Body Composition Changes in Patients with Oropharyngeal Squamous Cell Carcinomas Treated With Radiotherapy.

AIMS: Patients with oropharyngeal squamous cell carcinomas (OPSCC) are often malnourished at diagnosis, can lose weight during treatment, and many require nutritional support. Different modes of weight loss exist, caused by fat and/or muscle loss, but current practice relies on monitoring weight alone. We aimed to determine whether routinely collected data could be used to model changes in body composition in greater detail. MATERIALS AND METHODS: Clinical and imaging data were retrospectively collected for 337 patients with OPSCC treated with radiotherapy (RT) at two tertiary cancer centres.

The Role of Stereotactic Body Radiotherapy in Oligometastatic Non-Small Cell Lung Cancer.

Non-small cell lung cancer (NSCLC) is a major cause of mortality in Canada, with many patients presenting with metastatic disease. The oligometastatic state (OM-NSCLC) may be amenable to cure using aggressive local consolidative therapies. Stereotactic body radiotherapy (SBRT), which entails the utilization of a high dose of radiation in one or few fractions, has many benefits in this setting, including its applicability in varied patient populations to ablate lesions in varied anatomical locations. It has also been demonstrated to prolong the time to next-line systemic therapy, to reduce fina

Macrophages and fibroblasts in foreign body reactions: How mechanical cues drive cell functions?

Biomaterials, when implanted in the human body, can induce a series of cell- and cytokine-related reactions termed foreign body reactions (FBRs). In the progression of FBRs, macrophages regulate inflammation and healing by polarizing to either a pro-inflammatory or pro-healing phenotype and recruit fibroblasts by secreting cytokines. Stimulated by the biomaterials, fibrotic capsule is formed eventually. The implant, along with its newly formed capsule, introduces various mechanical cues that influence cellular functions. Mechanosensing proteins, such as integrins or ion channels, transduce ext

Whole-Body Lung Function Test-Derived Outcome Predictors in Allogenic Stem Cell Transplantation.

Despite clinical advances, late onset pulmonary complications in adult recipients of allogenic stem cell transplantation are a major cause of morbidity and mortality. Reported incidence and risk factors in the literature vary broadly and are partly contradictory. Identification of pretransplant factors associated with major complications would be helpful to define individual treatment strategies and early initiation of preventive measures. To evaluate incidence and risk factors of late onset noninfectious pulmonary complications, with special regard to small airways disease (SAD) and bronchiol

Treatment of Peripheral Non-Small Cell Lung Carcinoma with Stereotactic Body Radiation Therapy.

Stereotactic body radiation therapy (SBRT) is an effective and well-tolerated noninvasive treatment for medically inoperable patients with peripheral non-small cell lung carcinoma. The term "peripheral" refers to lesions that lie 2 cm or more from the mediastinum and proximal bronchial tree and was instituted based on results from a specific dose and fractionation schedule. Improvements in immobilization, respiratory motion management, and image guidance have allowed for SBRT's highly conformal and accurate delivery of large radiation doses per fraction. Results from prospective and retrospect

Aberrant cell cycle reentry in human and experimental inclusion body myositis and polymyositis.

Inclusion body myositis (IBM), a degenerative and inflammatory disorder of skeletal muscle, and Alzheimer's disease share protein derangements and attrition of postmitotic cells. Overexpression of cyclins and proliferating cell nuclear antigen (PCNA) and evidence for DNA replication is reported in Alzheimer's disease brain, possibly contributing to neuronal death. It is unknown whether aberrant cell cycle reentry also occurs in IBM. We examined cell cycle markers in IBM compared with normal control, polymyositis (PM) and non-inflammatory dystrophy sample sets. Next, we tested for evidence of r

Mouse oocytes within germ cell cysts and primordial follicles contain a Balbiani body.

The Balbiani body or mitochondrial cloud is a large distinctive organelle aggregate found in developing oocytes of many species, but its presence in the mouse has been controversial. Using confocal and electron microscopy, we report that a Balbiani body does arise in mouse neonatal germline cysts and oocytes of primordial follicles but disperses as follicles begin to grow. The mouse Balbiani body contains a core of Golgi elements surrounded by mitochondria and associated endoplasmic reticulum. Because of their stage specificity and perinuclear rather than spherical distribution, these clustere

Pattern analysis of human cutaneous mast cell populations by total body surface mapping.

BACKGROUND: Tuberculosis (TB) resulted in an estimated 1.7 million deaths in the year 2016. The disease is caused by the members of Mycobacterium tuberculosis complex, which includes Mycobacterium tuberculosis, Mycobacterium bovis and other closely related TB causing organisms. In order to understand the epidemiological dynamics of TB, national TB control programs often conduct standardized genotyping at 24 Mycobacterial-Interspersed-Repetitive-Units (MIRU)-Variable-Number-of-Tandem-Repeats (VNTR) loci. With the advent of next generation sequencing technology, whole-genome sequencing (WGS) has

A pH-sensitive chloride current in the chemoreceptor cell of rat carotid body.

1. Cardiorespiratory response to acidosis is initiated by the carotid body. 2. The direct effect of extracellular pH (pH(o)) on the chloride currents of isolated chemoreceptor cells of the rat carotid body was investigated using the whole-cell patch-clamp technique. 3. On applying intra- and extracellular solutions with a symmetrical high-Cl(-) content and with the monovalent cations replaced with membrane-impermeant ones, an inwardly rectifying Cl(-) current was found. 4. The current activated slowly and did not display any time-dependent inactivation. Current activation was present at membra

Hirano body in an inflammatory cell of leptomeningeal vessel infected by fungus Paecilomyces.

An intracytoplasmic microfilamentous lattice, ultrastructurally identical to Hirano body, was found in an inflammatory cell within a leptomeningeal vessel wall infected by fungus Paecilomyces javanicus. The structure was well preserved and not associated with phagosomes. This is the first report of Hirano body found in an inflammatory cell at the site of fungal infection. The present finding suggests that the formation of Hirano body is non-specific and not restricted to the cell of the neuro-muscular system.

Extensional flow of erythrocyte membrane from cell body to elastic tether. I. Analysis.

This is the first of two papers on an analytical and experimental study of the flow of the erythrocyte membrane. In the experiment to be discussed in detail in the second paper, preswollen human erythrocytes are sphered by aspirating a portion of the cell membrane into a small micropipette; and long, thin, membrane filaments or "tethers" are steadily withdrawn from the cell at a point diametrically opposite to the point of aspiration. The aspirated portion of the membrane furnished a "reservoir" of material that replaces the membrane as it flows as a liquid from the nearly spherical cell body

Comparison of Superoxide Dismutase Activity at the Cell, Organ, and Whole-Body Levels.

Superoxide dismutase (SOD) can be considered an antitoxic metalloenzyme that facilitates the production of oxygen and hydrogen peroxide from superoxide anions. Four classes have been identified depending on selective binding of metals, namely Cu,Zn-SOD, Fe-SOD, Mn-SOD, and Ni-SOD. The established isoforms are SOD1, SOD2, and SOD3 in various cells and tissues of eukaryotes. The relatively newer type Ni-SOD binds nickel and is observed in bacteria, including the genus Streptomyces. The Fe-SOD and Mn-SOD are also present in bacteria. Cu,Zn superoxide dismutase (SOD1) activity correlates with vari

A Prospective Study of Stereotactic Body Radiation Therapy in Oligometastatic Renal Cell Carcinoma.

PURPOSE: Metastasis-directed therapy for oligometastatic renal cell carcinoma (RCC) with stereotactic body radiation therapy (SBRT) has been shown to improve progression-free survival (PFS) and delay time to systemic therapy. Here, we present long-term follow-up of a prospective trial. METHODS AND MATERIALS: Patients with oligometastatic or recurrent RCC (1-5 lesions, with no therapies in prior month) were enrolled on a pilot study (NCT02542202). SBRT was delivered to all sites (preferred regimen of 50 Gy in 5 fractions). The primary endpoint was the rate of grade 4+ adverse events, and secon

Sporadic inclusion body myositis-derived myotube culture revealed muscle cell-autonomous expression profiles.

Sporadic inclusion body myositis (sIBM) is a muscle disease in older people and is characterized by inflammatory cell invasion into intact muscle fibers and rimmed vacuoles. The pathomechanism of sIBM is not fully elucidated yet, and controversy exists as to whether sIBM is a primary autoimmune disease or a degenerative muscle disease with secondary inflammation. Previously, we established a method of collecting CD56-positive myoblasts from human skeletal muscle biopsy samples. We hypothesized that the myoblasts derived from these patients are useful to see the cell-autonomous pathomechanism o

Considerations for Peripheral Blood Stem Cell Apheresis in a Low Body Weight Infant.

INTRODUCTION: Peripheral blood stem cell (PBSC) apheresis in infants (<10 kg body weight) requires specific precautions to prevent periprocedural complications. CASE REPORT: A 9 month old child was diagnosed with high-risk neuroblastoma and planned for autologous stem cell transplantation after induction chemotherapy. We illustrate the precautions and technical details observed while performing PBSC collection in this patient. DISCUSSION: Use of continuous flow devices, priming of apheresis circuits, appropriate flow rates and continuous monitoring can help to mitigate several procedure rela

Giant Cell Tumor of the Carotid Body: A Rare Tumor with Malignant Potential.

INTRODUCTION: Carotid body tumors (CBTs) are certainly unusual. They are vascular lesions originating from paraganglionic cells, located at the common carotid artery (CCA) bifurcation. They represent less than 0.5% of head and neck tumors, approximately 1-3 cases per million. Malignant CBTs are extremely rare; in the literature, published rates on average are < 10%. The diagnostic criteria for malignancy should be based on the finding of distant metastasis. Due to its unpredictable nature and its malignant potential, diagnosis before metastasis and complete surgical resection are the keys to a

Pilot study evaluating stereotactic body radiation therapy for feline facial squamous cell carcinomas.

OBJECTIVES: The use of stereotactic body radiation therapy (SBRT) has not been assessed in advanced-stage feline solar-induced facial squamous cell carcinomas (SCCs). The objective of this study was to provide preliminary data on the use and safety profile of coarse fractions administered with an SBRT regime to manage advanced-stage feline solar-induced facial SCCs. METHODS: This retrospective study assessed five cats diagnosed with advanced-stage solar-induced facial SCCs that received SBRT as their primary treatment or, in one cat, following failed surgical intervention. Current Use of Total Body Irradiation in Haploidentical Allogeneic Hematopoietic Stem Cell Transplantation.

Total body irradiation (TBI) is included in the conditioning regimen for allogeneic hematopoietic stem cell transplantation (HSCT), with unique advantages such as uniform distribution over the whole body and decreased exposure to cytotoxic chemotherapeutic agents. For individuals who lack matched sibling or matched unrelated donors, the use of haploidentical donors has been increasing despite challenges such as graft rejection and graft-versus-host disease (GVHD). Although a limited number of studies have been performed to assess the clinical role of TBI in haploidentical HSCT, TBI-based condi
