Heparan N-sulfatase: in vitro mutagenesis of potential N-glycosylation sites.

Heparan N-sulfatase cDNA contains five potential N-glycosylation sites at Asn positions 41, 142, 151, 264, and 413. We used site-directed mutagenesis, substituting the codon of asparagine for glutamine, to eliminate selected glycosylation sites and then performed expression studies in COS-7 cells to determine the influence on the catalytic activity, lysosomal targeting, and glycosylation-phosphorylation of the enzyme. Elimination of site 5 did not affect significantly enzyme activity; elimination of sites 2 and 4 gave a partial reduction, while elimination of sites 1 and 3 resulted in drastic

Enhanced stability of erythrocyte-entrapped glucocerebrosidase activity.

The stability of free glucocerebrosidase activity has been compared with the stability of glucocerebrosidase entrapped in hemolyzed, resealed erythrocytes. Encapsulation markedly stabilizes the enzyme, partly by providing an environment with a high protein concentration. In addition, in the presence of glucose, enzyme activity and GSH content remain constant, but in its absence, enzyme activity and GSH levels fall, indicating the existance of an additional, metabolically linked protective mechanism. Since the free enzyme is inactivated by p-CMB and is protected by DTT, it is likely that GSH ge

Bioenzymatic Approach to Cholesterol Detection Using Cholesterol Oxidase-Immobilized Copper-Incorporated Zeolitic Imidazolate Framework-8.

A significant challenge in creating enzyme-based electrochemical biosensors is the immobilization of enzymes onto the electrode surface while retaining their activity. High surface area, tunable porosity, biocompatibility, and stability of metal-organic frameworks (MOFs) make them a suitable matrix for the immobilization of enzymes. Besides acting as an effective immobilization platform, MOFs exhibit interesting enzyme-mimicking capabilities as well. In this study, we demonstrate dual functionality of copper-incorporated zeolitic imidazolate framework-8 (Cu@ZIF-8) both as a peroxidase mimic an

Enzymes metal-organic frameworks (MOFs) membrane: A review on design and applications.

Integrating enzyme-metal-organic frameworks (MOFs) with membranes offers a transformative approach to enhance biocatalytic applications. Combining the high porosity and stability of MOFs with the scalability and separation efficiency of membranes, these hybrid systems overcome challenges such as enzyme aggregation, recovery difficulties, and limited recyclability in traditional enzyme-MOF composites. This review examines strategies for synthesizing enzyme-MOF membranes, including bare MOF coating and interfacial diffusion methods. It evaluates their enhanced thermal and storage stability, kine

Interfacial Engineering Circumvents Activity-Stability Trade-Off in L-Threonine Aldolase for High-Yield L-threo-MTPS.

L-Threonine aldolase (LTA) is pivotal for β-hydroxy-α-amino acids (β-HAAs) synthesis but is limited by poor thermostability and activity-stability trade-off. Here, we applied interfacial engineering to Faecalimicrobium dakarense LTA, targeting nonconserved residues at subunit interfaces. The resulting variant M3 (L17F/V129R/L206W) exhibited 1.23-fold higher specific activity and significantly enhanced thermostability (T5060 + 20.5 °C; Tm + 13.1 °C). In a 1 L system, M3 produced the agrochemical precursor L-threo-4-methylsulfonyl-phenylserine in 2.49-fold higher yield than wild-type. Scaling up

Chemical Synthesis of the Mirror-Image Fast-PETase by the Enzyme-Cleavable Solubilizing-Tag Strategy.

Mirror-image poly(ethylene terephthalate) (PET) plastic-degrading enzymes have emerged as promising biocatalytic platforms due to their exceptional enzymatic stability and low immunogenicity. Currently, the sole reported mirror-image plastic-degrading enzyme, the D-form of 231-residue PET hydrolase ICCG (engineered leaf-branch compost cutinase variant), suffers from thermophilic activity requirements, which limits its practical applications. Here, the first total chemical synthesis of a mirror-image 271-residue D-Fast-PETase was presented by using an enzyme-cleavable solubilizing tag strategy.

Crosslinked Aggregates of Fusion Enzymes in Microaqueous Organic Media.

Baeyer-Villiger monooxygenases (BVMOs) are attractive for selectively oxidizing various ketones using oxygen into valuable esters and lactones. However, the application of BVMOs is restrained by cofactor dependency and enzyme instability combined with water-related downsides such as low substrate loading, low oxygen capacity, and water-induced side reactions. Herein, we described a redox-neutral linear cascade with in-situ cofactor regeneration catalyzed by fused alcohol dehydrogenase and cyclohexanone monooxygenase in aqueous and microaqueous organic media. The cascade conditions have been op

Screening of recombinant glycosyltransferases reveals the broad acceptor specificity of stevia UGT-76G1.

UDP-glycosyltransferases (UGTs) are a promising class of biocatalysts that offer a sustainable alternative for chemical glycosylation of natural products. In this study, we aimed to characterize plant-derived UGTs from the GT-1 family with an emphasis on their acceptor promiscuity and their potential application in glycosylation processes. Recombinant expression in E. coli provided sufficient amounts of enzyme for the in-depth characterization of the salicylic acid UGT from Capsella rubella (UGT-SACr) and the stevia UGT from Stevia rebaudiana (UGT-76G1Sr). The latter was found to have a remark

Phenylalanine hydroxylase from Legionella pneumophila is a thermostable enzyme with a major functional role in pyomelanin synthesis.

BACKGROUND: Legionella pneumophila is a pathogenic bacterium that can cause Legionnaires' disease and other non-pneumonic infections in humans. This bacterium produces a pyomelanin pigment, a potential virulence factor with ferric reductase activity. In this work, we have investigated the role of phenylalanine hydroxylase from L. pneumophila (lpPAH), the product of the phhA gene, in the synthesis of the pyomelanin pigment and the growth of the bacterium in defined compositions. METHODOLOGY/PRINCIPAL FINDINGS: Comparative studies of wild-type and phhA mutant corroborate that lpPAH provides the

Regulation of liver glutamate dehydrogenase by reversible phosphorylation in a hibernating mammal.

Glutamate dehydrogenase (GDH) is a key enzyme that links amino acid and carbohydrate metabolism in cells. Regulation is likely most important when organisms are confronted with extreme stresses such as the low environmental temperatures and lack of food associated with winter. Many small mammals, such as Richardson's ground squirrels, Spermophilus richardsonii, cope with these conditions by hibernating. Animals enter long periods of profound torpor where metabolic rate is greatly suppressed, body temperature drops to near-ambient and all metabolic needs must be met from fixed internal body sto

Crystallization and preliminary X-ray diffraction analysis of a thermostable D-hydantoinase from the mesophilic Bacillus sp. AR9.

D-hydantoinase catalyzes the conversion of DL-hydantoin derivatives to the corresponding optically pure N-carbamoyl amino acids, the first step in the industrial preparation of optically pure amino acids using biological catalysts. A thermostable D-hydantoinase from the mesophilic bacteria Bacillus sp. AR9 has been crystallized in three different crystal forms. The hexagonal faced crystals were the best looking, but did not diffract. One of the crystal forms is star-shaped and appeared to be twinned, but diffracted as a single crystal to a resolution of 2.3 A. These crystals belong to space gr

Discovery of a thermostable eugenol oxidase for vanillin biosynthesis from vanillyl alcohol via resting whole-cell catalysis.

Bacterial eugenol oxidases (EUGOs) belong to the 4-phenol oxidase subfamily of the VAO/PCMH flavoprotein family and are versatile biocatalysts offering potential for use in biotechnology and organic synthesis. In this work, a thermostable EUGO from Capillimicrobium sp. (CsEUGO) was identified and overexpressed in E. coli cells as a whole-cell biocatalyst for the biosynthesis of vanillin from vanillyl alcohol. The E. coli-CsEUGO cells oxidized vanillyl alcohol aerobically, producing only vanillin, indicating excellent reaction selectivity. Furthermore, the cells retained 63% and 57% of their ac

[Rational design for improving the thermostability of β-glucosidase from Bacillus thermoamylovorans].

The β-glucosidase from Bacillus thermoamylovorans (Bgl52) has been identified as a thermophilic and acidophilic β-glucosidase, which constitutes the most critical component in the cellulolytic enzyme system for cellulose hydrolysis. Although Bgl52 is a thermophilic enzyme, its poor thermal stability at high temperatures restricts its long-term industrial application under elevated-temperature conditions. In this study, rational design strategies, including two free energy calculation programs (Fireprot and FoldX) and homologous sequence alignment, were integrated to perform single-point and mu

Multi-substrate sequential optimization, characterization and immobilization of lipase produced by Pseudomonas plecoglossicida S7.

Lipases are important biocatalysts having the third largest global demand after amylases and proteases. In the present study, we have screened 56 potential lipolytic Pseudomonas strains for their lipolytic activity. Pseudomonas plecoglossicida S7 showed highest lipase production with specific activity of 70 U/mg. Statistical optimizations using Plackett Burman design and response surface methodology evaluated fourteen different media supplements including various oilcakes, carbon sources, nitrogen sources, and metal ions which led to a 2.23-fold (156.23 U/mg) increase in lipase activity. Contribution of subdomain structure to the thermal stability of the cholera toxin A1 subunit.

The catalytic A1 subunit of cholera toxin (CTA1) is an ADP-ribosyltransferase with three distinct subdomains: CTA1(1) forms the catalytic core of the toxin, CTA1(2) is an extended linker between CTA1(1) and CTA1(3), and CTA1(3) is a compact globular region. CTA1 crosses the endoplasmic reticulum (ER) membrane to enter the cytosol where it initiates a cytopathic effect. Toxin translocation involves ER-associated degradation (ERAD), a quality control system that exports misfolded proteins from the ER to the cytosol. At the physiological temperature of 37 °C, the free CTA1 subunit is in a partial

Properties of sialidase isolated from Actinomyces viscosus DSM 43798.

The cell-bound sialidase of Actinomyces viscosus DSM 43798 was solubilized by mechanical cell disruption and lysozyme treatment. The enzyme was enriched 30,000-fold by cation-exchange chromatography, gel-filtration, and FPLC ion-exchange chromatography, thus obtaining 10 micrograms sialidase protein from 26 g wet cells with a specific activity of 680 U/mg protein. Since sialidase activity was also found in the culture medium, this enzyme was isolated as well, requiring the additional application of FPLC gel-filtration. Both sialidase preparations were apparently homogenous on SDS-PAGE and have

Assessment of PEG-Asparaginase and Erwinia Asparaginase Activity Under Variable Storage Conditions.

Background: PEG-Asparaginase and Erwinia asparaginase are enzyme-based anticancer therapies used in the treatment of acute lymphoblastic leukaemia (ALL) and lymphoblastic lymphoma (LBL), where adequate plasma enzyme activity is required for therapeutic efficacy. In many study groups, therapeutic drug monitoring is routinely applied due to pharmacokinetic variability and the risk of hypersensitivity reactions followed by increased clearance and insufficient treatment. In clinical practice, samples may be exposed to prolonged transport and variable pre-analytical conditions. Knowledge on pre-ana

Prediction of improved therapeutics for fabry disease patients generated by mutagenesis of the α-galactosidase A active site, dimer interface, and glycosylation region.

Fabry disease is an X-linked lysosomal storage disorder caused by the deficiency of the enzyme, α-galactosidase A that induces the accumulation of the substrate globotriaosylceramide. Currently approved enzyme replacement therapy using recombinant human α-galactosidase A improves patient symptoms but a majority of patients experience adverse events due to the multiple infusions required for full therapeutic efficacy. Our approach is to use medicinal chemistry and phylogenic comparisons to introduce mutations into the human enzyme to increase catalytic activity and/or stability to generate an i

Heat inactivation and reactivation of broccoli peroxidase.

Heat inactivation characteristics differed for acidic (A), neutral (N), and basic (B) broccoli peroxidase. At 65 degrees C, A was the most heat stable followed by N and B. The activation energies for denaturation were 388, 189, and 269 kJ/mol for A, N, and B, respectively. Reactivation of N occurred rapidly, within 10 min after the heated enzyme was cooled and incubated at room temperature. The extent of reactivation varied from 0 to 50% depending on the isoenzyme and heating conditions (temperature and time). The denaturation temperature allowing the maximum reactivation was 90 degrees C for

Production and regulation of a thermostable protease by Pseudomonas sp. B45.

A Pseudomonas sp. produced an extracellular thermostable protease which required induction by peptone. Growth of the organism and the production of protease was optimum at 30 degrees C. The enzyme was subjected to catabolite repression by glucose. Both chloramphenicol and rifamycin completely abolished protease production indicating de novo synthesis of the enzyme. Leucine, lysine, histidine and glycine enhanced the protease production considerably and they were the most effective when added during the active period of production. Glucose repression could not be relieved by addition of leucine
