The stimulation of GLP-1 secretion and delivery of GLP-1 agonists via nanostructured lipid carriers.

Nanoparticulate based drug delivery systems have been extensively studied to efficiently encapsulate and deliver peptides orally. However, most of the existing data mainly focus on the nanoparticles as a drug carrier, but the ability of nanoparticles having a biological effect has not been exploited. Herein, we hypothesize that nanostructured lipid carriers (NLCs) could activate the endogenous glucagon-like peptide-1 (GLP-1) secretion and also act as oral delivery systems for GLP-1 analogs (exenatide and liraglutide). NLCs effectively encapsulated the peptides, the majority of which were only

Glucagon-Like Peptide-1 Agonist vs. Placebo and Pulmonary Decline After Open-Heart Surgery: A Substudy of the GLORIOUS Randomised Clinical Trial.

BACKGROUND: Postoperative pulmonary decline is an established complication of open-heart surgery extending beyond the immediate postoperative phase. Inflammation-mediated lung damage and ischaemia-reperfusion injury secondary to extracorporeal circulation is a proposed pathophysiological driver. GLP-1 receptor agonists (GLP-1RA) have emerged as promising protective agents in this setting. AIM: Investigate whether infusion of the GLP-1RA, exenatide during cardiopulmonary bypass and weaning thereof, can mitigate the decline in diffusing capacity and ventilatory performance 3 months postoperativ

Tumour Risk with Once-Weekly Glucagon-Like Peptide-1 Receptor Agonists in Type 2 Diabetes Mellitus Patients: A Systematic Review.

BACKGROUND AND OBJECTIVE: Once-weekly glucagon-like peptide-1 receptor agonists (GLP-1RAs) are a novel class of injectable antidiabetic drugs. Previous studies indicated that GLP-1RAs (exenatide and liraglutide) might increase the incidence of pancreatitis and pancreatic cancer. Here, we evaluated the clinical safety of once-weekly GLP-1RAs with respect to tumour risk. METHODS: Relevant studies were selected from ClinicalTrials.gov. Randomized controlled trials that reported the incidences of neoplasms were included in our research. Outcomes were calculated as the risk ratio using the Mantel-

Microbial expression of Exendin-4 analog and its efficacy in mice model.

Exendin-4 is a GLP 1 agonist incretin-mimetic peptide hormone comprising 39 amino acids. Exenatide (Byetta®) is a chemically synthesized version of Exendin-4 with an additional C-terminal amidation. Exenatide acts as a GLP-1 receptor agonist. This paper illustrates the method adopted for cloning, fermentation and purification of recombinant Exendin-4 analog expressed in Escherichia coli. The biologically expressed analog was extensively characterized using different orthogonal methods to confirm their biological activity and physicochemical properties. It was observed that the expressed analog

GLP-1 receptor agonists: effects on cardiovascular risk reduction.

Comorbid obesity, dyslipidemia, and hypertension place patients with type 2 diabetes (T2DM) at greatly increased risk of cardiovascular (CV) disease-related morbidity and mortality. An urgent need exists for effective treatment for patients with T2DM that encompasses glycemic control, weight loss, and reduction in CV risk factors. The glucagon-like peptide-1 receptor agonists (GLP-1 RAs) liraglutide and exenatide are incretin-based antidiabetes agents. This review examines CV-associated effects of liraglutide and exenatide in animal models and clinical trials with patients with T2DM. Studies s

GLP-1 Receptor Agonists: Practical Considerations for Clinical Practice.

PURPOSE: Type 2 diabetes (T2D) imparts an increased risk of adverse health outcomes in patients unable to achieve glycemic control. Patient education and individualization of treatment are important for effective management of T2D. Glucagon-like peptide-1 receptor agonists (GLP-1RAs) are a class of injectable glucose-lowering agents that lower A1C with added benefits of weight loss and improved cardiovascular risk markers. This review discusses the role of GLP-1RAs currently approved in the United States (exenatide, liraglutide, albiglutide, dulaglutide) for T2D management and characterizes th

Injection-site and dermatologic reactions associated with glucagon-like peptide-1 receptor agonists: Insights from meta-analysis of randomised controlled trials and real-world evidence.

AIMS: Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) are widely used for type 2 diabetes mellitus and obesity, with once-weekly dosing that supports adherence. However, injection-site reactions (ISRs) and dermatologic events have been recognised, ranging from mild local events to rare systemic hypersensitivity reactions that may cause discontinuation. To evaluate dermatologic and ISR safety of GLP-1 RAs through a meta-analysis of randomised controlled trials (RCTs) and disproportionality analysis of data from the United States Food and Drug Administration Adverse Event Reporting System

Development and validation of qualitative methods for the detection of GLP-1 receptor agonists in serum and dried blood spots by LC-HRMS/MS for doping control purposes.

The increasing use of glucagon-like peptide-1 (GLP-1) receptor agonists for weight management has raised concerns about their potential misuse in sports, where effects such as weight loss, glucose-dependent insulin secretion, and enhanced mitochondrial biogenesis may offer performance advantages. Although currently not prohibited, semaglutide and tirzepatide have been included in the World Anti-Doping Agency (WADA) Monitoring Program to assess potential misuse. In response to these concerns, this study aimed to develop detection methods for five GLP-1 receptor agonists as well as the endogenou

Glucagon-Like Peptide-1 Receptor Analogues in Type 2 Diabetes: Their Use and Differential Features.

Glucagon-like peptide-1 receptor analogues/agonists (GLP-1RAs) are well established as effective adjuncts to lifestyle modification in the treatment of type 2 diabetes (T2D) as monotherapy or in combination with oral glucose-lowering drugs ± insulin. The six subcutaneous GLP-1RA formulations (i.e. twice-daily exenatide, once-daily liraglutide and lixisenatide, and once-weekly dulaglutide, exenatide and semaglutide) currently available in the EU and USA have many similarities, but also some unique features and properties. By stimulating GLP-1 receptors, GLP-1RAs increase insulin secretion and s

Acute glucagon-like peptide-1 receptor agonist, semaglutide, attenuates cue-, drug-, and stress-induced fentanyl seeking in male Sprague-Dawley rats.

Opioid overdose deaths continue to be a significant public health problem worldwide, with fentanyl, the primary driver of these losses of life. Indeed, in the USA, overdose deaths continue despite the availability of three food and drug administration-approved medications for the treatment of opioid use disorder (OUD) due to minimal access to these medications and to stigma. Fortunately, a growing preclinical and clinical literature suggests that glucagon-like peptide-1 receptor (GLP-1R) agonists may serve as new treatments for OUD. Exendin-4/exenatide and liraglutide have been found to reduce

Potential of Glucagon-Like Peptide-1 (GLP-1) Receptor Agonists in Mitigating Metabolic Side Effects Associated With Clozapine or Olanzapine Therapy: A Literature Review.

Clozapine and Olanzapine can cause severe metabolic side effects, increasing cardiovascular risk. Glucagon-like peptide-1 (GLP-1) receptor agonists, primarily approved for use in patients with type 2 diabetes, may counter these effects by promoting weight loss, improving glucose metabolism, and enhancing cardiovascular health, offering a promising approach for managing antipsychotic-related metabolic complications. We conducted a literature review using the search terms "clozapine OR olanzapine AND GLP-1 agonist," "schizophrenia AND GLP-1 agonist," "antipsychotic AND GLP-1 agonist," and "psych
