AIMS: This study aimed to develop voriconazole-loaded microbubbles (MBs@VRC) combined with ultrasound-targeted microbubble destruction (UTMD) as a novel strategy to disrupt Candida albicans biofilms and enhance antifungal efficacy, addressing the critical challenge of biofilm-associated resistance in fungal periprosthetic joint infections (PJIs).
METHODS: MBs@VRC were produced using a thin-film hydration method, and their physicochemical properties were evaluated, including size, zeta potential, and drug loading efficiency. The penetration of DiI-labelled MBs@VRC into C. albicans biofilms was

Background: Pseudomonas aeruginosa combines intrinsic resistance, adaptive plasticity, and biofilm formation, making treatment failure common in veterinary and human medicine. We examined whether geraniol and Tris-ethylenediaminetetraacetic acid (Tris-EDTA) modulate marbofloxacin activity during serial exposure and whether whole-genome sequencing detects genomic correlates of the observed phenotypic changes. Methods: A biofilm-producing reference P. aeruginosa ATCC 27853 strain was serially passaged for 10 days with marbofloxacin, geraniol, Tris-EDTA, and their double and triple combinations.

BACKGROUND: Pseudomonas aeruginosa, a member of the "ESKAPE" pathogens, possesses a robust ability to form biofilms-a key factor that contributes to its antibiotic resistance and poses significant challenges for clinical management. Identifying potential therapeutic targets through bioinformatic analysis of genomic data is therefore critical for developing more effective treatment strategies.
METHODS: In this study, five gene expression datasets from the Gene Expression Omnibus were integrated to investigate transcriptional differences between planktonic and biofilm-associated P. aeruginosa p

Chronic wounds, diabetic foot ulcers, venous leg ulcers, and pressure injuries affect millions of patients worldwide and cost healthcare systems in the order of $150 billion annually, yet treatment options have changed less than the scale of the problem would suggest. Biofilm formation, documented in up to 78% of chronic wounds, is a central cause: bacteria embedded in extracellular polymeric matrices tolerate antimicrobial concentrations up to 1000-fold higher than planktonic cells and sustain a chronic inflammatory state that actively prevents tissue repair. Hydrogels, crosslinked polymer ne

OBJECTIVE: The effects of Candida albicans biofilm growth under treatment with cleaning agents on the deterioration of denture acrylic resin (PMMA) and a nano-filled restorative composite resin were examined in vitro.
MATERIALS: A total of 240 PMMA (Travelon™) and nano-filled composite (Filtek™ Z350XT) specimens were fabricated. Baseline surface hardness VHN, arithmetic mean surface roughness (Sa, in nm), and flexural strength (ISO 20795-1) were measured. The specimens underwent a Candida albicans ATCC 10231 biofilm challenge for 90 days and were then immersed in 1% sodium hypochlorite (NaOCl

INTRODUCTION: Treatment of biofilms is a priority in purulent surgery. A biofilm consists of 75-85% extracellular polymeric matrix and 15-20% microbial cells. Polysaccharides, proteins, and extracellular DNA within the matrix protect bacteria from adverse environmental factors (pH, antibiotics, phagocytosis). Communication between bacteria occurs through the "quorum sensing" system. Bacteria within biofilms are 100-1000 times more resistant to antibiotics compared to planktonic forms. This is explained by the limited penetration of antibiotics into the matrix and the reduced metabolic activity

UNLABELLED: Catheter-associated urinary tract infections (CAUTIs) are the most prevalent nosocomial infections. These infections impose a significant financial burden on the healthcare system and are responsible for approximately 15,000 deaths each year in the US. CAUTIs are caused by free-floating bacteria that adhere to the surface of Foley catheters and the patient's uroepithelium, forming protective biofilms that enhance bacterial survival and resistance. In this study, a nitric oxide (NO)-releasing liquid formulation is used to inflate the Foley catheter balloon, aiming to inhibit biofilm

OBJECTIVES: Sympathomimetic nasal solutions (SNS), containing oxymetazoline or phenylephrine, may be used directly onto dental pulp tissue and initiate hemostasis via activation of alpha-receptors that innervate arteriole smooth muscles. The aim of this study was to assess these SNS's biocompatibility with human dental pulp stem cells (hDPSCs) and ability to kill a deep caries-associated pathogen, Rothia dentocariosa.
MATERIALS AND METHODS: A Kirby-Bauer disk diffusion susceptibility test assessed SNS zones of inhibition (ZOI) toward R. dentocariosa growth on agar plates. Over-the-counter (OT

Histones possess significant antimicrobial potential, yet their activity against biofilms remains underexplored. Moreover, concerns regarding adverse effects limit their clinical implementation. We investigated the antibacterial efficacy of human recombinant histone H1 subtypes against Pseudomonas aeruginosa PAO1, both planktonic and in biofilms. After the in vitro tests, toxicity and efficacy were assessed in a P. aeruginosa PAO1 infection model using Galleria mellonella larvae. Histones were also evaluated in combination with ciprofloxacin (Cpx) and gentamicin (Gm). Our results demonstrate a

BACKGROUND: The increasing prevalence of invasive fungal infections in immuno-compromised patients is a considerable cause of morbidity and mortality. With the rapid emergence of antifungal resistance and an inadequate pipeline of new therapies, novel treatment strategies are now urgently required.
METHODS: The antifungal activity of the alginate oligosaccharide OligoG in conjunction with nystatin was tested against a range of Candida spp. (C. albicans, C. glabrata, C. parapsilosis, C. auris, C. tropicalis and C. dubliniensis), in both planktonic and biofilm assays, to determine its potential

The bacterial heat-shock response is regulated by the alternative sigma factor, σ32 (RpoH), which responds to misfolded protein stress and directs the RNA polymerase to the promoters for genes required for protein refolding or degradation. In P. aeruginosa, RpoH is essential for viability under laboratory growth conditions. Here, we used a transcriptomics approach to identify the genes of the RpoH regulon, including RpoH-regulated genes that are essential for P. aeruginosa. We placed the rpoH gene under control of the arabinose-inducible PBAD promoter, then deleted the chromosomal rpoH allele.

Fungal populations are commonly found in natural environments and present enormous health care challenges, due to increased resistance to antifungal agents. Paeonol exhibits antifungal activities; nevertheless, the antifungal and antibiofilm activities of paeonol against Candida albicans and Cryptococcus neoformans remain largely unexplored. Here, we aimed to evaluate the antifungal and antibiofilm activities of paeonol against C. albicans and/or C. neoformans (i.e., against mono- or dual-species). The minimum inhibitory concentrations (MICs) of paeonol for mono-species comprising C. albicans

Many microbes in their natural habitats are found in biofilm ecosystems attached to surfaces and not as free-floating (planktonic) organisms. Furthermore, it is estimated that nearly 80% of human infections are associated with biofilms. Biofilms are traditionally defined as three-dimensional, structured microbial communities that are attached to a surface and encased in a matrix of exopolymeric material. While this view of biofilm largely arises from in vitro studies under static or flow conditions, in vivo observations have indicated that this view of biofilms is essentially true only for for

Invasive fungal infections are an emerging problem worldwide, which bring huge health challenges. Candida albicans, the most common opportunistic fungal pathogen, can cause bloodstream infections with high mortality in susceptible hosts. At present, available antifungal agents used in clinical practice are limited, and most of them also have some serious adverse effects. The emergence of drug resistance because of the wide use of antifungal agents is a new limitation to successful patient therapy. Drug combination therapy is increasingly becoming a way to enhance antifungal efficacy, and reduc

Dermatophytosis is the most common mycosis worldwide, affecting approximately 20 to 25% of the population, regardless of gender, race, color, and age. Most antifungal agents used for the treatment of dermatophytosis belong to the azole and allylamine classes. Dermatophytes are reported to be resistant to most commercial drugs, especially microbial biofilms, in addition to their considerable toxicity. It should be emphasized the importance of looking for new molecules with reduced toxicity, as well as new targets and mechanisms of action. This work aims to incorporate nonyl 3,4-dihydroxybenzoat

A novel biscarbazol triphenylamine end-capped dendrimeric zinc(II) porphyrin (DP 5) was synthesized by click chemistry. This compound is a cruciform dendrimer that bears a nucleus of zinc(II) tetrapyrrolic macrocycle substituted at the meso positions by four identical substituents. These are formed by a tetrafluorophenyl group that possesses a triazole unit in the para position. This nitrogenous heterocyclic is connected to a 4,4'-di(N-carbazolyl)triphenylamine group by means of a phenylenevinylene bridge, which allows the conjugation between the nucleus and this external electropolymerizable

BACKGROUND: The quality and health safety of water used for refrigeration and flushing of the handpieces, water-syringes and other components of dental units is of considerable importance. Water crosses these devices by a system of intersected small plastic tubes (about 2 mm of diameter), named dental unit water lines (DUWLs). DUWLs may be heavily colonized by many bacterial species in a planktonic phase, adherent or in biofilm lifestyle, resulting in a potential risk of infection, not only for all professionals who routinely use these devices, but also for occasional-patients, especially immu

Nutrient limitation restricts bacterial growth in privileged sites such as the middle ear. Transient heme-iron restriction of nontypeable Haemophilus influenzae (NTHI), the major causative agent of chronic and recurrent otitis media (OM), promotes new and diverse phenotypes that can influence planktonic, biofilm, and intracellular lifestyles of NTHI. However, the bacterial responses to nutrient restriction that impact intracellular fate and survival of NTHI are unknown. In this work, we provide evidence for the role of transient heme-iron restriction in promoting the formation of intracellular
