Hemoglobin P(50) during a simulated ascent of Mt. Everest, Operation Everest II.

The amount of O(2) available to tissues is essentially the product of cardiac output, [Hb], and O(2) saturation. Saturation depends on P(O2) and the O(2)Hb dissociation curve. With altitude, increased [2,3-DPG] shifts the dissociation curve rightward, but hypocapnia and alkalosis move it leftward. We determined both standard and in vivo P(50) in 5 fit subjects decompressed over 42 days in an altitude chamber to the equivalent of the Mt. Everest summit (Operation Everest II). Arterial and venous blood was sampled at five "altitudes " (P(B) = 760, 429, 347, 282, 253 mmHg), and P(O2), P(CO2), pH,

A subchronic toxicity study of two inhaled aerosolized atropine sulfate formulations in rats and dogs.

The potential subchronic (21 days) toxicity of inhaled metered aerosol formulations (solution and suspension) of atropine sulfate was investigated in rats and dogs. The doses administered to rats were 0.78 and 2.5 mg/kg/day (solution) or 1.4 and 3.2 mg/kg/day (suspension). In the dog, the daily doses achieved were 0.5 and 1.3 mg/kg/day, regardless of formulation. In both species, sham control animals inhaled air only and vehicle control animals inhaled either placebo solution or placebo suspension. There was no mortality or other evidence of a toxic effect of atropine sulfate. The expected myd

Concentration-dependent immunologic response to toluene diisocyanate (TDI) following inhalation exposure.

Little is known concerning industrial exposure conditions which lead to development of allergic sensitivity in exposed workers. This study investigated the relationship between exposure concentration and the induction of antibodies and sensitivity in a guinea pig animal model for inhalation exposure to isocyanates (Karol et al., Toxicol. Appl. Pharmacol. 53, 260-270, 1980). Groups of guinea pigs were exposed, via inhalation, to TDI concentrations ranging from 0.12 to 10 ppm. Exposure was for 3 hr/day on 5 consecutive days. Beginning on Day 22, animals were evaluated for TDI-specific antibodies

Pituitary hormones and growth retardation in rats raised at simulated high altitude (3800m).

Growth and the endocrine status of the pituitary and thyroid glands were studied in rats born and raised in a hypobaric chamber at a simulated high altitude of 3800 m (SHA); comparisons were drawn with similar rats at sea level. From birth until 40 days of age, SHA rats weighed significantly less than controls with the most striking growth impairment found in female SHA rats. Relative organ weights of anterior pituitary glands, ovaries and uteri from 40-day-old female SHA rats were significantly less than controls. Pituitary content of growth hormone (GH) was reduced in 40-day-old female SHA r

Cerebral artery blood velocity in normal subjects during acute decreases in barometric pressure.

To investigate the effect of acute changes in barometric pressure on regional cerebral perfusion we studied the middle cerebral artery (MCA) blood velocity in five healthy male volunteers by means of a low-pressure chamber. The MCA blood velocity, arterial blood and respiratory gases were measured at the barometric pressures of 1, 0.8, 0.65, and 0.5 atmospheres. The observed blood velocity (Vo) showed no systematic changes. Decreases in barometric pressure induced hypoxia and hypocapnia. When normalizing the MCA blood velocity (Vn) to a standard P(CO2) (5.3 kPa), thereby correcting for the hyp

Toxicological responses in SW mice exposed to inhaled pyrolysates of polymer/tobacco mixtures and blended tobacco.

Modern cigarette manufacturing is highly automated and produces millions of cigarettes per day. The potential for small inclusions of non-cigarette materials such as wood, cardboard packaging, plastic, and other materials exists as a result of bulk handling and high-speed processing of tobacco. Many non-tobacco inclusions such as wood, paper, and cardboard would be expected to yield similar pyrolysis products as a burning cigarette. The aircraft industry has developed an extensive literature on the pyrolysis products of plastics, however, that have been reported to yield toxic by-products upon

Advancements in fire-related toxic gas detection and prophylactic strategies: A focus on cyanide concentration analysis and antidote efficacy in controlled smoke inhalation models.

Fire smoke inhalation represents a major cause of acute mortality in fire incidents, with hydrogen cyanide being a critical contributor to rapid systemic toxicity. This study aimed to establish a controlled smoke inhalation model to characterize cyanide-dominant exposure and to evaluate the prophylactic efficacy of a nebulized antidote combination under acute conditions. A reproducible smoke chamber system was developed to generate cyanide-rich toxic atmospheres. C57BL/6 mice were exposed to controlled cyanide concentrations, followed by prophylactic administration of a nebulized formulation c

Increased distensibility in dependent veins following prolonged bedrest.

Displacement of blood to the lower portion of the body that follows a postural transition from recumbent to erect is augmented by a prolonged period of recumbency (bedrest). Information is scarce as to what extent this augmented blood-volume shift to dependent veins is attributable to increased distensibility of the veins. Accordingly, we studied the effect of 5 weeks of horizontal bedrest on the pressure-distension relationship in limb veins. Elevation of venous distending pressure was induced by exposure of the body except the tested limb to supra-atmospheric pressure with the subject seated

Understanding the Early Biological Effects of Isoprene-Derived Particulate Matter Enhanced by Anthropogenic Pollutants.

INTRODUCTION: Airborne fine particulate matter (PM2.5; particulate matter ≤ 2.5 μm in aerodynamic diameter) plays a key role in air quality, climate, and public health. Globally, the largest mass fraction of PM2.5 is organic, dominated by secondary organic aerosol (SOA) formed from atmospheric oxidation of volatile organic compounds (VOCs). Isoprene from vegetation is the most abundant nonmethane VOC emitted into Earth's atmosphere. Isoprene has been recently recognized as one of the major sources of global SOA production that is enhanced by the presence of anthropogenic pollutants, such as ac

Oxygen transport during exercise at extreme altitude: Operation Everest II.

Eight male volunteers had rest and exercise measurement to determine the mechanisms of oxygen transport during a 40-day chamber decompression simulating high-altitude exposure equivalent to the summit of Mt Everest. Five subjects completing the study decreased their maximum oxygen uptake by 72%. During maximal or near-maximal exercise, arterial PCO2 fell as low as 8 mm Hg, defending the alveolar PO2 and confirming marked hyperventilation. Alveolar-arterial diffusion did not improve and V/Q worsened. Cardiac function was unimpaired. Circulatory oxygen transport resembled that at sea level. The

Working with C. difficile in a Hypoxic Chamber.

Clostridioides difficile is an enteropathogen responsible for diarrhea under conditions leading to a dysbiosis of the gut microbiota, in particular after an antibiotic treatment. It is a spore-forming obligate anaerobe, which is, as such, usually cultivated in anaerobiosis in laboratories. Yet the gastrointestinal tract (GIT), its natural niche, is not completely deprived of oxygen (O2) and the O2 tension in the gut even rises under conditions of dysbiosis. C. difficile is able to tolerate the low O2 tensions it encounters in the GIT. C. difficile spores germinate in the small intestine at 4%

Evaluation of a headspace solid-phase microextraction method for the analysis of ignitable liquids in fire debris.

The chemical analysis of fire debris represents a crucial part in fire investigations to determine the cause of a fire. A headspace solid-phase microextraction (HS-SPME) procedure for the detection of ignitable liquids in fire debris using a fiber coated with a mixture of three different sorbent materials (Divinylbenzene/Carboxen/Polydimethylsiloxane, DVB/CAR/PDMS) is described. Gasoline and diesel fuel were spiked upon a preburnt matrix (wood charcoal), extracted and concentrated with HS-SPME and then analyzed with gas chromatography/mass spectrometry (GC/MS). The experimental conditions--ext

Use of a physiologically based model to predict systemic uptake and respiratory elimination of perchloroethylene.

The pharmacokinetics of inhaled perchloroethylene (PCE) were studied in male Sprague-Dawley rats to characterize the pulmonary absorption and elimination of the volatile organic chemical (VOC). The direct measurements of the time course of PCE in the blood and breath were used to evaluate the ability of a physiologically based pharmacokinetic (PBPK) model to predict systemic uptake and elimination of PCE. Fifty or 500 ppm PCE was inhaled for 2 hr through a miniaturized one-way breathing valve by unanesthetized male Sprague-Dawley rats of 325-375 g. Serial samples of the inhaled and exhaled bre

Exposure to air pollution is associated with lung hyperinflation in healthy children and adolescents in Southwest Mexico City: a pilot study.

Air pollution produces adverse health effects. The consequences of lifelong daily exposures to atmospheric pollutants upon the respiratory apparatus of healthy children are of considerable clinical importance. We investigated the association between exposure to a highly polluted urban environment with a complex mixture of air pollutants-ozone and particulate matter the predominant ones-and chest x-ray abnormalities in 59 healthy Mexican children who are lifelong residents of Southwest Metropolitan Mexico City (SWMMC), with a negative history of tobacco exposure and respiratory illnesses. The effect of intravenous perfluorocarbon emulsions on whole-body oxygenation after severe decompression sickness.

INTRODUCTION: Decompression sickness (DCS) results from a decrease in ambient pressure leading to supersaturation of tissues with inert gas and bubble formation. Perfluorocarbons (PFCs) are able to dissolve vast amounts of non-polar gases. Intravenous (IV) PFC emulsions reduce both morbidity and mortality associated with DCS, but the mechanism of this protective effect has not yet been demonstrated. METHODS: Juvenile Dorper-cross sheep (n = 31) were anaesthetised and instrumented for physiological monitoring, IV fluid administration and blood sampling. Animals were compressed in air in a hype

An effort to test the embryotoxicity of benzene, toluene, xylene, and formaldehyde to murine embryonic stem cells using airborne exposure technique.

Benzene, toluene, xylene, and formaldehyde are well-known indoor air pollutants, especially after house decoration. They are also common pollutants in the working places of the plastic industry, chemical industry, and leather industry. It has been reported that these pollutants cause people to be irritated, sick, experience a headache, and be dizzy. They also have the potential to induce asthma, aplastic anemia, and leukemia, even cause abortion or fetus malformation in humans. In this study, the airborne toxicity of benzene, toluene, xylene, and formaldehyde to murine embryonic stem cells (mE

Effects of age and alcohol exposure during early life on pyramidal cell numbers in the CA1-CA3 region of the rat hippocampus.

We have previously shown that exposing rats to a relatively high dose of ethanol during early postnatal life can result in an alteration in spatial learning ability. The hippocampal formation is known to be involved in the control of this ability. The purpose of the present study was to determine whether exposure of rats to ethanol during early postnatal life had either immediate or delayed effects on the numbers of pyramidal cells in the CA1-CA3 subregion of the hippocampus. Wistar rats were exposed to a relatively high daily dose of ethanol at postnatal day 10-15 by placing them for 3 h/day

Corneal Structural and Response to Elevated CO2 Concentrations in Occupational Environment: FTIR and Chemometric Evidence of Proteins and Lipids Remodeling.

This study investigates the effects of occupational carbon dioxide (CO2) exposure on corneal tissue through Fourier transform infrared (FTIR) spectroscopy and chemometric analysis. Human activities have significantly increased atmospheric CO2 levels, leading to potential health risks, particularly in confined environments such as mining, submarines, and enclosed workspaces. While CO2 exposure is commonly associated with respiratory and cardiovascular effects, its impact on ocular health remains underexplored. Rats were exposed to three different CO2 concentrations (3%, 5%, and 10%) in a contro
