Prenatal diagnosis of cor triatriatum sinister in association with hypoplastic left heart syndrome.

Cor triatriatum sinister is a rare congenital heart defect related to incomplete common pulmonary vein resorption into the primitive heart. This lesion usually presents with pulmonary venous obstruction and can occur in association with left-sided obstructive lesions such as hypoplastic left heart syndrome (HLHS). In the context of HLHS, the presence of cor triatriatum sinister carries additional surgical and prognostic implications. Fetal diagnosis can enable appropriate counseling of the family and guide optimal peri- and postnatal management. The reported case represents the first prenatal

Jianwei Shoutai Pills alleviates miscarriage by modulating gut microbial production of BAs and NLRP3-inflammasome at the maternal-fetal interface of rats.

BACKGROUND: Miscarriage has the characteristics of recurrent attacks and complex etiology, so it is gradually attracted the wide attention of scholars in the fields of reproduction. Potential association between gut microbiome (GM) and pregnancy disorders has been investigated. Jianwei Shoutai pills (JWP), as a representative formula, have been proven to have protective effect in both clinical and experimental research in miscarriage. However, the specific mechanism of JWP in miscarriage through GM remains unclear. PURPOSE: To investigate the underlying mechanism of JWP against miscarriage th

The effect of acute maternal toxicity on fetal development in the mouse.

The effects of acute alterations in maternal health status upon fetal development were assessed following exposure of pregnant CD-1 mice on day 8 of gestation to one of ten chemicals at doses calculated to exert either a low or a moderate degree of maternal lethality. The dams were killed on day 18 of gestation, and the fetuses were examined by routine teratological techniques. The chemicals were cacodylic acid, caffeine, deltamethrin, dinoseb, ethylene bisisothiocyanate sulfide (EBIS), endrin, guthion, kepone, sodium salicylate, and toxaphene. Three (cacodylic acid, EBIS, and kepone) produced

Fetal outcome following maternal treatment with prostaglandin E2.

Pregnant Sprague-Dawley rats were treated subcutaneously with PGE2 at doses of 50, 100 and 200 micrograms on gestational days 5 through 8. Control animals received physiological saline in a similar manner. On day 20 of gestation the animals were anaesthetized and fetuses were removed by hysterotomy. All fetuses recovered were alive and appeared normal. The incidence of fetal resorptions was not significantly increased. Visceral abnormalities and skeletal defects were not observed in the offspring. Also, the mean weight of treated fetuses was not significantly different from that of the corresp

The embryotoxicity of gossypol.

In utero development was analyzed in pregnancies that resulted from matings between gossypol-treated male rats and untreated female rats and in pregnancies in which gossypol was administered to the pregnant rat only. Gossypol treatment of males had no effect on the outcome of pregnancy. There was no significant effect on resorption, fetal growth, or malformation rate. Similarly, gossypol administered to pregnant dams at stages during organogenesis had no observable effect on pregnancy. Under the conditions of this experiment, gossypol administered to either the breeding male rat or the pregnan

Amelioration of TCDD-induced teratogenesis in aryl hydrocarbon receptor (AhR)-null mice.

The aryl hydrocarbon receptor (AhR) mediates many of the biological effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and transcriptional activation of genes encoding a number of xenobiotic metabolizing enzymes. Prenatal exposure of mice to TCDD causes severe alterations in embryo and fetal development, including hydronephrosis and cleft palate. However, the mechanisms underlying these effects are unclear. In this work, the teratogenicity of TCDD in AhR-null mice was evaluated to determine if this effect is mediated by the AhR. Homozygous wild-type (+/+) or AhR-null (-/-) female mice were

Modulation of embryonic glutathione reductase and phenytoin teratogenicity by 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU).

The teratogenicity of phenytoin may be mediated through a reactive electrophilic and/or free radical intermediate which, if not detoxified, may interact with fetal cellular macromolecules and initiate teratologic effects. Glutathione (GSH) maintains cellular physiological processes and detoxifies xenobiotic reactive intermediates. The role of GSH in protection against phenytoin embryopathy was studied by altering GSH homeostasis using 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU), an inhibitor of glutathione reductase. BCNU was administered to pregnant CD-1 mice on gestational day 12, in doses r

Use of bone cell cultures to study skeletal pathology.

We describe procedures for the isolation, culture, and analysis of neonatal osteoblasts from osteopetrotic (toothless (tl) and osteopetrosis [op]) rats and normal littermates. Normal osteoblasts produce and mineralize an extracellular matrix indistinguishable from that of well-characterized fetal rat osteoblasts in vitro. Mutant (tl and op) cultures show an early abnormal pattern of cell proliferation and a later premature, extensive mineralization which mimic the mutant phenotype in vivo. In cocultures with normal osteoclasts, mutant (tl) osteoblasts also show a greatly reduced ability to orc

Papillary Traction Due to the Posterior Form of a Bilateral Persistent Fetal Vasculature Mimicking Papilledema.

Persistent fetal vasculature (PFV), or persistent hyperplastic primary vitreous (PHPV), is a congenital developmental disorder characterized by a failure of resorption of the hyaloid system. It typically presents unilaterally and has three forms: anterior, posterior, and mixed. In this case report, a seven-year-old patient, without specific personal or family medical history, was referred from the pediatric department for bilateral papilledema. The patient had a best-corrected visual acuity of 20/20 (Logarithmic Measure of Angle of Resolution (LogMAR): 0) in both eyes. Fundus examination of bo

Variable in vivo embryoprotective role for ataxia-telangiectasia-mutated against constitutive and phenytoin-enhanced oxidative stress in atm knockout mice.

Knockout mice lacking the ataxia-telangiectasia-mutated (Atm) protein exhibit impaired detection and repair of DNA damage and increased embryopathies from ionizing radiation in vivo, and vehicle or phenytoin in embryo culture. Here we determined if Atm-deficient mice are more susceptible in vivo to phenytoin embryopathies. Wild-type (+/+) or heterozygous (+/-) Atm knockout dams were mated with +/- males, pregnant dams were treated with phenytoin (65 mg/kg ip) or its vehicle, and resorptions and fetuses were genotyped and characterized. This strain proved resistant to phenytoin-initiated cleft

The pathogenic role of anti-phosphatidylserine antibodies: active immunization with the antibodies leads to the induction of antiphospholipid syndrome.

Primary antiphospholipid syndrome (pAPS) can be experimentally induced in mice by immunization with anti-cardiolipin (aCL) antibodies (Abs). Recently, we have pointed to the pathogenic role of antiphosphatidylserine (aPS) Abs by inducing an experimental model of pAPS in naive mice following passive transfer of human aPS to the tail vein of ICR mice. The aim of the present study was to induce experimental pAPS in mice following active immunization with aPS. Mice were immunized with IgG and IgM aPS, purified from two patients with pAPS. The sera of the mice were examined for the presence of diff

Toxic effects of 70% ethanol extract of Moringa stenopetala leaf (Baker f.) Cufod. (Moringaceae) on fetus and placenta of pregnant Wistar rats.

BACKGROUND: Moringa stenopetala leaves (Baker f.) Cufod. (Moringaceae) are used as a staple food and traditional medicine for treating various diseases like malaria, hypertension, stomach pain, diabetes, elevated cholesterol, and removing the retained placenta. Its prenatal toxicity study is minimal. Thus, this study aimed to assess the toxic effects of a 70% ethanol extract of Moringa stenopetala leaf on the fetuses and placentas of pregnant Wistar rats. METHOD: Fresh leaves of Moringa stenopetala were collected, dried at room temperature, ground to powder, and extracted using 70% ethanol. F

Experimental formation of cleft palate in mice with polychlorinated biphenyls (PCB).

The teratological effect of polychlorinated biphenyls (PCB) was examined using the ddY strain of mouse. PCB in 0.05 ml ethanol was injected daily, subcutaneously into the back of pregnant mice, for 10 days from day 6 of gestation. Cleft palates were found in fetuses at the 18th gestation, with a significant dose response between 10 mg and 50 mg as total body dose of PCB per pregnant mouse. Comparison of these results with simultaneous observations of body weight of dam or fetus, and number of resorbed and dead fetuses, indicated that cleft palate formation was due mainly to the specific effect

Thymic and extrathymic Aire-expressing cells in maternal-fetal tolerance.

Healthy pregnancy requires maternal immune tolerance to both fetal and placental tissues which contain a range of self- and non-self-antigens. While many of the components and mechanisms of maternal-fetal tolerance have been investigated in detail and previously and thoroughly reviewed (Erlebacher A. Annu Rev Immunol. 2013;31:387-411), the role of autoimmune regulator (Aire), a critical regulator of central tolerance expressed by medullary thymic epithelial cells (mTECs), has been less explored. Aire is known to facilitate the expression of a range of otherwise tissue-specific antigens (TSAs)

Functions of alpha-melanotropin and other opiomelanocortin peptides in labour, intrauterine growth and brain development.

In a number of animals and in humans, factors from the fetal hypothalamus function in intrauterine growth, in labour and in brain development. Peptides of the opiomelanocortin family are produced by the pituitary, brain and placenta and are probably involved in these developmental processes. In the rat, alpha-MSH stimulates fetal growth, protein synthesis, wound healing and liver regeneration and it reduces periosteal bone resorption. In chick embryos, alpha-MSH restores the corticosteroid-induced growth retardation. Thus alpha-MSH seems to possess general trophic properties. The fetal brain i

Citrinin and endosulfan induced teratogenic effects in Wistar rats.

Dietary exposures to food pollutants such as mycotoxin(s) or pesticide(s) are most significant due to their adverse effects on the production and reproduction in animals and the human population. The present investigation was conducted to evaluate the teratogenic potential of citrinin (CIT) and endosulfan either alone or in combination in pregnant rats during gestational days 6-20. Endosulfan (1 mg kg(-1) body weight, by oral intubation) and CIT (10 mg kg(-1) feed, through diet) when administered either alone or in combination in pregnant rats caused significant teratogenic effects in the deve

Increased accumulation of sorbitol in offspring of manifest diabetic rats.

The effects of maternal diabetes on somatic development and activity of the polyol pathway were investigated during early and late gestation in a rat model for diabetic pregnancy. We studied embryo-fetal growth, mortality, and malformation rate in the offspring of nondiabetic rats and in the offspring of diabetic rats either treated with an aldose reductase inhibitor during gestation or left untreated. The numbers of embryo-fetal resorptions and malformations were significantly increased in the diabetic groups compared with the controls despite maternal treatment with the aldose reductase inhi

Effects of low concentrations of nitrous oxide on rat fetuses.

The long-term inhalation of air polluted with nitrous oxide has been implicated in spontaneous abortions and fetal abnormalities in operating room personnel. Studies with 1% and 0.5% N2O in air have confirmed the adverse effects of dilute concentrations of N2O in pregnant rats. The present study was designed to determine the threshold concentration of nitrous oxide necessary to produce adverse responses in pregnant rats. Wistar albino rats were mated in an environmental chamber and conception confirmed by vaginal smears. Immediately thereafter, the rats were randomly divided into control and e

Relation of in vivo drug metabolism to stereoselective fetal hydantoin toxicology in mouse: evaluation of mephenytoin and its metabolite, nirvanol.

Anticonvulsant therapy during pregnancy with the hydantoin phenytoin carries a risk of teratologic sequelae and developmental retardation known as the fetal hydantoin syndrome. The putative teratogen is a highly reactive arene oxide intermediate produced during phenytoin metabolism. By using two structurally similar hydantoins, mephenytoin and its in vivo demethylated metabolite nirvanol, we examined the possibility for a stereoselective dissociation of fetal hydantoin toxicity from maternal anticonvulsant activity. The d-isomers (S-enantiomers) of mephenytoin and nirvanol are p-hydroxylated v
