[An experimental study of the psychostimulant action of amantadine].

According to previous experimental and clinical data, we have tested the hypothetic psycho-stimulating activity of amantadine on experimental sensitized models. We obtained a strong action on the spontaneous sleep of rats at a dose of 10 mg/kg and a less striking action in diminishing the effects of barbitol on the righting reflex of mice and in reducing the narcosis of rats induced by mebubarbital. This action does not seem to be due to a release of catecholamine because the lesions produced by electrocoagulation in the ascending aminergic pathways or pretreatment by alpha-methylparatyrosine

Midline (central) fluid percussion model of traumatic brain injury in pediatric and adolescent rats.

OBJECTIVE Experimental traumatic brain injury (TBI) models hold significant validity to the human condition, with each model replicating a subset of clinical features and symptoms. TBI is the leading cause of mortality and morbidity in children and teenagers; thus, it is critical to develop preclinical models of these ages to test emerging treatments. Midline fluid percussion injury (FPI) might best represent mild and diffuse clinical brain injury because of the acute behavioral deficits, the late onset of behavioral morbidities, and the absence of gross histopathology. In this study, the auth

Vitamin D intervention improves recognition memory and reduces hippocampal inflammation in rats submitted to an experimental model of cerebral palsy.

Cerebral Palsy (CP) comprises neurological conditions characterized by abnormalities in movement, body posture, and altered muscle tone. Vitamin D (VitD) is important in bone metabolism and has anti-inflammatory, immune, and neuromodulatory properties. The aim of this study was thus to investigate the effects of VitD supplementation on parameters associated with memory, hippocampal inflammation, and neuronal loss in male and female rats with CP. The experimental model of CP included postnatal anoxia (P0-P1) and sensory-motor restriction (P2-P28). The pups were treated intraperitoneally with ve

Hyperammonemia heightens anesthetic sensitivity in male rats with acute hepatic encephalopathy via affecting brain areas represented by the cerebellum.

BACKGROUND: Precise anesthesia is essential to ensure perioperative safety in patients with hepatic encephalopathy (HE) due to severe liver injuries. Hyperammonemia has been implicated as a possible trigger for HE. However, anesthetic sensitivity in HE and the role of hyperammonemia, in the context of anesthetic sensitivity of HE both remain uncertain. METHODS: We injected male Sprague Dawley rats with thioacetamide (TAA) to induce acute HE. The time to lose and recover the righting reflex induced by either the inhalation anesthetic isoflurane or the intraperitoneal anesthetic propofol, as we

In utero exposure to protease inhibitor-based antiretroviral regimens delays growth and developmental milestones in mice.

Antiretroviral therapy (ART) in pregnancy has dramatically reduced HIV vertical transmission rates. Consequently, there is a growing number of children that are HIV exposed uninfected (CHEUs). Studies suggest that CHEUs exposed in utero to ART may experience developmental delays compared to their peers. We investigated the effects of in utero ART exposure on perinatal neurodevelopment in mice, through assessment of developmental milestones. Developmental milestone tests (parallel to reflex testing in human infants) are reflective of brain maturity and useful in predicting later behavioral outc

Pentobarbital-morphine anesthetic interactions in terms of intensity of noxious stimulation required for arousal.

BACKGROUND: Previous reports suggest that the outcome (synergism, antagonism, summation) of opioid-barbiturate interactions may depend on the depth of anesthesia. One aim of the present study was to determine whether pentobarbital, alone and in combination with morphine, blocks awakening caused by noxious stimulation in a dose-related manner: the more intense the noxious stimulation, the more pentobarbital is required to suppress the response. A second aim of the study was to determine whether the pentobarbital-morphine anesthetic interaction depends on the depth of anesthesia measured in term

Prenatal Maternal Immune Activation with Lipopolysaccharide Accelerates the Developmental Acquisition of Neonatal Reflexes in Rat Offspring Without Affecting Maternal Care Behaviors.

Maternal immune activation (MIA)-infection with an immunogen during pregnancy-is linked to an increased risk of neurodevelopmental disorders (NDDs) in offspring. Both MIA and NDDs are associated with developmental delays in offsprings' motor behavior. Therefore, the current study examined the effects of MIA on neonatal reflex development in male and female offspring. Sprague Dawley rats were administered lipopolysaccharide (LPS; 50 μg/mL/kg, i.p.) or saline on embryonic day (E)15 of gestation. The offspring were then tested daily from postnatal day (P)3-P21 to determine their neonatal reflex a

Euthanasia efficacy and plasma biochemical implications of intraperitoneal thiamylal sodium in three rat strains.

Acceptable euthanasia methods must rapidly induce unconsciousness and death without pain or distress. We evaluated intraperitoneal thiamylal sodium (TM; 150 mg/kg; hereafter TM150) in male Sprague-Dawley, Wistar, and Fischer 344 rats. TM produced immobilization, loss of righting reflex, respiratory arrest, and cardiac arrest in all strains, with the shortest latencies in Fischer 344. Plasma chemistry showed strain-dependent shifts, including aspartate aminotransferase (AST) increases and variable alanine aminotransferase (ALT) responses; chloride and calcium showed small, non-uniform changes a

Prolonged hypothermia after phenobarbital-induced anesthesia in adrenalectomized rats.

Adrenalectomized rats are hypersensitive to the hypothermic as well as hypnotic effects of phenobarbital. The exaggerated hypnotic response is more pronounced in the chronic (10 day) than the acute (1 day) adrenalectomized rat and is reversed by glucocorticoid replacement. The prolonged hypothermia to a hypnotic dose of phenobarbital occurs in adrenalectomized but not adrenodemedullated rats. This hypersensitivity is characterized by an impaired ability to regain normal body temperatures for as long as 24 hours after regaining the righting reflex. The prolonged hypothermia is prevented by prio

Developmental Changes in Locomotion and Sensorimotor Reflexes Following Spinal Cord Transection.

The developmental trajectory of weight-bearing locomotion and sensorimotor reflexes following a spinal cord injury, as well as the mechanisms for plasticity, remain unclear. In rats, the second postnatal week is a critical period for the development and recovery of spinal sensorimotor function. The purpose of the present study was to characterize developmental changes during this time frame to provide a basis for potential interventions and future research. Rats underwent a complete low-thoracic (T8-T10) spinal cord transection surgery, or sham procedure, on postnatal day (P)1. Spontaneous loc

Effects of esketamine-based opioid-sparing anesthesia protocol in Bama miniature pigs undergoing robot-assisted nephrectomy.

The safety and efficacy of the esketamine-based opioid-sparing anesthesia protocol, though validated, are understudied in robotic surgeries. This study used robot-assisted nephrectomy in Bama miniature pigs to explore its application. Six healthy Bama miniature pigs were randomly assigned to esketamine group (K) and control group (C). In Group K, based on the anesthetics of Group C, 0.5 mg/kg esketamine was used for anesthesia induction, and anesthesia was maintained with 0.5 mg/kg·h esketamine. Results showed Group K exhibited more stable hemodynamics intraoperatively, particularly at 5 min a

DEXMEDETOMIDINE AND MIDAZOLAM INTRAMUSCULAR SEDATION IN BROWNBANDED BAMBOO SHARKS (CHILOSCYLLIUM PUNCTATUM).

Bamboo sharks are some of the most common elasmobranch species in zoos and aquaria and are frequently sedated for medical exams, treatments, and research. This study investigated the use of an IM sedation protocol of a single dose of dexmedetomidine (0.05 mg/kg) and midazolam (2.0 mg/kg) in brownbanded bamboo sharks (Chiloscyllium punctatum). Sharks were serially monitored every 5 min for heart rate, branchial beats, righting reflex, coelomic response, cloacal response, pelvic fin reflex, response to noxious stimulus, voluntary movement, and ability to swim. This sedation dose was effective at

Pharmacokinetics of propofol in mixed-breed dogs and greyhounds.

Pharmacokinetics and recovery characteristics of propofol in Greyhounds and mixed-breed dogs were compared. In all dogs, disposition of propofol was adequately described by a 2-compartment open model, with a rapid distribution phase followed by a slower elimination phase. When findings in Greyhounds were compared with those in mixed-breed dogs, significant differences were observed in mean concentrations of propofol in blood, recovery characteristics, and estimates for apparent volume of distribution, volume of distribution at steady state, and total body clearance. In addition, Greyhounds rec

Early motor development and cerebral cortical morphology in rats exposed perinatally to alcohol.

Specific tests of early motor development and cerebral cortical morphology were studied in rats born from alcohol-fed mothers, pair-fed mothers, and ad libitum controls (either liquid or pellet diets). Liquid Bio-Serv diets were used. Alcohol feeding began on day 5 of pregnancy and ended on day 10 postpartum. The weight gain of the pups after birth was drastically reduced compared with pair-fed and other control groups; however, removal of the alcohol on day 10 postpartum resulted in an upswing of the growth curve that paralleled control groups. Between days 2 and 21 (postnatal) three behavior

Non-traditional administration of remifentanil in an experimental setting.

Remifentanil is ultrashort-acting opioid with a unique pharmacokinetic profile. It is used exclusively intravenously. While considering its rapid onset of action and other pharmacokinetic properties, we decided to assess its effects following administration via non-traditional routes. Rabbits (n=10 per each group) were randomized into six groups: remifentanil 1 microg/kg and 3 microg/kg IM, 5.0 and 10.0 microg/kg conjunctivally, and 10 microg/kg and 25.0 microg/kg intranasally. Sedating effects were assessed via a loss of the righting reflex. Secondary, mean arterial blood pressure, arterial o

Dipyrone, a novel anticonvulsant agent? Insights from three experimental epilepsy models.

Dipyrone, a non-steroidal anti-inflammatory agent, was anticonvulsant in three experimental epilepsy models. At a dose of 300 mg/kg i.p., dipyrone blocked the maximal hind limb extension in the electroshock model in Wistar rats, the tonic-clonic component of acute sound-induced seizures and the limbic component of audiogenic kindling in genetically susceptible Wistar-derived rats, all in 100% of the animals. In the electroshock model higher doses (400 and 500 mg/kg) were also effective but lower doses (100 and 200 mg/kg) were not. In this model dipyrone had no effect on the recovery of the rig
