Quantitative assessment of Candida albicans, Staphylococcus aureus and Staphylococcus epidermidis in peri-implant health and disease: correlation with clinical parameters.

Introduction. Peri-implantitis poses a significant challenge in oral health due to its complex microbial profile and biofilm-related persistence, complicating both prevention and treatment strategies.Hypothesis/Gap Statement. We hypothesized that biofilm formation by Candida albicans, Staphylococcus aureus and Staphylococcus epidermidis plays a critical role in peri-implantitis by inducing an inflammatory response, contributing to bone loss and implant failure. This study aimed to investigate the presence and quantify C. albicans, S. aureus and S. epidermidis in health and peri-implantitis usi

Insertion sequence elements associated with Staphylococcus epidermidis evolution in persistent orthopaedic device-related infections.

BACKGROUND: Staphylococcus epidermidis is a major cause of orthopaedic device-related infections (ODRIs), which are often challenging to treat due to their extensive antimicrobial resistance (AMR) and biofilm formation. It has been hypothesised that S. epidermidis may rapidly adapt to the medical device niche, enhancing persistence, but direct evidence of within-host pathoadaptive evolution remains limited. RESULTS: To investigate within-host evolution during chronic infection by S. epidermidis, we analysed isolates from patients with confirmed ODRIs and used a rat infection model to examine

Anti-quorum-sensing activity of Plectranthus aliciae in multispecies Cutibacterium acnes-Staphylococcus epidermidis systems.

BACKGROUND: Quorum sensing (QS) is a bacterial communication process mediated by the production, release, and detection of autoinducers, regulating gene expression linked to virulence and biofilm formation. Cutibacterium acnes and Staphylococcus epidermidis exhibit opportunistic pathogenic behavior on the skin, complicating treatment strategies for inflammatory skin conditions such as acne vulgaris and wound-associated infections. The interspecies relationship between C. acnes and S. epidermidis, particularly within multispecies biofilms and under different oxygen conditions, remains insuffici

Active components of Patrinia scabiosifolia and mechanisms of action against methicillin-resistant Staphylococcus Epidermidis.

INTRODUCTION: Staphylococcus epidermidis is a significant cause of hospital bloodstream infections, often transmitted through medical devices. Methicillin-resistant Staphylococcus epidermidis (MRSE) exhibits multidrug resistance, challenging clinical treatment. Current options are limited, with vancomycin as the primary choice despite its negative effects. Traditional Chinese Medicine (TCM) is a valuable resource for novel antibiotics; Our research showed Patrinia scabiosifolia has potent anti-MRSE activity. TCM's efficacy stems from active components, but active components and mechanisms of P

Mapping Antimicrobial Resistance in Staphylococcus epidermidis Isolates from Subclinical Mastitis in Danish Dairy Cows.

Background/Objectives: Although Staphylococcus epidermidis is a key cause of subclinical mastitis in Danish dairy cows, its sensitivity to antimicrobials remains unexplored. Here, we analyzed sixty S. epidermidis isolates derived from 42 dairy cows across six conventional dairy herds in Denmark. Methods: Phenotypic resistance was measured by antimicrobial susceptibility testing and minimum inhibitory concentration (MIC) analysis, and genotypic resistance was examined through whole-genome sequencing and identification of antimicrobial resistance genes (ARGs). Correspondence between phenotypic a

Whole genome analysis and antimicrobial resistance assessment of Staphylococcus epidermidis isolated from food sources.

Coagulase-negative staphylococci (CoNS), including Staphylococcus epidermidis, are commonly occurrence in a variety of food products. Historically considered non-pathogenic, these microorganisms were excluded from routine food safety monitoring protocols. However, their increasing involvement in nosocomial infections underscores their pathogenic potential. Emerging evidence suggests that the food chain may serve as a reservoir and transmission route for antibiotic-resistant bacteria. In this study, 26 S. epidermidis isolates obtained from ready-to-eat food were subjected to whole-genome sequen

Increased incidence of teicoplanin-non-susceptible Staphylococcus epidermidis strains: a 6-year retrospective study.

Glycopeptide antibiotics (vancomycin and teicoplanin) are usually used for the treatment of Staphylococcus epidermidis infections owing to their increased oxacillin resistance. However, S. epidermidis strains with decreased susceptibility to teicoplanin have become increasingly incident in recent years. We aimed to identify the characteristics of teicoplanin-non-susceptible (Teico-NS) S. epidermidis isolated at our hospital and analyze its relationship with teicoplanin usage. We retrospectively evaluated 328 S. epidermidis strains isolated from clinical isolates between January 2016 and Decemb

Eradication of Staphylococcus epidermidis within Biofilms: Comparison of Systemic versus Supratherapeutic Concentrations of Antibiotics.

Biofilm-forming bacterial infections result in clinical failure, recurring infections, and high health care costs. The antibiotic concentrations needed to eradicate biofilm require further research. We aimed to model an in vitro prosthetic joint infection (PJI) to elucidate the activity of traditional systemic concentrations versus supratherapeutic concentrations to eradicate a Staphylococcus epidermidis biofilm PJI. We evaluated S. epidermidis high-biofilm-forming (ATCC 35984) and low-biofilm-forming (ATCC 12228) isolates in an in vitro pharmacodynamic biofilm reactor model with chromium coba

Extracellular Vesicles Influence the Growth and Adhesion of Staphylococcus epidermidis Under Antimicrobial Selective Pressure.

Staphylococcus epidermidis causes infections associated with orthopedic implants due to its ability to establish persistent biofilms, making infections chronic and hard to treat. Extracellular vesicles (EVs) are part of the bacterial communication system, but the role of S. epidermidis-derived EVs in biofilm formation processes and survival is completely unknown. The aims of this study were (i) to investigate the effect of subinhibitory concentrations of antibiotics on vesiculation in S. epidermidis and evaluate the role of EVs in bacterial survival and adhesion under antimicrobial selective p

Poly-N-Acetylglucosamine Production by Staphylococcus epidermidis Cells Increases Their In Vivo Proinflammatory Effect.

Poly-N-acetylglucosamine (PNAG) is a major component of the Staphylococcus epidermidis biofilm extracellular matrix. However, it is not yet clear how this polysaccharide impacts the host immune response and infection-associated pathology. Faster neutrophil recruitment and bacterial clearance were observed in mice challenged intraperitoneally with S. epidermidis biofilm cells of the PNAG-producing 9142 strain than in mice similarly challenged with the isogenic PNAG-defective M10 mutant. Moreover, intraperitoneal priming with 9142 cells exacerbated liver inflammatory pathology induced by a subse

Differential expression of methicillin resistance by different biofilm-negative Staphylococcus epidermidis transposon mutant classes.

Biofilm formation mediated by polysaccharide intercellular adhesin (PIA) is the major virulence factor of Staphylococcus epidermidis and is often associated with methicillin resistance. Transposon Tn917 insertions leading to a biofilm-negative phenotype in the biofilm-producing S. epidermidis strain 1457 (mecA-negative) were transferred into the methicillin-resistant, biofilm-producing S. epidermidis 1057 (mecA-positive) by transduction. According to their phenotypes and genotypes, the mutants could be separated into genetic classes I to IV (D. Mack, H. Rohde, S. Dobinsky, J. Riedewald, M. Ned

Lack of mecA transcription in slime-negative phase variants of methicillin-resistant Staphylococcus epidermidis.

Five phase variants (PV1 to PV5) of the well-characterized, slime-producing, methicillin-resistant, pathogenic strain of Staphylococcus epidermidis sensu strictu RP62A (ATCC 35984) were isolated by the Congo red agar method. In comparison with the parent strain, the phase variants showed a different colonial morphology on Congo red agar, a strongly reduced adherence capacity, and decreased levels of resistance to methicillin, oxacillin, and penicillin. All phase variants yielded biochemical reaction patterns and profiles in pulsed-field gel electrophoresis identical to those of parent strain R

A Molecularly Imprinted Polymer Nanobodies (nanoMIPs)-Based Electrochemical Sensor for the Detection of Staphylococcus epidermidis.

Methicillin-resistant Staphylococcus epidermidis (MRSE) contamination is commonly found on human skin and medical devices. Herein, we present a sensor utilizing molecularly imprinted polymer nanobodies (nanoMIP) for recognition and electrochemical impedance spectroscopy (EIS) to detect S. epidermidis. Sensor manufacturing involves synthesizing nanoMIP via solid-phase synthesis using whole bacteria as templates. Screen-printed gold electrode (AuSPE)-modified 16-mercaptohexadecanoic acid (MHDA) served to immobilize the nanoMIPs on the sensor surface through an amide bond, with the remaining func
