Shotgun Metagenomic Characterization of Skin Microbiome Shifts in Human Scabies Before and After Scabicidal Treatment.

Scabies, caused by Sarcoptes scabiei, is a globally prevalent ectoparasitic infestation associated with intense pruritus and secondary bacterial infection, yet the molecular composition of the skin microbiome during active infestation remains poorly characterized. We performed shotgun metagenomic sequencing of 41 skin samples collected from 18 patients at dry and moist anatomical sites before and after scabicidal treatment. In exploratory group-level comparisons, pretreatment moist-site samples had lower alpha diversity and higher bacterial and viral read-based burdens than post-treatment mois

Skin microbiome analysis of a junctional epidermolysis bullosa patient treated with genetically modified stem cells.

BACKGROUND AND OBJECTIVE: Junctional epidermolysis bullosa (JEB) is a subtype of epidermolysis bullosa caused by mutations in the LAMB3 gene. We treated a patient with JEB using genetically corrected autologous epidermal cultures retrovirally transduced with the functional LAMB3 gene sequence. The objective of this study was to analyze the skin microbiome of this patient, with a particular focus on transgenic skin, and to compare the findings to the skin microbiome of healthy controls and patients with atopic dermatitis and well-documented microbial dysbiosis. PATIENTS AND METHODS: Skin micro

Staphylococcus caprae and Staphylococcus epidermidis define the skin microbiome among different grades of acne vulgaris.

Acne vulgaris (AV) has been associated with Cutibacterium acnes (C. acnes) colonization in sebaceous follicles. However, recent studies have revealed the role of skin microbiome dysbiosis in acne pathogenesis. AV grading, which is classified by the sum of noninflammatory and inflammatory lesions, is essential in making clinical decisions about AV management. Henceforth, a better understanding of the skin microorganism profile in AV is needed. Our purpose was to compare microbiome profiles between different grades of AV severity. The microbiome samples were collected by swabbing from 108 partic

Skin Microbiome Variation Among Hosts and Batrachochytrium dendrobatidis Infection in Hyla meridionalis and Pelophylax perezi.

Chytridiomycosis, caused by the fungus Batrachochytrium dendrobatidis (Bd), is a key driver of global amphibian declines. The amphibian skin microbiome, which may include Bd-inhibitory bacteria, plays a crucial role in defence against infection, influencing host susceptibility and disease outcome. In this study, we investigated the skin microbiota and Bd infection in two frog species, Hyla meridionalis (Hm) and Pelophylax perezi (Pp), from northeastern Spain using full-length 16S rRNA gene sequencing. We found that microbiota composition differed significantly between frog species, with Pp har

Stage- and host-dependent microbiome remodeling and reciprocal changes between Haemaphysalis longicornis and host skin.

Ticks harbor diverse microbial communities that are crucial for their biology and capacity to transmit pathogens. Although interactions between tick and host skin microbiomes are likely to play critical roles in feeding and pathogen transmission, these reciprocal changes during tick-host interactions remain largely unexplored. Here, we used 16S rRNA-seq to investigate how blood feeding by Haemaphysalis longicornis (larvae, nymphs, and adults) influences both the tick microbiome and the host skin microbiome. We further characterized microbial distribution across major tick tissues. Results reve

Integrated Metagenomic and Lipidomic Profiling Reveals Dysregulation of Facial Skin Microbiome in Moderate Acne Vulgaris.

Acne vulgaris is a prevalent chronic inflammatory dermatosis primarily affecting the pilosebaceous units. Current therapeutic approaches often exhibit limited efficacy and high recurrence rates. To investigate the microbiome-related mechanisms of acne vulgaris, facial skin samples from 19 patients with moderate acne and 20 healthy individuals were analyzed using an integrated metagenomic and lipidomic profiling strategy. Metagenomic analysis revealed a significant reduction in microbial diversity (Chao index) in acne-affected skin compared to healthy controls (p < 0.001). The relative abundanc

Large-scale investigation for antimicrobial activity reveals novel defensive species across the healthy skin microbiome.

The human skin microbiome constitutes a dynamic barrier that can impede pathogen invasion by producing antimicrobial natural products. Gene clusters encoding for production of secondary metabolites, biosynthetic gene clusters (BGCs), that are enriched in the human skin microbiome relative to other ecological settings, position this niche as a promising source for new natural product mining. Here, we introduce a new human microbiome isolate collection, the EPithelial Isolate Collection (EPIC). It includes a large phylogenetically diverse set of human skin-derived bacterial strains from eight bo

The skin microbiome stratifies patients with cutaneous T cell lymphoma and determines event-free survival.

Mycosis fungoides (MF) is the most common entity of Cutaneous T cell lymphomas (CTCL) and is characterized by the presence of clonal malignant T cells in the skin. The role of the skin microbiome for MF development and progression are currently poorly understood. Using shotgun metagenomic profiling, real-time qPCR, and T cell receptor sequencing, we compared lesional and nonlesional skin of 20 MF patients with early and advanced MF. Additionally, we isolated Staphylococcus aureus and other bacteria from MF skin for functional profiling and to study the S. aureus virulence factor spa. We identi

The skin microbiome enhances disease through IL-1b and delays healing in cutaneous leishmaniasis patients.

Leishmania braziliensis infection results in inflammation and skin injury, with highly variable and unpredictable clinical outcomes. Here, we investigated the potential impact of microbiota on infection-induced inflammatory responses and disease resolution by conducting an integrated analysis of the skin microbiome and host transcriptome on a cohort of 62 L. braziliensis -infected patients. We found that overall bacterial burden and microbiome configurations dominated with Staphylococcus spp. were associated with delayed healing and enhanced inflammatory responses, especially by IL-1 family me

Integrated Human Skin Bacteria Genome Catalog Reveals Extensive Unexplored Habitat-Specific Microbiome Diversity and Function.

The skin is the largest organ in the human body. Various skin environments on its surface constitutes a complex ecosystem. One of the characteristics of the skin micro-ecosystem is low biomass, which greatly limits a comprehensive identification of the microbial species through sequencing. In this study, deep-shotgun sequencing (average 21.5 Gigabyte (Gb)) from 450 facial samples and publicly available skin metagenomic datasets of 2069 samples to assemble a Unified Human Skin Genome (UHSG) catalog is integrated. The UHSG encompasses 813 prokaryotic species derived from 5779 metagenome-assemble

Neuropeptides neurotensin and substance P accelerate diabetic wound healing by modulating immunity and the skin microbiome.

Diabetic foot ulcers (DFUs) are a common and debilitating complication of diabetes mellitus, typified by impaired healing, persistent inflammation, and skin microbiota dysbiosis. Although some neuropeptides are immunomodulatory, their effects on skin microbiota and wound resolution remain largely unexplored. We examined the therapeutic potential of the neuropeptides neurotensin (NT) and substance P (SP) in reshaping inflammatory and microbial dynamics and accelerating wound closure in a healthy and streptozotocin-induced diabetic mice model. Full-thickness cutaneous wounds were induced on both

Large-scale investigation for antimicrobial activity reveals newly-identified defensive species across the healthy skin microbiome.

The skin microbiome forms a protective barrier to pathogens, including through the production of antimicrobial metabolites. Here, we present EPICHHS, a large and taxonomically diverse skin microbiome culture collection of 968 strains from eight body sites. EPICHHS captures >95% of cumulative species-level abundance across 268 skin metagenomes. It includes isolates present at <0.1% relative abundance and the cultured representatives for eight species not previously isolated, markedly expanding current skin microbiome resources. A contact-independent screen assaying ~14,000 pairwise interactions

Gut and skin microbiome-metabolome pathways in vitiligo: from dysbiosis to immune activation and melanocyte dysfunction.

Vitiligo is an autoimmune depigmenting disorder driven by genetic susceptibility, oxidative stress, immune dysregulation, and progressive melanocyte loss. Emerging microbiome-metabolome evidence suggests that gut and skin dysbiosis may contribute to disease progression through the gut-skin axis. This review integrates current evidence linking microbial imbalance with immune activation and melanocyte dysfunction in vitiligo. Studies in patients and experimental vitiligo models show altered gut and skin microbial diversity, enrichment of inflammation-associated taxa, depletion of commensal bacte

Probiotic cells-encapsulated electrospun scaffolds accelerate diabetic wound healingviasynergistic anti-infection activity and skin microbiome modulation.

Diabetic wound healing is often hindered by persistent pathogenic infections and associated skin microbiome dysbiosis. The efficacy of conventional antibiotic drugs is significantly compromised by increasing drug resistance and further disruption of commensal microbiota. In this study, a probiotic-loaded, dry-state electrospun scaffold, Gel@LRNF@PLA, is developed for dual-functional diabetic wound healing. By maintaining probiotic viability while enabling localized antibacterial and microbiome-regulatory effects, the dressing exhibits potent activity against both planktonic Pseudomonas aerugin

Microbiome and Skin Health: A Systematic Review of Nutraceutical Interventions, Disease Severity, Inflammation, and Gut Microbiota.

Skin disorders are a major global cause of morbidities, and increasing evidence links several to gut microbiome dysregulation. Because of this the bidirectional gut-skin axis, nutraceuticals have been proposed as therapeutic adjuncts, but their clinical effects across skin conditions remain unclear. To understand how pro/pre/synbiotics can affect health, we conducted a systematic review to investigate disease severity indices, inflammatory and immunological markers, quality of life, and changes in gut microbiota composition. PubMed, Embase, and Web of Science were utilized to identify relevant

Skin transcriptome and microbiome profiling are associated with Atlantic salmon resistance/susceptibility to sea lice infestation.

Salmon aquaculture is continually threatened by sea lice infestations, which impact fish health, welfare, and productivity. Thus, breeding programs enable the selection of phenotypes to improve productive performance. This study uses a controlled, repeated lice infestation model to analyze the skin transcriptome and microbiome of Atlantic salmon concerning their resistance or susceptibility to sea lice infestation. The transcriptomic profile of skin from three phenotypes-resistant (R), susceptible (S), and one salmon family presumably associated with acquired resistance to sea lice (ALR)-was a

Skin microbiome and causal relationships in three dermatological diseases: Evidence from Mendelian randomization and Bayesian weighting.

BACKGROUND: Atopic dermatitis (AD), psoriasis (PSO), rosacea, and other related immune skin diseases are affected by multiple complex factors such as genetic and microbial components. This research investigates the causal relationships between specific skin microbiota and these diseases by using Mendelian randomization (MR), and Bayesian weighted Mendelian randomization (BWMR). METHODS: We utilized genome-wide association study (GWAS) data to analyze the associations between various skin bacteria and three dermatological diseases. Single nucleotide polymorphisms (SNPs) served as instrumental

Seven-day Green Tea Supplementation Revamps Gut Microbiome and Caecum/Skin Metabolome in Mice from Stress.

Green tea supplementation has beneficial health effects. However, its underlying mechanisms, such as effects on modulating the intestinal microbiome and endogenous metabolome, particularly following short-term supplementation, are largely unclear. We conducted an integrative metabolomics study to evaluate the effects of short-term (7-day) supplementation of green tea extract (GTE) or its components, epigallocatechin gallate, caffeine, and theanine, on the caecum microbiota and caecum/skin metabolome in mice. Further, we established an integrative metabolome-microbiome model for correlating gut

Microbiome dysbiosis is associated with disease duration and increased inflammatory gene expression in systemic sclerosis skin.

BACKGROUND: Infectious agents have long been postulated to be disease triggers for systemic sclerosis (SSc), but a definitive link has not been found. Metagenomic analyses of high-throughput data allows for the unbiased identification of potential microbiome pathogens in skin biopsies of SSc patients and allows insight into the relationship with host gene expression. METHODS: We examined skin biopsies from a diverse cohort of 23 SSc patients (including lesional forearm and non-lesional back samples) by RNA-seq. Metagenomic filtering and annotation was performed using the Integrated Metagenomi
