Bone-related diseases impact a large portion of the global population and, due to their high disability rates and limited treatment options, pose significant medical and economic challenges. Mesenchymal stem cells (MSCs) can differentiate into multiple cell types and offer strong regenerative potential, making them promising for treating various diseases. However, issues with the immune response and cell survival limit the effectiveness of cell transplantation. This has led to increased interest in cell-free stem cell therapy, particularly the use of exosomes, which is the most studied form of

Efficient nucleic acid enrichment is pivotal for deciphering epigenetic modifications and disease biomarkers, yet current methods are constrained by insufficient specificity, poor versatility, and high costs. We developed a universal strategy named "Click-IP-Seq" by leveraging the high-affinity binding between Protein A/G and Fc region of DBCO-modified IgG. This enabled the directional conjugation of DBCO-IgG with any azide-modified nucleic acids via copper-free strain-promoted azide-alkyne cycloaddition (SPAAC) click chemistry, achieving specific capture and enrichment of modified nucleic aci

In cancer immunotherapy, peptide-major histocompatibility complexes are increasingly exploited as precise therapeutic targets. For instance, T-cell receptor (TCR)-engineered T cells, peptide vaccines, and TCR-mimic antibodies leverage the direct recognition of tumor-specific or tumor-associated antigens presented on human leukocyte antigen (HLA) molecules, offering highly specific and effective approaches to eliminate malignant cells. Classical immunopeptidomic studies rely on tumor tissue samples to identify targetable peptides; however, tissue accessibility and tumor heterogeneity remain maj

Infertility affects millions of women worldwide, with causes ranging from ovarian dysfunction to endometrial disorders. Conventional treatments, such as hormone therapy and assisted reproductive technologies (ART), often have limited success and significant side effects. Stem cell-derived extracellular vesicles (EVs) have emerged as a promising alternative, offering therapeutic potential through their cargo of proteins, nucleic acids, and bioactive molecules that regulate tissue repair and regeneration. This review explores the role of EVs in addressing female infertility by enhancing ovarian

Preeclampsia is one of the leading causes of maternal mortality/morbidity and preterm delivery in the world, affecting 3% to 5% of pregnant women. The pathophysiology of preeclampsia likely involves both maternal and fetal/placental factors. Abnormalities in the development of placental vessels early in pregnancy may result in placental hypoperfusion, hypoxia, or ischemia. Hypoperfusion, hypoxia, and ischemia are critical components in the pathogenesis of preeclampsia because the hypoperfused placenta transfers many factors into maternal vessels that alter maternal endothelial cell function an

The presence of fetal cells in the maternal circulation was first noted by Georg Schmorl when he documented the presence of multinucleated syncytial giant cells of placental origin in the lung tissue of women who had died from complications of eclampsia. In the intervening century, advances in cellular and molecular biology further elucidated both the physiology and pathophysiology of communication within the fetomaternal unit. This concept is at the foundation of the rapidly expanding field of noninvasive prenatal diagnosis. However, the clinical utility of this phenomenon had been limited un

Monolithic chromatography media represent a novel generation of stationary phases introduced in the last 10-15 years providing a chromatography matrix with enhanced mass transfer and hydrodynamic properties. These features allow for an efficient and fast separation of especially large biomolecules like e.g., DNA and viruses. In this study, the enrichment of virus RNA on short monolithic columns prior to molecular detection of viruses is described. Measles and mumps viruses were chosen as model viruses. The results show that it is possible to bind viral RNA on monoliths and concentrate viral nu

Formaldehyde-based fixation is the most used chemical system for histopathological examination worldwide. However, its toxicity is well known, and preservation of gross features, proteins, and nucleic acids is not optimal. Alternative fixatives resulting in similar morphological tissue quality and costs, but with reduced toxicity and with better preservation of gross features, proteins, and nucleic acids would increase operator safety and application possibilities in pathology. This multi-institutional study aimed to compare the morphological, histochemical, immunohistochemical (IHC), and mole

Conventional single-stranded antisense oligonucleotides (ASOs) work by directly binding target mRNA, which limits their use to genes that need to be silenced without affecting healthy cells. To address this limitation and achieve conditional gene silencing specifically within target cells, we developed. This system activates its RNA-cleaving function only upon binding to a specific biomarker nucleic acid sequence followed by RNase H-dependent cleavage of the targeted RNA. Under cell-free conditions, ARC showed 3-fold greater RNA cleavage efficiency in the presence of the biomarker as compared

Single-microbe RNA sequencing (smRNA-seq) emerges as a powerful approach for resolving microbial heterogeneity at single-cell resolution, but current high-throughput workflows still depend on chemical fixation to preserve cell-of-origin transcriptomic information during lysis, permeabilization, and downstream processing. Although fixation is integral to current smRNA-seq workflows, its broader impact on overall performance remains unresolved because a practical alternative is lacking. Here, we present an smRNA-seq strategy that integrates aqueous two-phase system microcapsules with our previou

INTRODUCTION: Leukemia, a heterogeneous group of hematological malignancies, is characterized by abnormal proliferation of immature hematopoietic cells. Current diagnostics primarily rely on morphological evaluation for subtype classification, methods that are subjective and labor-intensive. To overcome these limitations, a High-Content Spectral Raman Pathology Imaging platform (H-SRPI) was introduced.
METHODS: H-SRPI imaging enables profiling of proteins, nucleic acids, saturated and unsaturated lipids in leukemia. We analyzed leukemia samples from 12 patients with six distinct subtypes, alo

Antiviral agents are a cornerstone of therapeutic strategies against viral infections; however, the early-stage discovery and optimization of viral inhibitors remain time-consuming, resource-intensive, and mechanistically opaque. Here, we introduce a mechanism-guided strategy for rapid antiviral candidate identification and evaluation using confocal Raman microscopy (CRM) imaging across cellular and animal models. CRM enables label-free, quantitative, and spatially resolved assessment of drug efficacy at the single-cell level by correlating subcellular biochemical distributions, specifically o

Cell-free nucleic acids (cfNAs) carried by exosomes released from UVB-damaged skin cells act as damage-associated molecular patterns (DAMPs) that amplify sterile inflammation and contribute to sunburn, photoaging, and skin tumorigenesis. We isolated exosomes and cfNAs from UVB-irradiated human dermal fibroblasts (HDFs) and epidermal keratinocytes (NHEKs) and demonstrated that exosomes mediate the release and propagation of cfDNA, cfRNA, and cfmiRNA. These cfNAs activate endosomal Toll-like receptors (TLR3 and TLR9) and downstream NF-κB signaling in both reporter and recipient cell assays. To s

The effector component of immune system was studied in 39 cases of fibroadenomatosis by morphometric and histochemical methods. Signs of immunosuppression (decreased fraction of free cells of stroma and disturbances in their cooperation) were identified in cases of precancerous changes and cancer. A considerable difference in nucleic acid level in fibroadenomatosis and cancer was established. It is suggested that quantitative morphological study of the effector component of immune system may provide more diagnostic criteria for identification of prognostically unfavorable forms of fibroadenoma

Almost 10 years have passed since the first attempts of liquid biopsy aimed at the characterisation of tumor cells present in the bloodstream from a regular sample of peripheral blood were performed. Liquid biopsy has been used to characterise tumor heterogeneity in various types of solid tumors including adrenocortical carcinoma. The development of molecular biology, genetics, and methodological advances such as digital PCR and next-generation sequencing allowed us to use besides circulating tumor cells a variety of circulating cell-free nucleic acids, DNAs, RNAs and microRNAs secreted by tum

Systemic lupus erythematosus (SLE) is a prototypic autoimmune disease that primarily affects young women and causes a wide range of inflammatory manifestations. The hallmark of SLE is the production of antibodies to components of the cell nucleus (antinuclear antibodies [ANAs]). These antibodies can bind to DNA, RNA, and protein complexes with nucleic acids. Among ANAs, antibodies to DNA (anti-DNA) are markers for classification and disease activity, waxing and waning disease activity in many patients. In the blood, anti-DNA antibodies can bind to DNA to form immune complexes with two distinct

Gene-silencing mechanisms using RNA interference (RNAi) and antisense oligomers have drawn attention to nucleic acid medicines. However, several challenges remain such as low in vivo stability and inadequate target selectivity. Here we report a versatile and highly selective method for suppressing gene expression with a short oligonucleotide. The oligonucleotide, a Staple oligomer, hybridizes specifically to a target mRNA and artificially induces a higher-order structure, an RNA G-quadruplex (rG4), on the mRNA. This results in the rG4 effectively suppressing the target protein's translation. T

Recent advances in cell-free synthetic biology have given rise to gene circuit-based sensors with the potential to provide decentralized and low-cost molecular diagnostics. However, it remains a challenge to deliver this sensing capacity into the hands of users in a practical manner. Here, we leverage the glucose meter, one of the most widely available point-of-care sensing devices, to serve as a universal reader for these decentralized diagnostics. We describe a molecular translator that can convert the activation of conventional gene circuit-based sensors into a glucose output that can be re

Plant virus-like particles (VLPs) are promising nanotechnology platforms, but their translation requires purification strategies suitable for large-scale production. Here, we developed a nuclease-free downstream process for Physalis mottle virus (PhMV) VLPs expressed in Escherichia coli, combining ammonium sulfate precipitation, low-pH extraction, ultrafiltration/diafiltration (UF/DF), and flow-through anion-exchange (AEX) chromatography. Ammonium sulfate precipitation separated the VLPs from bulk host nucleic acids, whereas extraction at low pH removed most host cell proteins (HCPs) while ret
