Dopamine D2 receptors in addiction-like reward dysfunction and compulsive eating in obese rats.

We found that development of obesity was coupled with emergence of a progressively worsening deficit in neural reward responses. Similar changes in reward homeostasis induced by cocaine or heroin are considered to be crucial in triggering the transition from casual to compulsive drug-taking. Accordingly, we detected compulsive-like feeding behavior in obese but not lean rats, measured as palatable food consumption that was resistant to disruption by an aversive conditioned stimulus. Striatal dopamine D2 receptors (D2Rs) were downregulated in obese rats, as has been reported in humans addicted

Physiopathology and Treatment of Obesity and Overweight: A Proposal for a New Anorectic.

The "new epidemic," as WHO calls obesity, is caused by overeating, which, having exceeded the body's actual needs, accumulates in the form of health-damaging fat deposits. Moving more and eating less is the main remedy, but eating belongs to vital instincts, which are beyond the control of reason. In this sense, eating is different from drinking and breathing because without food it is possible to survive for a few weeks, without water for a few days, without oxygen for a few minutes. The first part of this article provides an overview of obesity and its treatment, focusing on the new anorecti

Long-term changes on behavioral addictions symptoms among adults with attention deficit hyperactivity disorder treated with methylphenidate.

BACKGROUND: Attention deficit hyperactivity disorder (ADHD) and behavioral addictions (BAs) are highly comorbid but little is known about the effect of anti-ADHD medications on behavioral addiction symptoms. Thus, the aim of this naturalistic prospective study was to investigate the long-term changes on BAs symptoms among methylphenidate-treated adults with a primary diagnosis of ADHD. METHODS: 37 consecutive adult ADHD outpatients completed a baseline and follow-up assessment of ADHD, mood and BAs symptoms (internet, shopping, food, sex addictions and gambling disorder) after one year of met

Diet and opiate addiction: a quantitative assessment of the diet of non-institutionalized opiate addicts.

The diet, weight and height of 38 non-institutionalized chronic opiate addicts have been assessed and compared to those of a random sample of the 20-35 year old general population of Canton Geneva, Switzerland. A history questionnaire was administered by specially trained dietitians. Results show that, after adjustment for age and alcohol intake, male addicts have the same caloric intake as non-addicts. However, addicts tend to replace foods that are rich in fat and proteins with foods rich in sucrose and relatively poor in vitamins and minerals. Beer is the major component of their much large

The overlap between binge eating disorder and substance use disorders: Diagnosis and neurobiology.

BACKGROUND AND AIMS: Binge eating disorder (BED) is a relatively common condition, especially in young adult females, and is characterized by chronic over-consumption of food resulting in embarrassment, distress, and potential health problems. It is formally included as a disorder in DSM-5 for the first time, an acknowledgement to its debilitating nature. This article explores the overlap between binge eating disorder and substance use disorders (SUD). METHODS: The bibliographic search was a computerized screen of PubMed databases from January 1990 to the present. Binge eating disorder, subst

Similarities and Differences in Diagnostic Scales.

The scale as a diagnostic tool plays an important role in discriminating between addicts and normal subjects. At the beginning of this chapter, we will briefly introduce the development of substance and non-substance addiction scales, which not only include alcohol addiction, nicotine addiction, cannabis addiction, pathological gambling pathological Internet use, but also exercise addiction, sex addiction, and food addiction, which are in dispute. While we will find almost all the addiction scales contain the items on social impairment, preoccupied, withdrawal, and tolerance, the variability i

A narrative review of binge eating and addictive behaviors: shared associations with seasonality and personality factors.

Binge-eating disorder and seasonal affective disorder were first described as clinically relevant conditions in very close temporal proximity a few decades ago. Both disorders have a higher prevalence rate in woman than in men, are characterized by a high proneness-to-stress and manifest heightened responsiveness to high-calorie, hyper-palatable foods. In recent years, a compelling body of evidence suggests that foods high in sugar and fat have the potential to alter brain reward circuitry in a manner similar to that seen when addictive drugs like alcohol and heroin are consumed in excess. The

Pregnant Women Consume a Similar Proportion of Highly vs Minimally Processed Foods in the Absence of Hunger, Leading to Large Differences in Energy Intake.

BACKGROUND: The eating in the absence of hunger (EAH) experimental paradigm measures intake of highly palatable, highly processed foods when sated. However, no studies have examined EAH in pregnant women. OBJECTIVE: The objectives were to investigate whether EAH in pregnant women differs by level of food processing and to examine relationships of EAH with hedonic hunger, addictive-like eating, and impulsivity. DESIGN: EAH was assessed in a counterbalanced crossover feeding substudy in which participants completed two free-access eating occasions following a standardized meal during their sec

Predicting alcohol consumption and reward sensitivity using responses for natural sucrose reward in non-food-restricted rats.

RATIONALE: While around 50% of the risk for developing Alcohol Use Disorder (AUD) has been determined to be genetic, examining predictors of addictive-like behavior and individual variance in models without a family history of addiction is also important. Impaired reward sensitivity and response to natural reward may play a role in an individual's propensity to develop alcohol addiction. Little research has been done on predictors of alcohol addiction without food or water deprivation to coax animals into addiction. OBJECTIVES: This study seeks to examine psychological and behavioral predicto

Risky decision-making is associated with impulsive action and sensitivity to first-time nicotine exposure.

Excessive risk-taking is common in multiple psychiatric conditions, including substance use disorders. The risky decision-making task (RDT) models addiction-relevant risk-taking in rats by measuring preference for a small food reward vs. a large food reward associated with systematically increasing risk of shock. Here, we examined the relationship between risk-taking in the RDT and multiple addiction-relevant phenotypes. Risk-taking was associated with elevated impulsive action, but not impulsive choice or habit formation. Furthermore, risk-taking predicted locomotor sensitivity to first-time

mGluR5 antagonism attenuates methamphetamine reinforcement and prevents reinstatement of methamphetamine-seeking behavior in rats.

Addiction to methamphetamine is a significant public health problem, and there are currently no pharmacological agents that are approved for the treatment of addiction to this powerful psychostimulant. Chronic methamphetamine use leads to cognitive dysfunction as well as numerous psychiatric, neurological, and cardiovascular complications. There is a growing body of literature implicating an important role for glutamate neurotransmission in psychostimulant addiction. In the present study, we examined the effects of the selective type 5 metabotropic glutamate receptor (mGluR5) antagonist 3-((2-

Potential role of repetitive transcranial magnetic stimulation in obesity.

BACKGROUND: Repetitive transcranial magnetic stimulation (rTMS) is a non-invasive technique with mild side effects. Some forms of obesity may arise from eating disorders and recent data suggest that rTMS treatment could have beneficial effects in eating disorders. PURPOSE: The purpose of this comprehensive review is to hypothesize that rTMS may play a role in obesity. MATERIALS AND METHODS: Articles were selected based on a search on medline using the terms "rTMS", "food craving", "eating disorders", "addiction", and "obesity". They were published between 1995 and 2013. RESULTS: Repetitive

The sigma-1 receptor as key common factor in cocaine and food-seeking behaviors.

Addiction and eating disorders involve brain reward circuits. Binge eating predisposes to addictive behavior, while the cessation of exposure to drugs of abuse leads to reward activities, including intake of tasty foods. Cocaine use is associated with a decrease in food intake, with reversal after drug use is discontinued. Exciting new findings show that receptors for the 'hunger' hormone, ghrelin, directly interact with the sigma-1 receptor (σ1R), which is a target of cocaine. σ1Rs are key players in regulating dopaminergic neurotransmission and ghrelin-mediated actions. This review focuses o

Does food use disorder exist? Item response theory analyses of a food use disorder adapted from the DSM-5 substance use disorder criteria in a treatment seeking clinical sample.

BACKGROUND: Increased consumption of food that are high in energy and sugar have been pointed as a major factor in the obesity epidemic. Impaired control of food intake and the concept of food addiction has been developed as a potential contributor. Our objective was to evaluate the dimensionality and psychometric validity of diagnostic criteria for food addiction adapted from the 11 DSM-5 substance use disorder (SUD) criteria (i.e.: Food Use Disorder (FUD) criteria), and to evaluate the influence of age, gender, and body mass index (BMI). METHODS: Cross-sectional observational study includin

History of withdrawal modulates drug- and food-cue reactivity in cocaine dependent participants.

While the centrality of withdrawal in the diagnosis of addiction has been decreasing with each successive edition of the Diagnostic and Statistical Manual of Mental Disorders, psychometric and neurobiological evidence provides withdrawal a central role in the development and maintenance of addiction. The current study offers insight into these conflicting positions by using secondary analyses to assess how a history of DSM-assessed withdrawal influences the magnitude of bias in neural reactivity to drug- and/or food-related reward cues. To this end, we separated an existing sample of cocaine-d

Vaping and Smoking Cue Reactivity in Young Adult Nonsmoking Electronic Cigarette Users: A Functional Neuroimaging Study.

INTRODUCTION: The rapid growth in the use of electronic cigarettes (e-cigarettes) among nonsmoking young adults is concerning, as it raises the potential for chronic vaping and nicotine addiction. A key characteristic of drug addiction is the elevated neural response to conditioned drug-related cues (i.e., cue reactivity). Generalized reactivity to both vaping and smoking cues may signify an increased risk for smoking initiation in nonsmoking vapers. In this study, we used functional magnetic resonance imaging (fMRI) to evaluate brain responses to vaping and smoking cues in young adult nonsmok

Patterns of Addiction in Chronic Skin Diseases: A Comparative Analysis of Addictions and Influencing Factors in Atopic Dermatitis and Psoriasis.

Addictions seem to be more frequent in atopic dermatitis and psoriasis patients than in the general population. This cross-sectional observational study comparatively evaluated substance-related and behavioural addictions in atopic dermatitis and psoriasis patients and analysed possible addiction patterns. From October 2023 to April 2024, 100 atopic dermatitis and 104 psoriasis patients at a German university hospital completed an anonymous questionnaire, including sociodemographic and health-related parameters, along with validated assessment tools for common addictions (smoking, gambling, al
