Modern approaches to non-surgical biofilm management.

The subgingival dental plaque is a microbial biofilm consisting of highly variable bacterial microcolonies embedded within a self-produced matrix of extracellular polymeric substance. In contrast to microorganisms growing in a planktonic state, the inhabitants of a biofilm are effectively protected within this dense structure from host defense mechanisms and from therapeutic agents, including antimicrobials. The mechanical removal of the microbial biofilm and the establishment of meticulous plaque control measures comprise the key elements for the success of non-surgical periodontal treatment.

Comparative fermentation performance of biofilm and planktonic Saccharomyces cerevisiae under standard and high-carbon media.

Biofilms are structured microbial communities with wide-ranging industrial applications. In Saccharomyces cerevisiae, biofilm formation can influence fermentation, improve microbial stability, and enhance production efficiency. This study evaluates the fermentation performance of S. cerevisiae in both planktonic and biofilm states under standard and high-carbon media. During a 96-hour fermentation, planktonic cultures consumed glucose within 24 h, whereas biofilm-associated cells required 48 h due to diffusion constraints within the extracellular polymeric substance (EPS) matrix. Under standar

Antimicrobial and Antibiofilm Activity of Disubstituted Bis-benzimidazolium Salts.

The increased prevalence of antibiotic-resistant bacteria is a critical issue for human health. Developing new antibiotic agents is vital for fighting persistent infections and lowering mortality rates. In this study, we designed lutidine-disubstituted bis-benzimidazolium salts (lutidine-bis-benzimidazolium core with octyl, adamantyl, and cholesteryl lipophilic side chains), and tested their antimicrobial activity, their capacity to inhibit planktonic bacterial and fungal growth, and their ability to inhibit the formation of or disrupt mature methicillin-resistant Staphylococcus aureus (MRSA)

The roles of extracellular polymeric substances of Pandoraea sp. XY-2 in the removal of tetracycline.

In this study, the roles of extracellular polymeric substances (EPSs) excreted by Pandoraea sp. XY-2 in the removal of tetracycline (TC) were investigated. In the early stage, TC in the solution was mainly removed by the adsorption of EPSs, which accounted for 20% of TC. Thereafter, large amount of TC was transported into the intracellular and biodegraded. EPSs was extracted and the contents of polyprotein and polysaccharides reached their maximum values (30.84 mg/g and 11.15 mg/g) in the first four days. Fourier transform infrared spectroscopy analysis revealed that hydroxyl, methylidyne, met

Biofilm-Forming Methicillin-Resistant Staphylococcus aureus: A Comprehensive Phenotypic and Genotypic Review.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major global pathogen, capable of forming biofilms that confer enhanced antimicrobial tolerance, virulence, and persistence in both hospital and community settings. Biofilms are structured microbial communities encased in extracellular polymeric substances, composed of polysaccharides, proteins, and extracellular DNA. Complex genetic networks, including the ica operon, the agr quorum-sensing system, the sarA regulatory system, and stress-response genes such as sigB, regulate MRSA biofilm formation. Phenotypic methods (Congo red agar, micr

Sugar supplementation enhances biofilm formation and extracellular polysaccharides production in Sulfobacillus acidophilus.

Sulfobacillus is an acidophilic bacterial genus that plays a key role in bioleaching of metal sulfides at moderately thermophilic temperatures (45-55 °C), owing to its mixotrophic metabolism and capacity to form biofilms. Biofilm development depends on an extracellular polymeric substances (EPS) matrix, which is largely composed of polysaccharides. However, the composition and function of polysaccharides in sulfobacilli remain poorly characterized. A genome-wide survey identified a diverse repertoire of putative sugar uptake transporters, belonging to three distinct families. We demonstrate th

An update on biofilms in acute and chronic wounds: Incidence, clinical evidence, diagnosis, prevention, and treatment.

Pathogenic biofilms are a critical barrier to wound healing, driving infection persistence, delayed tissue repair, and antimicrobial resistance (AMR) and tolerance. This recognition has driven a paradigm shift from a traditional planktonic model of infection to a biofilm-mediated framework, where structured microbial communities are encased within a protective extracellular polymeric substance (EPS) matrix. These communities are highly prevalent in chronic wounds, with reported detection rates of 60%-100% across diabetic foot ulcers, venous leg ulcers, and pressure injuries. The establishment

Electro-biochemical profiles of the boosted CO2-to-acetate conversion in microbial electrosynthesis using FeS-Hydrochar modified cathodes.

To mitigate CO2 emissions, microbial electrosynthesis (MES) with rationally designed cathodes offers a promising route for value-added chemical production. Herein, an FeS-loaded algae-derived hydrochar (FeSHC) biocathode was developed by coupling redox-active FeS with a conductive and biocompatible hydrochar matrix rich in surface functional groups. This structural design enabled homogeneous FeS anchoring, enhanced interfacial electron transfer, and improved microbe-electrode interactions. Consequently, the FeSHC biocathode achieved an acetate concentration of 1212.52 mg·L-1, representing a 76

Weaponizing volatiles to inhibit competitor biofilms from a distance.

The soil bacterium Bacillus subtilis forms beneficial biofilms that induce plant defences and prevent the growth of pathogens. It is naturally found in the rhizosphere, where microorganisms coexist in an extremely competitive environment, and thus have evolved a diverse arsenal of defence mechanisms. In this work, we found that volatile compounds produced by B. subtilis biofilms inhibited the development of competing biofilm colonies, by reducing extracellular matrix gene expression, both within and across species. This effect was dose-dependent, with the structural defects becoming more prono

Unveiling the chemical composition of a microbiota-derived extracellular polymeric substance and its antivirulence potential against Staphylococcus aureus from bovine mastitis.

Bovine mastitis caused by Staphylococcus aureus represents a major economic and health challenge in the dairy industry due to the pathogen's virulence factors and biofilm-forming ability, which confer resistance to conventional antibiotics. Sustainable alternatives are therefore urgently needed in the context of rising antimicrobial resistance. This study evaluated the chemical composition, surface interaction properties, and antivirulence potential of an extracellular polymeric substance (EPS-H21) produced by commensal Bacillus sp. strain H21, previously isolated from the microbiota of health

Macrophage-membrane-camouflaged lipid-polymer hybrid nanoparticles for target-specific biofilm penetration and eradication of methicillin-resistant staphylococcus aureus.

Methicillin-resistant Staphylococcus aureus (MRSA) biofilms present a formidable therapeutic challenge due to the dense extracellular polymeric substance (EPS) matrix that restricts antibiotic penetration. Here, we engineered biomimetic macrophage-membrane-camouflaged lipid-polymer hybrid nanoparticles (M-LPHNs) designed for targeted delivery and enhanced eradication of MRSA biofilms. M-LPHNs were prepared via emulsification-sonication followed by extrusion-mediated cell membrane coating, yielding monodisperse core-shell spherical structures (∼150 nm, PDI < 0.20) with high colloidal stability

From micro to macro: The role of seawater in maintaining structural integrity and bioactivity of granules in treating antibiotic-laden mariculture wastewater.

Granular sludge (GS) has superior antibiotic removal ability to flocs, due to GS's layered structure and rich extracellular polymeric substances. However, prolonged exposure to antibiotics degrades the performance and stability of GS. This study investigated how a seawater matrix might help maintain the structural integrity and bioactivity of granules. The results demonstrated that GS had better sulfadiazine (SDZ) removal efficiency in a seawater matrix (85.6 %) than in a freshwater matrix (57.6 %); the multiple ions in seawater enhanced boundary layer diffusion (kiR1 = 0.0805 mg·g-1·min-1/2 a

Biodegradation of carbon tetrachloride from groundwater in an upflow solid-phase biofilm system.

In the present study, an upflow solid-phase denitrification biofilm reactor (US-DBR) was applied for simultaneous carbon tetrachloride (CT) and nitrate removal from groundwater by using poly(butylene succinate) (PBS) as carbon source and biocarrier. After 80 days continuous operation, the nitrate and CT removal efficiencies in the biofilm reactor were high of 98% and 94.3%, respectively. After PBS-biofilm formation, protein (PN) content in loosely bound extracellular polymeric substances (LB-EPS) and tightly bound EPS (TB-EPS) significantly increased 2.6 and 4.0 times higher in the presence of

Desorption of Hg(II) and Sb(V) on extracellular polymeric substances: effects of pH, EDTA, Ca(II) and temperature shocks.

Extracellular polymeric substances (EPS) existed ubiquitously in biological systems affect the mobility and availability of heavy metals in the environments. The adsorption-desorption behaviors of Hg(II) and Sb(V) on EPS were investigated. The sorption rates follow Sb(V) > Hg(II), and the desorption rates follow reverse order. Applications of ethylene diamine tetraacetic acid (EDTA), Ca(II) and pH shocks affect desorption rates and desorbed quantities of Hg(II) from EPS-Hg complex. Temperature shock minimally affects the desorption rate of Hg(II). Conversely, the EPS-Sb complex is stable subje

Phosphoenolpyruvate phosphotransferase system components positively regulate Klebsiella biofilm formation.

BACKGROUND/PURPOSE: Klebsiella pneumoniae is one of the leading causes of device-related infections (DRIs), which are associated with attachment of bacteria to these devices to form a biofilm. The latter is composed of not only bacteria but also extracellular polymeric substances (EPSes) consisting of extracellular DNAs, polysaccharides, and other macromolecules. The phosphoenolpyruvate (PEP):carbohydrate phosphotransferase system (PTS) regulates diverse processes of bacterial physiology. In the genome of K. pneumoniae MGH 78578, we found an uncharacterized enzyme II complex homolog of PTS: KP

Engineered bacterial biofloc formation enhancing phenol removal and cell tolerance.

A microbial floc consisting of a community of microbes embedded in extracellular polymeric substances matrix can provide microbial resistances to toxic chemicals and harsh environments. Phenol is a toxic environmental pollutant and a typical lignin-derived phenolic inhibitor. In this study, we genetically engineered Escherichia coli cells by expressions of diguanylate cyclases (DGCs) to promote proteinaceous and aliphatic biofloc formation. Compared with the planktonic E. coli cells, the biofloc-forming cells improved phenol removal rate by up to 2.2-folds, due to their substantially improved

Electron Lever-Assisted d-Band Center Engineering in Intermetallic Alloy Nanozymes for Efficient Marine Microbiologically Influenced Corrosion Inhibition.

Microbiologically influenced corrosion (MIC) remains a persistent global challenge, with traditional inhibition strategies often constrained by limited efficiency, poor durability, and ecological toxicity. Herein, we propose a rational design that utilizes a heteroatom as an electronic lever to circumvent these constraints. By integrating heteroatom Ag into an FePt matrix, the d-band center of the Pt active sites is upshifted, optimizing the adsorption energy of substrates and ensuring superior catalytic performance even in H2O2-limited marine environments. The resulting FePtAg (L-FPA) nanozym
