used atomistic molecular dynamics (MD) simulations to study polyelectrolyte brushes based on anionic α,L-glutamic acid and α,L-aspartic acid grafted on cellulose in the presence of divalent CaCl2 salt at different concentrations. The motivation is to search for ways to control properties such as sorption capacity and the structural response of the brush to multivalent salts. For this detailed understanding of the role of side-chain length, the chemical structure and their interplay are required. It was found that in the case of glutamic acid oligomers, the longer side chains facilitate attr
