A quantitative PCR-based assay reveals that nucleotide excision repair plays a predominant role in the removal of DNA-protein crosslinks from plasmids transfected into mammalian cells.

DNA-protein crosslinks (DPCs) are complex DNA lesions that induce mutagenesis and cell death. DPCs are created by common antitumor drugs, reactive oxygen species, and endogenous aldehydes. Since these agents create other types of DNA damage in addition to DPCs, identification of the mechanisms of DPC repair is challenging. In this study, we created plasmid substrates containing site-specific DPC lesions, as well as plasmids harboring lesions that are selectively repaired by the base excision or nucleotide excision repair (NER) pathways. These substrates were transfected into mammalian cells an

In vitro chromosomal aberrations induced by various shapes of multi-walled carbon nanotubes (MWCNTs).

OBJECTIVES: IARC has classified one type of multi-walled carbon nanotubes (MWCNTs), MWNT-7, as possibly carcinogenic to humans (Group 2B); however, other types of MWCNT were categorized as not classifiable as to their carcinogenicity to humans (Group 3). In vitro chromosomal aberration assays of MWNT-7 showed polyploid formation but not structural abnormalities. This study investigated the influence of the shape and size of MWCNT on in vitro induction of chromosomal aberrations. METHODS: Microscopic analysis and viable cell counting were used to assay for chromosomal aberrations and cytotoxic

Naturally occurring mutations of human corticosteroid-binding globulin.

CONTEXT: Corticosteroid-binding globulin (CBG) is encoded by SERPINA6. It is the major plasma binding protein of glucocorticoids and regulates plasma cortisol levels and bioavailability in humans. Several proteases target CBG and disrupt its steroid-binding properties. To date, most genetic deficiencies that alter plasma CBG levels or function have been identified in patients presenting with a variety of clinical conditions. OBJECTIVE: The objective of the study was to test 32 previously uncharacterized nonsynonymous, single-nucleotide polymorphisms in SERPINA6 for their ability to alter CBG

Proteomics analysis of altered cellular metabolism induced by insufficient copper level.

Insufficient copper level in the mammalian cell culture medium resulted in lactate accumulation while maintaining similar growth and culture viability profiles. Label-free, LC-MS/MS-based shotgun proteomics method was applied to compare the protein expression profiles obtained from the cultures exposed to suboptimal copper level to those provided with sufficient amount of copper. Under copper deficient condition, a substantial reduction of the protein levels of the multiple subunits of Complex IV, also known as cytochrome c oxidase, of the mitochondrial electron transport chain was observed fo

Addition of valproic acid to CHO cell fed-batch cultures improves monoclonal antibody titers.

Improving the productivity of a biopharmaceutical Chinese hamster ovary (CHO) fed-batch cell culture can enable cost savings and more efficient manufacturing capacity utilization. One method for increasing CHO cell productivity is the addition of histone deacetylase (HDAC) inhibitors to the cell culture process. In this study, we examined the effect of valproic acid (VPA, 2-propylpentanoic acid), a branched-chain carboxylic acid HDAC inhibitor, on the productivity of three of our CHO cell lines that stably express monoclonal antibodies. Fed-batch shake flask VPA titrations on the three differe

Investigation of N-aryl-3-alkylidenepyrrolinones as potential Niemann-Pick type C disease therapeutics.

A five-step synthesis of an array of N-aryl-3-alkylidenepyrrolinones, which are potential Niemann-Pick type C (NPC) disease therapeutics, is described. The synthetic route allows for the production of analogues, including photoaffinity and biotinylated derivatives. Compound 1a increased esterification by acyl-coenzyme A:cholesteryl acyltransferase in NPC1 mutant cells. It also decreased LDL uptake and increased cholesterol efflux in both NPC1-deficient and normal cells.

Toxic effects of paracetamol and related structures in V79 Chinese hamster cells.

Exposure of V79 Chinese hamster cells to non-cytotoxic concentrations of paracetamol (4-hydroxyacetanilide, 4-HAA) increased sister chromatid exchange (SCE) in the absence of an external activation system. Furthermore, a selective inhibition of DNA synthesis was observed at low 4-HAA concentrations. The inhibition could be counteracted by the addition of ascorbate, indicating that the effect is caused by an oxidation product of 4-HAA. In attempt to clarify possible relationships between cytotoxicity, inhibition of DNA synthesis and increased SCE, we studied the effect of 4-HAA and some related

Attenuation of Triple Selection Markers to Enhance mAb Expression in Tyrosine-Based Selection Systems via an Inducible CRISPRi Platform.

Chinese hamster ovary (CHO) cells are the predominant host for monoclonal antibody (mAb) production, where the choice of selection system is critical. While metabolic selection systems are favored over antibiotic-based ones to avoid purification and safety issues, traditional single-marker systems poorly regulate the light-to-heavy chain (LC/HC) ratio, which is key for yield. Although novel multi-marker systems can address this, they often lack strategies to enhance expression. To overcome these limitations, our study employed a triple-marker system based on the tyrosine biosynthetic pathway t

Multivariate phase-dependent optimization of bioprocesses boosts performance and quality-Why timing (of exposure) matters.

Applying a single parameter set to describe complex mammalian kinetics often is too simplistic, as it fails to capture sensitive cell-to-environment interactions that may be exploited to optimize production performance. To resolve this time dependency, intra-experimental parameter shifts as part of design of dynamic experiments (DoDE) can be performed to study mammalian growth and production kinetics in fed-batch processes. This enables growth phase-dependent optimization, aligned with cellular requirements. Here, we provide a comprehensive, head-to-head comparison of our phase-dependent optim

Nicotinamide Reverses the Warburg Effect in CHO Cell Culture.

The Warburg effect, the preferential conversion of glucose-derived pyruvat to lactate despite available oxygen, is a key feature of Chinese hamster ovary (CHO) cell culture. Lactate accumulation in recombinant protein-producing cell culture is an inefficient usage of glucose, as well as being deleterious to cells. Lactate accumulation lowers culture pH, requiring base addition to maintain bioreactor pH setpoint, which subsequently leads to hyperosmolarity, adversely impacting cell growth, productivity and product quality. A key driver for the Warburg effect, and hence lactate accumulation, is

Optimized NMDAR autoantibody detection via combined NR1-4a LCBA and NR1/NR2 fusion FCBA.

BACKGROUND: Autoimmune encephalitis (AE) is a group of neurological disorders mediated by autoantibodies targeting proteins within the central nervous system, of which anti-N-methyl-d-aspartate receptor (NMDAR) encephalitis is the most common. The current Cell-Based Assay (CBA) that is primarily used to detect NMDAR antibodies in clinical settings, however, exhibits several limitations, including both false-positive and false-negative results. The aim of this research is to improve the sensitivity and specificity of NMDAR antibody detection through optimization of the CBA detection model. MET

Codon and Signal Peptide Optimization to Enhance Therapeutic Antibody Production from CHO Cells.

A simple and rapid method for reproducible transient expression experiments to quickly identify the best conditions before the development of a stable CHO cell line is described, using adalimumab as a model antibody. Firstly, the signal peptide (SP) is optimized using different in silico programs, of which those having a D score of more than 0.85 will be selected. A list of useful SPs is provided. Next, guidelines for selecting the codon optimization algorithm is provided based on a CAI value of more than 0.95. After that, the codon optimized genes of the heavy chain (HC) and light chain (LC)

Native proteins from Galdieria sulphuraria to replace fetal bovine serum in mammalian cell culture.

The use of fetal bovine serum (FBS) in cell culture applications causes high costs and unacceptable animal suffering when FBS is extracted from fetal calves. Despite efforts, the exact composition of FBS still remains partially unresolved. Native proteins in FBS, such as growth factors, and their binding to cell receptors seem to be crucial for cell proliferation and differentiation. Recently, algal extracts with high protein content were considered to reduce the FBS demand. Algae extracts yielded promising results as growth serum in mammalian cell culture. Nevertheless, the dependence on resi

Engineering Chinese Hamster Ovary Cells to Mitigate Polysorbate Degradation in Biotherapeutics.

A recurring root cause for polysorbate degradation and associated particle observations in liquid biotherapeutics is the presence of residual host cell proteins (HCPs). Some of these HCPs can hydrolyze the polysorbate surfactant to release poorly-soluble fatty acids that accumulate over time and form particles. To tackle this industry-wide issue, we disrupted the genes for a plurality of potential polysorbate-degrading HCPs in CHO cells via two CHO cell engineering approaches. First, we sequentially knocked out six hydrolase genes in a stable recombinant mAb-expressing cell line and substantia

Integrated native digestion and molecular weight cutoff enrichment with DIA-MS: a sensitive and selective approach for quantitative analysis of HCP.

Residual host cell proteins (HCPs) in biopharmaceutical production processes not only compromise drug efficacy but also pose risks to patient safety and product stability, particularly when high-risk HCPs are present. Continuous monitoring of HCPs content and species during downstream purification is therefore critical. Although liquid chromatography-mass spectrometry-based proteomics has emerged as a promising approach for HCP identification, its application is hindered by the substantial dynamic range disparity between high-abundance therapeutic proteins and trace-level HCPs (at parts-per-mi

A Surface Plasmon Resonance-Based Integrated Assay for Quantification and Glycosylation Characterization of Monoclonal Antibodies in Crude Heterogeneous Samples.

The rise in cancer, autoimmune, inflammatory, and infectious diseases in recent decades has led to a surge in the development of monoclonal antibodies (mAbs) therapies, now the most widely used family of biologics. To meet the growing global demand, biopharmaceutical industries are intensifying their production processes. One approach to achieve more efficient production of effective mAbs is to develop tools for real-time quality monitoring. Specifically, the glycosylation profile of mAbs must be closely monitored, since it greatly impacts their therapeutic efficacy and innocuity, making it a

The Study of Effectiveness of Combined Proton-Neutron Irradiation of Tumor Cells In Vitro.

Combined proton-neutron therapy can be the best opportunity for neutron radiation therapy due to highly conformal proton irradiation and high relative biological effectiveness of neutrons. The study compares 4 schemes of sequential in vitro exposure of Chinese hamster fibrosarcoma cells B14-150 to 14.5 MeV neutrons and a scanning beam of protons. Treatment efficiency increased with increasing the contribution of the neutron component to the total dose from 30 to 40% and the delivery of the neutron dose as the first fraction in the two-fraction proton-neutron exposure.

Inhibition of Acid-Sensing Ion Channels by KB-R7943, a Reverse Na+/Ca2+ Exchanger Inhibitor.

KB-R7943, an isothiourea derivative, is widely used as a pharmacological inhibitor of reverse sodium-calcium exchanger (NCX). It has been shown to have neuroprotective and analgesic effects in animal models; however, the detailed molecular mechanisms remain elusive. In the current study, we investigated whether KB-R7943 modulates acid-sensing ion channels (ASICs), a group of proton-gated cation channels implicated in the pathophysiology of various neurological disorders, using the whole-cell patch clamp techniques. Our data show that KB-R7943 irreversibly inhibits homomeric ASIC1a channels het

Inhibition of Neuromyelitis Optica Immunoglobulin G Binding to Aquaporin-4 by the Small Molecule Blocker Melanthioidine.

BACKGROUND: Neuromyelitis optica (NMO) is a severe neurological demyelinating autoimmune disease affecting the optic nerves and spinal cord. The binding of neuromyelitis optica immunoglobulin G (NMO- IgG) and aquaporin-4 (AQP4) on the surface of astrocytes in the serum and cerebrospinal fluid is the main pathogenesis of NMO. Currently, therapeutic strategies for NMO include a reduction of the secondary inflammation response and the number of NMO-IgG, which can only alleviate clinical symptoms rather than fundamentally preventing a series of pathological processes caused by NMO-IgG binding to A

PgtE Enzyme of Salmonella enterica Shares the Similar Biological Roles to Plasminogen Activator (Pla) in Interacting With DEC-205 (CD205), and Enhancing Host Dissemination and Infectivity by Yersinia pestis.

Yersinia pestis, the cause of plague, is a newly evolved Gram-negative bacterium. Through the acquisition of the plasminogen activator (Pla), Y. pestis gained the means to rapidly disseminate throughout its mammalian hosts. It was suggested that Y. pestis utilizes Pla to interact with the DEC-205 (CD205) receptor on antigen-presenting cells (APCs) to initiate host dissemination and infection. However, the evolutionary origin of Pla has not been fully elucidated. The PgtE enzyme of Salmonella enterica, involved in host dissemination, shows sequence similarity with the Y. pestis Pla.
