Repurposing ibuprofen-loaded microemulsion for the management of Alzheimer's disease: evidence of potential intranasal brain targeting.

Studies have shown the use of non-steroidal anti-inflammatory drugs, such as ibuprofen could reduce the risk of Alzheimer's disease. The drug-repurposing strategy offers a bright opportunity for these patients. Intranasal administration through the olfactory pathway provides noninvasive and direct drug delivery to the target brain. A novel ibuprofen microemulsion was prepared, characterized and assessed the brain uptake in rats. The solubility of ibuprofen in various oils, surfactants, co-surfactants, and different ratios of surfactant/co-surfactant mixtures was screened and the phase diagrams

Emerging mechanisms of drug-induced phospholipidosis.

Drug-induced phospholipidosis is a lysosomal storage disorder characterized by excessive accumulation of phospholipids. Its cellular mechanism is still not well understood, but it is known that cationic amphiphilic drugs can induce it. These drugs have a hydrophilic amine head group that can be protonated in the endolysosomal compartment. As cationic amphiphiles, they are trapped in lysosomes, where they interfere with negatively charged intralysosomal vesicles, the major platforms of cellular sphingolipid degradation. Metabolic principles observed in sphingolipid and phospholipid catabolism a

Lysosomal glycogen storage mimicking the cytological picture of Pompe's disease as induced in rats by injection of an alpha-glucosidase inhibitor. I. Alterations in liver.

The present paper describes an animal model of lysosomal glycogenosis as induced by a competitive inhibitor of alpha-glucosidase. Rats received intraperitoneal injections of the inhibitor, a pseudotetrasaccharide (Acarbose, Bay g 5421); liver tissue was examined by light and electron microscopy. Substrate-histochemical and enzyme-cytochemical methods were used to demonstrate intralysosomal glycogen storage within hepatocytes and Kupffer cells. The cytological picture closely resembled that occurring in glycogenosis type II (Pompe's disease) of humans. After cessation of drug treatment, the gly

Understanding the influence of stabiliser selection and concentration on the physical stability of dual-drug co-suspensions.

In the treatment of e.g. infectious diseases, administration of multiple drugs over extended periods is common. Long-acting injectables improve patient compliance, and combining multiple drug compounds into a single suspension may provide additional benefits. This study investigated the physical stability of dual-drug co-suspensions prepared by wet bead milling, with particular focus on stabiliser selection and concentration. In total, nine stabilisers were screened in combination with itraconazole and carbamazepine as individual suspensions and in co-suspensions, followed by evaluation of sel

Bench-to-bedside: Feasibility of nano-engineered and drug-delivery biomaterials for bone-anchored implants and periodontal applications.

Nanotechnology and drug-release biomaterials have been thoroughly explored in the last few years aiming to develop specialized clinical treatments. However, it is rare to find biomaterials associated with drug delivery properties in the current dental market for application in oral bone- and periodontal-related procedures. The gap between basic scientific evidence and translation to a commercial product remains wide. Several challenges have been reported regarding the clinical translation of biomaterials with drug-delivery systems (BDDS) and nanofeatures. Therefore, processes for BDDS developm

Uropharmacology: X. Central nervous system stimulants and depressants.

Several drugs that are utilized primarily for their effects on the central nervous system also affect lower urinary tract function. Most of these effects are produced by the action of these drugs on adrenergic and cholinergic receptors or by direct action of lower urinary tract musculature. Central nervous system stimulants and depressants which are known to affect the storage or evacuation role of the lower urinary tract are discussed.

Toward preclinical evaluation of a thermosensitive PEG-poly(L-alanine) hydrogel: A study on sterilization, storage stability, and in vivo performance.

Poly(amino acid)-based thermosensitive hydrogels hold great potential for clinical translation. Herein, we employ a thermosensitive methoxy poly(ethylene glycol)-block-poly(L-alanine) (mPEG-PAla) hydrogel that undergoes a sol-to-gel transition upon heating as the model system to systematically evaluates its sterilizability, storage stability, in vivo degradation and in vivo drug release profiles-critical factors for clinical translation. mPEG-PAla copolymers are synthesized via ring-opening polymerization using the optimized amount of crown ether as the catalyst, ensuring controlled polymeriza

Harnessing dicarboxylate-based salts for enhancing the performance of amorphous solid dispersions: The case of olanzapine.

Previous studies have shown that the use of dicarboxylic acid salts can improve the performance of drug amorphous solid dispersions (ASDs), such as in the case of olanzapine (OLN). However, these studies focused only on the use of limited ASD matrices/carriers, e.g. polyvinyl alcohol, overlooking thus the critical impact of their selection on the drug's physical stability and dissolution performance. This study evaluates the performance of ASDs containing fumarate (FUM) and succinate (SUC) drug salts in comparison to ASDs prepared with the drug base, by utilizing the same model drug (i.e., OLN

Vaccine Handling Management in Korea, Japan, and Taiwan: A Comparative Study of Storage Standards, Implementation Practices, and Regulatory Approaches.

BACKGROUND: Robust vaccine cold-chain management safeguards potency and public trust. Korea, Japan, and Taiwan share core practices (2-8°C storage, continuous monitoring, first-expiry-first-out [FEFO] stock rotation) yet differ in implementation and governance. We compared standards and real-world practices across the three systems to identify actionable improvements for Korea. METHODS: From March-July 2025 we conducted structured interviews and onsite observations at public health centers, hospitals, and clinics in Korea, Taiwan (National Taiwan University Children's Hospital), and Japan (Ky

Review article: management of cyanide poisoning.

Cyanide poisoning is uncommon, but generates interest because of the presumed utility of an antidote immediately available in those areas with a high risk of cyanide exposure. As part of its regular review of guidelines, the Australian Resuscitation Council conducted a systematic review of the human evidence for the use of various proposed cyanide antidotes, and a narrative review of the relevant pharmacological and animal studies. There have been no relevant comparative or placebo-controlled human trials. Nine case series were identified. Treatment with hydroxocobalamin was reported in a tota

Structurally optimized photo-crosslinkable casein-PEG hydrogels for enhanced encapsulation and functional stability of resveratrol.

Casein-based gel delivery systems often exhibit limited loading efficiency and functional performance under conventional formulation approaches. We present a novel UV-induced photo-crosslinking strategy for generating porous casein-polyethylene glycol gels, enhancing resveratrol stabilization and enabling controlled delivery during processing and storage. Structural characterization by FTIR and 13C NMR supported the formation of an enhanced covalent network via ester and amide linkages. Formulation optimization showed that 10% casein achieved a crosslinking degree of 93.32 ± 0.43% (p < 0.05),

Hypoxic storage of red blood cells improves metabolism and post-transfusion recovery.

BACKGROUND: Blood transfusion is a lifesaving intervention for millions of recipients worldwide every year. Storing blood makes this possible but also promotes a series of alterations to the metabolism of the stored erythrocyte. It is unclear whether the metabolic storage lesion is correlated with clinically relevant outcomes and whether strategies aimed at improving the metabolic quality of stored units, such as hypoxic storage, ultimately improve performance in the transfused recipient. STUDY DESIGN AND METHODS: Twelve healthy donor volunteers were recruited in a two-arm cross-sectional stu

Efficacy of multipurpose solutions for rigid gas permeable lenses.

The use of multipurpose solutions for cleaning and disinfecting rigid gas permeable lenses has replaced single purpose solutions, but there are no reports of the efficacy of these multipurpose solutions, or of the effects of storage conditions on their disinfecting capacities. This study investigated activity against four bacterial and two fungal species, and the effects of storage in a refrigerator, at room temperature, at elevated temperature in both dry and humid conditions and with exposure to sunlight. The disinfecting solutions were challenged with the micro-organisms initially upon open

A dose-optimization trial of laronidase (Aldurazyme) in patients with mucopolysaccharidosis I.

Recombinant human alpha-l-iduronidase (Aldurazyme), laronidase) is approved as an enzyme replacement therapy to treat the lysosomal storage disorder, mucopolysaccharidosis type I (MPS I) at a dose of 0.58 mg/kg by once-weekly intravenous infusion. To assess whether alternate dosing regimens might provide a better reduction in lysosomal storage, a 26-week, randomized, open-label, multinational dose-optimization trial was conducted. The pharmacodynamic effect and safety of the approved laronidase dose was compared to three alternative regimens (1.2mg/kg every 2 weeks; 1.2mg/kg every week; 1.8 mg

Red blood cell storage time and transfusion: current practice, concerns and future perspectives.

Red blood cells (RBCs) units are the most requested transfusion product worldwide. Indications for transfusion include symptomatic anaemia, acute sickle cell crisis, and acute blood loss of more than 30% of the blood volume, with the aim of restoring tissue oxygen delivery. However, stored RBCs from donors are not a qualitative equal product, and, in many ways, this is a matter of concern in the transfusion practice. Besides donor-to-donor variation, the storage time influences the RBC unit at the qualitative level, as RBCs age in the storage bag and are exposed to the so-called storage lesion

Mephenytoin as a probe for CYP2C19 phenotyping:effect of sample storage, intra-individual reproducibility and occurrence of adverse events.

AIMS: To further evaluate mephenytoin as a probe for CYP2C19 phenotyping. METHODS: Healthy subjects (n = 2638) were phenotyped using the urinary (S)-mephenytoin to (R)-mephenytoin ratio. This method was evaluated for (a) the stability of the S/R-ratio following sample storage, (b) the intraindividual reproducibility of the ratio, and (c) the occurrence of adverse events. RESULTS: After prolonged storage, the S/R-ratio of samples from extensive metabolisers (EM) increased up to 85%. In 1.5% of the cases (1 out 66), this led to incorrect classification of phenotype. In EMs, but not in poor met

Preanalytical Factors Influencing the Stability of Ethanol in Antemortem Blood and Urine Samples.

The quantitative analysis of ethanol in blood and other biological specimens is a commonly requested service from forensic science and toxicology laboratories worldwide. The measured blood alcohol concentration (BAC) constitutes important evidence when alcohol-related crimes are investigated, such as drunken driving or drug-related sexual assault. This review article considers the importance of various preanalytical factors that might influence changes in the ethanol concentration in blood after collection and before analysis or reanalysis after various periods of storage.
