Use of the Ophtec Artificial Iris Model C1 in Patients with Aniridia/Aphakia.

In the present cases, the use of the Ophtec Arificial Iris model C1 is described in patients with traumatic or postoperative aniridia/aphakia. In one of the patients, it was combined with perforating keratoplasty because of corneal scarring. In both of the presented cases, improvement in visual acuity and a satisfactory aesthetic result without any serious complications can be shown. However, the short follow-up time must be emphasised.

A Novel PAX6 Heterozygous Mutation Found in a Chinese Family with Congenital Aniridia.

Purpose: Congenital aniridia is a kind of panocular disorder characterized by the absence of the iris in both eyes. Paired box 6 (PAX6) gene mutations have been identified as the most common cause of congenital aniridia. The aim of this study was to reveal the genetic defect in PAX6 in a Chinese family with congenital aniridia. Methods: Twelve individuals from a three-generation Chinese family were recruited. All the family members underwent comprehensive ophthalmologic examinations. The entire coding region of PAX6 was amplified by polymerase chain reaction, followed by direct Sanger sequenci

A novel PAX6 mutation causes congenital aniridia with or without retinal detachment.

BACKGROUND: Aniridia is a rare developmental eye disorder characterized by complete or partial iris hypoplasia often accompanied with other ocular changes that affect the cornea, anterior chamber, lens, retina, and optic nerve. Most cases of aniridia are inherited with an autosomal dominant mode of inheritance caused by PAX6 mutations or deletions. To reveal the underlying genetic defect in a four-generation Iranian family with aniridia, we carried out a genetic screening of PAX6. METHODS: Complete ophthalmic examinations were performed for available affected family members. All PAX6 exons an

Novel Intragenic PAX6 Deletion in a Pedigree with Aniridia, Morbid Obesity, and Diabetes.

Purpose: Aniridia is a rare congenital eye disease, characterized by a constellation of symptoms including hypoplastic irides, foveal hypoplasia, early cataract, corneal stem cell deficiency, and glaucoma. Large chromosomal deletions spanning the PAX6 gene cause WAGR syndrome (Wilms tumor, aniridia, genitourinary anomalies, and intellectual disability [formerly called mental retardation]). We describe clinical and genetic studies of a three-generation pedigree with aniridia along with additional systemic conditions (morbid obesity, diabetes) suggesting the possibility of a contiguous-gene synd

Molecular analysis of Cypriot families with aniridia reveals a novel PAX6 mutation.

The present study investigated the clinical and mutational spectrum of aniridia in a cohort of 17 affected individuals from six families from Cyprus. Each proband was initially evaluated for copy number variants at the PAX6 locus and subsequently underwent PAX6 mutation screening. Sequence analysis of FOXC1 and PITX2 was performed in patients who did not carry a PAX6 mutation. The most common clinical features in the group of aniridia patients associated with aniridia were nystagmus, cataracts and glaucoma. PAX6 pathogenic mutations were identified in five out of six families (a diagnostic yie

Case of a novel PAX6 mutation with aniridia and insulin-dependent diabetes mellitus.

We present a case of a novel PAX6 heterozygous mutation with aniridia and insulin-dependent diabetes mellitus. To the best of our knowledge, this is the first case of its mutation with a complete loss of insulin secretory capacity. We believe that our letter will add new knowledge to diabetes mellitus associated with PAX6 mutations and might help us to understand the role of PAX6 in beta-cell development.

Artificial iris implantation in a male with post-traumatic aniridia - case report.

Artificial iris implantation in a 38-year-old patient with post-traumatic aniridia is reported. The patient was assaulted in May 2013 and suffered a blunt trauma to his left eye. Directly after the assault, he was treated at the local hospital, where the upper eyelid wound and the lacerations of bulbar conjunctiva were sutured. One month later, he underwent a combined procedure of cataract extraction and anterior vitrectomy without the implantation of an intraocular lens. Next, he was hospitalised in our department, where apart from medical treatment of secondary, post-traumatic glaucoma, he w

Aniridia with a heterozygous PAX6 mutation in which the pituitary function was partially impaired.

We herein report the case of a woman with aniridia and with a heterozygous PAX6 mutation. Pax6 is a transcription factor involved in the development of several organs, including the eye, pancreas and pituitary. The patient had been diagnosed with aniridia in childhood and was found to have impaired glucose tolerance with a heterozygous PAX6 mutation 12 years prior to the current admission. Hormone stimulating tests revealed a slightly impaired pituitary function, including subtle hypogonadotropic hypogonadism and borderline growth hormone (GH) deficiency. The present case is the first report o

Mutation analysis of PAX6 in inherited and sporadic aniridia from northeastern China.

PURPOSE: Haplo-insufficiency at the paired box gene 6 (PAX6) locus causes aniridia, which is characterized by iris hypoplasia and other anterior and posterior eye defects leading to poor vision. This study aimed to identify novel PAX6 mutations that lead to familial and sporadic aniridia in northeastern China. METHODS: Two aniridia patients from a family and a sporadic patient underwent full ophthalmologic examinations. Genomic DNA was isolated from the affected individuals, 5 noncarriers in the family and 100 healthy normal controls. The coding regions and the adjacent intronic sequence of PA

[Glaucoma with aniridia and isolated congenital glaucoma in siblings: contribution and limits of genetics].

INTRODUCTION: Congenital glaucoma associated with aniridia and primary congenital glaucoma are regarded as different entities. Indeed, the abnormalities of the angle's structures as well as the genes involved are different. We report the observation of two sisters presenting these two types of glaucoma with particular attention paid to the importance and the difficulty of genetic counseling. OBSERVATIONS: Child L, with no particular family history, had presented bilateral aniridia complicated by bilateral glaucoma since birth. In addition to medical and surgical treatment, general and genetic

Aniridia as part of a WAGR syndrome in a girl whose brother presented hypospadias.

Aniridia can arise as part of the WAGR syndrome (Wilms tumour. aniridia, genitourinary anomalies, and mental retardation), due to a deletion or chromosomal region 11p13. We report a girl with a complete WAGR syndrome, whose brother presented hypospadias. Cytogenetic, FISH and molecular studies showed a deletion in one chromosome 11 of the patient. No cytogenetic rearrangement or deletion affecting the genes included in this region (PAX6 and WT1) were observed in her brother and parents. This excludes a higher risk than that of the general population for developing Wilms tumour in the brother a

Genomic structure, evolutionary conservation and aniridia mutations in the human PAX6 gene.

Aniridia is a semidominant disorder in which development of the iris, lens, cornea and retina is disturbed. The mouse mutation Small eye (Sey), which has been proposed as a model for aniridia, results from defects in Pax-6, a gene containing paired-box and homeobox motifs that is specifically expressed in the developing eye and brain. To test the role of PAX6 in aniridia, we isolated human cDNA clones and determined the intron-exon structure of this gene. PAX6 spans 22 kilobases and is divided into 14 exons. Analysis of DNA from 10 unrelated aniridia patients revealed intragenic mutations in t

Early Eye Organoids Demonstrate Cadherin and Semaphorin Disruption in PAX6-Related Aniridia.

The paired box transcription factor PAX6 is a master regulator of eye development. Pathogenic PAX6 variants predominantly cause aniridia, resulting in a wide range of ocular anomalies affecting both the anterior and posterior segments of the eye. However, a detailed examination of the transcriptomic alterations that occur with these variants during early eye development has not yet been undertaken. To investigate this, human-induced pluripotent stem cells (hiPSC)-derived 3D optic vesicle-like organoids were generated up to day 35 from an aniridia patient with a heterozygous nonsense PAX6R261X

Patient-Specific iPSC-Derived Cornea Organoids for Investigating Aniridia-Associated Corneal Disorders.

Aniridia is a rare congenital eye disorder marked by the partial or complete absence of the iris, and it is often associated with corneal opacification and limbal stem cell deficiency. The underlying genetic mutations, primarily affecting the PAX6 gene, disrupt both the development and function of the cornea. The most common ocular features besides iris defects are nystagmus, foveal hypoplasia, cataract, glaucoma and aniridia-associated keratopathy (AAK). Although in vivo models offer valuable insights into the pathology of aniridia, patient-derived in vitro models are crucial for investigatin

Central Corneal Epithelial Wounding Accelerates Aniridia-Associated Keratopathy in PAX6-Deficient Mice.

PURPOSE: To investigate corneal healing in a Pax6+/- mouse model of aniridia-associated keratopathy (AAK) when challenged with a mild epithelial wound. METHODS: Pax6+/- small-eye (Sey) mice (n = 15) and Pax6+/+ wild-type mice (n = 4) on the 129S1/SvImJ background underwent 1 mm central corneal mechanical epithelial debridement. Corneal status before and four weeks after wounding was evaluated by slit-lamp imaging. Longitudinal corneal thickness was measured using optical coherence tomography (OCT), and AAK was graded in Sey mice. Wounded corneas were additionally compared to unwounded contral
